Effect of feeding a by-product feed-based silage on nutrients intake, apparent digestibility, and nitrogen balance in sheep

  • Background: Literature is lacking on the effects of feeding by-product feed (BF)-based silage on rumen fermentation parameters, nutrient digestion and nitrogen (N) retention in sheep. Therefore, this study was conducted to determine the effect of replacing rye straw with BF-based silage as a roughage source on ruminal parameters, total-tract apparent nutrient digestibility, and N balance in sheep. Methods: The by-product feed silage was composed of spent mushroom substrate (SMS) (45 %), recycled poultry bedding (RPB) (21 %), rye straw (11 %), rice bran (10.8 %), corn taffy residue (10 %), protected fat (1.0 %), bentonite (0.6 %), and mixed microbial additive (0.6 %). Six sheep were assigned randomly to either the control (concentrate mix + rye straw) or a treatment diet (concentrate mix + BF-based silage). Results: Compared with the control diet, feeding a BF-based silage diet resulted in similar ruminal characteristics (pH, acetate, propionate, and butyrate concentrations, and acetate: propionate ratio), higher (p < 0.05) ruminal NH3-N, higher (p < 0.05) ether extract digestibility, similar crude protein digestibility, lower (p < 0.05) dry matter, fiber, and crude ash digestibilities, and higher (p < 0.05) N retention (g/d) Conclusion: The BF-based silage showed similar energy value, higher protein metabolism and utilization, and lower fiber digestion in sheep compared to the control diet containing rye straw.

Prediction of the Chemical Composition of Fresh Whole Crop Barley Silages by Near Infrared Spectroscopy

  • Near infrared reflectance spectroscopy (NIRS) has become increasingly used as a rapid and accurate method of evaluating some chemical compositions in forages and feedstuff. This study was carried out to explore the accuracy of near infrared spectroscopy (NIRS) for the prediction of chemical parameters of fresh whole crop barley silages. A representative population of 284 fresh whole crop barley silages was used as a database for studying the possibilities of NIRS to predict chemical composition. Samples of silage were scanned at 1 nm intervals over the wavelength range 680~2,500 nm and the optical data were recorded as log 1/Reflectance (log 1/R) and were scanned in fresh condition. NIRS calibrations were developed by means of partial least-squares (PLS) regression. NIRS analysis of fresh whole crop barley silages provided accurate predictions of moisture, acid detergent fiber (ADF), neutral detergent fiber (NDF), crude protein (CP) and pH, as well as lactic acid content with correlation coefficients of cross-validation ($R^2cv$) of 0.96, 0.81, 0.79, 0.84, 0.72 and 0.78, respectively, and standard error of cross-validation (SECV) of 1.26, 2.83, 2.18, 1.19, 0.13 and 0.32% DM, respectively. Results of this experiment showed the possibility of the NIRS method to predict the chemical parameters of fresh whole crop barley silages as a routine analysis method in feeding value evaluation and for farmer advice.

쌀 전분의 현탁액과 호화액의 유동 거동 (Flow Behaviors of Native and Gelatinized Rice Starch Solutions)

  • 5 wt% 쌀전분 현탁액 및 호화된 쌀전분 $3{\sim}9wt%$ 용액을 시료로 한 쌀전분 수용액계의 유동거동을 $30{\sim}80^{\circ}C$ 범위에서 관형 리오메타를 사용하여 조사였다. 생전분의 현탁액은 딜라탄트유체거동을 보인 반면 호화액은 농도증가에 따라 의가소성으로부터 빙햄의 가소성의 유동거동을 보였다. 호화액의 유동거동지수 값은 모든 시료에서 약 1.2정도였으며 현탁액에서는 $0.87{\sim}0.90$범위였다. 3 및 5 % 호화액에서는 항복응력값을 무시할수 있었으나 이들 농도이상에서는 무시할 수없는 정도였고 농도증가에 따라 증가하였다. 점조도 지수값은 지수함수식에 따르는 농도 및 온도의존성을 나타내었다. 쌀전분 수용액계의 $30{\sim}80^{\circ}C$에서 측정한 활성화에너지는 현탁액과 호화액에서 각각 $0.13{\sim}2.71$$5.39{\sim}9.57\;Kcal/g\;mole$ 범위였다.

근적외선 분광법을 이용한 이탈리안 라이그라스 사일리지의 품질 평가 (Evaluation of the quality of Italian Ryegrass Silages by Near Infrared Spectroscopy)

  • 본 연구는 근적외선분광법의 현장 이용성 및 이탈리안 라이그라스 원물 사일리지의 신속한 품질평가를 위한 근적외선 검량식을 개발하기 위하여 전국에서 이탈리안 라이그라스 사일리지 약 450여 점을 수집하였다. 각각의 시료는 근적외선 분광기를 이용하여 스펙트럼을 측정한 후 측정된 스펙트럼과 실험실 분석값 간에 상관관계를 이용한 다변량회귀분석법을 통하여 검량식을 유도한 다음 각 성분별로 예측 정확성을 평가하였다. 이탈리안 라이그라스 원물 사일리지의 수분 함량에 대한 검량식 개발 결과는 검량식 작성시 표준오차(SEC)가 0.94% ($R^2$=0.99)로 매우 우수한 정확성을 보였으며 개발된 검량식의 상호검증(SECV) 결과는 1.27% ($R^2$=0.98)로 나타났다. ADF, NDF 및 조단백질 함량 평가를 위해 개발된 검량식의 상호검증(SECV) 결과는 각각 1.26% ($R^2$=0.88), 2.0% ($R^2$=0.84) 및 0.96% ($R^2$=0.93)으로 나타났으며 조회분함량 평가에 대한 검량식 개발 결과는 다소 낮은 정확성(SECV 0.72%, $R^2$=0.62)을 나타내었다. 사일리지의 발효품질의 주요 평가항목인 pH와 젖산함량 평가에 대한 검량식 개발결과(SEC)는 각각 0.41 ($R^2$=0.85), 0.18% ($R^2$=0.92)로 나타났으며 개발된 검량식의 상호검증 결과(SECV)는 각각 0.56 ($R^2$=0.78), 0.31%(0.81)로 나타났다.

  • 본 연구는 각종 유기성 폐기물의 혐기소화시 혐기성 분해 특성에 대해 알아보고자 하였다. 실험에 적용된 유기성 폐기물의 종류는 음식폐기물, 축산폐기물, 농산부산물 이었으며 기질과 접종액을 1:1로 혼합하여 회분식으로 실험을 수행하였고, 이때 발효조내의 TS, VS, sCOD 변화 및 가스발생량을 측정하였다. 음식폐기물의 경우 실험초기에 혐기성 분해가 일어났으며, 농산부산물의 경우 실험 종반부에 혐기성 분해가 일어났다. 반면 축산폐기물의 경우 실험초기에 한차례 혐기성 분해가 이루어지고 종반부에서 다시 혐기성 분해가 이루어짐을 알 수 있었다.

Technical and clinical aspects of cortisol as a biochemical marker of chronic stress

  • Stress is now recognized as a universal premorbid factor associated with many risk factors of various chronic diseases. Acute stress may induce an individual's adaptive response to environmental demands. However, chronic, excessive stress causes cumulative negative impacts on health outcomes through "allostatic load". Thus, monitoring the quantified levels of long-term stress mediators would provide a timely opportunity for prevention or earlier intervention of stressrelated chronic illnesses. Although either acute or chronic stress could be quantified through measurement of changes in physiological parameters such as heart rate, blood pressure, and levels of various metabolic hormones, it is still elusive to interpret whether the changes in circulating levels of stress mediators such as cortisol can reflect the acute, chronic, or diurnal variations. Both serum and salivary cortisol levels reveal acute changes at a single point in time, but the overall long-term systemic cortisol exposure is difficult to evaluate due to circadian variations and its protein-binding capacity. Scalp hair has a fairy predictable growth rate of approximately 1 cm/month, and the most 1 cm segment approximates the last month's cortisol production as the mean value. The analysis of cortisol in hair is a highly promising technique for the retrospective assessment of chronic stress. [BMB Reports 2015; 48(4): 209-216]

  • This study was conducted to investigate the physicochemical changes during the aerobic composting of swine manure mixed with bulking agent, sawdust(v/v, 1:1), in a full-scale composting plant using rectangular escalator-aginated bed composting system. Physical and chemical properties were analyzed on the samples which were collected at 5, 15, and 25 day of composting, curing and final step. The results of this study were summarized as follows; 1. Moisture and K2O content, and pH of final step were higher than those of 5th day of composting (p<0.05). 2. Ammonium nitrogen, total organic corbon and organic matter content, and electrical conductivity(EC) were significantly decreased (p<0.05) but nitrate nitrogen, ash and P2O5 content increased(p<0.05) throughout the aerobic composting process. 3. Total organic carbon per total nitrogen(C/N) and total organic matter per total nitrogen(OM/N) ratio were significantly decreased throughout the aerobic composting process(p<0.05). 4. Physical and chemical properties of swine manure were varied by aerobic fermentation using rectangular escalor-aginated bed composting system.

Effects of Nitrogen and Oxygen Supply on Production of $Poly-{\beta}-Hydroxybutyrate$ in Azotobacter chroococcum

  • Production of $poly-{\beta}-hydroxybutyrate$ (PHB) in a strain of Azotobacter chroococcum, a nitrogen-fixing bacteria, was investigated at various levels of nitrogen and oxygen. Feeding nitrogen source increased both cell growth and PHB accumulation. Oxygen supply appeared to be one of the most important operating parameters for PHB production. Both cell growth and PHB accumulation increased with the sufficient supply of air in the fed-batch fermentation of the strain. However, it was also noted that keeping the oxygen level under limited condition was critical to achieve high PHB productivity. A high titer of PHB (52 g/l) with a high cellular content (60%) was obtained after 48 hr of fed-batch operation by controlling the oxygen supply. Dual limitation of nitrogen and oxygen did not further increase the PHB accumulation probably due to the greater demand for reducing power and ATP for nitrogen fixation.

  • Most medium formulations for improving culture of entomopathogenic nematodes (EPN) based on protein sources have used enriched media like animal feed such as dried egg yolk, lactalbumin, and liver extract, among other ingredients. Most results, however, showed unstable yields and longer production time. Many of the results do not show the detailed parameters of fermentation. Soy flour, cotton seed flour, corn gluten meal, casein powder, soytone, peptone, casein hydrolysates, and lactalbumin hydrolysate as protein sources were tested to determine the source to support optimal symbiotic bacteria and nematode growth. The protein hydrolysates selected did not improve bacterial cell mass compared with the yeast extract control, but soy flour was the best, showing 75.1% recovery and producing more bacterial cell number ($1.4{\times}10^9$/ml) than all other sources. The highest yield ($1.85{\times}10^5$ IJs/ml), yield coefficient ($1.67{\times}10^6$ IJs/g medium), and productivity ($1.32{\times}10^7$ IJs/l/day) were also achieved at enriched medium with soybean protein.

Enhanced Biofuel Production from High-Concentration Bioethanol Wastewater by a Newly Isolated Heterotrophic Microalga, Chlorella vulgaris LAM-Q

  • Microalgal biofuel production from wastewater has economic and environmental advantages. This article investigates the lipid production from high chemical oxygen demand (COD) bioethanol wastewater without dilution or additional nutrients, using a newly isolated heterotrophic microalga, Chlorella vulgaris LAM-Q. To enhance lipid accumulation, the combined effects of important operational parameters were studied via response surface methodology. The optimal conditions were found to be temperature of $22.8^{\circ}C$, initial pH of 6.7, and inoculum density of $1.2{\times}10^8cells/ml$. Under these conditions, the lipid productivity reached 195.96 mg/l/d, which was markedly higher than previously reported values in similar systems. According to the fatty acid composition, the obtained lipids were suitable feedstock for biodiesel production. Meanwhile, 61.40% of COD, 51.24% of total nitrogen, and 58.76% of total phosphorus were removed from the bioethanol wastewater during microalgal growth. In addition, 19.17% of the energy contained in the wastewater was transferred to the microalgal biomass in the fermentation process. These findings suggest that C. vulgaris LAM-Q can efficiently produce lipids from high-concentration bioethanol wastewater, and simultaneously performs wastewater treatment.
