Changes of Organic Acids and Free Fatty Acids During the Ripening of Emmental Cheese (에멘탈치즈의 숙성 중 유기산과 유리지방산의 변화)

  • The objective of this study was to characterize the lactate metabolism and lipolysis in Emmental cheese made of Korean raw milk throughout the ripening periods; 14 d at $10^{\circ}C$, 42 d at $23^{\circ}C$, and 30 d at $4^{\circ}C$. Emmental cheese was made using a commercial starter culture with propionic acid bacteria (PAB) and without PAB as a control on the pilot plant scale. Changes in the contents of five organic acids (citric, lactic, formic, acetic, and propionic acid) and individual free fatty acids (FFAs) were measured using HPLC/PDA and GC/FID. As a result of propionic fermentation by PAB, the concentration of acetic acid and propionic acid increased up to 1.5 and 6.1 g/kg, respectively and the most dramatic increased occurred when incubated in the hot room ($23^{\circ}C$). Lactic, citric, and formic acid contents were 2.6, 2.5 and 0.8 g/kg at the end of ripening, respectively. As a result of lipolysis, the amount of total FFAs was 6,628.2 mg/kg. Compared to the control, levels of individual FFAs from butyric (C6:0) to linoleic (C18:2) acids increased significantly (p<0.05) during the ripening period. Especially, 65.1% of total FFAs was released in the $23^{\circ}C$ room and the most abundant FFAs were palmitic (C16:0), stearic (C18:0) and oleic acid (C18:1). These results demonstrated that the lipolysis of Emmental cheese was strongly affected by bacterial lipase from PAB.

Effect of Hydrocolloids on Physicochemical Properties of Bread Flour Dough with Rice Flour (Hydrocolloids가 쌀을 첨가한 빵용 밀가루 반죽의 물성학적 특성에 미치는 영향)

  • This study was carried out to investigate the effect of hydrocolloids [hydroxypropylmethyl cellulose (HPMC), xanthan gum (XG), guar gum (GG), and glucomannan (GM)] on the physicochemical properties of bread flour dough containing rice flour. In amylograph analysis, the significantly lowest gelatinization temperature was obtained in dough with XG (P<0.05). XG revealed the highest maximum viscosity while GM revealed the lowest. In viscograph test, the lowest gelatinization temperature and maximum viscosity showed the same result as in the amylograph. Breakdown value was also highest in dough containing XG, but lowest in the control and dough containing HPMC. Setback value was highest in dough containing HPMC, but lowest in dough containing XG. In farinograph analysis, consistency was greatest in dough with HPMC and XG. Hydrocolloids affected water absorption, which was highest in dough containing GM. Development times of dough containing HPMC and XG were low. Stability was lowest in dough with XG. Degree of softening was reduced in dough containing HPMC and GG compared to the control but increased in dough containing XG and GM. Dough containing HPMC and GG showed the largest volume at 3 h of fermentation. Dough with HPMC showed the lowest pH value. Hydrocolloids in this study affected physicochemical properties of dough.

Effect of Neupectin-L on Ethanol Production from Raw Starch Using a Co-Immobilized Aspergillus awamori and Zymomonas mobilis (Aspergillus awamori와 Zymomonas mobilis로 구성된 혼합고정화 배양계의 에탄올 생산에 미치는 Neupectin-L의 영향)

  • In order to reduce energy input in direct ethanol production from raw starch by co-immobilized Aspergillus awamori(A) and Zymomonas mobilis(Z), A-Z 36 culture system which was changed to anaerobic after 36 h of aerobic fermentation without sterilization was investigated. This immobilized cell system can not be carried out under unsterile conditions because of growth of microbial contaminants from original medium. Among some food additives such as sorbic acid, benzoic acid, dehydroacetic acid, p-hydroxybenzoic acid, Vantocil IB and Neupectin-L, Vantocil IB and Neupectin-L were a potent antibacterial agent in A-Z 36 culture cell system and were not affected in hydrolysis of substrate as compared with the case of control. Ethanol yield(6.9 g/l) in system of addition of 0.1% Neupectin-L was slightly higher than that in control(6.4 g/l). When 2% starch was fed five times in fed-batch culture with 0.1% Neupectin-L, ethanol yield and productivity were 34 g/l and 2.0 g/l/day, respectively.

Effects of Germinated and Fermented Unmarketable Soybean on Laying Performance and Egg Quality in Laying Hens (발아, 발효 처리한 비상품성대두 급여가 산란계의 생산성과 계란의 품질에 미치는 영할)

  • This study was conducted to investigate the effects of germinated and fermented unmarketable soybean (GFS) on laying performance and egg quality in laying hens. A total of two hundred laying hens were divided into 5 groups (5 treatment $\times$ 4 replication $\times$ 10 birds each) and fed with the experimental diets for 8 wk as follows: control, GFS free; T1, GFS 0.15%; T2, GFS 0.3%; T3, GFS 1%; T4, GFS 2%. The laying performance, egg quality, blood profiles, cecal microbial population, isoflavone content in egg yolk were investigated. There were no significant differences laying performance, relative liver and spleen weights, egg yolk color, eggshell color among groups. Eggshell strength in groups fed with diets containing GFS increased, but not significantly. Eggshell thickness significantly increased in the GFS-supplemented group. No significant differences were observed in the blood profiles and intestinal microflora after supplementation. The decrease of Haugh unit during storage was alleviated by feeding of GFS (p<0.05). The concentrations of malondialdehyde in groups fed with GFS were decreased as compared with control (p<0.05). Isoflavones in the egg yolk were detected in group fed with diet containing 2% GFS. These results showed that unmarketable GFS could be used as a favorable feed additive and feedstuff for production of quality enhanced and isoflavone fortified eggs.

Studies on the Shelf-life of the Grain Shape Improved Meju (낱알형 개량메주의 품질수명에 관하여)

  • In order to investigate the index component and basis for the shelf-life of the grain share improved Meju, the effect of the storage time and package on the quality of Meju and soy sauce were studied during 90 days storage at $30^{\circ}C$. Also, sensory evaluation for the soy sauces from Meju with various storage time were carried out. During the storage period, moisture content, amylase and protease activity were decreased, on the other hand, the contents of amino-nitrogen and ammonia-nitrogen were increased. Among these components, protease activity was found the major index component for quality control because it was the most important component for soy sauce fermentation and the most changeable component of Meju during the storage period. According to the sensory test, the quality of soy sauce was agreed well with the protease activity of Meju, and the soy sauce from longer storage Meju was inferior organoleptic quality to that from shorter Meju. By the storage quality test, protease activity showed the highest value in 15 days storage Meju and decreased gradually with storage time passed. The basis of protease activity for quality control was 200 (O.D. at 660 nm/g). which was 50% of the initial activity And it was known that the shelf-life of the grain shape improved Meju was about 90 days . It was also shown that the storage in package did not affect noticeably to prolong the shelf-life of Meju on this study.

Replacement of Imported Timothy Hay with Domestic Italian Ryegrass Silage in a Horse Feedstuff (말에서 국내산 이탈리안 라이그라스 사일리지의 수입산 티모시 건초 대체 효과)

  • This study was conducted to investigate the possibility of replacing imported Timothy hay (TH) with domestic Italian ryegrass silage (IRGS) as a horse feed considering feed quality, nutrient digestibility and feed price. Two experimental diets (TH and IRGS) were fed to six-headed Thoroughbred (body weight, 475.7±33.3kg) of the Korea Racing Authority of Wondang Stud Farm. The 3 head animals were assigned to Control group (TH) and Treatment group (IRGS), respectively. The nutrient digestibility was determined by the total collection method. IRGS is enough for using as a horse feed because its Relative feed value(RFV) was higher than TH and its fermentation quality is suitable for horses. Although no difference was observed in nutrient digestibility, Total digestible nutrients(TDN), and Digestible energy(DE) between Control and Treatment group (p>0.05), the fact that price of IRGS was much lower (53.7~62.4%) than that of TH indicates IRGS has competitive advantage over TH as a horse forage feed. The present study indicates that IRGS can be fully replaced with TH due to its superior economic value even though the similarity of its nutrient digestibility, TDN, and DE to TH.

Effects of Dietary Fermented Apple Pomace and Cinnamon Addition on Meat Quality and Performance in Broiler (육계에 사과박 발효물, 계피의 첨가, 급여가 생산성 및 계육 품질에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary supplementation of fermented apple pomace (FAP) and cinnamon on performance, blood profile and meat quality in broiler chickens. A total of six hundred, day old male broiler chicks (Cobb) were randomly divided into 5 groups with 4 replicates of 30 birds each. The treatment groups were; negative control (antibiotic-free diet), positive control (basal diet with 0.05% antibiotics and 0.03% anticoccidials), FAP 1.0%, cinnamon 0.1% and FAP 1.0% + cinnamon 1.0%. The body weight of the broilers fed the diets containing FAP 1.0% was higher than the other treatments during overall period. No synergistic effect in growth performance (weight gain and feed conversion rate) was found when FAP with cinnamon were fed to broilers. No significant differences were observed on the concentration of serum blood chemical. the TBARS was lower in the chickens fed 1.0% FAP diet compared to those of negative group and positive group. These results suggest the possibility that fermented of apple pomace could be used as an alternative to antibiotic growth promoters to improve the performance and meat quality of broiler chickens.

Effect of lotus leaf on the quality characteristics and antioxidant properties of bread (연잎 첨가가 제빵 특성 및 항산화 활성에 미치는 영향)

  • This study was carried out to investigate the quality characteristics and antioxidative activity of bread containing lotus leaf powder (in ratios of 0, 1, 2, and 3% of the total flour). The pH of dough and bread were decreased as the concentration of lotus leaf powder increased. The volume of dough during fermentation did not show significant difference between 1% lotus leaf powder containing dough and control. The weight of bread increased by addition of lotus leaf powder. The volume, specific volume and baking loss rate of the bread decreased as lotus leaf powder levels increased. Redness and yellowness of the inner crumb were decreased by the addition of lotus leaf powder, but lightness was increased. 2,2-diphenyl-1-picrylhydrazyl-radical scavenging activity and total polyphenol contents were increased significantly as the concentration of lotus leaf powder increased. The overall acceptability of bread containing 1% lotus leaf powder showed no significant difference compared with control. The result exhibited that adding the lotus leaf powder into the bread increased antioxidant activity, and the highest quality improvement was obtained by incorporating 1% (w/w) lotus leaf powder into the bread formula.

On Preparation and Effects of Composts from Industrial Wastes via High Temperature and Aerobic conditions (유기성 산업폐기물의 고온·호기성 퇴비화 및 비효평가)

  • In order to find a way to utilize paper mill sludge, its composting was conducted with anaerobic waste of kraft paper sludge, raw kraft paper sludge, and CMC sludge(CMC : Sodium Carboxymethylcellulose) under aerobic condition at $50^{\circ}C$. It took 3 days for initial fermentation with anaerobic waste and CMC sludge, and six days with raw kraft paper sludge. Each compost was applied to radish(Rhaphanus Stativus L.), and absorption rate of staple nutrients increased 6.7~9.3 times higher in N, 17~21 times in P and 2~3 times in K than control at the harvesting stage. Also, organic matter contents were increased 1.5~2.3 times 4.5~5.3 times in CEC compared to control soil.

Chemosensitization of Fusarium graminearum to Chemical Fungicides Using Cyclic Lipopeptides Produced by Bacillus amyloliquefaciens Strain JCK-12

  • Fusarium head blight (FHB) caused by infection with Fusarium graminearum leads to enormous losses to crop growers, and may contaminate grains with a number of Fusarium mycotoxins that pose serious risks to human and animal health. Antagonistic bacteria that are used to prevent FHB offer attractive alternatives or supplements to synthetic fungicides for controlling FHB without the negative effects of chemical management. Out of 500 bacterial strains isolated from soil, Bacillus amyloliquefaciens JCK-12 showed strong antifungal activity and was considered a potential source for control strategies to reduce FHB. B. amyloliquefaciens JCK-12 produces several cyclic lipopeptides (CLPs) including iturin A, fengycin, and surfactin. Iturin A inhibits spore germination of F. graminearum. Fengycin or surfactin alone did not display any inhibitory activity against spore germination at concentrations less than 30 ug/ml, but a mixture of iturin A, fengycin, and surfactin showed a remarkable synergistic inhibitory effect on F. graminearum spore germination. The fermentation broth and formulation of B. amyloliquefaciens JCK-12 strain reduced the disease incidence of FHB in wheat. Furthermore, co-application of B. amyloliquefaciens JCK-12 and chemical fungicides resulted in synergistic in vitro antifungal effects and significant disease control efficacy against FHB under greenhouse and field conditions, suggesting that B. amyloliquefaciens JCK-12 has a strong chemosensitizing effect. The synergistic antifungal effect of B. amyloliquefaciens JCK-12 and chemical fungicides in combination may result from the cell wall damage and altered cell membrane permeability in the phytopathogenic fungi caused by the CLP mixtures and subsequent increased sensitivity of F. graminearum to fungicides. In addition, B. amyloliquefaciens JCK-12 showed the potential to reduce trichothecenes mycotoxin production. The results of this study indicate that B. amyloliquefaciens JCK-12 could be used as an available biocontrol agent or as a chemosensitizer to chemical fungicides for controlling FHB disease and as a strategy for preventing the contamination of harvested crops with mycotoxins.
