  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks. then they were fed with the diet containing lard 3.0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil, and antihyperlipidemic drugs for 2 weeks. Analysis of the fatty acid composition of the brain and heart lipids showed following results. In the fatty acid composition of brain lipids, C20:4 and C22:6 were the major fatty acids but showed little difference among the groups. In the fatty acid of heart lipids,C18:2 was the major fatty acid. The proportion of C20:4 decreased gradually as n-3P/n-6P ratio of the test lipids increased in groups 5 (linseed oil 12.0%) to 9 (sunflower seed oil 12.0%) while the proportion of C22:6 was not affected by the fatty acid composition of the test lipids.

Fatty Acid Analysis from Leech Skin

  • The lipids of a Korean native blood-sucking leech (Hirudo nipponia) were isolated and analyzed. Cholesterol, fatty acids, triglycerides, glycerylether diester and cholesteryl esters were found from epidennal extracts. The major fatty acids in the leech skin were $C_{16:0}$, $C_{18:0}$, $C_{20:1}$. $C_{20:3}$ components. These components were derived from cholesteryl esters, glycerides, fatty acids and other polar lipids. After 60 days fasting, cholesterol, fatty acids and triglycerides in the skin were reduced in number and the total fatty acid composition was changed slightly. The data of total fatty acids in fasted and fed leech showed that medium-chain fatty acids were more affected than long-chain fatty acids. Other trace fatty acids appeared to be decreased by fasting.

Discriminating Eggs from Two Local Breeds Based on Fatty Acid Profile and Flavor Characteristics Combined with Classification Algorithms

  • This study discriminated fatty acid profile and flavor characteristics of Beijing You Chicken (BYC) as a precious local breed and Dwarf Beijing You Chicken (DBYC) eggs. Fatty acid profile and flavor characteristics were analyzed to identify differences between BYC and DBYC eggs. Four classification algorithms were used to build classification models. Arachidic acid, oleic acid (OA), eicosatrienoic acid, docosapentaenoic acid (DPA), hexadecenoic acid, monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), unsaturated fatty acids (UFA) and 35 volatile compounds had significant differences in fatty acids and volatile compounds by gas chromatography-mass spectrometry (GC-MS) (p<0.05). For fatty acid data, k-nearest neighbor (KNN) and support vector machine (SVM) got 91.7% classification accuracy. SPME-GC-MS data failed in classification models. For electronic nose data, classification accuracy of KNN, linear discriminant analysis (LDA), SVM and decision tree was all 100%. The overall results indicated that BYC and DBYC eggs could be discriminated based on electronic nose with suitable classification algorithms. This research compared the differentiation of the fatty acid profile and volatile compounds of various egg yolks. The results could be applied to evaluate egg nutrition and distinguish avian eggs.

Effect of Growth on Fatty Acid Composition of Total Intramuscular Lipid and Phospholipids in Ira Rabbits

  • The changes in fatty acid composition of total intramuscular lipid and phospholipids were investigated in the longissimus dorsi, left-hind leg muscle, and abdominal muscle of male Ira rabbits. Changes were monitored at 35, 45, 60, 75, and 90 d. Analysis using gas chromatography identified 21 types of fatty acids. Results showed that the intramuscular lipid increased and the intramuscular phospholipids (total intramuscular lipid %) decreased in all muscles with increasing age (p<0.05). An abundant amount of unsaturated fatty acids, especially polyunsaturated fatty acids, was distributed in male Ira rabbits at different ages and muscles. Palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1), linoleic acid (C18:2), and arachidonic acid (C20:4) were the major fatty acids, which account to the dynamic changes of the n-6/n-3 value in Ira rabbit meat.

Determination of Fatty Acid Composition in Peanut Seed by Near Infrared Reflectance Spectroscopy

  • This study was conducted to develop a fast and efficient screening method to determine the quantity of fatty acid in peanut oil for high oleate breeding program. A total of 329 peanut samples were used in this study, 227 of which were considered in the calibration equation development and 102 were utilized for validation, using near infrared reflectance spectroscopy (NIRS). The NIRS equations for all the seven fatty acids had low standard error of calibration (SEC) values, while high R2 values of 0.983 and 0.991 were obtained for oleic and linoleic acids, respectively in the calibration equation. Furthermore, the predicted means of the two main fatty acids in the calibration equation were very similar to the means based on gas chromatography (GC) analysis, ranging from 36.7 to 77.1% for oleic acid and 7.1 to 42.7% for linoleic acid. Based on the standard error of prediction (SEP), bias values, and $R^2$ statistics, the NIRS fatty acid equations were accurately predicted the concentrations of oleic and linoleic acids of the validation sample set. These results suggest that NIRS equations of oleic and linoleic acid can be used as a rapid mass screening method for fatty acid content analysis in peanut breeding program.

한국 호박의 지방산 조성 (Fatty Acid Composition of Korean Pumpkins)

  • In order to investigate the fatty acid composition of Korean pumpkins, this study was designed. Three samples of Korean pumpkins, A(yellowish ripe), B(unripe, 30 days old) and C(unripe, 20 days old) were used for fatty acid analysis by gas chromatography. The results were as follows : Pumkin C, B and A showed 10, 12 and 15 kinds of fatty acids, respectively. In case of palmitic acid(C16:0), pumpkin C, B and A showed 22.5%, 22.9% and 26.6%, respectively. In case of u linolenic acid(C18:3), pumpkin C, B and A showed 22.6%, 23.6% and 44.8%, respectively. In case of palmitoleic acid(Cl6:1), pumpkin C. B and A showed 12.4%, 10.1% and 0.5%, respectively. In case of oleic acid(C18:1), pumpkin C. B and A showed 9.2%, 7.7% and 2.8%, respectively, In case of stearic acid(C18:0), pumpkin C, B and A showed B.S%, 3.7% and 3.5%, respectively. The ratio of P/S was 1.4, 1.6 and 1.9 for sample C, B and A, respectively. The ratio of w-3/w-6 was 1.1, 0.9 and 2.6 for sample C, B and A, respectively. Through this study, the ripe pumpkin(sample A) was thought to be good enough in nutritional aspects of fatty acids, particularly a-linoleic acid, Cl8 : 3(u-3) series.

지방산 조성이 다른 식이 지방이 흰쥐 간 미토콘드리아의 지질조성과 Adenine Nucleotide Translocase 활성도에 미치는 영향 (Effects of Different Dietary Oils on Hepatic Mitochnodial Lipid Composition and Adenine Nucleotide Translocase Activity in Rat)

  • This study was done to investigate whether dietary fats differing in their fatty acid compositions change hepatic mitochondrial lipid composition and thereby change adenine nucleotide translocase activity. Male Sprague-Dawley rats were fed 5 different wxperimental diets for 6 weeks, which were different in their fatty acid compositions. The dietary fats were beef tallow(BT), olive oil(OO), corn oil(CO), perilla oil(PO) and sardine oil(SO) as a source of saturated fatty acid, oleic acid, n-6 linoleic acid, n-3 $\alpha$-linolenic acid and n-3 eiocosapentaenoic acid+docosahexaenoic acid respectively. Body weight of PO group was significantly higher than that of either BT or SO group. This increase in body weight of PO group was due to the increase of food intake. Although there was no difference in liver weight, % liver weight per body weight of SO group was significantly higher than BT and OO groups. Analysis of mitochondrial lipid composition showed that dietary oils differing their fatty acid compositions altered mitochondrial fatty acid patterns, especially n-6/n-3 ratio, cholesterol/phospholipid ratio and phopsholipid composition. The n-6/n-3 ratio was highest in CO group but lowest in SO group whereas the ratio of Chol/PL was highest in SO group but lowest in CO group. Such changes in mitochondrial lipids did not lead to a significant alteration in the activities of adenine nucleotide translocase, which is embedded in mitochodrial inner membrane.

남극해 크릴 새우의 지방산조성과 무기질 및 비타민함량 (Fatty Acid Compositions, Mineral and Vitamin Contents of the Antarctic Krill (Euphausia superba))

  • Antarctic krill (Euphausia superba) is typical of a enormous biomass of marine zooplankton that could provide good nutrition in human body. This study was conducted to identify the nutrition of krill, a live in Antarctic Ocean. The analysis result of fatty acids of krill meal was as follow. The compositions of fatty acid were saturated fatty (SFA) acid 41.41%, monounsaturated fatty acid (MUFA) 21.69%, polyunsaturated fatty acid (PUFA) 36.89%, and p/s ratio was 0.89. The major fatty acids in all parts were eicosapentaenoic acid (EPA, 21.54%), palmitic acid (27.51%), oleic acid (13.35%) and docosahexaenoic acid (DHA, 12.42%). Especially, EPA and DHA were occupied 33.96% of polyunsaturated fatty acid. The mineral contents of krill meal were calcium 24477.21 mg $kg^{-1}$, sodium 14728.69 mg $kg^{-1}$, magnesium 6973.49 mg $kg^{-1}$, potassium 3981.67 mg $kg^{-1}$, iron 395.33 mg $kg^{-1}$ and manganese 5.74 mg $kg^{-1}$. The contents of major vitamin were retinol 86717.37 ${\mu}g$ RE, ${\beta}$-carotene 44.87 ${\mu}g$ RE, tocopherol 2.60 mg, pantothenic acid 1.61 mg, indicating that krill meal contains large amount of retinol and ${\beta}$-carotene.

The combination of canagliflozin and omega-3 fatty acid ameliorates insulin resistance and cardiac biomarkers via modulation of inflammatory cytokines in type 2 diabetic rats

  • The present study was carried out with the hypothesis that combination of canagliflozin and omega-3 fatty acid may have potential effect on insulin level, insulin resistance, cardiac biomarkers, inflammatory cytokines and histological studies in type 2 diabetes mellitus (DM). Type 2 DM was induced by injecting nicotinamide (120 mg/kg, i.p.) 15 min before STZ (60 mg/kg) injection. Canagliflozin (5 and 10 mg/kg) and omega-3 fatty acid (300 mg/kg) were given for 28 days after confirmation of diabetes. Biochemical estimations revealed elevated levels of glucose, insulin, HOMA-R and inflammatory cytokines in diabetic group. Daily dosing of alone canagliflozin and omega-3 fatty acid slightly reduced elevated levels of glucose, insulin, HOMA-R and inflammatory cytokines ($IL-1{\beta}$, IL-2, and $TNF{\alpha}$), whereas canagliflozin and omega-3 fatty acid combination has reduced these biochemical parameters significantly when compared with diabetic group. Similarly in diabetic group the levels of cardiac biomarkers such as lipid profile, LDH, CKMB and troponin were significantly increased. Elevated levels of cardiac biomarkers were significantly reduced after daily dosing of alone canagliflozin and omega-3 fatty acid. Canagliflozin and omega-3 fatty acid combination has offered better improvement in cardiac biomarkers compared to alone canagliflozin and omega-3 fatty acid. Histopathological analysis also supported the above hypothesis that combination therapy (canagliflozin and omega-3 fatty acid) offered better protection against degenerative changes in ${\beta}-cells$ of pancreas as compared to alone treatment with these drugs. Thus the present study revealed that canagliflozin and omega-3 fatty acid can be used as potential combination therapy in type 2 DM along with cardiac complication.

지방산대사이상질환 스크리닝을 위한 TBDMS 유도체화 후 GC-MS를 이용한 혈장 중 유리지방산의 분석 (A Sensitive Determination of Plasma Free Fatty Acids Following Tert-butyldimethylsilyl Derivatization using Gas Chromatography-Mass Spectrometry for Screening of Fatty Acid Oxidation Disorders)

  • Purpose: A sensitive gas chromatography mass spectrometry (GC-MS) method was developed for screening of fatty acid oxidation disorders. Methods: The assay utilized a simple protein precipitation with sulfosalicylic acid followed by tert-butyl dimethylsilyl (TBDMS) derivatization of hydroxyl functional group by N-tert-butyldimethylsilyl-N-methyltrifluoroacetamide (MTBSTFA). Results: Calibration curves of spiked pooled plasma showed a linear relationship in the range of 0.01 ng -2 mg with correlation coefficient value greater than 0.98. Limits of detection (LOD) and limits of quantification (LOQ) were found in the range of 0.9-8.8 ng and 9-88 ng, respectively. Conclusion: The new developed method might be useful for a rapid, sensitive screening of inherited fatty acid oxidation disorders. In addition, the method expected to be one of the alternative method for screening newborns of metabolic disorders in the laboratories where expensive MS/MS is unavailable.
