배추흰나비의 變態에 따른 血球細胞의 組織化學的 硏究 (Histochemical Study on the Hemocytes during Metamorphosis)

  • 1. 變態時期에 따른 배추흰나비(Pieris rapae)의 血球를 組織化學的方法 및 autoradiographic method 에 의거하여 硏究하였다. 2.幼蟲, 전용 및 용기에서 形態의變化와 細胞內容物을 土臺로 血球細胞를 prohemocyte, plasmatocyte, podocyte, granular hemocyte, spherule cell 및 oenocytoid 의 6型으로 分類했다. 3, 幼蟲基에서 용기까지의 血球를 組織化學的으로 硏究하고 血球細胞內의 蛋白質, 核酸, 多糖類 및 脂質을 檢出하였으며, 成長 및 變態에 따른 그들 物質의 消長을 밝혔다, 4. Granular hemocyte 와 spherule cell 의 大部分의 顆粒은 glycogen, neutral mucopolysaccharide, mucoprotein 으로 되었고 neutral fat와 phospholipid 의 顆粒도 나타났다, 5. 細胞分裂像(mitotic figure) 과 tritium-thymidine 攝取세포 (DNA 合成細胞)를 幼蟲 2齡부터 5齡까지의 各種 未分化血球에서 觀察하였고, 모든型의 血球가 prohemocyte에 起源함을 밝혔다. 6. Plasmatocyte 와 podocyte 의 細胞質突起가 전용기 와 용기에 매우 길게 伸張함을 보아 vermiform cell 은 plasmatocyte 의 더 分化한 型이라고 생각된다.

Ginsenoside Rg1 suppresses early stage of adipocyte development via activation of C/EBP homologous protein-10 in 3T3-L1 and attenuates fat accumulation in high fat diet-induced obese zebrafish

  • Background: Ginsenoside Rg1 is a class of steroid glycoside and triterpene saponin in Panax ginseng. Many studies suggest that Rg1 suppresses adipocyte differentiation in 3T3-L1. However, the detail molecular mechanism of Rg1 on adipogenesis in 3T3-L1 is still not fully understood. Methods: 3T3-L1 preadipocyte was used to evaluate the effect of Rg1 on adipocyte development in the differentiation in a stage-dependent manner in vitro. Oil Red O staining and Nile red staining were conducted to measure intracellular lipid accumulation and superoxide production, respectively. We analyzed the protein expression using Western blot in vitro. The zebrafish model was used to investigate whether Rg1 suppresses the early stage of fat accumulation in vivo. Results: Rg1 decreased lipid accumulation in early-stage differentiation of 3T3-L1 compared with intermediate and later stages of adipocyte differentiation. Rg1 dramatically increased CAAT/enhancer binding protein (C/EBP) homologous protein-10 (CHOP10) and subsequently reduced the $C/EBP{\beta}$ transcriptional activity that prohibited the initiation of adipogenic marker expression as well as triglyceride synthase. Rg1 decreased the expression of extracellular signal-regulated kinase 1/2 and glycogen synthase kinase $3{\beta}$, which are also essential for stimulating the expression of $CEBP{\beta}$. Rg1 also reduced reactive oxygen species production because of the downregulated protein level of nicotinamide adenine dinucleotide phosphate hydrogen (NADPH) oxidase 4 (NOX4). While Rg1 increased the endogenous antioxidant enzymes, it also dramatically decreased the accumulation of lipid and triglyceride in high fat diet-induced obese zebrafish. Conclusion: We demonstrated that Rg1 suppresses early-stage differentiation via the activation of CHOP10 and attenuates fat accumulation in vivo. These results indicate that Rg1 might have the potential to reduce body fat accumulation in the early stage of obesity.

Gramoxone이 단백질 level에 따라 흰쥐 간에 미치는 독성에 관한 연구 (Effect of Dietary Protein Levels on the Manifestation of Gramoxone Toxicity in Rat Liver)

  • 식이속의 단백질농도가 피리딘계 제초제인 gramoxone의 독성발현에 어떤 영향을 미치는가를 알기 위해 Wistar계 흰쥐 수놈 36마리 (6~7주령)를 6개의 실험사료군으로 나누어 2주간 사육하며 그 미치는 영향을 조사하였다. 체중증가 상황은 각단백-gramoxone군들의 경우 각 정상단백군에 비해 저조하였으며 저단백-gramoxone군에서 제일 심하였고 고단백-gramoxone군에서는 유의한 차가 없었다. 간의 지질함량변화는 각단백-gramoxone군들의 경우, 고단백-gramoxone군을 제외하고 각 단백군에 비해 증가하는 경향을 나타내었으며 특히 저단백-gramoxone군에서 가장 심하였다. 간장내 TBA가는 정상단백군, 저단백군, 고단백군 및 고단백-gramoxone군 사이에는 유의차가 없었으나 정상단백-gramoxone군과 저단백-gramoxone군의 경우 전자의 실험군들에 비해 매우 높았다. 정상단백군 및 고단백군의 간에서는 유의한 형태적인 변화가 없었으나 저단백군과 각단백-gramoxone군들에서는 정도의 차이는 이었으나 간조직의 변화가 나타났으며 간세포 지방변화 및 Kupffer 세포의 숫적인 증가가 관찰되었다. 특히 간세포 지방변화는 저단백-gramoxone군에서 심하였고 고단백-gramoxone군에서는 현저하지않았다. 간세포의 glycogen함량은 각단백-gramoxone군들에서 타군들에 비해 증가하는 경향을 보였으며 저단백-gramoxone군에서 제일 심하였다. 제일심하였다.

백서에서 식이내 열량 영양소의 배분이 인슐린 분비능과 인슐린 저항성에 미치는 영향 (The Effects of Dietary Caloric Distribution on Insulin Secretion and Insulin Resistance in Sprague Dawley Rats)

  • The prevalence of type 2 diabetes mellitus has been rapidly increased in parallel with the westernization of eating behavior in Korea. Increased consumption of animal fat and simple sugar can be potential contributors for insulin resistance. The purpose of the study was to determine whether Western-(WD) and Korean-style(KD) diets altered insulin secretion and insulin resistance in male Sprague Dawley rats. Rats weighing 98$\pm$5g were provided by KD(77 En% of starch, 5 En% of corn oil and 13 En% of gluten plus 5 En% of casein), WD(42 En% of starch, 40 En% of butter and 18% of casein) or control diet(62 En% of starch, 20 En% of corn oil and 18% of casein) for 12 weeks. Body weights were lower in KD compared to WD. Fasting blood glucose levels were not different among diets. Insulin secretion from the beta cells was higher by 2.2$\pm$0.4 folds in WD than KD at baseline. In hyperglycemic clamp insulin secretion was higher in WD than KD and CD. Whole body glucose disposal rates referred to the state of insulin sensitivity were lowest in WD among groups. Glycogen deposits in soleus and quadriceps muscles were lowest in WD among all groups, but their triglyceride contents were highest. GLUT4 contents and glycogen synthase were lowest in WD in both muscles. In conclusions, westernization of diets needed more insulin to normalization of blood glucose levels due to increased insulin resistance. Thus, WD would lead to increased prevalence of diabetes mellitus when increased insulin resistance could not be compensated by insulin secretion in the case of elevated blood glucose levels. 백미대맥(白米大麥) 및 속(粟) 사료(飼料)가 실험용(實驗用) 백서(白鼠)에 미치는 영양학적(營養學的) 및 생물화학적(生物化學的) 연구(硏究) (Biochemical and Nutritional Effects of Rice, Barley, and Millet on Rats)

  • Dividing into six(6) experimental groups; Stock Diet(SD) Group, Rice Diet(RD) Group, Rice Diet supplemented with Barley(RD-BD) Group, Barley Diet(BD) Group, Rice Diet supplemented with Millet(RD-MD) Group, and Millet Diet(MD) Group, biochemical and nutritional effects of rice, barley, and millet on rats are studied. The grains used in this experiment are polished ones and purchased from market. Six(6) weaning male albino rats are housed into the individual cages in each group and fed with each experimental diet and water ad libitum for 21 weeks. The composition of the experimental diets are shown in Table 1. At the end of experiment, the appearences of rats are observed and the bloods are sampled by heart puncture method following ether anesthesia for determinations of serum protein and serum cholesterol. And the livers are sampled immediately right after heart after heart puncture for determinations of liver weight, liver nitrogen, liver fat, and liver glycogen. And also liver tissues are observed histologically. The result of growth experiment is shown in Table 2 and Figure 7. The results of blood analysis are shown in Table 3 and the results of liver analysis are shown in Table 4. And the histological observations of liver tissue are shown in Figures 1, 2, 3, 4, 5, and 6. According to the above results, it could be summarized as follows; 1) The normal appearences are observed at SD Group and BD Group. Alopecia is observed at RD, RD-­BD, RD-MD, and MD groups and the wet appearences of hair are observed at RD-MD and MD groups. The color of hair is changed to yellowish white at RD, RD-MD groups. The maximum growth inhibition, is observed at MD group and the growths of RD, RD-BD, BD, and RD-MD groups are significantly lower than that of SD group. The growth of RD group is lower than those of RD-BD and BD groups, though this difference is not significant. 2) At the blood analysis, the highest serum protein content is shown at SD group and the lowest serum protein is observed at MD group. The serum cholesterol contents are not significantly different among the six(6) experimental groups. 3) The highest liver weight is observed at SD group and the lowest weight is shown at MD group. However, oppositely the highest liver weight per 100 grams of body weight at MD group and the lowest liver weight per 100 grams of body weight at SD group are observed as shown in Table 6. 4) The differences of liver nitrogen contents of RD, RB-BD, BD, RD-MD, and MD Groups are not significant except SD group showing the highest content. 5) The significant highest content of liver fat is observed at RD group and the lowest content of liver fat is shown at SD group. And no significant differences in contents of liver glycogen are observed among the six(6) experimental groups. 6) No significant abnormal histological changes of liver tissue are observed at SD, RD-BD, and BD groups. However, the fat metamorphosis at peripheral zone of liver tissue is observed at RD Group and the some histological abnormal changes are observed at RD-MD and MD groups. According to the above summaries, the followings might be estimated and recommended; 1) The BD group shows histological normality of liver tissue, improvement of liver fat and liver nitrogen, normality of appearence, and better growth compared with RD Group. This could be indicated that barley is nutritionally and biochemically better than rice as the feeding diet on rat. And it might be considered that there might be the species speciality for nutritionary evaluation of food. 2) Recently, Marshall indicated that the high consumption of carbohydrate causes fatty liver and Kondo reported that the higher consumption of rice causes the more heart disease and the shorter life span. Considering these reports, the higher consumption of rice as a main dish in our country should be considered as an important national nutritional problem. And this might be improved by supplementing with barley or other grains to rice.

도라지의 급이가 고콜레스테롤혈증 흰쥐의 간기능 및 간조직의 지질조성에 미치는 영향 (Effect of the Feeding Platycodon grandiflorum on Lipid Components of Liver and Liver Function in Hypercholesterolemia Rats)

  • 도라지의 급이가 고콜레스테롤혈증 흰쥐의 간기능과 지질대사에 미치는 영향 구명하기 위하여 SD계 숫흰쥐에게 동결건조한 도라지 분말 5%와 도라지 분말 5%에 해당하는 식물섬유, saponin, 에탄올 추출물을 각각 급이하여 3주간 실험 사육한 후 혈청효소 및 간장의 지질성분 분석, 간세포의 형태를 검토하였다. 혈청 중 AST, ALT효소활성은 대조군에 비해 전 실험군이 유의적으로 감소하였고 그 중 saponin 급이군이 가장 낮았고 그 다음이 식물 섬유군이었다. LDH효소 활성도 전 실험군에서 유의적으로 낮게 나타났으며 22년근 도라지의 식물 섬유군과 saponin군이 낮은 값을 보였다. 간장 중의 총 콜레스테롤 함량은 4년근과 22년근 도라지 분말 및 식물섬유 급이군이 비교적 낮았으며, 중성지질 함량은 대조군이 비해 전 실험군에서 낮았으나, 각 실험군간의 차이는 크지 않았고, 인지직 함량은 전 실험군에 있어 비슷한 수준이었다. 간장 조직세포의 전자현미경 관철 결과, 대조군은 지방구가 증가되어 있는 반면 도라지 급이군에서는 감소되었으며, 22년근 도라지 급이군은 간장 글리코겐 축적량이 증가되어 있었다.

Effects of Starvation on Lipid Metabolism and Gluconeogenesis in Yak

  • This research was conducted to investigate the physiological consequences of undernourished yak. Twelve Maiwa yak ($110.3{\pm}5.85kg$) were randomly divided into two groups (baseline and starvation group). The yak of baseline group were slaughtered at day 0, while the other group of yak were kept in shed without feed but allowed free access to water, salt and free movement for 9 days. Blood samples of the starvation group were collected on day 0, 1, 2, 3, 5, 7, 9 and the starved yak were slaughtered after the final blood sample collection. The liver and muscle glycogen of the starvation group decreased (p<0.01), and the lipid content also decreased while the content of moisture and ash increased (p<0.05) both in Longissimus dorsi and liver compared with the baseline group. The plasma insulin and glucose of the starved yak decreased at first and then kept stable but at a relatively lower level during the following days (p<0.01). On the contrary, the non-esterified fatty acids was increased (p<0.01). Beyond our expectation, the ketone bodies of ${\beta}$-hydroxybutyric acid and acetoacetic acid decreased with prolonged starvation (p<0.01). Furthermore, the mRNA expression of lipogenetic enzyme fatty acid synthase and lipoprotein lipase in subcutaneous adipose tissue of starved yak were down-regulated (p<0.01), whereas the mRNA expression of lipolytic enzyme carnitine palmitoyltransferase-1 and hormone sensitive lipase were up-regulated (p<0.01) after 9 days of starvation. The phosphoenolpyruvate carboxykinase and pyruvate carboxylase, responsible for hepatic gluconeogenesis were up-regulated (p<0.01). It was concluded that yak derive energy by gluconeogenesis promotion and fat storage mobilization during starvation but without ketone body accumulation in the plasma.

Study of the effect of diet supplemented with powdered Salicornia herbacea L. on glucose transporter and antioxidant capacities in rats

  • This study was conducted to examine the possibility of Salicornia herbacea L. powder as a functional food beneficially affecting carbohydrate metabolism and antioxidant capacity. Twenty-four, 6-week old, maleSprague-Dawleyrats were classified into three groups: normal diet control (CON), high-fat diet control (HFC) and high fat diet + Salicornia herbacea L. (SHF). Related feed was provided to each group for 4 weeks. Weight gain rate increased most in the HFC rats, and the concentration of glucose was significantly high in both the HFC and SHF groups, compared to the CON group. The SHF group showed a significantly high expression rate of Glut 4 (21.36%), compared to the CON and HFC groups. The glycogen content in muscle was significantly high in both the HFC and SHF groups, compared to the CON group. There were significant differences in the malondialdehydecontent in muscles between the groups, with the content in the CON and HFC groups being significantly higher than the SHF group. All the groups showed a similar tendency to each other in the liver tissue as well. Concerning the expression of Cu,Zn-super oxide dismutase andglutathione peroxidaseproteins, the SHF group was significantly higher than the CON and HFC groups. Overall, the experiment result above implies a possibility that an intake of Salicornia herbacea L. powder can regulate weight by decreasing the weight gain rate, further suggesting its effectiveness as a functional food before exercise by increasing the energy storage capacity and antioxidant capacity.

산양삼을 첨가한 고지방식이 흰쥐 골격근의 지질과산화 및 항산화 단백질 발현 효과 (Effects of Mountain Ginseng-added High Fat diet on Lipid Peroxidation and Antioxidant Protein Expression of Skeletal Muscle in Rats)

  • 본 연구에서는 산양삼을 첨가한 고지방식이를 하였을 때 흰쥐 골격근의 지질과산화와 항산화 단백질의 발현 정도를 분석하고자 하였다. 32마리의 웅성 흰쥐를 대상으로 난괴법을 사용하여 HS(고지방식이 대조군), MG1(0.5%의 산양삼 첨가식이 섭취군), MG2(1.0%의 산양삼 첨가식이 섭취군), 그리고 MG3(2.0%의 산양삼 첨가식이 섭취군)으로 각각 8마리씩 분류하였다. 식이는 4주간 섭취시켰다. 혈중 중성지방은 MG1과 MG2가 HS에 비하여 유의하게 낮았으며, 혈중 HDL-C는 MG3이 HS 및 MG2에 비하여 높게 나타났다. 근육 글리코겐 함량은 MG2, MG3이 HS와 MG1에 비하여 높았으며, MDA 함량은 MG1, MG2, MG3이 HS에 비하여 낮은 경향만이 나타났다. 비복근의 GPx 단백질 발현은 MG2가 HS에 비하여 높았으며, Cu,Zn-SOD은 MG1과 MG2가 높게 나타났다. Mn-SOD는 MG1, MG2, MG3이 HS에 비하여 높은 경향만이 나타났다. 이상의 결과, 고지방식이에 산양삼 첨가는 골격근의 MDA 함량을 저하시키고 항산화기능을 증가시키는 중요한 역할을 한다고 할 수 있다.

유산동(硫酸銅) 중독(中毒)이 랫트 간장(肝臟)의 미세구조(微細構造)에 미치는 영향(影響) (Effects of copper sulfate poisoning on the fine structure of rat liver)

  • This study was designed in order to investigate the effect of copper sulfate to the ultrastructural changes of the hepatocytes in Sprague Dawley rats. The animals were administered with copper sulfate (10mg/kg B.W.), which was dissolved in normal saline. The solution was injected into abdominal cavity every day. The animals were sacrificed at the 6th, 12th, and 24th day from the beginning of administration. The specimens obtained from the liver were observed with electron microscope and significant changes were as follows. 1. A prominent dilatation and disruption of the cisternae of rough endoplasmic reticulum were recognized. Also, the detachment of membrane bound ribosomes was shown. 2. The proliferation of smooth endoplasmic reticulum and the depletion of glycogen particles were noted. 3. The increase of primary lysosomes and autophagic vacuoles was obserbed. 4. The dilatation of mitochondrial cristae was obserbed. And it was irregulary scattered in the stroma of mitochondria. 5. The atrophy of microvilli in the bile canaliculi and space of Disse was prominent. 6. Membrane of hepatocytes was damaged and significant hydrophic degeneration was obserbed in the perisinusoidal regions. 7. The damage of Fat-storing cells was more significant than that of hepatocytes.
