Identification of polymorphic loci in the deiodinase 2 gene and their associations with head dimensions in geese

  • Objective: This study was conducted to clone and compare the molecular characteristics of the deiodinase 2 (DIO2) gene between Sichuan White geese and Landes geese, and to analyze the association between polymorphisms of the DIO2 gene and head dimensions in Tianfu meat geese. Methods: The coding sequence of the DIO2 gene was cloned by polymerase chain reaction and vector ligation and aligned by DNAMAN software. A total of 350 Tianfu meat geese were used to genotype the polymorphisms of the DIO2 gene and measure the head dimensions. Association analysis between the polymorphisms of the DIO2 gene and head dimensions was carried out. Results: An 840-bp coding sequence of the DIO2 gene was obtained and comparison analysis identified four polymorphic loci between Sichuan White geese and Landes geese. Further analysis showed that the dominant alleles for the four polymorphic loci were G, G, A, and T and the frequency of the heterozygous genotype was higher than that of the homozygous genotype in Tianfu meat geese. Compared to that in the population of non-knob geese of Tianfu meat geese, the head dimensions in the population of knob geese were significantly higher except for nostril height. However, in the non-knob geese, beak width 1, beak width 2, nostril length, cranial width 1, and maxillary length had significant differences among different genotypes or haplotypes/diplotypes. Conclusion: These results suggested that polymorphisms of the DIO2 gene could be considered molecular markers to select larger heads of geese in the population of non-knob geese.

  • Objective: Pigs, an ideal biomedical model for human diseases, suffer from about 50% early embryonic and fetal death, a major cause of fertility loss worldwide. However, identifying the causal variant remains a huge challenge. This study aimed to detect single nucleotide polymorphisms (SNPs) and candidate genes for the number of mummified (NM) piglets using the imputed whole-genome sequence (WGS) and validate the potential candidate genes. Methods: The imputed WGS was introduced from genotyping-by-sequencing (GBS) using a multi-breed reference population. We performed genome-wide association studies (GWAS) for NM piglets at birth from a Landrace pig populatiGWAS peak located on SSC11: 0.10 to 7.11 Mbp (Top SNP, SSC11:1,889,658 bp; p = 9.98E-13) was identified in cyclin dependent kinase on. A total of 300 Landrace pigs were genotyped by GBS. The whole-genome variants were imputed, and 4,252,858 SNPs were obtained. Various molecular experiments were conducted to determine how the genes affected NM in pigs. Results: A strong GWAS peak located on SSC11: 0.10 to 7.11 Mbp (Top SNP, SSC11:1,889,658 bp; p = 9.98E-13) was identified in cyclin dependent kinase 8 (CDK8) gene, which plays a crucial role in embryonic retardation and lethality. Based on the molecular experiments, we found that Y-box binding protein 1 (YBX1) was a crucial transcription factor for CDK8, which mediated the effect of CDK8 in the proliferation of porcine ovarian granulosa cells via transforming growth factor beta/small mother against decapentaplegic signaling pathway, and, as a consequence, affected embryo quality, indicating that this pathway may be contributing to mummified fetal in pigs. Conclusion: A powerful imputation-based association study was performed to identify genes associated with NM in pigs. CDK8 was suggested as a functional gene for the proliferation of porcine ovarian granulosa cells, but further studies are required to determine causative mutations and the effect of loci on NM in pigs.

Prepartal Decrease in Plasma Total Cholesterol Concentration in Dairy Cows Developed Subclinical Ketosis

  • A retrospective study was designed to determine whether there were any differences in prepartal blood biochemical parameters between cows developed subclinical ketosis (SCK) and non-SCK cows. Data on blood biochemistry from 33 Holstein parturient cows (15 SCK and 18 non-SCK cows) in one farm for one year, were compared. Plasma beta-hydroxybutyrate, non-esterified fatty acid and glucose concentrations were utilized in the diagnosis of SCK and showed significant differences between SCK and non-SCK cows after calving. Before calving, however, only plasma total cholesterol concentration in SCK cows was significantly lower than that in non-SCK cows. Consequently, it seems that lower plasma total cholesterol concentration before calving is related to the occurrence of SCK immediately after calving.

The Impact of Crossbred Cattle (Red Sindhi×Yellow Local) on Smallholder Households in the Mountainous and Lowland Zones of Quang Ngai, Vietnam

  • This research investigates the use of crossbreed cattle (Red Sindhi${\times}$Yellow Local cattle) at household level in the lowland and mountainous zones in Quang Ngai province, Vietnam. The internal and external inputs and outputs of mixed farming systems were analysed to quantify the productivity and efficiency of the use of the crossbred and yellow local cattle. In the mountainous zone, households with crossbred cattle had a lower crop and farm efficiency rate than households without crossbred cattle, but in terms of crop, livestock and farm productivity they did not differ. In the lowland zone, households with crossbred cattle had a higher crop, livestock and farm productivity and crop efficiency rate than households without crossbred cattle, but did not differ in terms of farm efficiency rate. The lowland zone had higher off-farm income, crop and household productivity, but lower livestock productivity, livestock and farm efficiency rate than the mountainous zone. Households with crossbred cattle had lower off-farm income than households without crossbred cattle. The results suggest that interactions between zone and kind of household occur at the households and show that the yellow local cattle is a better breed in the mountainous zone and more or less comparable with crossbred cattle in the lowland zone. The extrapolation of the use of crossbred cattle should be carefully considered in line with feeding practice and management.

  • This experiment was carried out to breed a very early maturing variety of Italian ryegrass (Lolium multiflorum Lam.) in Grassland and Forage Crops Division, National Institute of Animal Science, RDA, Cheonan from 2009 to 2010. A new variety, 'Green farm' is a diploid variety with green in leaf color and has erect and semi-erect growth habit in late autumn and early spring, respectively. 'Green farm' was on 28 April in heading date as a early-maturing variety. 'Green farm' was also wider by 0.4 mm in flag leaf width, longer by 0.8 cm in flag leaf length and shorter by 5 cm in plant height than those of control variety, 'Florida 80', respectively. 'Green farm' was thicker in stem thickness and stronger in winter hardness than those of the 'Florida 80'. Dry matter (DM) yield (11,790 kg/ha) of 'Green farm' was similar to that of 'Florida 80'. In vitro dry matter digestibility (IVDMD), total digestible nutrient (TDN) and crude protein (CP) of 'Green farm' were 68.7, 63.3 and 10.3% which are 1.9, 1.7 and 0.6% higher than those of 'Florida 80', respectively. Acid detergent fiber (ADF) and neutral detergent fiber (NDF) of 'Green farm' were 32.5 and 54.6% which are 2.2 and 4.3% lower than those of the 'Florida 80', respectively.

  • Considerable controversy exists about the trend of animal traction effects on crop production in dryland farming systems in sub-Saharan Africa (SSA). This problem arises on account of the failure of the few available empirical studies to recognise the important of technological experience of the individual adopting farmers. This study hence addresses this issue by examining the effects of experience in animal traction technology (ATT) on farm size, cropping emphasis, total crop output and farm productivity. It is based on farm management survey data on 42 small holder farm households fro Ghana. Thirty of these households used animal traction technology (ATT) fro crop cultivation and the rest, mainly hand-hoe. The animal traction sub-sample is classified into three groups according to farmers' years of experience with the technology , thus , those with 1-2, 3-10, and more than 10. Evidence from the study shows that the progression of years of experience with ATT leads to inten ification of labour and land use systems, enhancement of degree of motivation to enter into the market economy, increases in total crop output and farm productivity resulting for decreases in cultivated acreages. The implication of the findings is that institutioal and technical support that do accompany the introduction of such technologies should be structured to last for a relatively longer period to accomodate the learning process.

Comparison of miR-106b, miR-191, and miR-30d expression dynamics in milk with regard to its composition in Holstein and Ayrshire cows

  • Objective: Milk composition varies considerably and depends on paratypical, genetic, and epigenetic factors. MiRNAs belong to the class of small non-coding RNAs; they are one of the key tools of epigenetic control because of their ability to regulate gene expression at the post-transcriptional level. We compared the relative expression levels of miR-106b, miR-191, and miR-30d in milk to demonstrate the relationship between the content of these miRNAs with protein and fat components of milk in Holstein and Ayrshire cattle. Methods: Milk fat, protein, and casein contents were determined in the obtained samples, as well as the content of the main fatty acids (g/100 g milk), including: saturated acids, such as myristic (C14:0), palmitic (C16:0), and stearic (C18:0) acids; monounsaturated acids, including oleic (C18:1) acid; as well as long-, medium- and short-chain, polyunsaturated, and trans fatty acids. Real-time stem-loop one-tube reverse transcription polymerase chain reaction with TaqMan probes was used to measure the miRNA expression levels. Results: The miRNA expression levels in milk samples were found to be decreased in the first two months in Holstein breed, and in the first four months in Ayrshire breed. Correlation analysis did not reveal any dependence between changes in the expression level of miRNA and milk fat content, but showed a multidirectional relationship with individual milk fatty acids. Positive associations between the expression levels of miR-106b and miR-30d and protein and casein content were found in the Ayrshire breed. Receiver operating characteristic curve analysis showed that miR-106b and miR-30d expression levels can cause changes in fatty acid and protein composition of milk in Ayrshire cows, whereas miR-106b expression level determines the fatty acid composition in Holsteins. Conclusion: The data obtained in this study showed that miR-106b, miR-191, and miR-30d expression levels in milk samples have peculiarities associated with breed affiliation and the lactation period.

Application of tenotomy on Korean native cattle (Hanwoo) with spastic paresis symptoms in the field

  • Bovine spastic paresis (BSP) is a neuromuscular disorder characterized by hypertension and stiffness of hindlimb. Two Korean native cattle (Hanwoo) calves developed BSP or BSP-like symptoms, and a tenotomy of superficial tendon of medial head and deep tendon of lateral head of gastrocnemius muscle was performed for treatment. A cast was applied postoperatively to prevent muscle rupture and was removed three weeks later. The prognosis was evaluated at 3 weeks, 6 and 18 months postoperatively. Neither calf showed any other postoperative sequelae. This is the first case study to report the diagnosis, treatment, and prognosis of BSP in Hanwoo.

Overcoming Ethical Conflicts and Dilemmas in Farm Animal Welfare: Investigation of Correlation between Ethical Awareness Level and Compliance with Animal Welfare-Related Regulations in Korean Layer Farms (축산농장 동물복지의 윤리적 갈등과 딜레마 극복: 국내 산란계 농장에서의 윤리의식 수준에 따른 동물복지 관련법규 준수여부 상관관계 조사)

  • Animal welfare was introduced relatively late to Korea in comparison with Western countries. Nonetheless, the Korean government has continuously improved animal welfare-friendly regulations as policy instruments. Given the current situation, it is predicted that spontaneous settlement of the animal welfare policies will be difficult and may cause conflict in the farm animal industry. To identify and categorize conflicts caused by animal-welfare-related policies in the last five years, we investigated the awareness of animal welfare among Korean hen farms and the level of compliance with the animal welfare regulations. We collected a sample of 53 egg-laying chicken farm operators (e.g., owners or head managers) was collected through the on-site survey (90% confidence level (Z-score: 1.65) and 10.18% tolerance, based on a number of 797 egg-laying farms in 2020). Ethical conflicts on the farms were categorized into three different types according to the hen farm's ethical awareness level: passive, moderate, and active. Additionally, we investigated the correlation between compliance with regulations and ethical consideration. This study confirmed that compliance with animal welfare-related regulations significantly correlated to the level of ethical consideration of farm operators. Interestingly, we also observed that farm operators did not comply with the regulation despite their high level of awareness of animal welfare. This conflict implies contradiction and unresolved ethical dilemmas. Therefore, this study argues that the policies cause conflict in the field despite the certain level of effectiveness on animal welfare regulations.
