  • To develop new biologically active compounds produced by anaerobes, 677 soil samples were collected and used to isolate 1, 889 anaerobic bacteria. Among the isolates 427 strains were strict anaerobes and the remaining 1, 462 strains were facultative anaerobes. From 427 strictly anaerobic isolates, 88 strains showed antibacterial activities, and 21 strains were selected for the further studies.

  • The purpose of this presentation is to investigate whether the certain therapy resistant bacteria can impair the immune defense system in the pariapical tissue. Recent studies have reported that the facultative or obligatory anaerobic bacteria such as Fusobacterium nucleatum, Enterococcus faecalis and Actinomyces species and Gram positive facultative bacteria Enterococcus faecalis have been shown to dominate in persistent periapical lesion and usually recovered from failed root canal treated cases. Moreover, E. faecalis has been reported to withstand the antimicrobial agent and endure potential starvation and resist the antibacterial effect of calcium hydroxide intracanal medication.(omitted)

  • There are many of anaerobic culture methods and equipments for isolation and cultivation of anaerabic bacteria, but most of these methods are used without pre-reduced media. Gas-jet method is a recommend. able method for the culture of anaerobes, resently developed. Bacteriological studies were experimented of liver abscess of cattle by the use of gas. jet method. The results were summarised as follows; 1. Gas-jet method for anaerobic culture are expedient for the making of pre-reduced media, maintaining of oxygen free condition in the culture tube, picking of bacteria from colony and colony counting etc. 2. A 121 strains of facultative anaerobic and anaerobic bacteria were isolated from liver abscess of 27 head of cattle, and the isolated anaerobic bacteria were as follows. Peptostreptococcus spp. 7 strains Acid aminococcus fermentans 1 Veillonella spp. 1 Bacterioides spp. 6 Bifidobacterium spp. 4 Arachinia propionica 2 Lactobacillus spp. 4 Propionibacterium acnes 1 3. Liver abscess were infected with many of bacteria, about $10^3-10^9$ numbers per gram of abcessed tissue. Almost of abscess were mixed infection of various bacterial species rather than simple species.

A report of 22 unrecorded bacterial species in Korea, isolated from Namhangang

  • As part of a larger study of indigenous prokaryotic species diversity in South Korea, various samples from Namhangang were subjected to analyses. Fresh water, underwater sediment, and moss-inhabiting aerobic and anaerobic bacteria were isolated. 22 of the isolates were identified as unrecorded bacterial species in Korea that had ${\geq}98.7%$ 16S rRNA gene sequence similarity with published species. The aerobic strains isolated were Kurthia gibsonii and Massilia plicata. Also identified were four facultative anaerobic strains: Bacillus hisashii, Enterococcus rotai, Paenibacillus vini, and Pediococcus pentosaceus. 16 strictly anaerobic strains were identified as Bacteroides xylanolyticus, Carnobacterium maltaromaticum, Clostridium argentinense, Clostridium beijerinckii, Clostridium butyricum, Clostridium cavendishii, Clostridium diolis, Clostridium frigidicarnis, Clostridium perfringens, Clostridium saccharoperbutylacetonicum, Clostridium sphenoides, Clostridium subterminale, Cutibacterium acnes, Paraclostridium bifermentans, Prevotella paludivivens, and Romboutsia lituseburensis. Based on the examination of morphological, cultural, physiological, and biochemical properties of the isolates, descriptive information of these previously unrecorded species is provided here.

Isolation and Identification of an Anaerobic Dissimilatory Fe(III)-Reducing Bacterium, Shewanella putrefaciens IR-1

  • In order to isolate a Fe(III)-reducer from the natural environment, soil samples were collected from various patty fields and enriched with ferric citrate as a source of Fe(III) under anaerobic condition. Since the enrichment culture was serially performed, the Fe(III)-reduction activity was serially diluted and cultivated on an agar plate containing lactate and ferric citrate in an anaerobic glove box. A Gram negative, motile, rod-shaped and facultative anaerobic Fe(III)-reducer was isolated based on its highest Fe(III)-reduction activity, Bacterial growth was coupled with oxidation of lactate to Fe(III)-reduction, but the isolate fermented pyruvate without Fe(III), The isolate reduced an insoluble ferric iron (FeOOH) as well as a soluble ferric iron (ferric citrate). Using the BBL crystal enteric/non-fermentor identification kit and 16S rDNA sequence analysis, the isolate was identified as Shewanella putrefaciens IR-1.

Oxidative stress on anaerobes

  • A strict anaerobe, Prevotella melaninogenica is highly sensitive to oxidative stress. Oxidative stress such as exposure to oxygen or addition of hydrogen peroxide, increased 8-hydroxydeoxyguanosine (80HdG), a typical of oxidative DNA damage, and decreased the bacterial cell survival rate. We could detect the generation of reactive oxygen species in P. melaninogenica after exposure to oxygen. UVA irradiation also increased 80HdG in the bacterium. On the other hand, such oxidative stress did not increase 80HdG in a facultative anaerobe. These findings suggest that P. melaninogenica is a suitable material to study the biological effects of oxidative stress, to evaluate antioxidants, and to study the effects of oxygen or reactive oxygen species on molecular evolution.

  • Mycoplasma pneumoniae was examined for growth characteristics and morphology when cultivated in several media supplemented with a variety of sera and under different atmospheric conditions. different formula of the medium as well as different sources of lot numbers of the serum in the same medium exhibited varying effects on growth rate and adherence. When the organisms were cultivated in SSR-2 medium in a normal atmospheric environment or under a facultative anaerobic condition provided with carbon dioxide, they developed filamentous cells with heavy growth, whereas mainly round-shaped cells were produced under strict anaerobic conditions of hydrogen and carbon dioxide. Both morphologies of the organism were transformed by switching the incubation environments. An inverted phasecontrast microscopy using modified petri dishes was excellent to observe single cells and useful to follow the development of the cells. Growth, turbidity, and pneumoniae colonies developed on a solid medium and produced clear when overlaid with sheep blood agar.

Characterizations of Denitrifying Polyphosphate-accumulating Bacterium Paracoccus sp. Strain YKP-9

  • A denitrifying polyphosphate-accumulating bacterium (YKP-9) was isolated from activated sludge of a 5-stage biological nutrient removal process with step feed system. This organism was a Gram-negative, coccus-shaped, facultative aerobic chemoorganotroph. It had a respiratory type of metabolism with oxygen, nitrate, and nitrite as terminal electron acceptors. The 16S rRNA gene sequence of strain YKP-9 was most similar to the 16S rRNA gene sequence of Paracoccus sp. OL18 (AY312056) (similarity level, 97%). Denitrifying polyphosphate accumulation by strain YKP-9 was examined under anaerobic-anoxic and anaerobic-oxic batch conditions. It was able to use external carbon sources for polyhydroxyalkanoates(PHA) synthesis and to release phosphate under anaerobic condition. It accumulated polyphosphate and grew a little on energy provided by external carbon sources under anoxic condition, but did neither accumulate polyphosphate nor grow in the absence of external carbon sources under anoxic condition. Cells with intracellular PHA cannot accumulate polyphosphate in the absence of external carbon sources under anoxic condition. Under oxic condition, it grew but could not accumulate polyphosphate with external carbon sources. Based on the results from this study, strain YKP-9 is a new-type denitrifying polyphosphate-accumulating bacterium that accumulates polyphosphate only under anoxic condition, with nitrate and nitrite as the electron acceptors in the presence of external carbon sources.

Effect on the Concentration of Glucose and Sucrose on the Hydrogen Production using by the Facultative Anaerobic Hydrogen Producing Bacterium Rhodopseudomonas sp. MeL 6-2 (통성혐기성 수소생산균주 Rhodopseudomonas sp. MeL 6-2를 이용한 수소생산효율에 미치는 포도당 및 자당 농도의 영향)

  • Hydrogen producing bacterium, strain MeL 6-2 was isolated from the sludge of the factory areas in Anyang through the acclimation in basal salt medium (BSM) supplemented with 10 g/L of sucrose. Isolated strain MeL 6-2 was a facultative anaerobe which could grow in both aerobic and anaerobic environments. An aerobically grown pure culture isolated from enriched culture was analyzed by 16S rDNA sequencing and identified as Rhodopseudomonas sp. MeL 6-2. Effects of the concentrations of glucose and sucrose on the hydrogen production rate and the hydrogen production yield were investigated. When glucose in the range of 1~12 g/L was supplemented to the BSM, strain MeL 6-2 could grow without lag phase. An increased glucose concentration increased the specific hydrogen production rate linearly to $4.2\;mmol-H_2{\cdot}L^{-1}{\cdot}h^{-1}$ at 10 g/L, and $60\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$, but decreased slightly as the concentration increased to 12 g/L. The hydrogen production yield was maintained over a range from 2.6 to $3.1\;mol-H_2{\cdot}mol-glucose^{-1}$. When sucrose in the range of 1~12 g/L was supplemented to the BSM, strain MeL 6-2 could grow after ten hours. An increased sucrose concentration increased the specific hydrogen production rate and the hydrogen production yield to $163\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$ and to $4.5\;mol-H_2{\cdot}mol-sucrose^{-1}$, respectively.

Studies on the Preparation of Fermented Milk by Bifidobacterium longum and Lactobacillus acidophilus (Bifidobacterium longum 및 Lactobacillus acidophilus를 이용한 발효유 제조)

  • Yoghurt was prepared with Bifidobacterium longum TK-100 and Lactobacillus acidophilus TK-2070. The prepared yoghurt showed the increase of the titratable acidity under cold storage condition. This was derived from the active L. acidophilus TK-2070 on the logarithmic phase rather than from the B. longumn TK-2070. B. longum TK-100 grew well in the facultative anaerobic condition as well as in the strict anaerobic condition. Reinforced clostridial agar medium with 0.1% aniline blue was tried for the differential viable cell counts in the mixed culture and in the yoghurt. B. longurn TK-2070 had the light gray, blue-dotted colonies of about 2 mm diameter. L. acidophilus TK-2070 had the light gray colonies of about 1 mm diameter.
