A Study on the Transmission of a Transgene in the Offspring of Transgenic Mice (형질전환 생쥐의 후손에서 외래 유전자의 유전성에 대한 연구)

  • It is known that the incorporation of genes into transgenic mice is generally stable and is p passed on to succeeding generations in a Mendelian fashion. In this report, transgenic mice were set as a model to evaluate whether the transgenes are transmitted in a Mendelian principle in a successive generations and how they are tran s smitted into their offspring. A 3.0 kb linear DNA fragment, containing the MMTV LTR, bovine aSI casein cDNA and SV 40 splicing and polyadenylation site; was microinjected into fertilized mouse embryos. The tail DNAs of the resulting pups were subjected to dot and Southern hybridizations to screen transgenic founders. The DNAs of their offspring were anlyzed by PCR to confirm the transmission of the transgene from F0. Out of 72 live pups four pups (5.6%), 3 males and 1 female, were positive for the transgene. The rates of transmission from F0 into F1 were 33.3, 7.7, 0, and 62.5%. Those from F1 into F2 were 63.6, 5.9, and 68.8% and those from F2 into F3 were 85.7, and 88.2%. In this report, the transmission pattern of transgenes in transgenic mice into their offspring was demonstrated. It either follows or does not follow in a Mendelian fashion. Deletion or loss of the transgenes from F0 in some lines became apparant to the succeeding generations.

  • This study was conducted to estimate the heritabilities of height and stem diameter growth in 5- and 6-year-old Pinus koraiensis. Estimates of individual tree and family heritability for height growth were $h^2_I=0.40$, $h^2_F=0.77$ in 5-year-old and $h^2_I=0.57$, $h^2_F=0.89$ in 6-year-old, respectively. Heritabilities for diameter growth were estimated as $h^2_I=0.16$, $h^2_F=0.53$ and $h^2_I=0.26$, $h^2_F=072$ in 5-and 6-year-old seedlings.

  • In this study, a method of error compensation for channel gain imbalance, phase imbalance and local oscillator leakage in the modulator of frequency direct conversion is suggested. The compensation of channel imbalance can be carried out by using the received power after transmitting test signal. By applying this method, the phase imbalance conversion with frequency can be easily compensated since this method is rarely affected by the transmission channel. It is confirmed that the algorithm proposed in this study(iteration coefficient=11) converges faster than conventional algorithm(iteration coefficient=43). From the numerical results, the DC-offset, channel gain, phase imbalance compensation coefficient and iteration number converges into($f_1$=0.0199999, $f_2$=-0.050001, $C_{22}$=0.9133, $C_{12}$=-0.0524, N=13) when the local oscillator leakage is not considered. However, it converges into($f_1$=-0.02, $f_2$=-2.2476, $C_{22}$=0.9133, $C_{12}$=-0.0524, N=16) when the local oscillator leakage is considered.

The Yield and Wood Quality of 1-year-old Hybrid Poplars : Populas alba X P. glandulosa $F_1$ and Populus nigra var. italica X P. maximowiczii $F_1$ Clones (1년생 현사시 및 양황철의 물질생산 및 재질특성)

  • In order to investigate biomass yield of one-year-old hyblrid poplars, Pripulusalba x P. glandulosa $F_1$ and Populus nigra var. italica x P. maximowiczii $F_1$clones, as energy and fiber resources, dry matter yield, leaf area, leaf area index, dry matter production ability, specific gravity and fiber length and width were measured. Dry matter yield was 1.89 ton/ha for Poplus alba x P. glandulosa $F_1$ and 3.63 ton/ha for Populus nigra via.italica x P. merximowiczii $F_1$ clones in the planting density of 20,000 trees/ha and in the planting density of 40,000 trees/ha was 3.87 ton/ha for Populus alba x P. glandulosa $F_1$and 5.64 ton/ha for Populus nigra var. italica x P. maximowiczii $F_1$ clones. Leaf area index was 1.24mtim2 in the planting density of 20,000 trees/ha and 2.45 m31m3 in the density of 40,000 trees/ha for Populus alba x P. glandulosa $F_1$ clones and it was 1.96 m21m2 in the planting density of 20,000 trees/ha and 3.36 m21m2 in the density of 40,000 trees/ha for the hybrid $F_1$ Populus nigra var. italica x P. maximowiczii clones. The average specific gravity of the hybrid poplars was 0..36 when bark and pith were included and 0.31 when bark and pith were removed in the plot of 20,000 trees/ha and in the 40,000 trees/ha plot showed 0.35 and 0.31 respectively, for Populus alba x P. glandulosa $F_1$clones. It was 0.36 when bark and pith were included and 0.32 when bark and pith were removed in the 20,000 trees/ha plot and in the 40,000 trees/ha plot was 0.34 and 0.31 respectively for Populus nigra var. italica x P. maximowiczii $F_1$clones. The average fiber length was 0.57 mm in the 20,000 trees/ha plot and 0.58 mm in the 40,000 trees/ha plot for Poplus alba x P. glandulosa $F_1$clones and was 0.60 in both plots of 20,000 trees/ha and 40,000 trees/ha for Populus nigra var. italica x P. maximowiczii $F_1$ clones. There is a big clonal variation among those clones studied, showing high selection potential in both species.

  • From the previous studies, F-P-4 formula was found to be comparable to full fat dry milk in its nutritive value and feeding performance. However, an attempt was made in order to make sure whether or not any possibility might exist, by which further improvement of nutritive quality and simultaneous reduction of product costs may be achieved. Using F-P-4 as a control, modifications were made in new formulas, F-P-5, F-P-6 and F-P-7 by reducing FPC, eliminating yeast from the mixture, and by enriching with methionine as needed. In particular, F-P-7 is completely free of FPC, hydrogenated oil and yeast. Yet, levels of total protein and fat were kept equal to those of F-P-4 in all formulas. An animal feeding test for all formulas using 10 female rats per group for 8 weeks and an infant feeding trial for F-P-5 and F-P-6 with 5 of each female infants under age of one for one month were conducted along with F-P-4 as a control. Almost the same results were obtained with F-P-4, 5 and 6, but F-P-7 showed the lowest body weight gain. FER of F-P-5 and 6 was 0.20 as was with F-P-4, while that of F-P-7 was 0.16. Acceptability to infants was excellent; growth, appearance and biochemical data were normal. As an example F-P-4 packed in 0.04mm polyethylene bags was used for storage study at $25^{\circ}C$ and relative humidity of $65{\sim}85%$ for 8 months. Although viable bacterial counts and vitamin C contents were reduced, peroxide and TBA values were increased gradually during such storage. Since there are also significant changes in color and organoleptic quality, the expected shelf life under the given conditions is considered to be about 2 months and thus further works are needed both on the product and packaging in order to improve the storage stability. Either elimination of yeast form F-P-4, that is F-P-5, or partial replacement of FPC with methionine, that is F-P-6 may well reduce material costs about 10%. Considering blending process of ingredients, F-P-5 is thus found to be the best formula developed. While F-P-7 free of FPC is inferior in its nutritive quality than that of others, but significantly superior than of rice. Furthermore, the material cost of the product can be reduced about 20% from that of F-P-4. And thus this vegetable blend is considered to be useful as a low cost supplementary food mixture for growing children.

Quantitative Evaluation of Liver Fibrosis on T1 Relaxometry in Comparison with Fibroscan (Fibroscan과 비교를 통한 T1 MR Relaxometry를 이용한 간섬유화의 정량적 평가)

  • Purpose This study was performed to determine whether the T1 relaxation time of gadoxetic acid-enhanced liver MR imaging is useful for detecting and staging liver fibrosis in patients with chronic liver disease. Materials and Methods One hundred and three patients with suspected focal liver lesion underwent MR imaging and Fibroscan. Fibroscan was chosen as the reference standard for classifying liver fibrosis. T1 relaxation times were acquired before (preT1), 20 minutes after (postT1) contrast administration, and reduction rate of T1 relaxation time (rrT1) on transverse 3D VIBE (volumetric interpolated breath-hold examination) sequence using 3T MR imaging. The optimal cut-off values for the fibrosis staging were determined with ROC analysis. Results PreT1 and postT1 increased and rrT1 decreased constantly with increasing severity of liver fibrosis according to the METAVIR score (F0-F4). There were statistically significant differences between F2 and F3 in preT1 (F2, 836.0 ± 74.7 ms; F3, 888.6 ± 77.5 ms, p < 0.05) and between F3 and F4 in postT1 (F3, 309.0 ± 80.2 ms; F4, 406.6 ± 147.7 ms, p < 0.05) and rrT1 (F3, 65.4 ± 7.7%; F4, 57.3 ± 11.4%, p < 0.05). ROC analysis revealed that combination test (preT1 + postT1) was the best test for predicting liver fibrosis. Conclusion PreT1 and postT1 increased constantly with increasing severity of liver fibrosis. T1 mapping in gadoxetic acid-enhanced liver MR imaging could be a helpful complementary sequence to determine the liver fibrosis stage.

Consonantal and Vocalic Effects in Korean Stop Identification

  • This study investigates the contribution of vocalic information following the release of an initial stop to the identification of the three-way stop contrast (aspirated, lax, and tense) in Korean. Recent studies showed that there is a strong interaction between consonant types and tone. The findings raise questions concerning Korean listeners' use of tonal (or vocalic F0) variation in differentiation initial tense, lax, and aspirated stops. The above issues are addressed in the present study using a cross-splicing methodology. The overall results show that low vocalic F0 provided the most salient information for lax stops; tense and aspirated stop identification depended on a combination of VOT, F0, and H1-H2 characteristics. The perceptual dominance of F0 over VOT for lax stops is consistent with the size of the F0 difference in utterance-initial position, as well as their prominent role in Korean intonational phonology.
