생쥐배아의 부화와 탈각에 미치는 Pronase의 영향 (Effect of Pronase Treatment on Mouse Embryos: Improving Hatching and Hatched Rates)

  • Objective: Hatching of the blastocyst from the zona pellucida (ZP) is a key event in mammalian implantation. In vivo, two factors have been identified as possible mediators of hatching: lysis of the ZP by substances elaborated either from the embryo or female reproductive tract and pressure exerted on the zona by expansion of the blastocyst. Two methods of zona manipulation were already in use to enhance the ability of embryos to hatch: mechanical PZD and chemical ZD by acidic Tyrode's solution. But several controversies of each method have been reported. The purpose of this study was to investigate the effect of pronase for mouse embryo hatching. Methods: Mouse embryos were obtained following ovulation induction of $F_1$ animals. Fresh and cryo-thawed morula embryos were exposed to 0.5, 1.0, 2.0, 5.0 ${\mu}g/ml$ pronase in Ham's F10 for 72 hrs. Main outcome measures were the rates of partial hatching and completely hatched blastocysts, and cell number of it. Results: In fresh and cryo-thawed group, the rates of completely hatched blastocyst were significantly higher in 5 ${\mu}g/ml$ pronase treatment group than control group. There was no difference in completely hatched blastocyst total cell number between pronase treatment group and control group. This suggest that pronase treatment did not harmful in mouse embryo development. In pronase treatment group, zona pellucida were thinner than control group. Conclusion: The addition of pronase to culture media may accelerate the hatching of embryo. So, enzymatic treatment of the zona may provide a valuable and effective assisted hatching technique for human in-vitro fertilization-embryo transfer.

수술에 사용하는 생리식염수의 오염수준 변화 (A Study on the Contamination of Saline Used in the Operation)

  • Post-operative wound infections have been the serious problems in nursing care in the operating room and appear to be strongly related to the infection occurring during the operation. The purpose of this study is to identify the level of contamination in saline used in the operation and also examine the correlation between the contaminated saline and the length of the operation, and unclean atmospheric factor. Subjects for this study include 13 cases of operation performed at the operative theatre of a hospital in Seoul area. Test samples and related data were collected from this medical facility between Oct. 6 through Dec. 10, 1994 by the author and anurse who worked in the operating room. For the study, multiple batches of saline sample were collected at the various time intervals duringthe operation and filtered through the membrane filters. Viable microorganisms retained on the filters were cultured on the appropriate culture media and the levels of existing cells in saline were enumerated according to Koch's method. In the analyses of the data, Pearson's correlation coefficient was obtained for the examination of relationship between the length of operation and numbers of microorganisms existing in saline and for the comparison of the differences in numbers if microrganisms in saline sample collected at the various operative stages, e. g. pre-incision, excision and skin suturing stages, ANOVA and Scheff Tests were performed. The results of this study are summarized as follows. 1) The lenth of the operation and numbers of microorganisms in the saline used in the operation appeared to be significantly correlated (r=0.5467, P<0,001). 2) In case of saline exposed to air, but not used in the operation, the length of exposure to the air and the numbers of microorganisms present in saline also showed an apparent correlation(r=0.5087, P<0. 001). 3) The frequencies of occurrence of microorganisms in saline used in the operation and in saline exposed only to the air in the given time showed significant differences(t=3.73, p=.0000). 4) In case of saline used in the operation, there is significant differences in its numbers of contained microorganisms between the operative stages ; pre-incision, excision, and skin suture (F=17.7500, p=.0000). 5) In case of saline exposed only to the air in the given time, there is significant differences in its numbers of contained microorganisms between the operative stages . pre-incision, excision, and skin suture(F=6.3807, p=.00031).

람세균 Synechocystis sp. PCC 6803 PTX의 주광성 운동에 미치는 몇가지 대사 억제제의 효과 (Effects of Some Metabolic Inhibitors on Phototactic Movement in Cyanobacterium Synechosystis sp. PCC 6803 PTX)

  • 최근에 Synechocystis sp. PCC 6803 중에 한 균주가 고체 한천 배지상에서 일정한 조명(300-1000 lux) 방향을 따라 활주 운동하는 것을 관찰하여 이 종을 S. 6803 PTX라고 명명하고 이의 주광성 운동에 대한 생리학적 특징을 이해하기 위하여 몇 가지 대사 억제제와 신호 전달 차단제의 주광성 운동에 미치는 효과를 조사하였다. DCMU는 광계 II로부터 광계 I의 일차 전자 수용체인 플라스토퀴논으로의 비순환성 광합성 전자전달을 억제하는 억제자로서 $100\;\mu\textrm{M}$의 농도에서는 주광성 운동을 억제하지 못하였다. 그러나 호흡에 의한 전자전달 억제제인 sodium azide를 처리하였을 경우에는 S. 6803 PTX에서 심하게 장해를 받았다. 이러한 관찰 결과는 주광성 운동의 주동력원이 광인산화 과정보다는 호흡에 의한 산화적인 인산화과정에 주로 연관되어 있음을 보여주었다. 또한, 세포를 CCCP나 DNP와 같은 막상의 uncoupler를 처리하였을 때, 세포내 ATP 농도를 저하시키거나 세포질막에 수소 이온의 전기화학구배($\Delta\mu_{H}+$)를 제거시키나, 이러한 화합물들은 주광성 운동에 뚜렷한 영향은 주지 못하였다. 이러한 결과와는 달리, H+-F0F1 ATPase에 민감하게 억제 작용을 나타내는 DCCD나 NBD의 처리는 세포내 ATP만 고갈시키고 막상에서 $\Delta\mu_{H}+$는 그대로 유지시키는 작용을 하는데, 이러한 DCCD나 NBD는 주광성 운동에 대해서는 심하게 억제 현상을 나타내었다. 또한, 특이성 calcium ionophore 중의 하나인 A23187의 처리는 양성 주광성에 심하게 장해를 주었다. 아마도 Ca2+ 유동은 주광운동 방향성의 신호전달 과정에 중요하게 관련되어 있는 것으로 나타났다. 마지막으로 S-adenosyl methionine과 같은 메틸 공여체의 고갈이 S. 6803 PTX 균주의 주광성 반응에 영향을 주는지를 알아보기 위하여 에티오닌을 BG11을 한천 배지에 첨가하였다. 이 생물종의 광운동은 에티오닌의 농도가 증가됨에 따라 일정하게 억제되다가 0.5mM에서 주광성 운동을 완전히 억제시켰다. 이것은 광수용 기작이 Escherichia coil나 Salmonella typhimurium에서 발견된 메틸기 수용 주화성 단백질과 같은 메틸화/탈메틸화 과정에 의하여 조절될 가능성을 보여주고 있음을 의미한다.

The Effects of TWEAK, Fn14, and TGF-$\beta1$ on Degeneration of Human Intervertebral Disc

  • Objective: The purpose of this study is to explain the effect and reciprocal action among tumor necrosis factor (TNF) like weak inducer of apoptosis (TWEAK), fibroblast growth factor-inducible 14 (Fn14), and transforming growth factor-$\beta1$ (TGF-$\beta1$) on degeneration of human intervertebral disc (IVD). Methods: Human intervertebral disc tissues and cells were cultured with Dulbecco's Modified Eagle's Medium/Nutrient F-12 Ham (DMEM/F-12) media in $37^{\circ}C$, 5% $CO_2$ incubator. When IVD tissues were cultured with TWEAK, Fn14 that is an antagonistic receptor for TWEAK and TGF-$\beta1$, the level of sulfated glycosaminoglycan (sGAG) was estimated by dimethyl methyleneblue (DMMB) assay and sex determining region Y (SRY)-box 9 (Sox9) and versican messenger ribonucleic acid (mRNA) levels were estimated by reverse transcriptase polymerase chain reaction (RT-PCR). Results: When human IVD tissue was cultured for nine days, the sGAG content was elevated in proportion to culture duration. The sGAG was decreased significantly by TWEAK 100 ng/mL, however, Fn14 500 ng/mL did not change the sGAG production of IVD tissue. The Fn14 increased versican and Sox9 mRNA levels decreased with TWEAK in IVD tissue TGF-$\beta1$ 20 ng/mL elevated the sGAG concentration 40% more than control. The sGAG amount decreased with TWEAK was increased with Fn14 or TGF-$\beta1$ but the result was insignificant statistically. TGF-$\beta1$ increased the Sox9 mRNA expression to 180% compared to control group in IVD tissue. Sox9 and versican mRNA levels decreased by TWEAK were increased with TGF-$\beta1$ in primary cultured IVD cells, however, Fn14 did not show increasing effect on Sox9 and versican. Conclusion: This study suggests that TWEAK would act a role in intervertebral disc degeneration through decreasing sGAG and the mRNA level of versican and Sox9.

팽이 신품종 '백이' 육성 (Breeding of a new cultivar of Flammulina velutipes: 'Baeke')

  • 팽이의 신품종을 개발하기 위하여 모균주 ASI 4216(한솔)과 4217(백중)으로부터 단포자를 분리하여 단핵균주를 얻고 이들의 교잡을 통해서 꺽쇠연결체가 확인된 교잡주총 129점을 선발하였다. 교잡주를 대상으로 3회에 걸쳐 재배시험 결과, 고온에서 배양이 가능하며 수량성이 우수한 계통 'Fv14a51'을 최종선발되었으며, 생산력검정 시험을 통해 2017년 농작물 직무육성 품종 심의회에서 '백이'로 명명되었다. 주요 특성은 균사생장이 $30^{\circ}C$의 고온에서도 균활력을 유지할 정도로 강한 온도적응성 품종이고 대조품종에 비해 갓이 두껍고 대가 얇는 형태를 띠는 것 외에는 갓색깔이 순백색이며, 갓모영은 반구형, 수량은 $257.4{\pm}13.5g$으로 대조품종과 비교해서 대체적으로 유사하였다. RAPD primer을 통해 유전적 다형성을 검정한 결과, 대조구인 '오키노메구미', 2016년에 육성된 '백승'과 신품종이 유전적으로 구별되는 것을 확인하였다. 전반적으로 '백이'는 수량과 품질이 외국품종과 거의 유사하였으며, 버섯의 균일한 형태로 상업적 가치가 있었다. 신품종의 현장실증을 확대하여 국산품종이 국내에 확산되기를 기대한다.

과불화화합물 구조적 속성에 따른 흡착 특성 연구 (Study on Adsorption Characteristics of Perfluorinated Compounds(PFCs) with Structural Properties)

  • Perfluorinated compounds(PFCs), an emerging environmental pollutant, are environmentally persistent and bioaccumulative organic compounds that possess a toxic impact on human health and ecosystems. PFCs are distributed widely in environment media including groundwater, surface water, soil and sediment. PFCs in contaminated solid can potentially leach into groundwater. Therefore, understanding PFCs partitioning between the aqueous phase and solid phase is important for the determination of their fate and transport in the environment. In this study, the sorption equilibrium batch and kinetic experiment of PFCs were carried out to estimated the sorption coefficient(Kd) and the fraction between aqueous-solid phase partition, respectively. Sorption branches of the PFDA(Perfluoro-n-decanoic acid), PFNA(Perfluoro-n-nonanoic acid), PFOA(Perfluoro-n-octanoic acid), PFOS(Perfluoro-1-octane sulfonic acid) and PFHxS(Perfluoro-1-hexane sulfonic acid) isotherms were nearly linear, and the estimated Kd was as follow: PFDA(1.50) > PFOS(1.49) > PFNA(0.81) > PFHxS(0.45) > PFOA(0.39). The sorption kinetics of PFDA, PFNA, PFOA, PFOS and PFHxS onto soil were described by a biexponential adsorption model, suggesting that a fast transport into the surface layer of soil, followed by two-step diffusion transport into the internal water and/or organic matter of soil. Shorter times(<20hr) were required to achieve equilibrium and fraction for adsorption on solid(F1, F2) increased with perfluorinated carbon chain length and sulfonate compounds in this study. Overall, our results suggested that not only the perfluorocarbon chain length, but also the terminal functional groups are important contributors to electrostatic and hydrophobic interactions between PFCs and soils, and organic matter in soils significantly affects adsorption maximum capacity than kinetic rate.

치위생(학)과 학생의 감성지능과 문화 역량과의 관련성 (The relationship between emotional intelligence and cultural competency of dental hygiene students)

  • Objectives: The study was a cross-sectional research to examine dental hygiene students' multi-cultural experiences, emotional intelligence and cultural competency and to understand the correlations among them. Methods: The study was conducted from September $1^{st}$ to October $31^{st}$ 2016, based on the survey of 449 students in the department of dental hygiene at 7 Universities. The questionnaire consisted of 57 questions including general characteristics (n=7), multi-cultural experiences (n=7), emotional intelligence (n=16) and cultural competency (n=27). Results: Each score of students' emotional intelligence and cultural competency is 3.43 and 3.01 respectively in 5-point scale. An analysis of correlations between emotional intelligence and cultural competency shows that the higher the emotional intelligence, the higher the cultural competency (r=0.342). The factors affecting the cultural competency include use of emotions (${\beta}=0.327$, p<0.001), control of emotions (${\beta}=0.254$, p=0.001), frequency of multi-cultural media (${\beta}=0.221$, p<0.001) and experience of multi-cultural class (${\beta}=0.221$, p=0.002). The modified explanatory power in this model is 28.2% (F=10.856, p<0.001). Conclusions: There was a positive correlation between emotional intelligence and cultural competency, and the contacts with multi-culture and experience of class are identified as the affecting factors. Dental hygiene students should acquire theoretical experiences regarding the multi-culture through curriculum or continuous educations and it is necessary to promote such educations in order to develop and apply the programs for the enhancement of emotional intelligence.

Molecular characterization of yeast Snf1 homologue (sucrose non-fermenting gene) from Magnaporthe grisea

  • Magnaporthee grisea causes the devastating blast disease of rice. Entensive research has been conducted on infection mechanisms, particularly on appressorium formation and penetration, of this fungus during the last decade. However, the role(s) of cell-wall-degrading enzymes (CWDEs) on pathogenesis is not clearly demonstrated at molecular level. Many CWDES in plant pathogenic fungi including M. grisea are redundant; that is, there are multiple genes encoding enzymes with a similar or overlapping spectrum of activities. It is laborious to isolate all of the genes encoding related enzymes and to construct mutants lacking all 9f them. Thus, we considered alternative strategies to address the role of CWDEs in pathogenesis. Since expression of CWDE genes Is repressed by a simple sugar, as the first step, we cloned a Snfl (sucrose non-fermenting) gene (MgSnf1) from M. grisea. The predicted amino acid sequence showed a high identity with other Snf1 genes from various fungi. To elucidate molecular function of MgSnf1, a transformant lacking MgSnf1 was created by targeted gene replacement. En glucose, sucrose, and xylan the MgSnf1 mutant grew normally but in pectin and complex media, it grew slower than wild type. Expression of various CWDEs in MgSnf1 mutant was investigated and found that expression of some CWDEs is repressed. However, no significant difference was observed in conidial germination, appressorium formation, and pathogenicity in MgSnf1 mutant. However, MgSnf1 functionally complemented a yeast MgSnf1 mutant. These results suggest that MgSnf1 is involved in regulation of CWDEs and MgSnf1 is dispensable in pathogenicity of M. grisea.

삼출성 중이염 환자의 경과 관찰 시(時) tympanometry 사용의 유용성 (Usability of Tympanometry in Oriental Medical Treatment of OME)

  • Background: Otitis media with effusion(OME) is one of the most common disease in childhood. Patient suffers from OME in many quarters. For example hearing loss, effects on speech, language, learning, behaviouralchanges etc. But the diagnosis of OME is difficult using only medical history and otoscopy. So, many other diagnostic tools are developed and tympanometry is one of them. When OME is treated by oriental medical Tx, tympanometry would be useful instrument. Objectives: To gain insight into the usability of tympanometry in F/U of OME treated by oriental medial Tx. Methods: Data was collected from 123 ears of 74 children who was treated by oriental medical Tx. Data includes sex, age, period of Tx, elapsed time to be improved, tympanograms and accompanied symptoms. The relationship between items was analysed by statistical methods. Results: There is no relationship between age, sex and period of Tx/elapsed time to be improved. When cough and phlegm is accompanied, the period of treatment and elapsed time to be improved are expanded. When improvement was seen on tympanogram, the period of treatment is expanded but the improvement was seen within 30days, the period of treatment is reduced. Conclusion: The application of tympanometry in oriental medical treatment of OME would provide us many informations about the status of patient so it will be helpful to predict the history of a case and to make a decision whether keep up treatment or not.

문화 코드의 변화에 따른 패션 트렌드 경향 연구 - 2005년 패션 트랜드를 중심으로 - (A Study on the Fashion Trend according to the Changes of Cultural Code - Focusing on 2005 Fashion Trends -)

  • Modern society is a multi-cultural consumer society, and there are multiple trends to cater to the tastes of diverse consumers with different sociocultural background. To grasp fashion trends in fast-changing society, how consumer life is changing and what sort of trend is prevailing should be understood above all. A major fashion trend keeps on changing in every season, and that is an extensive and compound measure of what affects the lives and values of cultural receivers who take the lead in it. The purpose of this study was to delve into what sorts of trends were presented in the 21st century's different cultures, how those cultures were reflected in fashion trends, and how design elements predicted by fashion trends could serve as the sources of design that could create a new fashion. The findings of the study were as follows: First of all, the theories of popular culture and trends were reviewed to describe how general receivers found meaning and delight in the products of cultural industry in their own way and how the products were converted into diverse cultural media. Secondly, consumer styles were discussed by classifying consumers into six groups, twixter, duppie, Ubi-Nomad, NONOS, LOHAS and chav, who were generated by changing cultural codes. Thirdly, sociocultural trends and consumer changes brought a lot of diverse changes to fashion trends. The visual materials about the 2005 S/S, F/W Collection were examined to track how changing trends affected fashion style.
