Improvement of RT-PCR Sensitivity for Fruit Tree Viruses by Small-scale dsRNA Extraction and Sodium Sulfite

  • Woody plant tissues contain great amounts of phenolic compounds and polysaccharides. These substances inhibit the activation of reverse transcriptase and/or Taq polymerase in RT-PCR. The commonly used multiple-step protocols using several additives to diminish polyphenolic compounds during nucleic acid extraction are time consuming and laborious. In this study, sodium sulfite was evaluated as an additive for nucleic acid extraction from woody plants and the efficiency of RT-PCR assay of commercial nucleic acid extraction kits and small-scale dsRNA extraction was compared. Sodium sulfite was used as an inhibitor against polyphenolic oxidases and its effects were compared in RNA extraction by commercial extraction kit and small-scale double-stranded RNA (dsRNA) extraction method for RT-PCR. During nucleic acid extraction, addition of 0.5%-1.5%(w/v) of sodium sulfite to lysis buffer or STE buffer resulted in lighter browning by oxidation than extracts without sodium sulfite and improved the RT-PCR detection. When commercial RNA extraction kit was used, optimal concentrations of sodium sulfite were variable according to the tested plant. However, with dsRNA as RT-PCR template, sodium sulfite 1.5% in STE buffer improved the detection efficiency of Apple chlorotic leaf spot virus (ACLSV) and Apple stem grooving virus (ASGV) in fruit trees, and reduced the unspecific amplifications signi-ficantly. Furthermore, when viruses existed at low titers in host plant, small-scale dsRNA extractions were very reliable.

  • In order to reconstruct the traditional technology of Korean golden varnish coatings, this study was carried out to separate and determine some main coloring components of the exudates of D. morbifera and its traditionally refined golden varnishes using a process of solvent extractions, chromatographies and spectrometries. The results obtained are as follows: 1. The exudate and its traditional-refined golden varnishes appear to have a kind of natural polyacetylenes because it has some triple bond peaks in FT-IR spectrometry. 2. Some yellowing spots of the polar-solvent extrats from the exudates and refined varnishes separated on TLC appeared under natural drying condition, but those of non-polar solvent extract such as hexane did not. 3. A traditional refining method for reconstructing a Korea golden varnishes was thought to be better than solvent separation because the former had higher triple-bond peaks than the latter in FT-IR spectrometry. 4. One of main conponents in the hexane-extracts of the traditional-refined varnishes and the exudates had the same molcular weighr of 204, but the fragmentation patterns was a little different between the exudate and the refined. in LC-MS soectrometry.

  • The study area is located in the southern part of the M'aider Paleozoic basin in the Moroccan Eastern Anti-Atlas. It is an arid region, characterized by minimal vegetation cover, which can provide an ideal environment to apply remote sensing. In this study, remote sensing and field investigations were integrated for lithological and structural lineaments mapping. The Landsat 8 OLI data were processed in order to understand the role of lithology and geological structures in the distribution of mineral deposits in the study area. To achieve this purpose, the Color Composite (CC), the Principal Component Analysis (PCA) and Band Rationing transformation (BR) tests were performed. The results of remote sensing techniques coupled with field investigations have shown that the zones of high lineaments densities are highly correlated with the occurrences of barite mineralization. These findings depict a spatial relationship between structural lineaments and the mineralization distribution zones. Therefore, the barite and Iron oxides mineralization veins, which occur mainly in the Ras Kammouna district, seem to have a structural control. The methodological approach used in this study examining lithological mapping and lineament extractions can be used to explore mineral deposits in arid regions to a high degree of efficiency.

Development of the Building Boundary Detection for Building DEM Generation (건물 DEM 생성을 위한 경계검출법 개발)

  • It is estimated that, in the twenty-first century, 70% of global citizens will live in urban areas. This accelerated urbanization will require a greater need for the building DEM and orthoimagery along with Geographic Information System for urban management. The building DEM requires the detection of outlines showing building shapes. To do this, automatic and semiautomatic building extractions are usually used. However, in cases where automatic extraction is performed directly from the aerial images, accurate building outline extraction is very difficult because of shadow, roof color, and neighboring trees making it hard to discern building roofs. To overcome this problem semiautomatic building extraction was suggested in this paper. When a roof texture was homogeneous, building outline detection was performed by mouse-clicking on a part of the roof. To construct the building outlines when the texture was not homogeneous, a computer program was developed to search out corner points by clicking spots near corner points. The building DEM was generated by taking into account building outlines and heights calculated by image matching.

Image Retrieval Method Based on IPDSH and SRIP

  • At present, the Content-Based Image Retrieval (CBIR) system has become a hot research topic in the computer vision field. In the CBIR system, the accurate extractions of low-level features can reduce the gaps between high-level semantics and improve retrieval precision. This paper puts forward a new retrieval method aiming at the problems of high computational complexities and low precision of global feature extraction algorithms. The establishment of the new retrieval method is on the basis of the SIFT and Harris (APISH) algorithm, and the salient region of interest points (SRIP) algorithm to satisfy users' interests in the specific targets of images. In the first place, by using the IPDSH and SRIP algorithms, we tested stable interest points and found salient regions. The interest points in the salient region were named as salient interest points. Secondary, we extracted the pseudo-Zernike moments of the salient interest points' neighborhood as the feature vectors. Finally, we calculated the similarities between query and database images. Finally, We conducted this experiment based on the Caltech-101 database. By studying the experiment, the results have shown that this new retrieval method can decrease the interference of unstable interest points in the regions of non-interests and improve the ratios of accuracy and recall.

[Retracted] Optimization of Jirisan Mountain Cudrania tricuspidata leaf substance extraction across solvents and temperatures

  • Objective: The aim of this study is to optimize the extraction of beneficial substance from Cudrania tricuspidata leaves grown at Jirisan Mountain in South Korea by three different solvents depending on extraction time and at different temperature. Methods: The total phenolic contents were determined by the method reported by $S{\acute{a}}nchez$-Moreno et al. The total flavonoid contents were analyzed by Slinkard and Singleton. The DPPH radical scavenging activity was determined according to the method reported by Blois Results: The extraction yield for each solvent is 9.05-14.1%, 2.17-5.67%, and 2.3-3.9% for D.W., ethanol, and hexane, respectively. The overall results were maximized for the extract obtained with D.W. for 5 min at $100^{\circ}C$. The average phenol contents were 77.11, 45.64, and 0.343 mg/g at $100^{\circ}C$ in water, $78^{\circ}C$ in ethanol, and $68^{\circ}C$ in hexane, respectively. The flavonoid contents were the highest in the materials extracted with D.W., and were increased with increasing temperature, regardless of the extraction solvents, whether water (green), polar organic ethanol, or nonpolar organic hexane. In the ethanol extract, the flavonoid contents are increased gradually from 5.66 mg/g to 7.73 mg/g. The total flavonoid contents were proportional to the concentrations of the water extracts, ranging from 4.14 mg/g to 48.89 mg/g. The antioxidative activities of the water-extracted compounds are generally increased with increasing temperature from 42.5% to 85.5%. Those of the hexane extracts are increased slowly from 3.79% to 8.8%, while those of ethanol extracts are increased from 29.8% to 47.4%. Conclusion: The extraction yields were dependent upon solvents for extraction as well as extraction time and the temperature. The optimal extraction time was 5 min and the extraction yields were increased with increasing temperature excepted hexane. Of the three tested extraction solvents, the greenest solvent of water shows excellent results, suggesting that water is among the most effective solvents for natural sample extractions for general medicinal, pharmaceutical, and food applications.

Screening for DPPH Free Radical Scavenging Activities of Autogenous Seaweeds in Jeju Island Using a Electron Spin Resonance (ESR) Spectroscopy (Electron Spin Resonance을 이용한 제주 자생 해조류의 DPPH Free Radical 소거활성 검색)

  • Extracts which were prepared by four different extractions - 80% methanol extracts (ME) at high ($70^{\circ}C$) and a room temperature ($20^{\circ}C$), respectively and aqueous extracts (AE) at both temperatures with the residue after the methanol extracts - of 10 green, 19 brown and 25 red seaweeds collected in Jeju Island coast were examined for their DPPH free radical scavenging activity using a ESR (electron spin resonance) spectroscopy. A variety of the extracts showed positive scavenging effect against DPPH free radical (except the green seaweeds). Among the extracts, the brown seaweed extracts exhibited the highest scavenging activity. Especially, Sargassum spp. of the brown seaweeds have remarkable scavenging activities - both methanolic and aqueous at the both temperatures ($20^{\circ}C$ and $70^{\circ}C$). On the other hand, ME showed better scavenging activity than AE in the red seaweed extracts. These results indicate that autogenous seaweeds in Jeju will be potential natural antioxidants for functional food compounds.

  • Eight kinds of foreign wheat were used for the assessment of milling quality, and the following results were obtained. (1) Straight and patent flour extractions were, $3{\sim}5%$ and $8{\sim}11%$ lower in Australian wheat than American & Canadian wheats, respectively. (2) There were no significant differences in clear flour extraction between the clases of wheat. (3) Flour extraction from impact finisher was $5{\sim}6%$ higher in Australian wheat than American and Canadian wheats. (4) Shorts yields were $6{\sim}8%$ in American and Canadian wheats and $14{\sim}16%$ in Australian wheat. (5) Bran yields were $2{\sim}6%$ lower in Australian wheat than American and Canadian wheats.

Comparative evaluation of salivary alpha amylase level for assessment of stress during third molar surgery with and without piano music and co-relation with pain catastrophizing scale: an in vivo study

  • Objectives: Preoperative nervousness and anxiety are frequently encountered by individuals who undergo extractions of impacted wisdom teeth. The aim of the present study is to evaluate salivary alpha amylase (sAA) level in patients for assessment of stress during third molar surgery while listening to piano music and to determine its co-relation with pain catastrophizing scale (PCS). Materials and Methods: Seven patients (four males and three females) indicated for surgical extraction of bilaterally impacted mandibular third molars were included. Pre-surgical patient assessments were completed, and three samples of saliva were collected during surgery-one at baseline, one 30 minutes after commencement of surgery, and one after suturing. Assessment was performed on both sides separately with and without piano music, and the samples were assessed for sAA level and correlated with the patient's self-reported PCS. Results: Statistically significant results were obtained in patients who underwent surgical extraction while listening to piano music (P=0.046). The correlation of sAA level with PCS was not significant. Conclusion: Music demonstrated a beneficial effect on lowering the levels of stress and anxiety that a patient exhibits during any surgical procedure, and sAA can be a useful biomarker for similar assessments.

A CLINICAL AND RADIOGRAPHIC STUDY OF THE CONSERVATIVE TREATMENT OF THE NECROTIC AND INFECTED PRIMARY MOLARS (괴사유치(壞死乳齒)의 보존적 처치에 관한 연구)

  • The conserative treatment of the necrotic and infected primary molars is often necessary in order to avoid extractions which may lead to inability to masticate or loss of space and subsequent problems concerning the development of the occlusion in the permanent dentition. The purpose of this study was to evaluate the clinical significance of the conservative treatment of the infected primary molars with necrotic pulps employing the formocresol pulpotomy technique same as in vital teeth and surgical intervention in the cases with a parulis, fistula or cellulitis. The materials consisted of 25 primary molars of 9 boys and 4 girls ranging in age from 4 to 7 years. This study included clinical and radiographic observation at 3-month intervals postoperatively. The following results were obtained. 1. Clinically, 20 cases of 25 were successful, exhibiting no clinical problems such as fistula, mobility, toothache and tenderness to percussion, etc. 5 cases were regarded as failures. 2. Radiographiclly, 14 cases revealed normal features, in 6 cases slight rarefaction was observed in the periapical and interradicular region, and marked periapical and interradicular rarefaction was observed in 5 cases. 3. Marked bony deposition was observed in the periapical and interradicular regions in all the remainder except for 5 failurese. 4. Pathologic root resorption was observed in 2 cases. 5. No alveolodental ankylosis was observed. 6. Regeneration of the dental sac of the permanent tooth was observed in 1 case of 2 cases which had revealed loss of the dental sac.
