Extraction Characteristics of Polysaccharide from Fomitopsis pinicola Jeseng Mushroom (소나무 재생버섯(Fomitopsis pinicola Jeseng) 다당류의 추출 특성)

  • This study was conducted to investigate the extraction characteristics of the polysaccharide from Jeseng mushroom (Fomitopsis pinicola Jeseng). Yields of the polysaccharide extracted from powdered mushroom by autoclaving(120, 30 min) with water at different pH and salt concentration were 8.2~9.2% in pH 5~11, 4.7~5.5% in 1~5% salt solution, respectively. The yield by the 0.05~1.0 N KOH-extraction was ranged 3.45~13.20%, while that by HAS-extraction(homogenizing after KOH swelling) using 1~2.5 N KOH 73.6~78.4%. Content of carbohydrate, protein, lipid and ash of the crude polysaccharide extracted from fruits body and its cultured mycelium by method of water extraction, KOH extraction(0.005~1N) and HAS-extraction were ranged 86.5~92.6%, 2.3~13.1%, 0.1~4.2% and 0.1~1.7%, respectively. The polysaccharide were composed of 62.0~77.8 g/g of pentose, 138.0~187.8 g/g of hexose and 21.2~117.3 mg/g of protein. From these results, the polysaccharide extracted was supposed to be a protein-bound polysaccharide.

CO-SEPARATION OF Am AND RARE EARTH ELEMENTS FROM A HIGHLY ACIDIC RADWASTE SOLUTION BY A SOLVENT EXTRACTION WITH (DIMETHYLDIBUTYL TETRADECYLMALONAMIDE-DIHEXYLOCTANAMIDE)/N-DODECANE

  • This study was carried out to investigate the high-acidity co-separation of Am and RE from a simulated radwaste solution by a solvent extraction using a mixture of Dimethyldibutyltetradecylmalonamide (DMDBTDMA, as an extractant) and dihexyl octanamide (DHOA, as a phase modifier) diluted with n-dodecane (NDD). All the experiments were conducted as a batch type. First, the environmentally friendly DMDBTDMA and DHOA composed of only CHON atoms were self-synthesized. Then, the conditions for the prevention of a third phase, generated in the organic phase were examined. In addition, the effects of the concentration of nitric acid, DHOA, oxalic acid and $H_2O_2$ on the co-extraction of Am and RE were elucidated. Consequently, the optimum condition of (0.5M DMDBTDMA+0.5M DHOA)/NDD-0.3M $C_2H_2O_4-4.5M$ $HNO_3$ and O/A=2 was obtained through experimental work. Under this condition, the extraction yields were found to be about 80% for Am, more than 70% for RE such as La, Eu, Nd, Ce, etc., 3% for Cs and Sr, 69% for Fe and less than 11% for Mo and Ru. For the co-extraction of Am and RE, Fe should be removed in advance or prevented from a co-extraction with Am by controlling the different extraction rates of Am and Fe. About 95% of the Am and RE in the organic phase were stripped using a 0.5M $HNO_3$.

Development of Radiochemical Analysis of Uranium Isotopes in Soil Samples with Extraction Chromatography (크로마토 그래피 추출법을 사용한 토양시료중 우라늄 동위원소 화학분석법 개발)

  • An accurate and rapid analytical technique of uranium isotopes in highly contaminated soil samples was developed and validated by application to the IAEA-Reference samples. For overcoming the demerits of the TBP extraction method, sample materials were decomposited with $HNO_3$ and HF, and uranium isotopes were purified by an anion exchange resin and a TRU Spec resin. With the extraction chromatography method, the hindrance elements were completely removed from the uranium fraction. The chemical yields with the extraction chromatography method were more 10% higher than those with the TBP extraction method. The concentrations of uranium isotopes in soil samples using the extraction chromatography method were consistent with the reference values reported by the IAEA.

  • Centella asiatica (C. asiatica) extracts, including asiaticoside and madecassoside, are used in ointments to treat the wound and atopic dermatitis due to their antibacterial and skin-regenerating effects in Asia. Therefore, research on the cultivation and extraction efficiency of C. asiatica is being actively conducted to increase commercialization efficiency. In this study, various deep eutectic solvents (DESs) were prepared and used as the extraction solvents according to the mole ratio between the hydrogen bond acceptor (HBA) and hydrogen bond donor (HBD). And then, the extraction yields in distilled water (DW) and methanol (MeOH), commonly used extraction solvents for C. asiatica, were compared and analyzed by HPLC in the optimized operating condition. As a result, a mixture of DW and DES at a ratio of 3:7 showed about 1.4 times higher extraction efficiency than MeOH only. Conversely, the extraction efficiency in a mixture of MeOH and DES at a ratio of 3:7 was about 6 % lower than that in MeOH only.

Heavy metals leaching behavior and ecological risks in water and wastewater treatment sludges

  • Single (0.005 M DTPA), sequential (six-step) and kinetic (0.05 M EDTA) extractions were performed to assess Cd, Cr, Cu, Ni, Pb, and Zn mobilization and their potential ecological risks in Abuja (Nigeria) water (WTS) and wastewater (WWTS) treatment sludges. Total metal levels (mg/kg) in WTS and WWTS, respectively were: Cd(3.67 and 5.03), Cr(5.70 and 9.03), Cu(183.59 and 231.53), Ni(1.33 and 3.23), Pb(13.43 and 17.87), Zn(243.45 and 421.29). DTPA furnished metal extraction yields (%) in WTS and WWTS, respectively as: Cd(11 and 6), Cr (15 and 7), Cu(17 and 13), Ni(23 and 3), Pb(11 and 12), and Zn(37 and 33). The metals were associated with the soluble/exchangeable, carbonate, Mn/Fe-oxide, organic matter and residual forms to varying degrees. Kinetic extractions cumulatively leached metal concentrations akin to the mobilizable fractions extracted sequentially and the leaching data fitted well into the Elovich model. Metal mobilities were concordant for the three leaching procedures and varied in the order:WTS>WWTS. Calculated ecological risk indices suggested moderate and considerable metal toxicity in WTS and WWTS, respectively with Cd as the worst culprit. The findings may be useful in predicting heavy metals bioavailability and risks in the sludges to guide their disposal and use in land applications.

  • We present a high speed pitch estimation algorithm that is based on peak detection and average magnitude difference function (AMDF). A few pitch candidates are first estimated from the low-pass filtered (800 Hz) speech by a peak detection algorithm. AMDF values of the pitch candidatestare then calculated, and the pitch candidate that yields the minimum AMDF value is chosen as the desired pitch period. The new method requires far less computation time than other pitch estimation algorithms, while it yields fairly accurate results.

  • Effects of enzymatic hydrolysis of polysaccharides with using three commercial mixed enzymes (Ultrazyme, Celluclast, Viscozyme) were investigated on supernatant ratio, solid yields and viscosity of sea mustard extracts. The result showed that enzymatic hydrolysis prior to extraction increased the solids concentration up to 27.3% and the solids yield up to 14.0%. However the supernatant ratio after centrifugation of the sea mustard suspension was rather reduced. The viscosity of the extracts was significantly increased during initial enzymatic hydrolysis.

  • The chiral amine auxiliary mediated stereoselective alkylation reactions of glycinamides 1-6 and 15-17 using phase transfer catalyst (PTC) for liquid-solid extraction are described. The secondary N-(diphenylmethylene) glycinamides 1, 2 and 3 give better selectivities and yields than tertiary N-(diphenylmethylene) glycinamides 4, 5 and 6. Alkylation of the glycinamide 1 and 2 using 18-Crown-6 as a PTC in toluene at $-40^{\circ}C$ gives best selectivities and yields. Alkylations of N-(4-chlorophenylmethylene)glycinamides 15, 16 and 17 under same PTC conditions give $\alpha$, $\alpha-disubstituted$ amino acid derivatives 18, 19 and 20 with low diastereoselectivities.

Comparison of the Efficiency from Raw and Processed Corns by Five Different DNA Extraction Methods (다섯 가지 DNA 추출방법에 의한 옥수수 원료 및 가공시료의 DNA 추출 효율의 비교)

  • In this study, the effects of five extraction methods for raw and processed corns were compared with respect to the integrity, yields and quality of DNA extracted from them and the results were assessed by PCR analysis. From the comparison of five extraction methods, DNA integrity showed a similar pattern. Amounts of genomic DNA obtained from the five extraction methods varies from $0.25{\mu}g\;to\;234{\mu}g$ per 1 g sample. The DNA yield extracted with CTAB method and DNeasy Plant Maxi kit is greater than that obtained from other extraction methods. These results would be applicable for the selection of an adequate extraction method for specific samples.

Extraction Characteristics of Rubi Fructus in Relation to Drying Methods and Extraction Solutions (복분자의 포제방법에 따른 추출물 특성)

  • This study was observed the effects of drying methods on the extraction characteristics of Rubi fructus (fruits of Rubus coreanus). Extraction yields of soluble solids and total sugar were high in the Rubi fructus dried by freeze drying, followed by infrared drying and sun drying. Extraction yield of phenolic compounds and DPPH radical-scavenging activity of extracts were in the following order; the Rubi fructus dried by freeze drying, the Rubi fructus dried by infrared drying, the Rubi fructus dried by sun drying. L value was the highest in the Rubi fructus dried by freeze drying, and a and b value were lowest in the fruit dried by freeze drying. These results suggest that freeze drying has an beneficial effect to enhance the quality of Rubi fructus. Water and ethanol extractions was more effective in the extraction of soluble solids and the antioxidative components.
