Optimization of Extraction Conditions and Quantitative Analysis of Isoquercitrin and Caffeic Acid from Aster scaber

  • To determine the optimum extraction conditions that give the highest yield of isoquercitrin and caffeic acid from Aster scaber, the effects of four extraction variables (solvent concentrations, extraction time, number of repeated extraction, and solvent volumes) on isoquercitrin and caffeic acid yield was examined via HPLC-UV. Our results showed that the highest extract and isoquercitrin yield were observed when A. scaber was extracted with 450 mL distilled water for 8 hr repeatedly for three times. In case of caffeic acid, the content was higher in the two repeated extracts. Also, content analysis of isoquercitrin in Aster species was performed in which A. fastigiatus, A. ageratoides, and A. scaber exhibited the highest isoquercitrin content at 6.39, 5.68, and 2.79 mg/g extract, respectively. In case of caffeic acid, the highest content of A. scaber and A. glehni was 0.64 and 0.56 mg/g extract, respectively. This study reports an optimized method for extraction of isoquercitrin and caffeic acid from A. scaber and evaluates potential sources of the compounds.

Effect of Soxhlet Extraction Condition on Dewaxing Efficiency of the Beeswax-Treated Paper (속실렛 추출법에 의한 밀랍지의 탈랍처리 효과분석)

  • In this study, the effect of soxhlet extraction on dewaxing of the aged beeswax-treated paper was examined. To do this, soxhlet extraction of the aged beeswax-treated paper performed with different reflux conditions and then measured the dewaxing efficiency and change in the various properties such as tensile strength, folding endurance, color and brightness. Also, an artificial aging test was executed in order to investigate the effect of soxhlet extraction on the preservability of the dewaxed paper. As the result of FT-IR analysis, a peak area of CH and COOH group, which are main band of beeswax, was decreased or removed after soxhlet extraction. The decreasing rate of each peaks were increased depending on reflux time. After dewaxing, the physical property such as MIT type folding endurance and tensile strength was decreased. And $L^*$ value and brightness were increased while $a^*$ and $b^*$ value were decreased due to the remove of color compounds in beeswax. The more color deviation (${\Delta}E$) is expected with higher reflux times. As the result of artificial aging, preservability of two dewaxed papers except of DP_SE(C) sample was higher than that of untreated paper i.e. beeswax-treated paper.

Simultaneous Quantitation of Nine Constituents of Fraxinus rhynchophylla using High Performance Liquid Chromatography - Diode Array Detector

  • A high-performance liquid chromatography-diode array detector (HPLC-DAD) method was established for quantitative evaluation of nine constituents of Fraxinus rhynchophylla such as four coumarins, esculin (1), fraxin (2), esculetin (3), fraxetin (4), three lignans, syringaresinol 4,4'-O-${\beta}$-diglucoside (5), pinoresinol 4-O-${\beta}$-glucoside (6), pinoresinol (9), one secoiridoid, oleuropein (7), and one coumarinolignan, cleomiscosin C (8). The preferred chromatographic condition was obtained on Phenomenex Gemini-NX (3 ${\mu}m$, C18 110A, $150{\times}4.60$ mm) and the mobile phase was composed of water and acetonitrile using a gradient elution. The wavelength was set at 220 nm. Extraction condition of these constituents in F. rhynchophylla was also optimized through extraction time, extraction solvent and extraction method using established method. From this study, extraction at $70^{\circ}C$ with the mixture of ethanol and water for more than 12 h was suggested to be good extraction condition for these constituents. Quantitation of nine constituents in different F. rhynchophylla samples was also successfully accomplished with the newly established method.

Comparison of Methanol with Formamide on Extraction of Nitrogen Heterocyclic Compounds Contained in Model Coal Tar Fraction (모델 콜타르 유분 중에 함유된 질소고리화합물의 추출에 관한 메탄올과 포름아마이드의 비교)

  • The separation of nitrogen heterocyclic compound (NHC) contained in a model coal tar fraction was compared by the methanol and formamide extraction. The model coal tar fraction comprising four kinds of NHC (NHCs : quinoline, iso-quinoline, indole, quinaldine) and three kinds of bicyclic aromatic compound (BACs : 1-methylnaphthalene, 2-methylnaphthalene, dimethylnaphthalene), biphenyl and phenyl ether was used as a raw material. The aqueous solution of methanol and formamide were used as solvents. A batch-stirred tank was used as the raw material - a solvent contact unit of this work. Independent of the solvent used, the distribution coefficient of NHCs sharply increased by decreasing the initial volume ratio of water to the solvent and increasing the equilibrium operation temperature, whereas, the selectivity of NHCs in reference to BACs decreased. Decreasing the initial volume ratio of solvent to feed resulted in deteriorating distribution coefficients, but the selectivity of NHCs in reference to BAC was almost the constant. The distribution coefficient of NHCs by the methanol extraction was 3~5 times higher than that of NHCs by the formamide extraction, inversely, the selectivity of NHCs based on BACs by the formamide extraction was 3~7 times higher than that of NHCs by the methanol extraction. Furthermore, two different solvent extraction methods by adding the extraction processing speed to the balance between solvency and selectivity of NHCs were compared.

Acceleration of Viewport Extraction for Multi-Object Tracking Results in 360-degree Video (360도 영상에서 다중 객체 추적 결과에 대한 뷰포트 추출 가속화)

  • Realistic and graphics-based virtual reality content is based on 360-degree videos, and viewport extraction through the viewer's intention or automatic recommendation function is essential. This paper designs a viewport extraction system based on multiple object tracking in 360-degree videos and proposes a parallel computing structure necessary for multiple viewport extraction. The viewport extraction process in 360-degree videos is parallelized by composing pixel-wise threads, through 3D spherical surface coordinate transformation from ERP coordinates and 2D coordinate transformation of 3D spherical surface coordinates within the viewport. The proposed structure evaluated the computation time for up to 30 viewport extraction processes in aerial 360-degree video sequences and confirmed up to 5240 times acceleration compared to the CPU-based computation time proportional to the number of viewports. When using high-speed I/O or memory buffers that can reduce ERP frame I/O time, viewport extraction time can be further accelerated by 7.82 times. The proposed parallelized viewport extraction structure can be applied to simultaneous multi-access services for 360-degree videos or virtual reality contents and video summarization services for individual users.

Optimization of Extraction Process for Mass Production of Paclitaxel from plant Cell Cultures (Paclitaxel 대량생산을 위한 추출공정 최적화)

  • Several solvents or combinations of solvents were tested for the extraction of wet or dried biomass at different extraction mode from plant cell cultures. Methanol gave the highest paclitaxel recovery with the least amount of solvent usage. before extraction drying of biomass wass helpful to decrease solvent usage in extraction step./ in this case drying method was very important to obtain high yield from dried biomass. In thid mode of operation counter-current extraction process can be able to decrease solvent usage but paclitaxel recovery was almost same with both batch and counter-current mode of operation. The number of extraction times was at least four to obtain high yield(>99%) from cell and one to obtain highyield(>96%) from cell debris in batch mode. Equilibrium (i.e. the ratio of paclitaxel in biomass to paclitaxel in the extraction solvent) was reached within 5 minutes. The minimum methodal concentration (90%) and solvent amount(biomass : solvent=1 Kg : 1L) are enough to obtain high yield(>98%) for extraction from biomass.

Solvent Extraction of Tb(III) from Hydrochloric Acid Solution with Cyanex 272, Its Mixture and Ionic Liquid (염산용액에서 Cyanex 272 및 혼합용매와 이온성 액체에 의한 Tb(III)의 용매추출)

  • Cyanex 272 shows the highest separation factor for the rare earth elements from hydrochloric acid solution among the organophosporus acidic extractants, D2EHPA and PC 88A. Solvent extraction of Tb(III) from weak hydrochloric acid solution with an initial pH 3 to 6 was compared with Cyanex 272, its mixture with Alamine 336, and ionic liquid with Aliquat 336. The solvent extraction reaction of Tb(III) using Cyanex 272 was the same as that of light rare earth elements. Synergism was observed for the extraction of Tb(III) by the mixture with Alamine 336 when the initial concentration ratio of Cyanex 272 to Alamine 336 was higher than 5. Use of the ionic liquid led to a great increase in the extraction percentage of Tb(III) from the same initial extraction conditions. While the equilibrium pH of the mixture was always lower than the initial pH, under some conditions extraction with the ionic liquid resulted in a higher equilibrium pH than the initial pH. The loading capacity of the mixture and the ionic liquid was the same and 2.6 times larger than that using Cyanex 272 alone. Ionic liquid was recommended as a suitable extractant for the extraction of Tb(III) from hydrochloric acid solution based on the ease of handling and higher extraction percentage.

Solvent Extraction of Trace Amount of Ni(II) in Sea Water by using Salen[N,N'-bis(salicylidene)ethylenediamine] (Salen[N,N'-bis(salicylidene)ethylenediamine]을 이용한 해수 중 극미량 니켈의 용매추출)

  • Solvent extraction of Ni(II) into a chloroform by using salen[N,N'-Bis (salicylidene)-ethylenediamine] as a ligand has been studied. Salen was synthesized from ethylenediamine and salicylaldehyde by simple condensation reaction in an ethanol. Salen formed a 1 : 1 complex with Ni(II) and its extraction constant was $10^{5.12}$. For the determination of Ni(II) in sea water samples, some experimental conditions such as pH of solution, amount of salen, acid type and concentration for back extraction, extraction time, and influence of foreign ions were optimized by using a synthetic sea water. The sea water of which the composition was similar to a natural sea water was synthesized in this laboratory. Trace Ni(II) was extracted into the chloroform in the weak basic solution above pH 8. And the nickel could be quantitatively extracted with the concentration of salen higher than $1.2{\times}10^{-4}mol/L$. This concentration was more than 180 times of Ni(II) in the solution with a mole ratio. Real samples of Korean coastal sea water were analyzed under optimized conditions. Even though Ni(II) was not detected in these samples. Recoveries more than 98% were obtained in the samples which 40 ng/mL of Ni(II) was spiked. And detection limit of proposed method was 1.3 ng/mL. From these results, it could be known that salen of this type would be applied for the determination of trace metals as an organic chelating reagent.

Spectrophotometric Determination of Trace Amount of Sulfide by Formation of Iodide and Its Solvent Extraction with Mehtylene Green (요오드이온 생성 및 Methylene Green과의 용매추출에 의한 미량 황이온의 분광광도법 정량)

  • The iodide formed stoichiometrically for sulfide by its oxidation with iodate was extracted as an ion-pair with methylene green into 1,2-dichloroethane and the extract was measured spectrophotometrically at 656nm for the determination of sulfide. Hydrogen sulfide separated from the sample matrix was introduced into a solution containing pH 3.5 acetate buffer and iodate, in which the hydrogen sulfide was completely converted into iodide. A linear calibration graph was obtained over the range $3{\times}10^{-7}{\sim}1.2{\times}10^{-5}M$ sulfide($0.0096{\sim}0.384{\mu}g$ of $S^{2-}/ml$) and the detection limit was $0.0032{\mu}g/ml$. The apparent molar absorptivity and a correlation coefficient(r) were $6.7{\times}10^4L\;mole^{-1}\;cm^{-1}$ and 0.999, respectively. When applied to the stream water samples, the proposed method gave a relative standard deviation of 1.59% at $5{\times}10^{-6}M$ sulfide level.

  • Organic acids were produced from wastes streams in food industries by cell-recycle fermentation using Propionibacterium acidipropionici ATCC 4965. As a results of continuous fermentation, maximum productivity was 3.32g organic acid/L/hr at the dilution rate of 0.2/hr. Compared to batch fermentation, maximum productivity was improved by as much as 13 times and cell mass production was increased by as much as 22 times. The diluted organic acids in the fermenter were selectively separated by liquid-liquid extraction using 30%(w/w) trioctylamine(TOA) dissolved in methylisobutylketone(MIBK). The degree of extraction was reached above 90% for both acetic and propionic acid through repeated extraction of organic acids in fermentation broth.
