  • damages of cotton cloth and characteristics of fabroid degradation were studied by Chaetomium globosum and Aspergillus niger which presupposed as powerful erosive fungi to cellulose fiber by means of tensile strength. The results obtained are as follows: 1. the growth(rate) of fungi in malt extract agar was superior to potato agar for two weeks. 2. Chaetomium globosum showed mostly severe damage t the cotton cloth in malt extract agar media at pH 4.5. 3. Tensile strength was reduced with time by Aspergillus niger-coenzyme and Chaetomium globosum-coenzyme reaction. In comparison with Chaetomium globosum and Aspergillus niger, the former weaken tensile strength about 15.8% and the latter enfeebled 10.0% after 124 hours. 4. after 30 days the breeding of fungi in pH 4.5 malt extract agar media, critical damage of cotton cloth was observe, I. e., 92.4% damage by chaetomium globosum and 74.9% lose by aspergillus nige respectively.

Monascus purpureus에서 화색식용색소의 분리 및 화학적 특성 (Isolation and Chemical Characterization of Yellow Food Pigments from Monascus Purpureus)

  • The isolation and chemical characterization of yellow food pigments from Monascus purpureus were studied according to the compositions of media. Monacus yellow pigments were isolated and purified by solvent fractionation, silicagel column chromatography, TLC and HPLC. The retention time of Monascus yellow pigments isolated by HPLC was respectively 5 min(I) and 9 min(II) as the yeast malt extract agar(YMA) media and was respectively 4.6 min(III), 5 min(I) 5.7 min(IV), 8.3 min(V), 9 min(II) and 10.7 min(Ⅵ) at the malt extract agar(MEA) media. The structure of monascin(I), ankaflavin(II), 6,11-dihydrorubropunctatin(III), 6,11-dihydromonascorubrin(V) and unknown compounds(IV,Ⅵ) was elucidated by EI-Mass, H and C NMR, UV-visible spectrometer. Therefore, it was suggested that 6,11-dihydrorubropunctatin(III) and 6,11-dihydromonascorubrin(V) are new intermediates of Monascus yellow pigments.

  • 본 연구는 Lentinus edodes의 액체배양을 통하여 물리 ${\cdot}$ 화학적인 방법을 이용, 고농도의 탄수화물 복합효소를 얻어 hemicellulose와의 반응을 거쳐 고수율의 아라비 녹실란을 얻는데 그 목적이 있다. 온도 $24^{\circ}C$, pH 5.5. working volume 100mL, 접종비 5%. 200rpm이며 가장 적합한 C-source 로서는 glucose가 선택되었으며 N - source로서는 yeast extract가 선택되었다.

한약재 추출물의 Streptococcus mutans에 대한 항균효과 (Antimicrobial Activity of Streptococcus mutans by Herbal Medicine Extracts)

  • 본 연구에서는 국내에서 유통되는 63종의 한약재 열수 추출물의 S. mutans에 대한 항균효과를 paper disc method로 검색하여, 항균효과가 우수한 지부자, 황금의 S. mutans 생장에 관한 영향을 평가하였다. 1. 63종의 한국산 한약재 열수 추출물의 향균효과를 탐색한 결과 S. mutans에 대한 향균력이 우수한 순서는 지부자, 황금순이었다. Paper disc method에 의한 항균활성 측정(20mg/ml 투여)결과는 지부자가 6.3mm, 황금이 3.5mm이었다. 2. 한약재 추출물의 S. mutans 생장에 대한 효과는 대조군이 8시간에서 1.05로 최대 생장률을 보였으나, 지부자, 황금 추출물 2mg/ml을 투여한 배지에서는 16시간 만에 0.91, 0.87로 최대 생장을 보였다. 3. S. mutans를 8시간 동안 배양한 후 배지내 pH를 측정한 결과 대조군은 5.08 이었으며, 지부자, 황금 추출물이 투여된 배지는 5.80, 5.74이었다. 4. S. mutans를 8시간 동안 배양한 후 배지내 탄수화물 변화량을 측정한 결과 대조군은 0.81mg/ml이었으며, 지부자, 황금 추출물이 투여된 배지는 1.70mg/ml, 1.84mg/ml 이었다. 5. S. mutans를 8시간 동안 배양한 후 배지내 단백질 변화량을 측정한 결과 대조군은 8.39mg/ml 이었으며, 지부자, 황금 추출물이 투여된 배지는 9.52mg/ml, 9.28mg/ml 이었다. 6. S. mutans의 다당류 생성에 미치는 효과는 대조군의 경우 배양 8시간에 300mg/100ml을 생성하였으나, 지부자, 황금 추출물이 투여된 배지에서는 200mg/100ml, 220mg/100ml을 생성하였다.

Beginning of a New Standard: Internet of Media Things

  • Recently, Internet of Things (IoT) drives a large variety of research, development, and new type of markets. All type of devices and sensors will be part of the Internet of Things and will be able to communicate not only plain data, but also audio-visual, olfactory, and haptic media data. In addition, as the devices and sensors getting smarter, it is highly probable that they can process acquired media and metadata to extract higher level of information (e.g., semantics). To support such enhanced functionalities, ISO/IEC SC29 WG11 (MPEG) starts a new standard project, ISO/IEC 23093, called Internet of Media Things (IoMT) to provide standard data formats and APIs for media things. This paper presents the standardization activities of IoMT focusing on explaining terms, standard scopes, and major media things with their use cases. One of the use cases, an IoT system for a blind pedestrian navigation assistance, is evaluated to prove its effectiveness.

표고 교잡균주들의 세포외 laccase 활성 검출에 미치는 배지성상과 발색반응 시약의 영향 분석 (Analysis of the Effect of Media Types and Chromagenic Chemicals on the Detection of Extracellular Laccase Activity among Lentinula edodes Strains)

  • 본 연구에서는 표고 교잡균주들의 생화학적 특성을 이해하기 위한 하나의 시도로서 교잡균주간의 laccase 효소의 활성을 비교하는 빠르고 간편한 방법을 모색하고자 균사체 배양 배지의 성상과 염색물질이 laccase 활성 검출에 미치는 영향을 조사 비교하였다. 고체배지와 액체배지 모두에서 발색시약이 Remazol Brilliant Blue R(RBBR)일 경우는 potato dextrose 성분 보다 malt extract 성분 배지를 사용하는 것이 표고버섯균 균사가 분비하는 세포외 laccase 활성 검출이 용이하였다. Guaiacol을 발색시약으로 사용하는 경우는 potato dextrose 성분과 malt extract 성분 배지 모두 세포외 laccase 활성 검출이 용이하였다. 2ml microfuge tube를 이용한 액체 배양 방법은 RBBR와 Guaiacol 모두 malt extract 성분배지에서 3일만에 표고 교잡균주간의 세포외 laccase 활성 검출과 정량적 비교를 가능케 하였다.

Saccharomyces cerevisiae를 이용한 효율적인 호흡저해제 검정법 (Efficient assay for respiration inhibitor using Saccharomyces cerevisiae)

  • Saccharomyces cerevisiae를 이용하여 효율적인 호흡저해 스크리닝 방법을 개발하고자 실험하였다. S. cerevisiae균을 glucose 발효와 미토콘드리아 호흡이 가능한 yeast extract-peptone-dextrose (YPD) 배지와 단지 미토콘드리아 호흡만이 가능한 non-fermentable carbon source-yeast extract (NFY) 배지로 수확하였다. 96-well plate의 각 well에 균 현탁액을 분주한 다음 다양한 작용기작의 46개 살균제를 여러 가지 농도로 처리하였다. NFY배지에서의 non-fermentable carbon source로는 ethanol (NFY-E배지) 및 glycerol (NFY-G배지), lactate (NFY-L배지)를 이용하였다. 접종 후 $1{\sim}3$일 동안 배양한 다음 최소억제농도 (minimum inhibitory concentration, MIC)를 결정한 결과, 4개의 호흡억제 살균제인 azoxystrobin, kresoxim-methyl, metominostrobin, trifloxystrobin은 YPD 배지에서 균의 생육을 전혀 억제하지 못하였으나, 세가지 NFY배지에서는 높은 항균활성을 보였다. 이와는 반대로 5개의 N-trihalomethylthio계 살균제는 NFY배지보다 YPD 배지에서 높은 활성을 보였다. 그리고 11개 살균제는 두 배지 모두에서 같은 항균활성을 나타내었고, 나머지 26개의 살균제는 모든 배지에서 전혀 항균활성을 보이지 않았다. 그러므로 S. cerevisiae와 96-well plate를 이용한 호흡저해제 검정법은 신속하고 편리하게 호흡저해제를 스크리닝 할 수 있는 방법으로 여겨진다.

Antioxidant Activity of Indigenous Trametes Species in Korea

  • The fungal genus Trametes is globally distributed and comprises various wood-decay species, including the well-known medicinal mushroom Trametes versicolor, a popular remedy in traditional Asian medicine. Trametes species produce antioxidants, which have a wide range of health benefits. The pressent study evaluated seven indigenous Trametes species from Korea, which were cultivated in three different media (dextrose-yeast extract, DY; malt extract-yeast extract, MY; malt extract broth, MEB) and tested for antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assays. We found that the medium consumption rate did not significantly differ between the media and among the strains (72-76%). However, the T. versicolor strains had a relatively low consumption rate (14-65%). The 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) (ABTS) tests demonstrated that culture filtrates of T. cf. junipericola, T. orientalis, T. suaveolens, and T. versicolor possessed antioxidant activity against damage from free radicals. In particular, T. cf. junipericola (DY) and T. versicolor (MY) had >80% activity in the DPPH and ABTS assays, compared with that of the positive control (ascorbic acid). Thus, our study identified promising candidates with substantial antioxidant activity among the indigenous strains of Trametes spp. from Korea.

미디어 아트에 나타난 시공간 개념의 표현 특성에 관한 연구 (A Study on the Characteristics of Expression of Time-Space Concept in Media Art)

  • The concept of time-space to be seen in the modem society has complex features at the points of scientific, philosophical, artistic, religious point of view. It has been brought up at a points of neo centric religious, philosophical view and then after, phenomenal approach and scientific approach were rendered through Newton and Einstein and so many scholar approaches affected the artistic field significantly. With such visual expression, it has affected the paintings for a long time till the 19th century, and then has affected various fields such as photography, film, video, computer, architecture and has currently affected the various space expressive of our daily lives and virtual space of digital. This study is to extract a visual and expressionistic characteristic through historical examination and multi-scholar analysis of time-space concept. Especially, by expanding the previous time and space concept, experimental cases and expressionistic characteristics has come up on the media art that offers various experience and by analyzing the expressionistic characteristics of the time-space concept expressed in media art, it is to extract the basic factors for the diversity of space experience.

인삼이 Aspergillus flayus의 생육 및 Aflatoxin 생성에 미치는 영향 (Effect of Panax ginseng on the Growth and Production of Aflatoxin by Aspergillus flayus)

  • This study was performed to investigate the effect of the Panax ginseng C. A. Meyer on the growth and production of aflatoxin by Aspergillus flayus ATCC 15517. Asp. fiavus with 10$^6$ conidia was incubated at 30$\circ$C for 7 days on YES broth containing 0.1%, 0.5%, and 1.0% of ginseng extract. After incubation, dry mycelial weight, pH, and production of aflatoxin were investigated. The results were as follows:There was no significant difference in dry mycelial weight by the addition of 0.1% and 0.5% ginseng extract. However, it was decreased to the rate of 13.7% by the addition of 1.0% ginseng extract in 7 days. pH changes in cultures were similar regardless of the concentration of ginseng extract. The pH values decreased to minimum in 5 days and again increased. Aflatoxin production was reduced as the concentration of ginseng extract increased. When compared to the control, the production of total aflatoxin significantly reduced to 56.7%, 54.0%, 53.3% in the media of 0.1%, 0.5% and 1.0% of ginseng extract, respectively. No significant difference was observed among ginseng extract groups.
