Effect of Paeonia lactiflora Extracts on ${\alpha}-Glucosidase$

  • This study was carried out to investigate inhibitory effect of extracts from the root of Paeonia lactiflora on postprandial hyperglycemia. Organic solvent (hexane, ethyl acetate, butanol, aqueous) extracts from the crude drug were fractionated by high performance liquid chromatography. These fractions were examined to evaluate ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) inhibition by microplate colorimetric assay. Among the fractions examined, the ethyl acetate fraction from the roots of Paeonia lactiflora showed potent inhibitory effects on ${\alpha}-glucosidase$. Therefore, further fractionation of the fraction was carried out to isolate the active principles. Finally, we isolated and Purified 1,2,3,4,6-penta-O-galloyl-beta-D-glucose (PGG) as a active principle by activity-guided fractionation. These results suggest that the extract from the root of Paeonia lactiflora can be used as a new nutraceutial for inhibition on postprandial hyperglycemia and PGG might be a candidate for developing an ${\alpha}-glucosidase$ inhibitor.

Study on the Alkaloids from Thalictrum fauriei

  • To explore the biological activities of Thalictrum alkaloids, a study was initiated to investigate the alkaloidal constituents from T. fauriei, a species indigenous to Taiwan. Extensive fractionation of the ethanolic extract provided three known aporphines, (+)-oconovine (Ia), (+)-isocorydine (Ib), and (+)-corydine (Ic) from nonphenolic tertiary base fraction. From quaternary base fraction two new protoberberinium salts, thalifaurine (IIa), dehydrodiscretine (IIb), in addition to magnoflorine (III), were isolated. The structure of both new compounds were confirmed by total syntheses. Fractionation of phenolic bases yielded a dihydromorphinandienone, (-)-ocobotrine (IV), as well as a novel aporphine-pavine dimer, tentatively named EP-10. The structure of EP-10 was established by means of 2D NMR studies. Preliminary study indicated a weak activity of lyzing Hela cells, in vitro, for EP-10.

포포나무 잎 추출물에 존재하는 Acetogenin 확인을 위한 액체크로마토그래피 질량분석기의 활용 (Utilization of Liquid Chromatography Mass Spectroscopy for Screening Analysis of Acetogenins in Extracts of Asimina triloba Leaves)

  • Asimina triloba contains many kinds of annonaceous acetogenins known to have toxicity and/or activity. In this study, analysis of acetogenins was carried out on the extract and its solvent fractions of A. triloba leaves using a liquid chromatography mass spectroscopy(LC-MS). In the extract, the ethanolic extract was significantly higher acetogenin contents than hot water extract. In the analysis of solvent fraction from the ethanolic extract, n-hexane, ethylacetate, and aqueous fraction were contained 85.17~90.92%, 8.96~14.52% and 0.12~0.35% of total acetogenin in the extract. Based on these results, the analytical method is useful for comparing annonaceous acetogenins contents in plant samples. Moreover, when research or product development aiming at the physiological activity of acetogenin such as anticancer activity using A. triloba leaves is performed, a fractionation using a nonpolar organic solvents and alcohol extract may be useful methods. This method can also be used for the purpose of eliminating toxicity as well.

넙치(Paralichthys olivaceus) 알로부터 Serine Protease Inhibitors의 분획 특성 (Recovery and Fractionation of Serine Protease Inhibitors from Bastard Halibut Paralichthys olivaceus Roe)

  • Protease inhibitors (PI) of trypsin and papain as target proteases from the roe of bastard halibut Paralichthys olivaceus were fractionated out using ammonium sulfate precipitation (A), DEAE 650M anion exchange chromatography (D), and Sephacryl S-300 gel filtration (S). The recovery percentages of the fractions with the strongest inhibitory activity for each fractionation method were 13% for the A4 fraction, 21.2% for the D3 fraction, and 21.3% for the S2 fraction, with specific inhibitory activities of the fractions toward trypsin and casein of 168, 139, and 218 U/mg, respectively, while no inhibition of papain was observed. The $IC_{50}$ for the trypsin-specific substrate $N{\alpha}$-benzoyl-$\small{L}$-arginine-p-nitroanilide (BAPNA) was 0.65, 1.55, 2.26, and 2.85 mg/mL for the A4, S2, A3, and D3 fractions, respectively. These results suggest that chromatographic fractionation methods (D and S) based on the molecular mass and charge of the protein were more effective at fractionating PI than was ammonium sulfate precipitation based on protein solubility, and that the bastard halibut roe extract acts as a serine protease inhibitor. Therefore, the PI fraction from fish roe might be useful for inhibiting proteases in foodstuffs, and could constitute an alternative food-grade inhibitor for the surimi industry.

황련 추출물의 분획화 및 BV2 microglial cells에서 LPS에 의해 유도되는 nitric oxide 생성억제효과 검정 (Effects of subfractions of Coptidis Rhizoma extract on the nitric oxide production in LPS-stimulated BV2 microglial cells)

  • Objectives : Uncontrolled activation of microglia may directly toxic to neurons by releasing various substances such as inflammatory cytokines, nitric oxide(NO), prostaglandin E2 and superoxide. In this study, the effects of the several subfractions isolated from Coptidis Rhizoma extract were investigated on NO production in LPS-stimulated BV2 microglial cells, Methods : Coptidis Rhizoma extract prepared with 80% methanol, and then fractionated with ethylacetate, chloroform, n-butanol and water. BV2 cells were pretreated four subfractions of Coptidis Rhizoma with various concentrations, and then stimulated with LPS. Cytotoxicity of each fraction was measured by MTT assay. NO production was determined in culture surpernatants by Griess reagent. Results : Ethylacetate, chloroform and butanol fractions of Coptidis Rhizoma extract significantly decreased LPS-induced NO production in BV2 cells as a dose-dependent manner without cytotoxicity. Ethylacetate fraction of Coptidis Rhizoma extract was most effective on inhibition of NO production in LPS-stimulated BV2 cells compared with other fractions. Conclusion : This data indicates that Ethylacetate fraction of Coptidis Rhizoma extract shows strong antiinflammatory effects through inhibition of LPS-induced microglial activation.

바나나 잎 추출물의 주름개선 효과 (Antiaging Effects of Musa sapientum L. (Banana) Leaf Extract)

  • To examine the possibility of using Musa sapientum L. (Banana) leaf extract as a cosmetic raw material, banana leaves grown in Jeju Island were extracted with 70% ethanol. Polysaccharides present in banana leaf extract were discarded by precipitation with cold ethanol. Polysaccharide-discarded banana leaf extract promoted procollagen and COL1A1 gene expression, but inhibited matrix metalloproteinase (MMP)-1 and MMP-2 gene expression in human skin fibroblasts when examined by real-time reverse-transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). The active compound in banana leaves was identified by fractionation with various solvents. The chloroform fraction showed the highest anti-wrinkle efficacy and the active compound of chloroform fraction was identified as corosolic acid by NMR, FT-IR, EA, and HPLC-MS. In addition, banana leaf extract showed anti-oxidative efficacy with an IC50 value of 67.91 ppm, as determined by DPPH free radical scavenging assay. Finally, the anti-wrinkle efficacy of banana leaf extract-containing cream was confirmed by clinical tests. Based on these results, banana leaves could have an application as a cosmetic raw material with anti-wrinkle efficacy.

비파엽 품질 비교 분석을 위한 Triterpenic Acid의 추출 방법 (Development of an Effective Extraction Method for the Quality Control of Eriobotrae Folium : Determination of Triterpenic Acids)

  • In result of analysis for the content determination of ursolic acid(UA) and oleanolic acid(OA) in extract of Eriobotryae folium using HPLC with UV detector, UA in chloroform extract of Moo-mok variety was showed highest content(2.7843 mg/g). And OA in ethyl acetate extract of Dae-bang variety was showed highest content(0.5898 mg/g). These result suggest that direct extraction using organic solvent(chloroform or ethyl acetate) was useful method for rapid quantitative analysis of UA and OA without preprocessing such as drying or fractionation.

사상자의 항염증작용 (Anti-inflammatory Activities of Torilis japonica Fruit)

  • The methanol extract of Torilis Fructus showed potent anti-edematous effect in carrageenan-induced paw edema in rats. Systematic fractionation of the extract with hexane, chloroform, ethyl acetate and butanol resulted in potent anti-edematous action only in hexane fraction at an oral dose of 237 mg/kg which is corresponding to 4.8 g of raw Fructus. This hexane fraction showed inhibitory effects at 77 mg/kg p.o on vascular permeability in mice, at 8mg/pouch on leucocyte emigration in rats, and at 120 mg/kg p.o. on adjuvant arthritis model in rats. These results demonstrate that hexane fraction of the extract possesses potent anti-inflammatory effect which may support its traditional uses.

연교(連翹)추출물의 일반(一般) 약리작용(藥理作用) (Pharmacological Studies on Forsythiae Fructus)

  • The Forsythiae fructus is described to be used as an antiinflammatory drug, diuretics, antidotes and antibacterials in oriental literatures. In order to investigate the efficacy of Forsythiae viridissima (Oleaceae), the methanol extract and its fraction have been evaluated for the acute toxicity, antiinflammatory, analgesic and spasmolytic action in animals. The methanol extract of Forsythiae fructus was found to have significant antiinflammatory activity in the acute and subacute antiinflammatory model in rats, but have no analgesic action. Furthermore, through fractionation procedure, it was found that the active compounds were easily soluble in chloroform and butanol. It is also noted that the extracts had spasmolytic activities in the rat fundus and uterus and had low acute toxicity in mice.

오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향 (Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours.
