Genetic Manipulation and Transformation Methods for Aspergillus spp.

  • Species of the genus Aspergillus have a variety of effects on humans and have been considered industrial cell factories due to their prominent ability for manufacturing several products such as heterologous proteins, secondary metabolites, and organic acids. Scientists are trying to improve fungal strains and re-design metabolic processes through advanced genetic manipulation techniques and gene delivery systems to enhance their industrial efficiency and utility. In this review, we describe the current status of the genetic manipulation techniques and transformation methods for species of the genus Aspergillus. The host strains, selective markers, and experimental materials required for the genetic manipulation and fungal transformation are described in detail. Furthermore, the advantages and disadvantages of these techniques are described.

  • Now a days, large volumes of data is accumulating in every field due to increase in capacity of storage devices. These large volumes of data can be applied with data mining for finding useful patterns which can be used for business growth, improving services, improving health conditions etc. Data from different sources can be combined before applying data mining. The data thus gathered can be misused for identity theft, fake credit/debit card transactions, etc. To overcome this, data mining techniques which provide privacy are required. There are several privacy preserving data mining techniques available in literature like randomization, perturbation, anonymization etc. This paper proposes an Enhanced Hybrid Privacy Preserving Data Mining(EHPPDM) technique. The proposed technique provides more privacy of data than existing techniques while providing better classification accuracy. The experimental results show that classification accuracies have increased using EHPPDM technique.

  • Understanding the chemical composition of the brain helps researchers comprehend various neurological processes effectively. Understanding of the fundamental pathological processes that underpin many neurodegenerative disorders has recently advanced thanks to the advent of innovative bioanalytical techniques that allow high sensitivity and specificity with chemical imaging at high resolution in tissues and cells. Mass spectrometry imaging [MSI] has become more common in biomedical research to map the spatial distribution of biomolecules in situ. The technique enables complete and untargeted delineation of the in-situ distribution characteristics of proteins, metabolites, lipids, and peptides. MSI's superior molecular specificity gives it a significant edge over traditional histochemical methods. Recent years have seen a significant increase in MSI, which is capable of simultaneously mapping the distribution of thousands of biomolecules in the tissue specimen at a high resolution and is otherwise beyond the scope of other molecular imaging techniques. This review aims to acquaint the reader with the MSI experimental workflow, significant recent advancements, and implementations of MSI techniques in visualizing the anatomical distribution of neurochemicals in the human brain in relation to various neurogenerative diseases.

  • Experimental design in simulation provides an efficient way of economizing simulation runs since a considerable number of simulation runs that originally were planned can be reduced by this approach. This experimental design method is an active area of research together with the output analysis and so no single panacea seems to exist so far. Thus, selection of techniques of experimental design and output analysis more lithely depends upon the objective of simulation analysis, budget constraint and sometimes the analysts subjective judgment. This paper attempts to describe an experimental design methodology for port investment analysis using a case study in a bulk terminal in Korea. Detailed display will be focused on simulation period, warm-up period, the number of replications needed in production runs after brief explanation on the system configuration.

  • Recently, cable-supported long-span bridge are widely constructed due to improved quality of cable and development of design and construction techniques. In this study, an existing cable-stayed bridge, Dolsan Bridge, has been evaluated based on the cable forces measured using vibration method. And the finite element model using in this study for the dynamic analysis has been found to be quite comparable with dynamic mode shapes and natural frequencies estimated from experimental data induced by ambient traffic excitations.

  • In this dissertation, experimental research was carried out to study the hysteretic behavior of reinforced concrete frame designed by high performance techniques, using carbon fiber plate, diagonal bracing system with or without steel frame. Experimental programs were carried to evaluate the structural performance of such test specimens, such as the hysteretic behavior, the maximum horizontal strength, crack propagation, and ductility etc. Specimens(RFCP, RFXB, RFXB-F), designed by the improvement of earthquake-resistant performance, were attained more load-carrying load-carrying capacity stable hysteretic behavior.

  • In this study, the experimental results were simulated by using a nonlinear analysis programs IDARC 2D and RUAUMOKO 2D. These programs use a global Takeda-like model. The objectives of this study is to verify the correlation between the experimental and analytical responses of reinforced concrete (RC) frame and to provide the calibration to the available static inelastic analysis techniques. The evaluation of the accuracy of analytical simulation by IDARC 2D and RUAUMOKO 2D leads to the conclusion that the global behaviors can be, in general, simulated with limited accuracy in the linear analysis as detailing.

Analysis of Fatigue Durability on Seam Weldment using Notch Stress Approach (노치응력접근법을 이용한 차량구조재 용접이음부의 피로내구성 해석)

  • Fatigue life at seam weldment of thick plate was estimated using the finite element and FEM-FAT(an exclusive fatigue solver). Finite element meshing at toe and root of weldment was based oil Radaj's theory. Also, the results of FE analysis were compared with experimental results in the point of Miner's Rule. The results of FE and FEM-FAT analysis were in accord with experimental results within 60% confidence. This result reveals that above techniques is useful in assessment of seam weldment and to be an alternative method instead of an object experiment.

  • The transient flow fields in direct injection engine was analyzed by using the STAR-CD CFD code during the intake/compression processes. The grids are generated by using the IC3M. The CFD results were compared with experimental data. The results showed that the used techniques were well suited for the flow analyses on any internal combustion engines.

  • As the system complexity increases, it is important to develop high-level verification techniques for fast and efficient design verifications. In this research, fast verification techniques for hardware and software co-design systems have been developed by using logic emulation and algorithm-level simulation. For faster and superior functional verification, we partition the system being designed into hardware and software parts, and implement the divided parts by using interface modules. We also propose several hardware design techniques for efficient hardware emulation. Experimental results, obtained by using a Reed-Solomon decoder system, show that our new verification methodology is more than 12,000 times faster than a commercial simulation tool for the modified Euclid's algorithm block and the overall verification time is reduced by more than 50%.
