A Experimental Study on the Effect of Evening primrose on Hyperlipidemia (월견초(月見草)가 고지혈증(高脂血症)에 미치는 영향(影響))

  • In order to study the anti-hyperlipidemic effects of Evening primrose, the changes of body weight, serum total cholesterol, serum HDL-cholesterol, serum triglyceride, serum LDL-cholesterol, serum total lipid and organ weight were observed after the liquid extracts of Single-dosage Evening primrose and Double-dosage Evening primrose were administered p.o to the hypercholestemic and hypertriglyceremic rats induced by 1% cholesterol diet during 10, 20, 30 days. The result were summarized as follows ; 1. The contents of body weight compared with control group was significantly decreased in single-dosage Evening primrose group during 10, 30 days and in double-dosage Evening primrose group did not show significant value. 2. The contents of serum total cholesterol with control group tend to be decreased in single-dosage Evening primrose group, but did not show significant value. Double-dosage Evening primrose group showed significant value during 20, 30 days. 3. The contents of serum HDL-cholesterol compared with control group was significantly increased in single-dosage Evening primrose group during 10, 20, 30 days. Double-dosage Evening primrose group showed significantly value during 30 days. 4. The contents of serum triglyceride compared with control group was significantly decreased in single-dosage Evening primrose group during 10, 30 days. Double-dosage Evening primrose group showed significant value during 20 days. 5. The contents of serum LDL-cholesterol compared with control group was significantly decreased in single-dosage Evening primrose group during 10 days. Double-dosage Evening primrose group showed significant value during 10, 20, 30 days. 6. The contents of serum total lipid compared with control group was significantly decreased in single-dosage Evening primrose group during 20 days. Double-dosage Evening primrose group showed significant value during 20, 30 days. 7. The contents of liver weight compared with control group was significantly decreased in single-dosage Evening primrose group and double-dosage Evening primrose group. The contents of kidney weight compared with control group was significantly decreased in single-dosage Evening primrose group. The contents of spleen weight compared with control group was significantly decreased in single-dosage Evening primrose group and double-dosage Evening primrose group. The contents of testis weight compared with control group tend to decreased in single-dosage Evening primrose group and double-dosage Evening primrose group, put did not show a significant value. From the above results, it was thought that Evening primrose could be applied effectively to the Hyperlipidemia.

Effect of Rehmanniae Radix and Pear Phenolic Compound on the STZ-Treated Mice for Induction of Diabetes (생지황(Rehmanniae Radix)과 배의 Phenolic Compound가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향)

  • This study has been carried out to investigate effect of Rehmanniae Radix (RR) and pear phenolic compound (PC) on the hyperglycemic mice induced with streptozotocin (STZ). For this purpose, male mice were fed with a 0.2 mL RR extract (S group) and the pear PC (90 mg/kg/day) dissolved in a 0.2 mL RR extract (SPC group) while the control group received the same commercial diet for 6 weeks. The blood glucose contents were examined from tail vein blood once a week for 6 weeks. Samples of pancreas removed after the experimental period were processed for the immunohistochemical identification of $\beta$-cells. The levels of serum glucose were decreased significalntly (p<0.05) in the S and SPC groups compared with the control group. The BUN and creatinine levels were significantly (p<0.05) decreased in SPC group compared with the control group. Intraperitoneal glucose tolerance tests peformed at 24 hours before that period revealed that glucose tolerances in S and SPC group were ameliorated. Immunohistochemical analyses of the pancreases revealed that a lot of insulin- positive $\beta$-cells were contained in a Langerhas's islets of S and SPC groups compared with the control group, and the number of Langerhas's islets were significalntly increased in S (p<0.01) and SPC (p<0.05) groups. These results suggest that RR extract and pear PC could recover the damages induced by STZ in the hyperglycemic mice.

Effect of Dietary Supplementation of β-Carotene on Lipid Peroxide Level and Antioxidative Vitamins of Diabetic Rats (β-Carotene의 보충 급여가 당뇨쥐의 지질과산화물과 항산화비타민 함량에 미치는 영향)

  • Diabetic vascular complications such as atherosclerosis have been reported as one of significant obstructions in treatment of diabetes. There has been a significant increase in recognition of the importance of antioxidative nutrients such as vitamin E, for the prevention of diabetic vascular complication by oxidative stress. This study focused on the effect of dietary $\beta$-carotene on the levels of lipid peroxide and antioxidative vitamins of diabetic rats. The plasma glucose level, hepatic level of lipid peroxide and contents of antioxidants such as vitamins A and E were determined in STZ-induced diabetic rats. Dietary supplementation of B-carotene did not reduce the blood glucose in diabetic rats. Hepatic level of lipid peroxide tended to increase in diabetic rats, but $\beta$-carotene intake reduced the value. Plasma levels of retinol and retinol/lipid were not changed by dietary supplementation of $\beta$-carotene. There was no significant difference among experimental groups in plasma level of $\alpha$-tocopherol. Hepatic levels of retionl and retinyl palmitate were increased by dietary supplementation of $\beta$-carotene in diabetic rats. These results suggest that the supplementation of $\beta$-carotene to the normal diet of diabetics may reduce the incidence of the diabetic vascular complications through the improvement of antioxidants depletion.

Effects of Raw Soy Flour and Magnesium on Serum Glucose and Lipid Metabolism in Streptozotocin-Diabetic Rats (대두와 Magnesium 급여가 당뇨쥐의 혈당 및 지질대사에 미치는 영향)

  • Effects of raw soy flour(RSF) and RSF with magnesium supplementation(RSF+Mg) feeding on lipid metabolism and serum glucose concentration of diabetic and nondiabetic rats were studied. Male rats(Sprague-Dawley, body weight $332.8\pm19.3g)$ were assigned to four dietary groups and fed for 4 weeks. For each experimental diet some rats were injected with streptozotocin in intraperitonium (i.p.) to induce diabetes, and other rats were injected with buffer i.p. as a nondiabetic group. The diabetic rats fed diets of control, RSP and RSF+Mg showed significant body weight decrease(-67~-79g) compared with nondiabetic group. Pancreas weights in diabetic RSF and RSF + Mg groups were heavier than those of the control and nondiabetic groups. , Fasting serum glucose level of diabetic RSF and RSF+Mg groups significantly decreased by 73 and 44% compared with the diabetic control group, but they significantly increased by 77 and 111% compared with the nondiabetic group. Serum triglyceride, phospholipid and total cholesterol concentration of diabetic RSF and RSF + Mg groups significantly decreased compared with the diabetic control group. Serum HDL-cholesterol, HDL-cholesterolitoal cholesterol ratio and liver total cholesterol were not affected by RSF and RSP+Mg feeding, Liver total lipid and triglyceride concentration were significantly lower in diabetic groups than those in the nondiabetic group. The results indicated that RSF and RSF+Mg feeding did not produce hyperlipemia and somewhat improved the glucose levels in diabetic rats.

Optimum Dietary Protein and Lipid Levels on Growth in Parrot Fish (Oplegnathus fasciatus) (돌둠사료의 적정 단백질 및 지질 함량)

  • A feeding trial was conducted to investigate optimum dietary protein and lipid levels in diets for parrot fish (Oplegnathus fasciatus). Three groups of 25 fish averaging 7.0g were fed each of the experimental diets containing 30, 40, 50 or 60% crude protein (CP) with 8 or 16% crude lipid level ($4{\times}2$factorial desing) for 8 weeks. Weight gain and feed efficiency increased significantly (P<0.05) with dietary protein level up to 50% CP when diets contained 16% lipid level, and increased with dietary protein level up 60% CP when diets contained 8% lipid level (P<0.05). The broken-like model showed that an optimum dietary protein level was 46% when the diet contained 16% dietary lipid. Fish fed diets containing 16% lipid level showed significantly (P<0.05) higher weight gain, feed efficiency and protein efficiency ratio than did fish fed diets containing 8% lipid level when diets contained 40 or 50% dietary protein (P<0.05).

Effects of Isoflavone-Rich Bean Sprout on the Lipid Metabolism of the Ethanol-Treated Rats (고 Isoflavone 콩나물이 만성 에탄올 투여 흰쥐의 지질대사에 미치는 영향)

  • The present study evaluated the effects of isoflavone-rich bean sprout on the lipid metabolism in ethanol-treated rats. Fifty male Sprague-Dawley rats were randomly divided into five groups: normal control, alcohol control, low soybean sprout+ethanol (low SS), high soybean sprout+ethanol (high SS) and isoflavone extract+ethanol (IE). They were fed experimental diets based on Lieber-DeCarli liquid diet for 40 days. Body weight, food intake and feed efficiency ratio (FER) of ethanol-treated groups were significantly suppressed compared with that of the normal control group. Among the ethanol-treated groups, high SS group showed in significant increase in the body weight, food intake and FER. Supplementation of isoflavone-rich soybean sprout powder or isoflavone extract significantly decreased plasma triglyceride (TG), total cholesterol (TC), atherogenic index (AI) and increased the ratio of HDL-cholesterol to TC. Supplements also significantly decreased total lipid, TG and TC in liver tissue compared with that of alcohol control. There was a significant decrease in hepatic lipid peroxidation products in IE group compared with other ethanol-treated groups. This results suggest that supplementation of isoflavone-rich bean sprout powder may exert beneficial effects on lipid metabolism in chronically alcohol-treated animals by improving lipid profiles in plasma and liver tissues.

Effects of Dietary Fructose and Glucose on Hepatic Steatosis and NLRP3 Inflammasome in a Rodent Model of Obesity and Type 2 Diabetes (비만 및 제2형 당뇨병 쥐 모델에서 과당과 포도당의 섭취가 지방간과 NLRP3 염증조절결합체에 미치는 영향)

  • This study is carried out to assess the relative effects of different doses of dietary glucose or fructose on non-alcoholic fatty liver disease (NAFLD) and hepatic metaflammation in a rodent model of type 2 diabetes. KK/HlJ male mice were fed experimental diets as follows: 1) control (CON), 2) moderate glucose (MG, 30% of total calories as glucose), 3) high glucose (HG, 60% of total calories as glucose), 4) moderate fructose (MF, 30% of total calories as fructose), and 5) high fructose (HF, 60% of total calories as fructose) for three weeks. Food intake was not affected by treatments. Compared with HF, HG not only increased serum fasting glucose and area under the curve during oral glucose tolerance test, but also decreased the levels of serum insulin and adiponectin. It indicated that glucose control was complicated via high glucose intake. High fructose treatment led to increased triglyceride in the serum and liver. In comparison to HG, high fructose diet activated NOD-like receptor family, pyrin domain containing 3 (NLRP3) inflammasome consisting of apoptosis-associated speck-like protein containing a CARD (ASC), NLRP3 and caspase 1, which increases interleukin (IL)-$1{\beta}$ maturation and secretion. The activation of NLRP3 inflammasome was accompanied by increased levels of tumor necrosis factor alpha (TNF-${\alpha}$) and IL-6. However, the expression of NLRP3 inflammasome components and pro-inflammatory cytokines did not differ between CON and HG. These data suggested that dietary fructose triggers hepatic metaflammation accompanied by NLRP3 inflammasome activation and has deleterious effects on NAFLD.

Effects of Cheonghyul-San on the Generation of Redox Status and on the Expression of NF-${\kappa}$B Dependent Proteins (청혈산(淸血散)이 Redox Status 및 NF-${\kappa}$B 의존성 단백질에 미치는 영향)

  • The aim of this study was to investigate the effects of Cheonghyul-san on the generation of peroxynitrite ($ONOO^-$), nitric oxide (NO) and superoxide anion radical ( ${\cdot}\;O_2^-$), and on the expression of NF-${\kappa}$B-dependent proinflammatory proteins in ob/ob mice. Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57BL/6J black mice) and control obese (ob/ob mice) groups have received the standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein-2 (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blot was performed using anti-IKK-${\alpha}$, anti-phospho I${\kappa}$B-${\alpha}$, anti-NF-${\kappa}$B (p50, p65), anti-COX-2, anti-iNOS, anti-VCAM-1 antibodies, respectively. Cheonghyul-san prevented $H_2O_2$-induced cell death. Cheonghyul-san inhibited the generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ in the $H_2O_2$-treated LLC-$PK_1$ cells. The generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ were inhibited in the Cheonghyul-san-administered ob/ob mice groups. The GSH/GSSG ratio was decreased in the ob/ob mice, whereas the ratio was improved in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the protein expression levels of phospho-I${\kappa}$B-${\alpha}$, IKK-${\alpha}$, NF-${\kappa}$B (p50, p65), COX-2, iNOS and VCAM-1 genes. These results suggest that Cheonghyul-san is an effective scavenger of $ONOO^-$, ${\cdot}\;O_2^-$ and NO, and has an inhibitory effect on the expression of NF-${\kappa}$B-dependent inflammatory genes in ob/ob mice. Therefore, Cheonghyul-san might be used as a potential therapeutic drug against the diabetes- and obesity-related proinflammatory diseases.

Effects of Mori Fructus Extract on Hyperglycemia, Hyperlipidemia, Polyol Pathway, AGE and RAGE in ob/ob Mice (상침자(桑椹子)추출물이 ob/ob mouse의 혈당, 혈청 지질, Polyol Pathway, AGE 및 RAGE에 미치는 영향)

  • Etiological studies of diabetes and its complications showed that oxidative stress might play a major role, Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Mori Fructus extract has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs, The aim of this study was to investigate the effect of Mori Fructus extract in male ob/ob mouse with severe obesity, hyperinsulinemia, hyperglycemia, hyperlipidemia. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57BL/6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Mori Fructus extract per 1 kg of body weight for 14 days. The effects of Mori Fructus extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Western blotting was performed using anti-AGE, anti-RAGE respectively. Mori Fructus extract lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Mori Fructus extract treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Mori Fructus extract-administered groups. Mori Fructus extract inhibited the generation of ROS in the kidney. MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Mori Fructus extract-administered groups. Mori Fructus extract inhibited the expression of AGE, RAGE in the ob/ob mice. The results suggested that Mori Fructus exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging the ROS, decreasing the MDA+HAE level, increasing the GSH level and inhibiting the expression of AGE, RAGE in the ob/ob mice.

Effects of the Prunus persica Batsch var. davidiana Max. Extract on the Blood Glucose and Serum Lipid Components in Streptozotocin-Induced Diabetic Rats (돌복숭아(Prunus persica Batsch var. davidiana Max.) 추출액이 Streptozotocin 유발 당뇨성 흰쥐의 혈당 및 지질 성분에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Prunus persica Batsch var. davidiana Max. extract on the improvement of the blood glucose, lipid compositions in the serum of streptozotocin(STZ)-induced diabetic rats fed the experimental diets for 5 weeks. Concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride(TG) and phospholipid (PL) in serum were significantly higher in the STZ (55 mg/kg B.W.)-induced diabetic group (group 2) and STZ(I.P.)+Prunus persica 5.0 g% extract group(group 3) than those in the control group (group 1, basal diet + water). But the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG and PL in serum were remakably lower in the group 3 than those in the group 2. In the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Prunus persica 5.0 g% extract administration group(group 3) were higher percentage than in the group 2. The activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), lactate dehydrogenase (LDH) and alkaline phosphatase(ALP) in serum were rather lower in the Prunus persica 5.0 g% extract administration group(group 3) than in the STZ- induced diabetic group (group 2). From the above results, it was suggested that the Prunus persica Batsch var. davidiana Max. were effective on the improvement of the blood glucose, lipid compositions in serum of STZ-induced diabetic rats. Moreover, in Prunus persica Batsch var. davidiana Max. was effective therapeutic regimen for the control of metabolic derangements in adult disease.
