Modification of Functional Properties of Soy Protein Isolate by Proteolytic Enzymes (단백분해효소에 의한 대두단백의 기능적 특성변화)

  • The effects of enzymatic modification with pepsin and actinidin was studied on molecular weight distributions and functional properties of hydrolysates from soy protein isolate (SPI) differing in degree of hydrolysis. The hydrolyzed SPI by pepsin showed 41.5% degree of hydrolysis after 5 min, and maximum hydrolysis was obtained after 2 hours. Actinidin hydrolyzed SPI 26.71% degree after 1 hour. On SDS-PAGE, native SPI showed 9 distinguishable bands on SDS-PAGE gel. Pepsin treated SPI showed one broad band in the lower part of gel. This band was shifted further to the bottom of the gel and became faint as hydrolysis time increased. While actinidin treated SPI showed different SDS-PAGE pattern from pepsin. However PAGE patterns were similar with pepsin and actinidin treated groups. With pepsin treatment, solubility of SPI distinctively increased around isoelectric point(pI). Emulsifying activity (EA) and emulsifying stability (ES) showed marked increase over pH range of $3.0{\sim}8.0$. 5 min modified group had most excellent foam expansion (FE). Foam stability (FS) was increased as pepsin treatment time increased at pI. With actinidin treatment, solubility was increased. 60 min modified SPI had the most effective EA at pH 4.5. However ES was not effected by actinidin treatment. 5 min modified group was most effect in FE. FS was higher at alkaline pH.

Sensory and Quality Evaluation of Aseptic-Packaged Cooked Rice by Cultivar (벼 품종에 따른 무균포장밥의 식미특성 및 품질 변이)

  • We carried out this experiment on the purpose to investigate the quality properties of aseptic-packaged cooked rice by cultivars in 2006. Brown rice was milled for white rice to 89.6% weight of it. Based on cluster analysis of acceptance of sensory evaluation, eleven rice cultivars of 29 cultivars had superior palatability. Whiteness(r=0.42, p<5%), lightness(r=0.39, p<5%), Toyo value(r=0.35, p<10%), and moisture content(r=0.33, p<10%) of milled rice were correlated positively with acceptability. Protein content(r=-0.40, p<5%), expansion rate(r=-0.68, p<1%) and water absorption rate(r=-0.42, p<5%) of milled rice, and yellowness(r=-0.45, p<5%) of aseptic-packaged cooked rice were negatively correlated. In sensory evaluation, correlation coefficients of taste and texture with acceptability were higher than those of appearance and flavor. During storage, hardness and cohesiveness of aseptic-packaged cooked rice before reheating were increased and decreased, but those after reheating had no difference based on storage period and cultivar. On the contrary, whiteness of reheated aseptic-packaged cooked rice with no distinct differences before reheating was decreased during storage. There were no significant differences of texture property, color characteristics and moisture contents by cultivar during storage.

  • Smart work is deeply embedded in our contemporary lives. This is called Smart Life. The expansion of communication, caused by smart life is utilized for the political purposes of politicians, political candidates, and political policies. Chiefly, smart life includes improvement in awareness, personal relations, and policy advocacy. Socially, smart life leads the proliferation of issues and civil movement. In particular, smart life has a significant effect on culture. In the case of K-POP, it is disseminated going beyond space time. It enhances economic value added by joining and sharing at the same time. Smart life has numerous advantages such as improving the quality of life, but it might cause inconvenience depending on users because there are differences in acceptance according to generations. Due to the heavy weight on the device, there are some side-effects such as lacking in human thinking, disclosure in private life, lacking in basic securities, increase in information addicts who are anxious when they do not get on the Internet or SNS, and interference with work. However, this inconvenience can change into convenience when users are accustomed to it. Like this, smart life influences politics, economy, society, and culture a lot. Working environment of smart life is a new paradigm, which can generate high performance through working in an exciting and bright atmosphere with easy economy and putting value on results.

Analysis of Physical Properties and Mechanical Energy Input of Cornmeal Extrudates Fortified with Dairy Products by Carbon Dioxide Injection (탄산가스 주입에 의한 유제품 강화 옥수수 압출성형물의 성질과 기계적 에너지 투입량의 분석)

  • Selected physical properties of cornmeal extrudates fortified with dairy products and extrusion process by $CO_2$ gas injection were analysed. Dairy products including whole milk powder, whey protein concentrate non-fat dry milk, and sodium caseinate were tested at the addition of 10% and 20%, based on cornmeal weight. $CO_2$ gas was injected to the barrel at the pressure of 0.7 and 1.4 MPa. Specific mechanical energy (SME) input was decreased by the addition of dairy products. Sodium caseinate had a little effect on decreasing the SME input, however whole milk powder tremendously reduced SME input when the concentration increased. An increase in milk product content resulted in increasing the piece density at the injection pressure of 0.7 MPa. At both 10% and 20% milk product content, the piece density was lowest at the injection pressure of 0.7 MPa. The sectional expansion index was highest at the injection pressure of 0.7 MPa. However, the specific length was constantly increased with the increase in $CO_2$ injection pressure. Water absorption index was decreased and water solubility index was increased by the addition of milk products. The injection pressure of $CO_2$ was optimum at 0.7 MPa. The addition of whole milk powder limited to puff the melt, but the other milk products tested resulted in puffing with $CO_2$ injection to 1.4 MPa.

Effect of Ionizing Radiation of Physiological Characteristics of Fresh Mushrooms (Agaricus bisporus L.) (양송이 버섯의 생리적 특성에 대한 감마선 조사 영향)

  • Fresh mushroom (Agaricus bisporus L.) was irradiated (0, 1, 2, 3kGy) and kept for 20 days at 9{\pm}1^{\circ}C\;and\;80{\pm}7%$ RH. Parameters of qualities were investigated on the physical and physiological characteristics. The pileus and stipe on nonirradiated mushroom were expanded and elongated from the 3rd day of storage, there by losing the acceptability as edible samples. After 5 days of storage, 2 to 3kGy of gamma irradiation were especially effective for controllong natural maturation and senescence of fresh mushrooms and so irradiated mushrooms were acceptable more than 20days storage. The texture of irradiated samples was superior to that of nonirradiated samples, even though softening of the tissue occurred during storage. Weight loss was greatest in the nonirradiated sample due to evaporation from an increased surface area resulting from expansion and ripending, which were retarded in the 2 to 3kGy irradiated samples after 5days of storage. These results suggest that the irradiation dose of 2 to 3kGy is apparently effective to extend the shelf life of fresh mushrooms stored at the above-mentioned condition.

Compound K attenuates hyperglycemia by enhancing glucagon-like peptide-1 secretion through activating TGR5 via the remodeling of gut microbiota and bile acid metabolism

  • Background: Incretin impairment, characterized by insufficient secretion of L-cell-derived glucagon-like peptide-1 (GLP-1), is a defining step of type 2 diabetes mellitus (T2DM). Ginsenoside compound K (CK) can stimulate GLP-1 secretion; however, the potential mechanism underlying this effect has not been established. Methods: CK (40 mg/kg) was administered orally to male db/db mice for 4 weeks. The body weight, oral glucose tolerance, GLP-1 secretion, gut microbiota sequencing, bile acid (BA) profiles, and BA synthesis markers of each subject were then analyzed. Moreover, TGR5 expression was evaluated by immunoblotting and immunofluorescence, and L-cell lineage markers involved in L-cell abundance were analyzed. Results: CK ameliorated obesity and impaired glucose tolerance in db/db mice by altering the gut microbiota, especially Ruminococcaceae family, and this changed microbe was positively correlated with secondary BA synthesis. Additionally, CK treatment resulted in the up-regulation of CYP7B1 and CYP27A1 and the down-regulation of CYP8B1, thereby shifting BA biosynthesis from the classical pathway to the alternative pathway. CK altered the BA pool by mainly increasing LCA and DCA. Furthermore, CK induced L-cell number expansion leading to enhanced GLP-1 release through TGR5 activation. These increases were supported by the upregulation of genes governing GLP-1 secretion and L-cell differentiation. Conclusions: The results indicate that CK improves glucose homeostasis by increasing L-cell numbers, which enhances GLP-1 release through a mechanism partially mediated by the gut microbiota-BA-TGR5 pathway. Therefore, that therapeutic attempts with CK might be useful for patients with T2DM.

  • A significant cause of fall or overturning accidents in the construction industry, including landscaping construction and management, is work at heights using portable ladders. Portable ladders are classified as A-type or triangular support ladders depending on the number of supporting leg and support conditions. The tripod mobile ladder, which supports itself with only three supporting legs, is unstable and more prone to overturning compared to the A type ladders. Therefore, using the specifications of the tripod mobile ladder and the stability regulations of EN131-Part 7, overturning and resistance moment calculation formulas were derived for all directions in which overturning could occur. The moments calculated using these equations, and the overturning stability in each direction were evaluated. According to the calculation results, although there are differences depending on the direction, most are unstable for overturning at 8 or more steps. Based on these results, this study proposed measures to increase the moment of resistance by changing the weight, depth, and width, and using outriggers to ensure stability against the overturning of ladder. However, when changing the specifications of these measures, the size increases are excessive and the applicability is insufficient. On the other hand, outriggers are an applicable measure as they can ensure stability against overturning with only a minimum expansion length.

The Effect of Silica binder content ans Sintering condition on the Strength of Zircon-based Shell Mold (실리카 바인더 함량과 소결조건이 지르콘계 주형의 강도에 미치는 영향)

  • The effect of silica binder content on the mechanical properties of zircon shell mold was investigated. Content of binder silica sol to refractory powder in weight[$R_W$] was adjusted from 0.18 to 0.43. Sintering of the shell mold was carried out in the temperature range of $871^{\circ}C$ to $1400^{\circ}C$. Green strength of the shell mold at room temperature increased with increasing $R_W$ and sintering temperature up to $1300^{\circ}C$. However, the mold with $R_W$ of 0.43 that sintered at $1400^{\circ}C$ for 3 hours showed relatively low strength and large level of porosity. The mechanical behavior of the shells is supposed to attributed to the difference in thermal expansion coefficient between refractory powder and binder silica. The optimum value of $R_W$ for zircon-based shell molds was found to be 0.33.

Structures and Double Layer Performances of Carbons Pyrolized from Carbon Oxides (산화탄소로부터 열분해한 탄소의 구조 및 전기이중층 거동)

  • Structural features and electrochemical performances of cokes pyrolized from oxidized cokes were examined, and compared with KOH-activated coke. Needle cokes ($d_{002}=3.5{\AA} $), having a graphene layer structure, were changed to a single phase of graphite oxide after oxidation treatment with an acidic solution having an $NaCLO_3$/needle coke composition ratio of above 7.5, and the inter-layer distance of the oxidized coke was expanded to $6.9{\AA} $ with increasing oxygen content. After heating at $200^{\circ}C$, the oxidized coke was pyrolized to the graphene layer structure with inter-layer distance of $3.6{\AA} $. However, the change of the inter-layer distance of the needle coke was not observed in the KOH activation process. On the other hand, an intercalation of electrolyte ions into the pyrolized coke, observed at first charge, occurred at 1.0 V, in which the value was lower than that of KOH-activation coke. The cell capacitor using pyrolized coke exhibited a lower internal resistance of $0.57{\Omega}$ in 1 kHz, and a larger capacitance per weight and volume of 30.3 F/g and 26.9 F/ml at the two-electrode system in the potential range 0~2.5 V than those of the cell capacitor using KOH-activation of coke. This better electrochemical performance may be associated with structure defects in the graphene layer derived from the process of the inter-layer expansion and shrinkage.

Evaluation of Shear Strength of Unreinforced Masonry Walls Retrofitted by Fiber Reinforced Polymer Sheet (FRP로 보강한 비보강 조적 벽체의 전단강도 산정)

  • Unreinforced masonry buildings represent a significant portion of the existing and historical buildings around the world. Recent earthquakes have shown the need for seismic retrofitting for these types of buildings. Various types of retrofitting materials (i.e., shotcrete, ECC and Fiber Reinforced Polymer sheets (FRPs)) for unreinforced masonry buildings (URM) have been developed. Engineers prefer to use FRPs, because these materials enhance the shear strength of the wall without expansion of wall sectional area and adding weight to the total structure. However, the complexity of the mechanical behavior of the masonry wall and the lack of experimental data from walls retrofitted by FRPs may cause problems for engineers to determine an appropriate retrofitting level. This paper investigate in-plane behavior of URM and retrofitted masonry walls using two different types of FRP materials to determine and provide information for the retrofitting effect of FRPs on masonry shear walls. Specimens were designed to idealize the wall of a low-rise apartment which was built in 1970s in Korea with no seismic reinforcements with an aspect ratio of 1. Retrofitting materials were carbon FRP and Hybrid sheets which have different elastic modulus and ultimate strain capacities. Consequently, this study evaluated the structural capacity of masonry shear walls and the retrofitting effect of an FRP sheet for in-plane behavior. Also, the results were compared to the results obtained from the evaluation method for a reinforced concrete beam retrofitted with FRPs.
