The Anti-complementary Activity of Exo-polymers Produced from Submerged Mycelial Cultures of Higher Fungi with Particular Reference to Cordyceps militaris

  • The anti-complementary activity (immuno-stimulating activity) was tested for the exo-polymers (extra-cellular polymer) produced from submerged mycelial cultures of 21 types of higher fungi. Anti-complementary activity of the exo-polymer from Cordyceps militaris showed the highest (69.0%) followed by Pleurotus ostreatus (63.7%) and Trametes suaveolens (61.4%). The mycelial growth rate and biomass doubling time of C. militaris in a 5 I air-lift fermenter were 0.0255 $h^{-1}$ and 27.2 h, respectively. The yield of the exo-polymer produced from the culture broth of C. militaris was 2.95 mg of dry weight/ml of culture broth. Sugar and amino acid compositions of this exo-polymer were analyzed.

  • New antitumor active polymers, poly(methacryloyl-2-oxy-1,2,3-propanetricarboxylic acid-co-exo-3,6-epoxy-l,2,3,6-tetrahydrophthalic acid) [poly(MTCA-co-ETAc)], poly(methacryloyl-2-oxy-l,2,3-propanetricarboxylic acid-co-hydrogen ethyl-exo-3,6-epoxy-l,2,3,6-tetrahydrophthalate) [poly(MTCA-co-HEET)], and poly(methacryloyl-2-oxy-l,2,3-propanetricarboxylic acid-co-a-ethoxy-exo-3,6-epoxy-1,2,3,6-tetrahydrophthaloyl-5-fluorouracil) [poly(MTCA-co-EETFU)] were synthesized and characterized. Their antitumor activity, inhibition of DNA replication and antiangiogenesis were examined. The structures of the polymers were identified by FT-IR, $^1H$ and $^{13}C$-NMR spectroscopy. The number average molecular weights of the fractionated polymers determined by GPC ranged from 9,400 to 14,900, and polydispersity indices were less than 1.7. The in vitro cytotoxicity of these polymers was determined and their antitumor activity was evaluated. The $IC_{50}$ values (the drug concentration at inhibition of 50% tumor growth) indicated that the synthesized polymers were much better inhibitors of cancer cells and showed lower cytotoxicity than the free 5-FU. The in vivo antitumor activity of the conjugates was examined using mice bearing the sarcoma 180 tumor cell line. The life spans (TIC) of the mice treated with the conjugates were higher than those treated with the free 5-FU. In addition, the synthesized conjugates showed excellent antiangiogenic activity based on an embryo chorioallantoic membrane assay.

A Preliminary Study on the Hypoglycemic Effect of the Exo-Polymers Produced by Five Different Medicinal Mushrooms

  • The Hypoglycemic effect of exo-polymers (EPs) produced from submerged mycelial cultures of five types of mushrooms on streptozotocin (STZ)-induced diabetic rats were investigated in this study. The five experimental groups were fed with EPs (50 mg/kg body weight) for 7 days. Significant reduction in plasma glucose, total cholesterol (TC), and triglyceride (TG) levels were observed in rats fed with Lentinus edodes and Cordyceps militaris EPs. Plasma glucose and TC were also reduced by administration of Phellinus linteus EPs, but the TG level was not changed significantly. The EPs of three mushroom species also demonstrated a marked reduction in the level of plasma glutamate-pyruvate transminase (GPT). The result proves the hypoglycemic activity of EPs of three fungal group in STZ-induced diabetic rats and indicates their potential in the control of diabetes mellitus.

  • Hypolipidemic effect of exo-polymers(EPs) form submerged mycelial culture of Cordyceps militaris was investigated in male Sprague-Dawley rats. For a dose-dependent study, EPs were administered at the level of 50-100 mg/kg body weight (BW) and the data was compared with the saline administered control group. A significant reduction of both the plasma total cholesterol (TC) and triglyceride (TG) was registered under the influence of EPs. It reduced the lowe density lipoprotein (LDL) cholesterol level as much as 40.5%. Levels of high density lipoprotein (HDL) choloesterol did not vary significantly within the different experimental groups, but the HDL: TC ratio showed consistently a high value with the increasing dose. The effects of cultural conditions (pH and temperature) in mycelial growth and EPs production were studied. Both the biomass and EPs were produced in a wide range of pH and temperature.

  • Palladium(II) 촉매를 이용한 norobornene carboxylic acid estsers의 중합 시 단량체의 알킬 작용기의 종류, endo/exo 비율이 촉매의 활성도 및 중합 특성에 미치는 영향을 조사하였다. Norbornene esters 단량체는 5-norborene-2-carboxylic acid와 다양한 알코올을 반응시켜 합성하였다. 중합 촉매로는 di-$\mu$-chloro-bis(-methoxybicyclo[2,2,1]-hept-2-ene)palladium(II)(DCBMP)를 합성하여 사용하였고, 짝음이온으로 실버 헥사플루오로안티모네이트($AgSbF_6$)를 이용하였다. 고분자의 분석을 위해 젤 투과 크로마토그래피(gel permeation chromatography, GPC), 열 중량 분석법(thermogravimetric analysis, TGA), 시차주사 열량측정법(differential scanning calorimetry, DSC), 화학구조 분석을 위해 $^1H$ NMR spectroscopy를 이용하였다. 분자량 분석 결과 모든 작용기의 경우 endo-이성체의 비율이 exo-이성체의 비율보다 높을 경우 촉매의 구조적 방해로 인하여 반응성이 감소됨을 보였다. 또한 단량체와 촉매의 비율이 중합 거동에 미치는 영향을 조사하기 위해 단량체와 촉매의 몰비율을 100:1, 200:1, 300:1로 변화시켜 실험을 진행하였으며, 이 때 exo-norbornene carboxylic acid octyl ester의 경우 300:1 촉매비에서 필름형성이 가능한 높은 분자량($M_n=27500g/mol$)의 고분자를 합성할 수 있었다.

Hypolipidemic Effect of Exo-Polymer Produced in Submerged Mycelial Culture of Five Different Mushrooms

  • The hypolipidemic effect of exe-polymer produced in submerged mycelial culture of Hericium erinaceus (HE), Auricularia auricula-judue (AA), Flammulina veluripes (FV), Phellinus pini (PP), and Grifola frondosa (GF) was investigated in dietary-induced hyperlipidemic rats. The animals were administered with exe-polymers at the level of 100 mg/kg body weight daily for four weeks. Hypolipidemic effect was achieved in all the experimental groups, however, HE exo-polymer proved to be the most potent one, which significantly reduced the plasma triglyceride ($28.9\%$), total cholesterol ($29.7\%$), low-density lipoprotein (LDL) cholesterol ($39.6\%$), phospholipid ($16.0\%$), and liver total cholesterol ($28.9\%$) level, when compared to the saline administered (control) group. The results of the present investigation strongly demonstrate the potential of HE exe-polymer in combating hyperlipidemia in the experimental animals.

Synthesis, Antitumor Activity and Release Rate of Polymers Containing Anionic Group and 5-Fluorouracil

  • Poly(exo-3,6-epoxy-1,2,3,6-tetrahydrophthalic anhydride)s [poly(ETA)s] and poly($\alpha$-ethoxy-exo-3,6-epoxy-1,2,3,6-tetrahydrophthaloyl-5-fluorouracil)s [poly(EETFU)s ] with various average molecular weights were prepared by photopolymerizations. The number average molecular weights of the fractionated poly(ETA)s and poly(EETFU)s determined by GPC were in the range of 3,600∼21,000 and 3,600-33,400, respectively. The release rate of 5-FU from poly(EETFU) decreased with increasing average molecular weight. The in vitro cytotoxicity of poly(ETA) against a normal cell line was lower than that of 5-fluorouracil(5-FU), The in vivo antitumor activities of the synthesized samples at dosage of 0.8 mg/kg against mice bearing sarcoma 180 tumor cell line decreased in the following order: poly(EETFU) > poly(ETA) > EETFU > ETA > 5-FU. The antiangiogenic activities of the poly(ETA)s were better than those of 5-FU.

Hydrolysis of Arabinoxylo-oligosaccharides by α-ʟ-Arabinofuranosidases and β-ᴅ-Xylosidase from Bifidobacterium dentium

  • Two α-ʟ-arabinofuranosidases (BfdABF1 and BfdABF3) and a β-ᴅ-xylosidase (BfdXYL2) genes were cloned from Bifidobacterium dentium ATCC 27679, and functionally expressed in E. coli BL21(DE3). BfdABF1 showed the highest activity in 50 mM sodium acetate buffer at pH 5.0 and 25℃. This exo-enzyme could hydrolyze p-nitrophenyl arabinofuranoside, arabino-oligosaccharides (AOS), arabinoxylo-oligosaccharides (AXOS) such as 32-α-ʟ-arabinofuranosyl-xylobiose (A3X), and 23-α-ʟ-arabinofuranosyl-xylotriose (A2XX), whereas hardly hydrolyzed polymeric substrates such as debranched arabinan and arabinoxylans. BfdABF1 is a typical exo-ABF with the higher specific activity on the oligomeric substrates than the polymers. It prefers to α-(1,2)-ʟ-arabinofuranosidic linkages compared to α-(1,3)-linkages. Especially, BfdABF1 could slowly hydrolyze 23,33-di-α-ʟ-arabinofuranosyl-xylotriose (A2+3XX). Meanwhile, BfdABF3 showed the highest activity in sodium acetate at pH 6.0 and 50℃, and it has the exclusively high activities on AXOS such as A3X and A2XX. BfdABF3 mainly catalyzes the removal of ʟ-arabinose side chains from various AXOS. BfdXYL2 exhibited the highest activity in sodium citrate at pH 5.0 and 55℃, and it specifically hydrolyzed p-nitrophenyl xylopyranoside and xylo-oligosaccharides (XOS). Also, BfdXYL2 could slowly hydrolyze AOS and AXOS such as A3X. Based on the detailed hydrolytic modes of action of three exo-hydrolases (BfdABF1, BfdABF3, and BfdXYL2) from Bf. dentium, their probable roles in the hemiceullose-utilization system of Bf. dentium are proposed in the present study. These intracellular exo-hydrolases can synergistically produce ʟ-arabinose and ᴅ-xylose from various AOS, XOS, and AXOS.

Anion-Dependent Exocyclic Mercury(II) Coordination Polymers of Bis-dithiamacrocycle

  • Synthesis and structural characterization of mercury(II) halides and perchlorate complexes (1-4) of bis-$OS_2$-macrocycle (L) are reported. L reacts with mercury(II) chloride and bromide to yield an isostructural 2D coordination polymers with type $[Hg(L)X_2]_n$ (1: X = Cl and 2: X = Br). In 1, each Hg atom which lies outside the cavity is six-coordinate with a distorted octahedral geometry, being bound to four adjacent ligands via monodentate Hg-S bonds and two remaining sites are occupied by two terminal chlorido ligands to form a fishnet-like 2D structure. When reacting with mercury(II) iodide, L afforded a 1D coordination polymer $\{[Hg_2(L)I_4]{\cdot}CHCl_3\}_n$ (3) in which each exocyclic Hg atom is four-coordinate, being bound to two sulfur donors from different ligands doubly bridging the ligand molecules in a head-to-tail mode. The coordination sphere in 3 is completed by two iodo terminal ligands, adopting a distorted tetrahedral geometry. On reacting with mercury(II) perchlorate, L forms solvent-coordinated 1D coordination polymer $\{[Hg_2(L)(DMF)_6](ClO_4)_4{\cdot}2DMF\}_n$ (4) instead of the anion-coordination. In 4, the Hg atom is five-coordinate, being bound to two sulfur donors from two different ligands doubly bridging the ligand molecules in a side-by-side mode to form a ribbon-like 1D structure. The three remaining coordination sites in 4 are completed by three DMF molecules in a monodentate manner. Consequently, the different structures and connectivity patterns for the observed exocyclic coordination polymers depending on the anions used are influenced not only by the coordination ability of the anions but also by anion sizes.

자몽 추출물로부터 분리된 항균성 Chitinase의 특성 (Characterization of an antimicrobial Chitinase Purified from the Grapefruit Extract)

  • An antimicrobial chitinase was purified from grapefruit extract and its properties were characterized. The chitinase was purified with a single step chromatography on regenerated chitin affinity gel column. The molecular weight of the purified chitinase was 29 kDa. The grapefruit extract contained the chitinase protein more than 50% of its total soluble proteins measured by coomassie stained protein bands. When the purified chitinase was incubated with polymers of N-acetylglucosamine (NAG), such as mycelia of Fusarium oxysproum and swollen chitin, they were degraded to oligosaccharides, and the oligosaccharides were then further hydrolyzed by the same enzyme to monomer and dimer of NAG. This result suggests that the chitinase contained both endo- and exo- chitinase activities. The chitinase was stable to heat and pH treatment; its activity was not diminished by the heat treatment upto 7$0^{\circ}C$ for 1 hr, and it showed a pH stability in the range of pH 4.0 to 12.0.
