  • An analytical method the determination of benzo(a)pyrene (BaP) and its hydroxylated metabolites, 1-hydroxybenzo(a)pyrene (1-OHBaP), 3-hydroxybenzo(a)pyrene (3-OHBaP), benzo(a)pyrene-4,5-dihydrodiol (4,5-diolBaP) and benzo(a)pyrene-7,8-dihydrodiol (7,8-diolBaP), in rat urine and plasma has been developed by HPLC/FLD and GC/MS. The derivatization with alkyl iodide was employed to improve the resolution and the detection of two mono hydroxylated metabolites, 1-OHBaP and 3-OHBaP, in LC and GC. BaP and its four metabolites in spiked urine were successfully separated by gradient elution on reverse phase ODS $C_{18}$ column (4.6 mm I.D., 100 mm length, particle size 5 ㎛) using a binary mixture of MeOH/H₂O (85/15, v/v) as mobile phase after ethylation at 90 ℃ for 10 min. The extraction recoveries of BaP and its metabolites in spiked samples with liquid-liquid extraction, which was better than solid phase extraction, were in the range of 90.3- 101.6% in n-hexane for urine and 95.7-106.3% in acetone for plasma, respectively. The calibration curves has shown good linearity with the correlation coefficients (R²) varying from 0.992 to 1.000 for urine and from 0.996 to 1.000 for plasma, respectively. The detection limits of all analytes were obtained in the range of 0.01-0.1 ng/mL for urine and 0.1-0.4 ng/mL for plasma, respectively. The metabolites of BaP were excreted as mono hydroxy and dihydrodiol forms after intraperitoneal injection of 20 mg/kg of BaP to rats. The total amounts of BaP and four metabolites excreted in dosed rat urine were 3.79 ng over the 0-96 hr period from adminstration and the excretional recovery was less than 0.065% of the injection amounts of BaP. The proposed method was successfully applied to the determination of BaP and its hydroxylated metabolites in rat urine and plasma for the pharmacokinetic studies.

  • That different mechanisms are involved in the secretory processes by the liver and the kidney of various dyes has been indicated by Sporter (1959), Kim and Hong (1963). Andrews (1958). suggested that a striking difference in the dye-secretory mechanism existed even in the same organ from species to species. Hence, the attempt has been made to study in the rabbit the secretory processes by the live. and the kidney of either phenol red (PSP), bromsulfalein (BSP) or green in the presence of Na-acetate, Na-taurocholate, P-Aminohippurate (PAH) or Benemid. In 37 rabbits, weighing about 2kg., anesthetized with ether, a dye was administered in such 8 manner that the plasma concentration was kept at a relatively constant level throughout the whole experimental period. Hepatic bile sad urine samples were quantitatively collected through the canulae which were previously inserted into the common bile duct (with the cystic duct ligated) and the urinary bladder, respectively, while arterial samples were taken from a femoral artery. After 50 min from the onset of dye administration, these samples were obtained every 10 mit for a period of 40 min. This was followed by the administration of either Na-acetate, Na-tauro-cholate, PAH or Benemid with a repetition of the same sample collecting procedures just stated. The results may be summarized as follows: 1) Na·acetate augmented urinary clearance of PSP by nearly 300 per cent, but lowered urinary BSP clearance by about 50 per cent. It enhanced biliary BSP clearance by 40% and had no effect on biliary psp clearance. 2) Na-taurocholate lowered biliary and urinary clearance of PSP by 10 per cent and 30 per cent respectively, and had no effect on both biliary and urinary clearance of BSP. 3) PAH lowered both biliary and urinary excretion of BSP and PSP, while it lowered the biliary excretion of indocyanine green which was excreted only in the bile. 4) Benemid suppressed BSP excretion by the liver and the kidney. 5) raper chromatographic analysis of PSP and of BSP in the bile and urine samples gave the following results: a) PSP Ivas excreted in the urine and bile only in free forms, and no modification in the excretory pattern was brought about by Na-taurocholate. b) BSP was excreted in the urine in 4 different conjugated froms and in the bile in both 3 different conjugated forms and in a free form. Na-taurocholate modified the excretory pattern of the urinary BSP.

  • The existence of oxytocin in the pituitary gland of chicken has been ascertained, but its physiological roles are still obscure. In the study the action of oxytocin on renal function of the chicken was investigated during water diuresis, utilizing clearance and the Sperber technique. The results obtained are summarized as follows: Oxytocin, like in many species of mammals, elicited a profound diuretic response in the chicken. Urine flow, excretion of electrolytes, as well as glomerular filteration rate increased, with intravenous infusion of $3{\sim}10\;m{\mu}/kg/min$. Oxtocin, infused into the renal portal circulation via hindleg vein in a dose of $3{\sim}13\;m{\mu}/kg/min$. elicited marked increase in urine flow, glomerular filteration rate and sodium excreted in the urine. The diuretic effect was more pronounced in the infused side. It is suggested that diuretic response to oxytocin in the chicken results from dual action of oxytocin: increase of GFR and inhibition of sodium reabsorption on the renal tubule. The possibility that oxytocin might act through some endogenous substances could be ruled out.

  • Human chorionic gonadotropin hormone(H.C.G.) is secreted from the villus tissues of the placenta and excreted in lange amount into the urine. Its isolation is chiefly made from the urine of a pregnant woman. Recently, Matsushima attempted isolation of H.C.G. directly from the placenta itself. In order to prepare H.C.G. from human placenta, general method of extractiag and purifying proteins was applied. Its way was as follow: Crude H.C.G. was extracted from placenta with pH 9.0 and pH 5.0 aqua ammonia, and purified with pH 8.0 ammonia and 50% ethanol at pH 4.8. The purified H.C.G. showed two moving bands on the anode by paper electrophoresis. On the other hand, the H.C.G. from pregnancy urine (Standard. Pharm. Co.) showed same two bands but their moving ratio were different. The purified H.C.G. showed gonadotropin effect when it was injected young fomale rats 40r/cc per day for 5 days and weighted the increased ovary weight.

  • Polyporus would used as diuretics. Then, for the purpose of experimentally certifying the above mention, the effect on the renal function of dog was investigated, utilizing clearance technique. Water and alcohol extracts, when injected intravenously, produced significant increases of urinary sodium and potassium, osmolar and free water clearances, and urine flow, while glomerular filtration rate and renal plasma flow remained unchanged. During diuresis produced by furosemide, addition of water extract reduced the action of furosemide and markedly renal plasma flow. It would appear that these compounds are capable of action by a different mechanism or a different site. water extract, when infused directly into a renal artery, reduced the urine flow of experimental kidney as well as renal plasma flow, and the contralateral kidney exhibited diuresis, whereas amounts of sodium and potassium excreted in urine increased on both kidney. It is surmised from those observations that Polyporus induces diuresis by inhibition the reabsorptive mechanism of renal tubules through some endogeneous humoral substances, in addition, directly reduces the renal plasma flow.

  • The metabolism of carbinoxamine, 2-[(4-chlorophenyl)-2-pyridinyl-methoxy]-N, N-dimethylethaneamine, was studied in adult male volunteers after an oral dose of 15 mg. Solvent extracts of urine obtained with or without enzyme hydrolysis were analyzed by gas chromatography-mass spectrometry after derivatization with MSTFA/TMSCl (N-methyl-N-trimethylsilyltrifluoroacetamide/trimethyl chlorosilane). The structures of metabolites were determined based on the electron impact (EI) and chemical ionization (CI) mass spectra. Nonconjugated metabolites identified in the urine were carbinoxamine, nor-carbinoxamine, and bits-nor-carbinoxamine. Parent drug, nor-carbinoxamine, and bits-nor-carbinoxamine were also detected as conjugated forms. These metabolites observed in human urine were different from those previously reported in the rat. Urinary excretions of carbinoxamine were reached to maxima in 4 hours after drug administration with 4.9%-8.1% and 2.5-4.2% of the dose excreted during 24 h as carbinoxamine and its glucuronide, respectively.

  • The metabolism of dromostanolone (2α-methyl-5α- androstan-17β-ol-3-one) was studied in three adult volunteers after oral dose of 20 mg. Solvent extracts of urine obtained after enzyme hydrolysis were derivatized with MSTFA/TMCS and MSTFA/TMIS. The structures of intact drug and its metabolites were determined by gas chromatography/mass spectrometry (GC/MS) in electron impact (EI) mode. The major metabolite (2α-methyl-5α- androstan-3α-ol-17-one), its 3β-epimer, parent compound, and several hydroxylated metabolites including intact drug were detected by comparing total ion chromatograms of control urine with that of the administered sample. Two epimers of 2α-methyl-5α- androstan-3,17β-diol were detected using selected ion monitoring. The maximum excretion of dromostanolone and 2α-methyl-5α- androstan-3α-ol-17-one was reached in 6.2-15 hr. The half-life of intact dromostanolone was 5.3 hr. About 3.0% of the administered amount was found to be excreted within 95 hr as unchanged form.

  • In summary, progesterone is probably the only naturallly occurring progestational agent of any significance. Small amounts may be synthesized by the cells of the follicle in the provulatory swelling phase; however, the major production is by the corpus luteum cells of the ovary during the luteal phase of the cycle. It is constantly produced in small amounts by the adrenal gland and by the testis in the male. In the adrenal and the testis, it probably serves as the precursor for corticoids and androgens. It is transported in the blood by a specific binding protein and metabolized and conjugated in the liver into sodium pregnanediol glucuronide which also circulates in the blood. Approximately 20% is excreted in the urine as sodium pregnanediol glucronide; pregnanolone represents a minor metabolic product. The pregnanediol which is excreted in the bile is enzymically hydrolyzed by the gut so that the pregnanediol recovered in the feces is in the free form.

  • This study was undertaken to investigate the effects of age and of the dietary protein sources & levels on urinary calcium excretion and bone metabolism in the rats. Two experiments were conducted . In experiment U, 6& 20 weeks old rats were fed 8 & 36%, casein & ffish protein diet for 5 weeks . In experiment II, 16& 52 weeks old rats were fed 40 % protein diets, protein sources were casein, fish and gluten. High-protein diet group excreted more caldium in urine than low-protein group. Urinary calcium excretion was affected by the sources of protein ; gluten group excreted more Ca, followed by fish & casein group. Total bone Ca & Ca proportion in ash were higher in 20 weeks old rats than 6 weeks old rats, but 16 & 52 weeks old rats showed no differences. bone composition showed that water proportion was high and ash proportion was low in 6 weeks old rats than in 20 weeks old rats. However, these tendencies were not observed between 16 weeks and 52 weeks old rats. And bone composition was affected by protein sources ; higher ash proportions were noted in one order of casein, fish, and gluten water proportion were lower in one same order.
