New Record of Two Apokeronopsis Species (Ciliophora: Urostylida: Pseudokeronopsidae) from Korea

  • The morphology of the two marine hypotrichous ciliates Apokeronopsis bergeri and A. ovalis, isolated from the Yellow Sea, Korea, are described based on live and protargol-impregnated specimens. It is the first time that these species have been recorded in Korea. In addition, the small subunit ribosomal RNA gene was sequenced for comparison with the public database. The genus Apokeronopsis has recently been established in the family Pseudokeronopsidae, and the two congeners of the Korean population share the following characteristics: one row of one or more buccal cirri; usually two frontoterminal cirri; midventral complex composed of two distinctly separated rows; one left and one right marginal row; number of transverse cirri, more than eight; absence of caudal cirri; two types of cortical granules. Apokeronopsis bergeri differs from A. ovalis primarily in body shape (fusiform vs. oval form), size (usually $260{\times}80{\mu}m$ vs. $160{\times}55{\mu}m$), type II cortical granules (oval vs. round shape; yellow-green vs. mostly colourless and only a few yellow-green in colour), and morphometric data (75-106 vs. 53-70 in adoral membranelles; 37-47 vs. 24-36 in frontal cirri; 9-15 vs. 1-2 in buccal cirri), as well as molecular data (2.87% of pairwise distance).

Molecular Characterization of a Novel Putative Partitivirus Infecting Cytospora sacchari, a Plant Pathogenic Fungus

  • Three double-stranded RNAs (dsRNAs), approximately 1.85, 1.65 and 1.27 kb in size, were detected in an isolate of Cytospora sacchari from Iran. Partial nucleotide sequence revealed a 1,284 bp segment containing one ORF that potentially encodes a 405 aa protein. This protein contains conserved motifs related to RNA dependent RNA polymerases (RdRp) that showed similarity to RdRps of partitiviruses. The results indicate that these dsRNAs represent a novel Partitivirus that we tentatively designate Cytospora sacchari partitivirus (CsPV). Treatment of the fungal strain by cyclohexamide and also hyphal tip culture had no effect on removing the putative virus. Phylogenetic analysis of putative RdRp of CsPV and other partitiviruses places CsPV as a member of the genus Partitivirus in the family Partitiviridae, and clustering with Aspergillus ochraceous virus 1.

  • 모래무지아과에 대한 계통분류학적 연구의 일환으로 한국 고유종인 쉬리 Coreoleuciscus splendidus Mori의 두골 및 두부표면근육을 기재하고 잉어과에 해당하는 다른 어류들과 비교해부학적으로 검토하였다. 본 조사의 결과 본 종은 설이골의 측돌기와 미설골이 잉어과 어류들 중에서도 매우 특이하게 나타났다. 한편 사부의 형태, 두부측선감각관의 구조, 부설골과 익설골 및 전이골의 관계, 미설골의 형태, 기설골의 형태, 하새골의 형태 및 adductor mandiblulae complex의 조성 등에 있어서 모래무지아과의 진정 저루성 어류와는 뚜렷한 차이를 보였다. 본 연구에서는 이러한 형질들과 관련지어 본 종이 잉어과 내에서 차지하는 분류학적 위치에 대해 논의하였다.

Four New Records of Two Genera Balanophyllia and Cladopsammia (Anthozoa: Hexacorallia: Scleractinia: Dendrophylliidae) from Korea

  • The four species of the family Dendrophylliidae are newly recorded in Korea: Balanophyllia (Balanophyllia) cumingii Milne Edwards and Haime, 1848, Balanophyllia (Balanophyllia) vanderhorsti Cairns, 2001, Cladopsammia eguchii (Wells, 1982), and Cladopsammia gracilis (Milne Edwards and Haime, 1848). The two genera of Balanophyllia and Cladopsammia, to which the four species belong, are newly recorded in Korea. They were collected from the subtidal zones in Jeju-do Island, Korea by SCUBA diving from 1987 to 2012. This study aims to identify the four dendrophyllid species based on external and internal morphological characters including growth form, size, budding, and color of colonies, shape and size of corallites, columella, theca, and septa. Balanophyllia (Balanophyllia) cumingii is distinguished by its solitary growth form, small and low subturbinate corallite with enlarged calice, and expanded basal part, exsert first and second septa, and Pourtal$\acute{e}$s plan. Balanophyllia (Balanophyllia) vanderhorsti is characterized by its quasi-colonial growth form, subturbinate corallites with compressed calice, thick theca, and Pourtal$\acute{e}$s plan. Cladopsammia eguchii is characterized by its phaceloid growth form of compressed corallites basally united with common coenosteum, flat spongy columella, thick theca, and Pourtal$\acute{e}$s plan. Cladopsammia gracilis is distinguished by its phaceloid growth form of corallites basally united with common coenosteum, and pronounced Pourtal$\acute{e}$s plan forming flower patterns.

Gene Duplications Revealed during the Process of SNP Discovery in Soybean[Glycine max(L.) Merr.]

  • Genome duplication(i.e. polyploidy) is a common phenomenon in the evolution of plants. The objective of this study was to achieve a comprehensive understanding of genome duplication for SNP discovery by Thymine/Adenine(TA) cloning for confirmation. Primer pairs were designed from 793 EST contigs expressed in the roots of a supernodulating soybean mutant and screened between 'Pureunkong' and 'Jinpumkong 2' by direct sequencing. Almost 27% of the primer sets were failed to obtain sequence data due to multiple bands on agarose gel or poor quality sequence data from a single band. TA cloning was able to identify duplicate genes and the paralogous sequences were coincident with the nonspecific peaks in direct sequencing. Our study confirmed that heterogeneous products by the co-amplification of a gene family member were the main cause of obtaining multiple bands or poor quality sequence data in direct sequencing. Counts of amplified bands on agarose gel and peaks of sequencing trace suggested that almost 27% of nonrepetitive soybean sequences were present in as many as four copies with an average of 2.33 duplications per segment. Copy numbers would be underestimated because of the presence of long intron between primer binding sites or mutation on priming site. Also, the copy numbers were not accurately estimated due to deletion or tandem duplication in the entire soybean genome.

Local Rule of Đại Việt under the Lý Dynasty: Evolution of a Charter Polity after the Tang-Song Transition in East Asia

  • Empirical research into Đại Việt before the $14^{th}$ century has made little progress since the 1990s. To improve this situation, I here examine how the L$\acute{y}$ dynasty (1009-1226), the first long-lasting dynasty of Đại Việt, established stable local ruleafter the "Tang-Song Transition" in China that changed the entire picture of East Asia (including both Southeast and Northeast Asia). This paper focuses on two issues. First are the local administrative units and their governors. The nature of both higher units like lộ(circuits), phủ and ch$\hat{a}$u (provinces), and basic units like hươg and gi$\acute{a}$p (districts?) will be examined. Second, I examine non-institutional channels of local rule by the imperial family. By combining such administrative and non-administrative means, the L$\acute{y}$ central court enforced a considerably stable local rule for two centuries. Finally, I attempt some preliminary comparisons with the local rule of Goryeo (918-1392) in the Korean peninsula, a polity that shared many features with Đại Việt in the process of localization of the Tang and Song models. I hope this approach of viewing small empires from the standpoint not of their "goal" (modern states) but of their "start" (charter polities), will enrich the discussion of East Asian small empires.

서양 아동복의 역사적 변천에 관한 연구 (A Research on the Changes of Western Children's Clothing)

  • Based upon literature survey, this research aims to analyze western children's clothing and characteristics over centuries. It was not until the 18th century that children were given serious consideration and that fabrication of clothing began to be designed only for children. The very first clothes which revolutionized children's wear was 'a la matelot' in the 18th century which freed children from physical restriction, recognized gender difference, and sailor suit became popular among all sexes and adopted as school uniform. And then children's clothing was shortened in length and adopted tubular silhouette, which allowed greater comfort and movement. The 20th century saw children's wear reform to be carried out after the World War II due to invention of synthetic fiber, easy care dress material, mass production system and sophistication of marketing methods. Further evidence of improvement of children's social status can be found in contemporary designs: for example, 'casualized' wear, 'character design' which reflects children's psychology and 'family look' which is designed to be worn with mothers. This evolution of children's clothing within western dressing support the view that children's clothing is not a miniature of adult wear but it reflects idiosyncrasies of the era and exerts children's dignity.

Taxonomy of the Golovinomyces cynoglossi Complex (Erysiphales, Ascomycota) Disentangled by Phylogenetic Analyses and Reassessments of Morphological Traits

  • The name Golovinomyces cynoglossi s. lat. is traditionally applied to a complex of morphologically similar powdery mildews on hosts of the plant family Boraginaceae. The current species-level taxonomy within this complex is ambiguous due to the lack of phylogenetic examinations. The present study applied phylogenetic methods to clarify the taxonomy of G. cynoglossi s. lat. Phylogenetic analysis of rDNA ITS sequences retrieved from Asian, European and North American specimens revealed that G. cynoglossi s. lat. collections from different hosts involved several species in five clearly separated lineages. Clade I consists primarily of Golovinomyces cynoglossi s. str. on Cynoglossum. Clade III consists of Golovinomyces sequences retrieved from the host genera Symphytum and Pulmonaria. The taxa within clade III are now assigned to G. asperifoliorum comb. nov. Clade V encompasses G. cynoglossi s. lat. on the host genera Bothriospermum, Buglossoides, Echium, Myosotis, and Trigonotis. The taxa within clade V are now assigned to G. asperifolii comb. nov. The species concerned in this study were lecto- and epitypified to stabilize their nomenclature.

Identification and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in Cancer Cells

  • The long terminal repeats (LTRs) of human endogenous retrovirus (HERV) have been found to be coexpressed with sequences of closely located genes. It has been suggested that the LTR elements have contributed to the structural change or genetic variation of human genome connected to various diseases and evolution. We examined the HERV-W LTR elements in various cancer cells (2F7, A43l , A549, HepG2, MIA-PaCa-2, PC-3, RT4, SiHa, U-937, and UO-31). Using genomic DNA from the cancer cells, we performed PCR amplification and identified twelve new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity (88-99%) with HERV-W LTR (AF072500). A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-W LTR elements could be mainly divided into two groups through evolutionary divergence. Three HERV-W LTR elements (RT4-2, A43l-1, and UO3l-2) belonged to Group 1, whereas nine LTR elements (2F7-2, A549-1, A549-3, HepG2-3, MP2-2, PC3-1, SiHa-8, SiHa-10, and U937-1) belonged to Group 11. Taken together, our new sequence data of the HERV-W LTR elements may contribute to an understanding of tissue-specific cancer by genomic instability of LTR integration.

Genetic tests by next-generation sequencing in children with developmental delay and/or intellectual disability

  • Developments in next-generation sequencing (NGS) techogies have assisted in clarifying the diagnosis and treatment of developmental delay/intellectual disability (DD/ID) via molecular genetic testing. Advances in DNA sequencing technology have not only allowed the evolution of targeted panels but also, and more currently enabled genome-wide analyses to progress from research era to clinical practice. Broad acceptance of accuracy-guided targeted gene panel, whole-exome sequencing (WES), and whole-genome sequencing (WGS) for DD/ID need prospective analyses of the increasing cost-effectiveness versus conventional genetic testing. Choosing the appropriate sequencing method requires individual planning. Data are required to guide best-practice recommendations for genomic testing, regarding various clinical phenotypes in an etiologic approach. Targeted panel testing may be recommended as a firsttier testing approach for children with DD/ID. Family-based trio testing by WES/WGS can be used as a second test for DD/ID in undiagnosed children who previously tested negative on a targeted panel. The role of NGS in molecular diagnostics, treatment, prediction of prognosis will continue to increase further in the coming years. Given the rapid pace of changes in the past 10 years, all medical providers should be aware of the changes in the transformative genetics field.
