Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury.   • 사과는 에틸렌에 의해 호흡량이 일시적으로 상승하는 호흡급등형 과실이다. 에틸렌 발생은 세포벽분해효소 활성화와 세포벽 연화를 진행시켜 사과의 상품성과 저장성을 떨어뜨리는 원인이 된다. 사과의 에틸렌 생합성 과정에는 Md-ACS1 및 Md-ACO1 유전자가 연관되어 있으며, 두 유전자는 과실의 에틸렌 발생량과 경도에 영향을 미치는 것으로 알려져 있다. 본 연구는 사과 국내육성 28품종의 Md-ACS1 및 Md-ACO1 대립유전자형을 분석하고, 'Fuji(FJ)', 'RubyS(RS)', 'Hongro(HR)', 'Arisoo(AS)', 'Summer King(SK)', 'Greenball(GB)', 'Golden Supreme(GS)'을 대상으로 수확 후 25일까지의 에틸렌 발생량 및 상온저장($20^{\circ}C$) 20일 동안의 경도 연화율을 조사하였다. 그 결과, 낮은 에틸렌 발생량과 관련 된 대립유전자(favorable alleles0(FA)) ACS1-2, ACO1-1이 많을수록 에틸렌 발생량과 경도 연화율이 낮은 경향을 보였다. GS은 ACS1-1/1, ACO1-1/2(FA 1)으로 모든 품종 중 가장 높은 에틸렌 발생 수치와 경도 연화율를 보였다. SK와 GB은 ACS1-1/2, ACO1-1/2(FA 2)으로 ACS1-2/2, ACO1-1/2(FA 3)인 HR와 AS 보다는 높고 GS 보다 낮은 에틸렌 발생량과 경도 연화율을 보였다. ACS1-2/2, ACO1-1/1(FA 4)인 FJ는 RS를 제외한 모든 품종 중에 에틸렌 발생량과 경도 연화율이 가장 낮았다. 본 실험의 결과 Md-ACS1 및 Md-ACO1 대립유전자형과 사과의 에틸렌 발생량 및 경도 연화율은 상관성이 있는 것으로 확인되었다. 따라서, Md-ACS1 및 Md-ACO1 분자표지를 사과 국내육성품종의 저장성 예측과 육종 효율 향상을 위한 Marker-assisted selection에 활용할 수 있을 것으로 생각 된다.

Exogenous Sugars Involvement in Senescence and Ethylene Production of Tree Peony 'Luoyang Hong' Cut Flowers

  • Sugars play important roles in petal senescence of cut flowers. In the Expt. 1 of this study, the effects of different concentrations of glucose (60, 90, and $120g{\cdot}L^{-1}$) and sucrose (30, 60, and $90g{\cdot}L^{-1}$) application on the vase life, rate of flower diameter increase, rate of flower weight increase and ethylene production of cut tree peony (Paeonia suffruticosa 'Luoyang Hong') were evaluated. At the earlier stage, treatments of different concentrations of glucose and sucrose all retarded the process of flower opening and inhibited the increase of flower diameter and weight, while senescence of flowers fed with different concentrations of glucose was delayed at later stage. Flowers treated with $90g{\cdot}L^{-1}$ glucose displayed the longest vase life, which showed significant difference (P < 0.05) from those of flowers with the control and sucrose treatments. All treatments with glucose or sucrose not only retarded the decrease of flower diameter and weight, but also suppressed the ethylene production at the earlier stage and delayed the peak of ethylene evolution. In order to study the effect of exogenous sugar on the postharvest response of cut tree peony to ethylene, Expt. 2 was conducted. Cut flowers were treated with $90g{\cdot}L^{-1}$ glucose for 4 hours before (GE) or after (EG) exposed to $10{\mu}L{\cdot}L^{-1}$ ethylene for 4 hours. Generally, the opening process of flowers with GE and EG treatments was similar to that of the control, however GE treatment delayed flower senescence. Both GE and EG treatments improved flower diameter and weight, and GE treatment delayed the time of flower weight decrease. Besides, GE delayed climacteric ethylene evolution for 8 hours. All above suggest that exogenous sugars delay tree peony 'Luoyang Hong' cut flower senescence and extend flower vase life through their roles in the decrease of water loss and the suppression of sensitivity to ethylene and ethylene production.

Biodegradation of Ethylene in an Activated Carbon Biofilter

  • The objective of this study was to investigate the biodegradation of ethylene in an activated carbon biofilter inoculated with immobilized microbial consortium. The biofilter performance was monitored in terms of ethylene removal efficiency and carbon dioxide production. The biofilter was capable of achieving ethylene removal efficiency as much as 100% at a residence time of 14 min and an inlet concentration of 290 ppm. Under the same conditions, carbon dioxide with a concentration of up to 546 ppm was produced. Its was found that carbon dioxide was produced at a rate of 87 mg day$\^$-1/, which corresponded to a volume of 0.05 L day$\^$-1/. During operation with an inlet ethylene of 290 ppm, the maximum elimination capacity of the biofilter was 34 g of C$_2$H$_4$m$\^$-3/ day$\^$-1/. The biofilter could provide an attractive treatment technology for removing ethylene, an extremely volatile and slowly adsorbed compound.

Biofiter를 이용한 에틸렌 분해 (Degradation of Ethylene by a Biofilter)

  • The objective of this study was to investigate the biodegradation of ethylene in an biofilter inoculated with ethylene-oxidizing microorganisms. The biofilter performance was monitored in terms of ethylene removal efficiency and carbon dioxide production. The biofilter was capable of achieving the ethylene removal efficiency as much as 100% at a residence time of 14 min and an inlet concentration of 290 ppm. Under the same conditions, carbon dioxide with a concentration of up to 546 ppm was produced. It was found that carbon dioxide was produced at a rate of 87 mg/day, which corresponded to a volume of 0.05 L/day. Observable features of the ethylene-oxidizing microorganisms, meaning microbial activity occurrence in the biofilter, were investigated with the microscopy analysis.

Changes in flavor-relevant compounds during vine ripening of tomato fruit and their relationship with ethylene production

  • Flavor quality is import for determining consumer perception and acceptance of tomato products. In this study, 'Fendou' tomato fruit were harvested at six ripening stages and sampled to investigate the development of flavor-relevant compounds during vine ripening. Results showed that upon the initiation of ripening there was an increase in respiration rate and concomitant ethylene evolution that was associated with increased membrane permeability. In accordance with these physiological changes, flavor-relevant compounds demonstrated different expression patterns as fruit ripened, which contributed to 'red-ripe' flavor characteristics of red-ripe fruit. Based on correlation analysis between ethylene evolution and the flavor-relevant compounds during 'Fendou' tomato ripening and the other researchers' reports, the activation of System 2-dependent autocatalytic ethylene production plays an important role in the development of most flavor-relevant compounds during tomato vine ripening. Overall, our results suggested that most flavor-relevant compounds that accumulated the most during tomato fruit ripening at red stage could be under ethylene regulation and were among the most important contributors to the 'red-ripe' flavor. Due to the development of these compounds, the flavor quality at late ripening stages is different from that of fruit at early ripening stages.

MA저장 중 파프리카 과실의 품종별 품질 및 저장성 비교 (A Comparison of the Storability in MA Storage and the Quality of Paprika Fruit among Cultivars)

  • 본 연구는 장기 저장 유통을 위해 MA 저장에 적합한 품종 선발을 위해 2011년에 9개, 2012년에 12개의 파프리카 품종을 대상으로 품질과 저장성을 비교하였다. 2011년도 실험에서는 빨간색 품종 중에 과중, 경도 그리고 당도이 높고, 에틸렌 발생률이 낮으며 저장수명도 23일로 두 번째로 길었던 'Mosanto 7044'이 노란색 품종에서는 경도와 당도가 높고, 에틸렌 발생률이 낮았던 'Stayer'가 우수하였다. 2012년도 실험에서는 빨간색 품종 중에 과중, 과피두께, 경도가 높고, 부패율과 에틸렌 발생률이 낮으며 저장 수명이 28일로 세 번째로 길었던 'Nagano'가, 노란색 품종에서는 과피두께, 당도, 비타민 C 함량이 상대적으로 높았고, 에틸렌 발생률이 낮으며 저장수명이 26일로 가장 길었던 'Freestar'가 가장 우수하였다. 과실 품질 특성과 저장수명과의 상관관계를 비교한 결과 에틸렌(r = -0.5504)과 당도(r = 0.6112)가 고도의 상관관계를 보였다.

옥수수 뿌리에서 굴중성 반응과 에틸렌 생성에 미치는 Phorbol 12-myristate 13-acetate 조절 작용 (Regulation of Phorbol 12-Myristate 13-Acetate in the Gravitropic Response and Ethylene Production in Primary Roots of Maize)

  • 암을 유발하는 phorbol ester로 알려진 Phorbol 12-myristate 13-acetate (PMA)는 동물세포에서 신호전달 효소의 하나인 protein kinase C (PKC)를 활성화시킨다. 본 연구에서는 옥수수 일차뿌리에서 PMA가 에틸렌 생성을 통하여 굴중성 반응을 조절하는 효과를 연구하였다. PMA는 8시간 동안 $10^{-6}$ M과 $10^{-4}$ M에서 농도 의존적으로 뿌리 생장과 굴중성 반응을 촉진시켰다. 이러한 촉진 효과는 PKC의 억제제인 staurosporine (STA)에 의해 상쇄되었다. 이 결과는 굴중성 반응이 신호전달 체계에 관여하는 protein kinase C를 통하여 조절될 가능성을 제시하고 있다. 식물호르몬인 에틸렌도 뿌리 생장과 굴중성 반응에 중요한 역할을 한다고 알려져 있다. 에틸렌 생성은 $10^{-6}$ M과 $10^{-4}$ M PMA에 의하여 각각 26%와 37% 증가하였다. PMA는 또한 ACC synthase (ACS) 활성을 촉진시켰다. 또한 이 증가 효과는 STA에 의하여 상쇄되었다. 이 결과는 옥수수 뿌리에서 굴중성 반응은 에틸렌 생성을 거쳐 protein kinase를 통하여 조절될 가능성을 제시하고 있다.

Pre-harvest ethylene control affects vase life of cut rose 'Carola' by regulating energy metabolism and antioxidant enzyme activity

  • We studied the role of ethylene control in regulating energy metabolism, antioxidant enzyme activity, and vase life of cut rose Rosa hybrida 'Carola'. Rose flowers at stage II were sprayed with one of the following solutions: water (control), $10{\mu}L\;L^{-1}$ 1-methylcyclopropene (1-MCP), or $0.5g\;L^{-1}$ 2-chloroethanephosphonic acid (ethephon). After harvest, ethylene production rate, respiration intensity, energy charge (EC), activities of energy metabolism-related and antioxidant enzymes, and malondialdehyde (MDA) content were measured. Results showed that 1-MCP enhanced the activities of superoxide dismutase, $H^+$-adenosine triphosphatase, $Ca^{2+}$-adenosine triphosphatase, succinic dehydrogenase, and cytochrome c oxidase, increased adenosine triphosphate (ATP) content, maintained high EC levels, inhibited respiration intensity, reduced peroxidase (POD) and polyphenol oxidase (PPO) activity and MDA accumulation, and prolonged vase life. Ethephon promoted ethylene production and respiration intensity, increased POD and PPO activity, reduced ATP content and EC levels, and accelerated senescence. Our results support a novel role for ethylene control in regulating senescence of 'Carola'.

시비법의 벼의 탈입과 Ethylene 생성에 미치는 영향 (Effects of Fertilization Methods on Ethylene Evolution and Shattering in Rice Grains)

  • 시비법을 달리하여 재배된 벼가 태풍(벼 등숙기인 8월 28~29일) 피해로 인하여 일어났던 탈리정도의 차이를 분석코자 가 시험구의 출수기, 수량 및 에틸렌 생성량 등과의 관계를 검토한 결과는 다음과 같다. 1. 질소첨가 시비구(질소단용구, NP구, NK구, NPK+SiO$_2$구, NPK+퇴비구)는 질소무첨가 시비구(무비구, 무비+퇴비구, 무질소구, P단용구, K단용구)보다 탈입율이 낮았고 에틸렌 생성량도 낮았다. 2. 수도의 시비법에 따른 에틸렌 생성량과는 정의 유의상관(r=0.718*)을 곡실수량과는 부의 유의상관(r=-0.858**)을 보였다.
