Determination and Ethylene Oxide(EO) Separation of Polysorbates by RP-HPLC (역상 HPLC에 의한 polysorbates의 산화에틸렌(EO) 분리 및 정량분석)

  • The EO separation and quantitative determination of polysorbate 20, polysorbate 40, polysorbate 60 and polysorbate 80 was carried out by reversed phase HPLC. The water/acetonitrile was used for the mobile phase of gradient conditions. An YMC Pack Ph ($250mm{\times}4.6mm$ i.d., $5{\mu}m$) and Phenomenex C4 ($250mm{\times}4.6mm$ i.d., $5{\mu}m$) and the selected ELSD detector was applied. The analysis results of HPLC showed good linearity with correlation coefficient of $r^2$=0.997 in the rage of $180.2{\sim}980.5{\mu}g/mL$ and detection limit.

Extraction Properties of Constituents in Ginseng Leaf Tea As Influenced by Decontamination Methods (인삼엽록차 성분의 추출특성과 살균방법에 따른 영향)

  • Extraction properties were investigated on ginseng leaf tea as functions of the current ethylene oxide fumigation and gamma irradiation at 2.5 to 10 kGy. In the extraction of tea constituents at boiling conditions, water soluble matters were the highest (83% yield for 5 min) in the time dependence, followed by tannin (82%), sugar (73%) and saponin (66%). No significant difference was observed in the extraction yield between the 5 kGy-irradiated and control samples, whereas both EO fumigation and 10 kGy irradiation reduced the extraction time of soluble matter and tannin, leading to the decrease in Hunter L value of tea extracts.

  • Physico-chemical properties were investigated on ginseng leaf tea which was decontaminated by the current ethylene oxide fumigation and gamma irradiation, respectively. Chemical components such as soluble matter, saponin and polyphenols were found to be stable to both treatments, while ascorbic acid and chlorophylls were relatively labile to fumigation and irradiation more than 5 kGy. Instrumental analysis has shown that EO fumigation causes apparent changes in pH and color of extracts and volatile profile of the sample, as compared to negligible changes in the irradiated sample at a decontaminating dose, 5 kGy.

  • The objective of this work is to investigate the effect of molecular weight of poly(ethylene oxide) (PEO) and release medium on the release of nifedipine (NP) from PEO tablets containing NP and to get some mechanistic insights into the release of NP. The tablets containing NP were prepared by direct compression, using a flat-faced punch and die. The molecular weights of PEOs used were 200K, 900K, 2000K and 7,000K. The release kinetics were studied for 24 hours in aqueous ethanol solution, using a dissolution tester at $36.5^{\circ}C$ and 100 rpm. Drug release rate increased, as the concentration of ethanol in the dissolution medium increased, due to the increased solubility of NP. As the molecular weight of PEO increased, release rate decreased, due to the slower swelling and dissolution of PEO. The power values obtained by fitting data to the power law expression $(M_t/M_{\infty}=kt^n)$ indicated that, at low ethanol concentration, the release of NP is governed by anomalous diffusion. However, as the ethanol concentration increases, diffusional release becomes to prevail over anomalous or zero-order release. Overall, these results provided some insights into the release of NP from PEO tablet.

Thermosensitive Block Copolymers Consisting of Poly(N-isopropylacrylamide) and Star Shape Oligo(ethylene oxide)

  • Thermosensitive block copolymers of ethylene oxide and N-isopropylacrylamide (NIPAM) were synthesized. A five armed star shape oligo(ethylene oxide) initiator with a cyclotriphosphazene core was prepared and used for the atom transfer radical polymerization (ATRP) of NIPAM. The lower critical solution temperatures (LCSTs) of the copolymers were 36 to 46 ${^{\circ}C}$, higher than that of PNIPAM (32 ${^{\circ}C}$), depending on their molecular weights. The copolymers were soluble in water below the LCSTs but formed micelles above the LCSTs. The thermosensitive micellization behaviors of the polymers were investigated by fluorescence spectroscopy. With increasing the temperature of an aqueous solution of P2 and pyrene above the LCST, the peak of 333 nm red-shifted to appear around 339 nm and its intensity increased significantly, indicating the micelle formation. The transfer of pyrene into the micelles was also confirmed by a confocal laser scanning microscope. The fluorescence image obtained from P2 in an aqueous pyrene solution exhibited a green emission resulting from the pyrene transferred into the micelles. Salt effects on the solubility of the copolymers in an aqueous solution were investigated. The LCST of P2 decreased sharply as the concentration of sodium chloride increased, while decreased slowly with potassium chloride.

A Study on Expiration Date on Ethylene Oxide Gas Sterilization Products - Related to Storage Environment - (Ethylene Oxide 가스 멸균물품의 유효기간에 관한 연구 - 보관환경을 중심으로 -)

  • Purpose: This was a study on the expiration date of Ethylene Oxide (EO) gas sterilization and effects of the environmental factors of temperature, humidity and type of cabinet in sterile goods storage area on the expiration date. Methods: Sterile goods storage areas from 13 departments in one hospital were selected and 455 EO gas sterilization samples were prepared and kept in those areas over the 14 months of the study. Each sample was tested with a microbiological culture in the laboratory every week. If the result was positive, the sample was regarded as contaminated. The researcher visited once a month to check the temperature, humidity and type of cabinet. Results: With the exception of 1 sample which was positive at 56th week. 454 samples were confirmed as negative. The environment of the samples storage area was measured monthly. The annual average temperature was $24.2{\pm}1.6^{\circ}C$, and the mean relative humidity $34.7{\pm}15.2%$. The types of cabinet were 7 open and 6 closed. Conclusion: The results of the microbiological culture at 13 months showed that none of the samples were contaminated. Therefore the hospital's existing Expiration Date can be extended from 6 months to 13~14 months.

Polymer-supported Zinc Tetrahalide Catalysts for the Coupling Reactions of CO2 and Epoxides

  • Homogeneous zinc tetrahalide complexes, highly active catalysts for the coupling reactions of alkylene oxide and CO2 produce alkylene carbonates, were heterogenized due to their tendency to decompose produced alkylene carbonates during the distillation process. Heterogenization of homogeneous zinc tetrahalide complexes was achieved by polymerizing 1-alkyl-3-vinylimidazolium zinc tetrahalides. These polymerized zinc tetrahalide catalysts displayed similar activities to their corresponding monomeric analogues for the coupling reactions of carbon dioxide with ethylene oxide (EO) or propylene oxide (PO) to produce ethylene carbonate (EC) or propylene carbonate (PC). TGA studies showed that the polymer-supported zinc tetrahalide catalysts are thermally stable up to 320 oC. The catalyst recycle test showed that the supported catalysts could be reused over six times. After removal of the polymer-supported catalyst through a simple filtration, EC was able to be isolated without decomposition.

Preparation and Characterization of pH-Sensitive Poly(ethylene oxide) Grafted Methacrylic Acid and Acrylic Acid Hydrogels by ${\gamma}-ray $ Irradiation

  • pH-sensitive hydrogels were studied as a drug carrier for the protection of insulin from the acidic environment of the stomach before releasing it in the small intestine. In this study, hydrogels based on poly(ethylene oxide) (PEO) networks grafted with methacrylic acid (MAA) or acrylic acid (AAc) were prepared via a two-step process. PEO hydrogels were prepared by ${\gamma}-ray $ irradiation (radiation dose: 50 kGy, dose rate: 7.66 kGy/h), grafted by either MAA or AAc monomers onto the PEO hydrogels and finally underwent irradiation (radiation dose: 520 kGy, dose rate: 2.15 kGy/h). These grafted hydrogels showed a pH-sensitive swelling behavior. The grafted hydrogels were used as a carrier for the drug delivery systems for the controlled release of insulin. Drug-loaded hydrogels were placed in simulated gastric fluid (SGF, pH 1.2) for 2 hr and then in simulated intestinal fluid (SIF, pH 6.8). The in vitro drug release behaviors of these hydrogels were examined by quantification analysis with a UV-Vis spectrophotometer.

  • A novel preparation method for core/shell nanoparticles with protein drug-loaded lipid core was designed and characterized. The lipid core is composed of lecithin and protein drug and the polymeric shell is composed of Pluronics (poly (ethylene oxide)-poly (propylene oxide)-poly(ethylene oxide) triblock copolymer, F-127 For the application of core/shell nanoparticles as a protein drug carrier, lysozyme and Vascular Endothelial Growth Factor (VEGF) were loaded into the core/shell nanoparticles by electrostatic interaction and the drug release pattern was observed by manipulating the polymeric shell.

  • The phase behavior of chlorinated hydrocarbon/mixed surfactants/water microemulsion systems were investigated for dry cleaning solvent properties. With appropriate surfactant mixtures, Winsor type I-III-II microemulsions were generated which is the same as hydrocarbon systems. For perchloroethylene(PCE) with mixed Tween systems, the optimum salinity(S*) decreases and the optimum solubilization parameter(o*) increases with decreas- ing HLB. For PCE with mixed Aerosol MA and ethoxylated alcohol systems, S* and o* both increase with increasing ethylene oxide moles. For dichlorobenzene(DCB) with mixed Aerosol MA and ethoxylated or propoxylated sulfate systems, S* and o* both increase with increasing ethylene oxide moles or propylene oxide moles.
