Precursors for the Ethylene Evolution of Pseudornonas syringae pv. Phaseolicola (Pseudomonas syringae pv. Phaseolicola에 의한 Ethylene 생성에서의 전구물질)

  • - The purpose of this work is to investigate the effects of various substrates on biosynthesis of ethylene by the Kudzu strain of Pseudomonas syn'ngae pv. Phaseolicola causing halo blight. In the intact cell of P. sym'ngue, optimal condition for ethylene production was achieved at p1-I 7.5 and $30^{\circ}C$ for 9 to 10 hours of culture. Ethylene was most effectively produced from amino acids such as Asn, Gln, Asp ans Glu, compared to those of various kinds of sugars. While ethylene production from $\alpha$-ketoglutarate ($\alpha$-KG) was gradually increased throughout 51 hours incubation period tested. Ethylene production derived from citrate, $\alpha$-KG and oxalacetate as well as a few amino acids was further enhanced by the addition of histidine or arginine. In cell-free ethylene-forming system, ethylene was most effectively produced from $\alpha$-KG, compared to those from citrate, oxalacetate, Glu, Arg, or Asp, at 0.5 mM among the range from 0.25 mM to 5 mM. Anlinooxyacetate, an inhibitor of a pyridoxal phosphate-linked enzyme, completely inhibited ethylene evolution derived from Glu but not affect that derived from $\alpha$-KG. The results obtained in this work suggest that $\alpha$-KG might be a direct precursor of ethylene production in this organism than any other substrates tested.

  • This experiment was conducted to evaluate the effect of fruit quality, ethylene evoluoon and storage in apple 'Tsugaru' as influenced by aminoethoxyvinylglycine(AVG) and several surfactants. When treated with AVG+Silwet L-77, there were few significant differences in soluble solid and acidity as compared with control, but dichlorprop treatment significantly increased in soluble solid and decreased in acidity. Color development was decreased when treated with AVG+Silwet L-77. AVG+Silwet L-77 treatment decreased in ethylene evolution, and increased in storage. It can be concluded that fruit can be left in trees longer and still maintain storage, and more fruit is able to go to fresh market into long-term storage, which increase the market-ability of apple.

Changes in flavor-relevant compounds during vine ripening of tomato fruit and their relationship with ethylene production

  • Flavor quality is import for determining consumer perception and acceptance of tomato products. In this study, 'Fendou' tomato fruit were harvested at six ripening stages and sampled to investigate the development of flavor-relevant compounds during vine ripening. Results showed that upon the initiation of ripening there was an increase in respiration rate and concomitant ethylene evolution that was associated with increased membrane permeability. In accordance with these physiological changes, flavor-relevant compounds demonstrated different expression patterns as fruit ripened, which contributed to 'red-ripe' flavor characteristics of red-ripe fruit. Based on correlation analysis between ethylene evolution and the flavor-relevant compounds during 'Fendou' tomato ripening and the other researchers' reports, the activation of System 2-dependent autocatalytic ethylene production plays an important role in the development of most flavor-relevant compounds during tomato vine ripening. Overall, our results suggested that most flavor-relevant compounds that accumulated the most during tomato fruit ripening at red stage could be under ethylene regulation and were among the most important contributors to the 'red-ripe' flavor. Due to the development of these compounds, the flavor quality at late ripening stages is different from that of fruit at early ripening stages.

  • Experiments were carried out to clarify changes of intercellular gas composition, respiration and ethylene evolution by controlled external atmospheric pressure and external gas composition on apples. Jonathan apples, harvested 25, in September and Rolls apples, harvested 25, in October, 1971 and 1972 were used. Results obtained were as follows: 1. Intercellular gas volumes were proportionally changed by the given pressure, and the given pressure remarkably affected internal gas composition and volume of apples. As intercellular gas volumes were reached in their equilibrium within 5 min. after treatment, the internal atmospheric conditions became constant rapidly. 2. The increase of internal $CO_2$ production was co-related with an amount of internal $O_2$ consumption, therefore, the decreasing period of internal $O_2$ consumption was equalled to the period of climacteric rise in respiration. 3. The increasing of $CO_2$ production followed evolution of $C_2$H$_4$ and this phenomenon on SAP part. was subsequent to NAP part 4. In sub-atmospheric storage, CA effect was also obtained by control of low $O_2$ and high $CO_2$ concentration.

  • A series of experiments were carried out to know the effect of growth retardants on the growth of rice seedlings and ethylene evolution from the rice seedlings. The results suggested that the reduction rate in order of plant height was S-327 > BAS-106 > NTN-821 > PP-333 > CGR-811. The heigher concentration levels of growth retardants applied. the more effective reductions of plant height were found. Ethylene evolution from the rice seedlings was significantly negative correlated with plant hight of of seedlings in all treatments.

Ecological Features of 'Pingguoli'(Pyrus sp.) Fruits Produced in Korea and China (한국산과 중국산 '사과배' 과실의 생태적 특성)

  • Postharvest characteristics of 'Pingguoli' fruit produced between China and Korea were compared to find an ecological difference. There was a difference in fresh weight per fruit, thus, fruit weight produced in Korea was much higher than those in China. However, no significant differences were confirmed in the level of soluble solid content and flesh firmness. The acidity of Korean 'Pingguoli' was higher than Chinese ones. The external appearance of Korean 'Pingguoli' was not attractive due to the rough and rugged surface. Thus, the prospect of 'Pingguoli' cultivation in Korea seems not to be promised. The general trend of respiration and ethylene evolution during storage did not significantly differed but the amount of $CO_2$ and ethylene production was higher in fruits produced in China. There was a strong similarity in respiration and ethylene evolution pattern to typical climacteric fruits. Thus, 'Pingguoli' is considered as a climacteric fruit. Even though the rate of ethylene production of 'Pingguoli' was relatively high, the storability of 'Pingguoli' showed a strong potential. The further studies in postharvest physiology of pear using 'Pingguoli' and in its breeding potentials as a source of long storage life are needed.

  • This experiment was carried out to determine the effect of nitrogen application on the cold tolerance of rice plant, with treatment of three levels of nitrogen and three times of application under the low temperature at tillering and panicle initiation stages. The higher cold tolerance variety was increased in plant height and number of tillers on high nitrogen level during the low temperature treatment. Nitrogen content of leaf blade was increased, but carbohydrate content was decreased during the low temperature treatment at tillering stage. Ethylene evolution from leaf was remark-ably increased just after low temperature treatment. Highly significant negative correlation was observed bet-ween the nitrogen content of leaf blade and percentage of filled grain under low temperature condition at reproductive growth stage.

  • The effect of ethylene on the proliferation of rhizomes and plant regeneration were investigated from rhizome segment culture of Cymbidium niveo-marginatum. Ethylene levels in the rhizome culture vessels were reached a maximum after 8 days of culture; total amount of ethylene evolution was much on the initiation of shoot induction than of rhizome proliferation. The treatment with ethephon on rhizomes was inhibited in the proliferation of rhizome and the growth of shoot length; however, the treatment was effective on shoot induction from rhizomes. Aminoethoxyvinylglycine(AVG) 1mg/L was effective on the proliferation of rhizomes and shoot induction from them; however, the proliferation of them was inhibited, and the growth of shoot length was significantly promoted at the concentration of 10mg/L AVG. The presence of $\textrm{AgNO}_{3}$ inhibited in the proliferation of rhizomes and shoot induction from them.

  • This experiment was conducted to investigate the effect of ethylene adsorbent removing ethylene gas produced from apple fruits in poly ethylene film bag storage. The treatment of ethylene adsorbent was not effected the change in soluble solids and organic acid content of fruits. The fruit softening was remarkably delayed by the absorbent treatment, and the more amount of it was the more effective. The ethylene evolution and respiration of fruits reduced or decreased by this treatment during storage. The ethylene adsorbent for 'Fuji' apple fruit was effective more than 30g per 10kg fruits.

The evolution characteristics of incipient soot particles in ethylene/air inverse diffusion flame (에틸렌/공기 역확산 화염에서의 초기 매연 입자의 성장 특성)

  • The evolution of incipient soot particles has been examined by high resolution electron microscopy (HRTEM) and elemental analyzer in ethylene-air inverse diffusion flames. Laser Induced Incandescence(LII) and laser scattering methods were introduced for examining the change of soot volume fraction and morphological properties in combustion generated soot qualitatively. Soot particles, collected by thermophoretic sampling were analyzed by using HRTEM to examine the nano structure of precursor particles. HRTEM micrographs apparently reveal a transformation of condensed phase of semitransparent tar-like material into precursor particles with relatively distinct boundary and crystalline which looks like regular layer structures. During this evolution histories C/H analysis was also performed to estimate the chemical evolution of precursor particles. The changes of C/H ratio of soot particles with respect to residence time can be divided into two parts: one is a very slowly increasing regime where tar-like materials are transformed into precursor particles (inception process) the other is an increasing region with constant rate where surface growth affects the increase of C/H ratio dominantly (surface growth region). These results provide a clear picture of a transition to mature soot from precursor materials.
