This study was carried out to investigate the mutagenic, antimutagenic, cytotoxicity and antitumor effect of Codonopsis lanceolata (CL). CL was extracted with 70% ethanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water. Antimutagenic, cytotoxicity and antitumor effects of CL extracts were measured by using Ames test, SRB method, and the tumor growth inhibition test. CL extracts did not show any mutagenicity in the Ames test; however, 70% ethanol extracts and its fractions had strong antimutagenic effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-nitroquinoline-1-oxide (4NQO). The ethyl acetate fraction of CL (200 ${\mu}g$/plate) showed approximately 72.1% inhibitory effect on the mutagenesis induced by 4NQO against TA98 strain, whereas 69.6% and 67.0% inhibitions were observed on the mutagenesis induced by MNNG and 4NQO against TA100 strain. In anticancer effects, the cytotoxicity of CL extract and its fractions against cancer cell lines including human cervical adenocarcinoma (HeLa), human hepatocellular carcinoma (HepG2), human breast adenocarcinoma (MCF-7), human lung carcinoma (A549) and transformed primary human embryo kidney (293) were investigated. The treatment of 1 mg/mL CL ethyl acetate fraction had the highest cytotoxicity of 74.5%, 70.7% and 80.3% against HeLa, MCF-7 and A549 cells, respectively. In contrast, the extract and its fractions showed only 2$\sim$31% cytotoxicity for a normal human kidney cell line (293). In vivo anticancer effect of CL extract was tested using Balb/c mice transplanted sarcoma-180 cells. CL ethyl acetate fraction showed the highest inhibition rate of 56.4% at the 50 mg/kg concentration.

In this study, we compared the antioxidant activities of three loquat (Eriobotrya japonica Lindl.) by cultivars (Daebang, Bubang and native cultivar). The leaves were extracted by 80% ethanol and then fractionated with various solvents (n-hexane, ethyl acetate, n-butanol, and water) for biological activities and bioactive compounds analysis. Total polyphenol content of extracts and fractions was in the range of 84.93~478.50 mg/g gallic acid equivalent (GAE). From highest to lowest GAE, the fractions were n-butanol> ethyl acetate> n-hexane> 80% ethanol> water. Among the three cultivars, the highest polyphenol content was found from native cultivar. As for DPPH radical scavenging activity, the n-butanol fraction showed the highest activity, and native cultivar was the highest on the $IC_{50}$ values (0.18 mg/mL). In the nitrite scavenging activity, the ethyl acetate fraction (54.99~60.86%) showed the most effective activity on the Bubang cultivar was higher than others. The ursolic acid content of the ethyl acetate fraction showed the highest (51.41 mg/g) in the Daebang cultivar. Based on all these results;the Bubang cultivar showed relatively higher antioxidant and nitrite scavenging activities, but the ursolic acid content was higher in the Daebang cultivar. These results suggest that extracts from loquats (Eriobotrya japonica Lindl.) can be used as bioactive and functional materials that could be important information for industrial use in the future.
This study compared and analyzed the antioxidant activities of various organic solvent fractions from the leaves and roots of Peucedanum insolens Kitagawa. For this study, the dried leaves and roots of P. insolens Kitagawa were first extracted using 70% ethanol. The extracts were sequentially sub-fractionated in the order of hexane, chloroform, ethyl acetate, n-butanol, and water. The results revealed that the distribution of total phenolic contents by organic solvent fractions showed the same pattern in both the leaves and roots, with the highest in the ethyl acetate fraction (101.1±1.0 mg vs 71.2±3.4 mg of GAE/mg), but the lowest content in the hexane fraction (9.5±0.2 mg vs 7.5±2.1 mg of GAE/mg). The distribution of total flavonoid content in the organic solvent fractions showed the same pattern as that of total phenolic content. The results of DPPH, ABTS, and FRAP assays showed that the leaf and root extracts exhibited free radical scavenging activity in the same pattern, particularly, the ethyl acetate fraction had the highest activity. These results indicate that not only the roots of P. insolens Kitagawa but also the leaves possess potential substances that exhibit strong antioxidant activity. Significant correlations (R=0.903, p<0.0001, DPPH radical; R=0.891, p<0.001, ABTS radical; R=0.745, p<0.05, FRAP radical) between total phenolics and radical scavenging activities, but also significant correlations (R=0.867, p<0.001, DPPH vs. ABTS radicals; R=0.882, p<0.0001, DPPH vs. FRAP radicals; R=0.973, p<0.0001, ABTS vs. FRAP radicals) between radical scavenging activities were found in the organic solvent fractions. Therefore, as in the roots of P. insolens Kitagawa, the leaves possess strong antioxidant capacity and can be used as the main antioxidant material.
The aim of this research was to investigate the industrial application of Sedum sarmentosum. Antibacterial activities of the n-hexane, methylene chloride (MC), ethyl acetate, and n-butanol fractions of S. sarmentosum were tested against Escherichia coli, Staphylococcus aureus, and Helicobacter pylori. The MC fraction showed the strongest antibacterial activity against E. coli, with an inhibition zone greater than 13 mm in disc assays. At $100{\mu}g/mL$, all fractions scavenged more than 50% of 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radicals (${\cdot}OH$). In particular, the MC fraction showed the strongest scavenging activity against DPPH and ${\cdot}OH$. In addition, we found that treatment with the MC fraction inhibited the growth of H. pylori and gastric adenocarcinoma cells. The present results suggest that the MC fraction of S. sarmentosum would play the promising protective role against pathogenic bacteria and free radicals.

This study was designed to examine the in vitro antioxidant and anti-inflammatory effects of kohlrabi (Brassica oleracea var. gonglodes) extract. Kohlrabi was extracted using 70% ethanol and then fractionated sequentially with n-hexane, ethyl acetate and butanol. Antioxidative ability was evaluated by bioassays using total polyphenol contents and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid diammonium salt) radical scavenging activity. Ethyl acetate fraction of kohlrabi was best on total polyphenol contents ($27.33{\pm}0.26mg\;GAE/g$) and ABTS radical scavenging effects ($IC_{50}\;172.9{\pm}1.6{\mu}g/mL$).For the anti-inflammatory activity in RAW 264.7 cells, the EtOAc fraction showed the highest inflammatory effect. Dose response studies were performed to determine the inhibitory effect of EtOAc fraction of kohlrabi on pro-inflammatory mediators in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. The EtOAc fraction of kohlrabi inhibited the NO and $PGE_2$ production and the protein level of iNOS and COX-2, and protein expression of pro-inflammatory cytokines (TNF-${\alpha}$, IL-6 and IL-$1{\beta}$), in a dose-dependent manner. These results suggest that kohlrabi has considerable potential as a ingredient with antioxidative and anti-inflammatory effects.

In this study, the antioxidative effects and inhibitory effects on tyrosinase and elastase of Sorbus commixta (S. commixta) twig extracts were investigated. The aglycone fraction of S. commixta twig extract showed the prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$, $13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities of S. commixta twig extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated by the luminol-dependent chemiluminescence assay. The 50 % ethanol extract among extracts showed the most prominent ROS scavenging activity ($OSC_{50}$, $0.189{\mu}g/mL$). The cellular protective effects of extract/fractions of S. commixta twig on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The 50 % ethanol extract and ethyl acetate fraction showed the cellular protective effects against ROS in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The inhibitory effect of S. commixta twig extract on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate ($IC_{50}$, $113.2{\mu}g/mL$) and aglycone fraction($IC_{50}$, $105.3{\mu}g/mL$) on tyrosinase showed more remarkable inhibitory effect than arbutin($IC_{50}$, $226.88{\mu}g/mL$), known as the whitening agent. The inhibitory effect of aglycone fraction ($IC_{50}$, $6.9{\mu}g/mL$) on elastase was simillar to quercetin($IC_{50}$, $6.1{\mu}g/mL$), flavonoid known as reference compound. These results indicate that extract/fractions of S. commixta twig can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. S. commixta twig extracts can be applicable to new functional cosmetics for anti-aging products.
In order to develop antimicrobial substances, many kinds of medicinal herbs were extracted with absolute ethanol and then antimicrobial activities against various microorganisms were investigated. Ethanol extract from cinnamon bark showed the strongest antimicrobial activity on the growth of almost all submitted microorganisms. Specially, molds such as Aspergillus sp. and Pencillium sp. were inhibited strongly. Therefore, the crude antimicrobial substance from the ethanol extract was fractionated with various solvents such as n-hexane, chloroform, ethyl acetate, and n-butyl alcohol and then their antimicrobial activities were tested. Among the various solvent fractions from the ethanol extract, n-hexane fraction was the best in antimicrobial activity especially against molds. There were no significant changes in antimicrobial activity of the n-hexane fraction by heat treatment at $100^{\circ}C$ for 60 min or $121^{\circ}C$ for 15 min and by the change of pH 4.0~10.0. We could get the results that the n-hexane fraction of cinnamon bark extract showed not only antimutagenicity but also no mutagenicity by Ames test with Salmonella typhimurium TA 98 and TA 100.
Zizyphus jujube leaf fractions (ZLFs) showed no cytotoxic effects of up to $100{\mu}g/mL$, while the anti-inflammatory effects of ZLFs were analyzed by checking the productions of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), cyclooxygenase-2 (COX-2), and inflammatory cytokines TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 in the lipopolysaccharide (LPS)-stimulated Raw264.7 macrophage up to the concentration of $100{\mu}g/mL$. ZLFs ($100{\mu}g/mL$) demonstrated a strong anti-inflammatory activity that reduced 61~85% of NO and 71~100% of $PGE_2$ production in the LPS-stimulated Raw264.7 macrophage. Even the low ZLFs concentration of $1{\mu}g/mL$ have reduced NO and $PGE_2$ production by 34~64%. Expressions of COX-2 protein were also effectively inhibited by the ZLFs. Furthermore, the TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 production were significantly suppressed through the treatment of ZLFs at concentrations of 1, 10, and $100{\mu}g/mL$. In the order of the Zizyphus jujube leaf water fraction (ZLWF) < buthanol fraction (ZLBF) < ethyl acetate fraction (ZLEF) showed anti-inflammatory activity. In particular, the ethyl acetate fraction ZLEF at $100{\mu}g/mL$ showed an excellent anti-inflammatory activity by reducing the production of NO, $PGE_2$, COX-2, and inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6) in the level of Raw264.7 macrophage without LPS-stimulation or even better. The results of our study suggest the potential of ZLEF for use as an excellent ant-inflammatory inhibiting mediator and may be used as a therapeutic approach to various inflammatory diseases.
The bulbil of the $Dioscorea$$species$ is produced, the amount of which is 2,000 tons annually, but it has been discarded without specific use. In this study the antioxidant and antimutagenicity of bulbil of the $Dioscorea$, which compared to bulbil of Danma($Dioscorea$$japonica$ Decaisne) and Jangma($Dioscorea$$batatas$ Decaisne), a major domestic cultivation species. The study was done by extracting bulbil of the Dioscorea methanol and the methanol extracts was re-extracted with chloroform, ethyl acetate, $n$-butanol and water. In methanol extract of Danma and Jangma, polyphenolic compounds contained 2.2 and 3.9 mg/g extract, respectively. The ethyl acetate fraction of Danma and Jangma had higher polyphenolic contents of 33.9 and 39.1 mg/g, whereas water fractions were much lower at 2.4 and 5.8 mg/g. Determination of antioxidant activity showed that the ethylacetate fraction strong DPPH radical scavenging activity and ABTS radical scavenging activity. The inhibitory effects of methanol extracts and chloroform, ethylacetate, $n$-butanol, water fraction from bulbil of Danma and Jangma on the mutagenicity in 1-NP, $AFB_1$, Trp-P-1 were investigated using $S.$$typhimurium$ TA98. Danma and Jangma cultivars decreased the reverse mutation induced by 1-NP, $AFB_1$, Trp-P-1 in $S.$$typhimurium$ TA98. The fraction of chloroform and ethylacetate showed strong inhibitory effects, in a dose dependant manner against the mutagenicities induced by 1-NP, $AFB_1$, Trp-P-1 in $S.$$typhimurium$ TA98.
This study was conducted to investigate the antioxidative effects of Paeonia lactiftora Pall. (PL) seed extracts and Forsythia viridissima Lindl. (FVL) extracts on the antioxidative defense system and lipid peroxidation in the erythrocytes of rats fed a high-cholesterol diet. Sprague-Dawley male rats weighing 100 10 g were randomly assigned to nine experimental groups and fed 0.5 % cholesterol. The HC group did not receive any supplement, while the MP group was supplemented with 0.1 % methanol extract of PL seed, the MP2 group with 0.2 % methanol extract of PL seed, the EP1 group with 0.05 % ether-soluble fraction of PL seed, the EP2 group with 0.1 % ether-soluble fraction of PL seed, the MS1 group with 0.05 % methanol extract of FVL, the MS2 group with 0.1 % methanol extract of FVL, the ES group with 0.025% ethyl acetate-soluble fraction of FVL, and the ES2 group with 0.05 % ethyl acetate-soluble fraction of FVL. The experimental diets were fed ad libitum for 3 weeks. The erythrocyte SOD activity in the EP1 and EP2 groups increased 38% and 59%, respectively, when compared with the HC group, while the erythrocyte GSHpx activity in the EP1, EP2, and ES2 groups increased 30%, 31 %, and 29%, respectively, when compared with the HC group. The level of erythrocyte TBARS was significantly lower in the MP2, EP1, and EP2 groups than in the HC group, yet the level of serum TBARS was significantly lower in the all supplemented groups than in the HC group. The level of serum HDL- TBARS was significantly lower in the EP1 and EP2 groups than in the HC group, while the level of serum LDL- TBARS was significantly lower in the all the supplemented groups than in the HC group. Accordingly, the results indicated that the PL seed extracts and FVL extracts reduced oxidative damage by activating the antioxidative defense system in the erythrocytes of rats fed a high-cholesterol diet.
