Influence of Ruminally Protected Fat and Urea Treated Corncobs Ensiled with or without Corn Steep Liquor on Nutrient Intake, Digestibility, Milk Yield and Its Composition in Nili-Ravi Buffaloes

  • Sixteen early lactating Nili-Ravi buffaloes, four animals in each group, were used in a Completely Randomized Design to evaluate the effect of varying levels of both ruminally protected fat and urea treated corncobs ensiled with or without corn steep liquor (CSL) on feed intake, digestibility and milk production and its composition. Four experimental diets were formulated. The control (C) diet was balanced to contain 0% fat and 35% urea treated corncobs ensiled with 0% CSL. The low fat (LF), medium fat (MF) and high fat (HF) diets had 45, 55 and 65% urea treated corncobs ensiled with 9% CSL and 2, 4 and 6% ruminally protected fat, respectively. Dry matter, crude protein (CP) and neutral detergent fiber (NDF) intakes by buffaloes remained similar across all treatments. However, DM and NDF as a percent of body weight and digestible DM intakes were higher in HF diet when compared to C, LF and MF diets. Digestible NDF intakes were also significantly higher in HF diet as compared to all other diets. The intakes of ADF and digestible ADF were higher in MF and HF than C and LF diets. The significant variation in digestible DM, ADF and NDF intakes may be attributed to the ammoniation of corncobs along with CSL that caused significant changes in the degradability and digestibility of the diets. Ether extract and digestible EE intakes differed significantly (p<0.05) among all treatments. Intakes of EE were the highest in animals fed HF diet, which was because of added fat. Apparent DM digestibility was the highest in animals C diet and was the lowest in those fed LF diet. Neutral detergent fiber and ADF digestibilities were higher in animals fed diets containing urea treated corncobs ensiled with 9% CSL when compared to those fed diets containing urea treated corncobs ensiled without CSL. Apparent digestibility of CP was noted highest (71.47%) in animals fed HF diet when compared to those fed MF (67.75%), LF (67.04%) and C (65.39%) diets. Milk yield (4% FCM) was the higher in buffaloes fed HF, MF and LF diets than those fed C diet. These results indicated that increasing levels both of fat and urea treated corncobs ensiled with CSL elevated the negative effects of poor quality fibrous feed on milk production by buffaloes.

Effects of physically effective neutral detergent fiber content on dry matter intake, digestibility, and chewing activity in Korean native goats (Capra hircus coreanae) fed with total mixed ration

  • Objective: This experiment was to determine proper physical traits in the diet for goats by investigating the effects of physically effective neutral detergent fiber (peNDF) content on dry matter intake (DMI), digestibility, and chewing activity in black goats fed with total mixed ration (TMR). Methods: Six growing wethers of Korean native black goats (Capra hircus coreanae) aged 8 months and weighing between 26.9 kg and 27.1 kg ($27.03{\pm}5.05kg$) were used in this experiment. Three diets of varying peNDF content were obtained by original TMR (T1), 12,000 rpm grinding (T2), and 15,500 rpm grinding (T3) of the same TMR diet. The $peNDF_{1.18}$ content of the experimental diets was 23.85%, 21.71%, and 16.22% for T1, T2, and T3, respectively. Results: Average daily gain (ADG) was higher in T2 group compared to those of the control and T3 groups, but ADG and DMI were not affected by the dietary particle size and peNDF content. Also, there was no difference between apparent nutrient digestibility of dry matter, crude fiber, ether extract, neutral detergent fiber, and acid detergent fiber. Although there was no significant difference, rumination and total chewing time were associated with decreased peNDF content. Conclusion: The feeding of peNDF-based TMR showed no impact on apparent nutrient digestibility and nitrogen balance. Further studies are required with a wider range of dietary peNDF level and particle size to better identify the effect of dietary peNDF and particle size on chewing activity and performance in goats.

Regulatory Mechanisms of Angiotensin II on the $Na^+/H^+$ Antiport System in Rabbit Renal Proximal Tubule Cells. II. Inhibitory Effects of ANG II on $Na^+$ Uptake

  • Many reports represent that angiotensin II (ANG II) caused a dose dependent biphasic effects on fluid transport in the proximal tubule. However, respective roles of different signaling pathways in mediating these effects remain unsettled. The aim of the present study was to examine signaling pathways at high doses of ANG II on the $Na^+$ uptake of primary cultured rabbit renal proximal tubule cells(PTCs) in hormonally defined serum-free medium. High concentrations of ANG II $(>10^{-9}\;M)$ inhibited $Na^+$ uptake and increased $[Ca^{2+}]_i\;level$ in the PTCs. However, low concentrations of $(<10^{-11}\;ANG\;II)$ stimulated $Na^+$ uptake and did not affect $[Ca^{2+}]_i\;level$. 8-(N, N-diethylamino)-octyl-3,3,5- trimethoxybenzoate (TMB-8), ethylene glycol-bis$({/beta}-amino\;ethyl ether)-N,N,N'$, N'-tetra acetic acid (EGTA), and nifedifine partially blocked the inhibitory effects of ANG II on $Na^+$ uptake. When ANG II and bradykinin (BK) were treated together, $Na^+$ uptake was further reduced $(88.47{\pm}1.98%\;of\;that\;of\;ANG\;II,\;81.85{\pm}1.84%\;of\;that\;of\;BK)$. In addition, W-7 and KN-62 blocked the ANG II-induced inhibition of $Na^+$ uptake. Arachidonic acid reduced $Na^+$ uptake in a dose-dependent manner. When ANG II and arachidonic acid were treated together, inhibitory effects on $Na^+$ uptake significantly exhibited greater reduction than that of each group, respectively. When PTCs were treated by mepacrine $(10^{-6}\;M)$ and AACOCF3 $(10^{-5}\;M)$ for 1 hr before the addition of $(<10^{-9}\;ANG\;II)$, the inhibitory effect of ANG II was reversed. In addition, econazole $(>10^{-6}\;M)$ blocked ANG II-induced inhibition of $Na^+$ uptake. In conclusion, the $[Ca^{2+}]_i$ (calcium-calmodulin-dependent kinase) and phospholipase $A_2\;(PLA_2)$ metabolites are involved in the inhibitory effects of ANG II on $Na^+$ uptake in the PTCs.

An Experimental Study on Single-dose and 4 Weeks Repeated-dose Toxicity of Aconitum ciliare Decaisne Pharmacopuncture (초오약침의 단회 및 4주 반복 투여 독성에 관한 실험적 연구)

  • Objectives : This study was performed to examine the toxicity of Aconitum ciliare Decaisne pharmacopuncture(ADP). Methods : The toxicity was evaluated for lethal dose for 50 percent kill(LD50), single dose and repeated dose for 4 weeks. Toxic symptoms, weight measurement, hematological test, blood biochemical test, visual examination and weight measurement of major organs and histopathological test were observed for 4 weeks. Dose of 300, 600, 1,200, 2,400, 3,600, 4,800, 6,000, 7,200 mg/kg in the LD50 experiment, 300, 600, 1,200 mg/kg/day in the single experiment, 150, 300, 600 mg/kg/day in 4 weeks experiments were injected into BALB/c mice. The ADP was injected into ST36 of the right leg. Normal saline solution of same volume was used for control group. In 24 hours after the last treatment, blood samples were taken after anesthesia by inhalation of ethyl ether. After that, the BALB/c mice were euthanized. Their heart, lungs, kidneys, liver and reproductive organs were removed and weighed. Histopathological evaluation was also performed. Results : ADP's LD50 was measured at 6,000 mg/kg. In both single and repeated dose toxicity test, no BALB/c mouse died during the experiments. ADP treatment for 4 weeks did not show any significant changes in toxic symptoms, weight measurement, hematological test, blood biochemical test, visual examination and weight measurement of major organs and histopathological test. Conclusions : As a result, ADP's LD50 was 6,000 mg/kg and repeated dose at a concentration of 600 mg/kg or less is considered to be not harmful for clinical treatment.

Synthesis and Characterization of Interfacial Properties of a Cationic Surfactant Having Three Hydroxyl Groups (세 개의 히드록실기를 가진 양이온 계면활성제 합성 및 계면 특성에 관한 연구)

  • In this study, a cationic surfactant BHMAS (N,N-bis-(3'-n-dodecyloxy-2'-hydroxypropyl)-N-methyl-2-hydroxyethylammonium methyl sulfate) having two lauryl and three hydroxyl groups was synthesized by the reaction of n-dodecyl glycidyl ether and 2-aminoethanol followed by the quarternization with dimethyl sulfate. The structure of the product was elucidated by $^{1}H-NMR$ and FT-IR. The CMC (critical micelle concentration) and surface tension of BHMAS at CMC condition were found to be $9.12\;{\times}\;10^{-4}$ mol/L and 28.71 mN/m respectively. Dynamic surface tension measurements using a maximum bubble pressure tensiometer indicated that a relatively long time was required to saturate the interface between air and aqueous surfactant solution. The interfacial tension measured between 1 wt% surfactant solution and n-decane reached an equilibrium value of 0.045 mN/m in 5 min. The adsorption capacity of the synthesized surfactant was observed to be excellent, which suggests that the surfactant can be used as a softening agent during a laundry process.

Effects of Roasted Cassia tora L. Extracts on the Chemical Changes and Microbial Growth (열처리가 결명자의 화학성분 변화 및 추출물의 균체증식에 미치는 영향)

  • The effects of roasting Cassia tora L. were investigated for proximate composition, color, volatile flavor profile, microbial growth and alcohol fermentation. While moisture, protein and fat contents decreased with increasing roasting temperature, fiber and ash contents increased. The L, a and b values of Cassia tora L. extracts decreased with increasing temperature, and only a small difference in total color difference$({\Delta}E)$ was observed. A little difference in major flavor components between raw and roasted treatment was found during roasting. Furfuryl alcohol, a major component of coffee flavor, was separated from Cassia tora L. extracts extracted with ethyl ether. The yeast growth was the highest on the water-extract of Cassia tora L. roasted at $160^{\circ}C$. With increased levels of water-extract at $160^{\circ}C$, S. cerevisiae grew rapidly for 24 hr incubation and the growth rate was higher than the unroasted control group. The growth rate of Bacillus subtilis was the highest in a treatment of 0.5% concentration. Little differences in ${\alpha}-amylase$ produced from Bacillus subtilis were observed among the treatment groups. The total alcohol content increased with increasing roasting temperature during alcohol fermentation.

  • Amphipathic compounds (bis-sulfonate Gemini type) with double or triple long chain alkyl groups were prepared by the reaction of N-(long chain acyl)diethanolamine diglycidyl ethers with fatty alcohols, followed by the reaction with propanesultone. All these new Gemini type surfactants were soluble in water and showed much better micelle forming ability and lowering surface tension than sodium dodecyl sulfonate with one sulfonate group. cmc and ${\Upsilon}$ cmc values of the triple-chain compounds were still much smaller than those of the corresponding double-chain compounds with two common alkyl groups. The efficiency of adsorption at the water/air interface ($pC_{20}$) of these surfactants was very high. Their foaming properties, wetting ability toward a felt chip, and lime-soap dispersing requirement (LSDR) were measured. Their initial foaming properties were high but showed good low foam stability, wettability and LSDR.

The Oxidative Stability of Solvent Extracts of Sea Tangle Powder(STP)and Maejakgwa Made with STP (다시마 용매추출물과 다시마 분말 첨가에 의한 매작과의 산화안정성)

  • The purpose of this study was to investigate the antioxidative effects of Sea tangle powder(STP) solvent extracts as well as Maejakgwa made with STP. The STP solvent extracts were added to soybean oil at a quantity of 0.05%. The solvents used for extraction were methanol, ethanol, ethyl acetate, and petroleum ether. Soybean oil without added STP was used as a negative control, and soybean oil samples containing 0.02% butylated hydroxy toluene(BHT) and $\alpha$-tocopherol were used as positive control, respectively. Each sample was stored at $50^{\circ}C$ for 30 days. The oxidation levels of these samples were determined by measuring their acid values, peroxide values, and thiobarbituric acid(TBA) values. The soybean oil samples containing the STP extracts had lower oxidation levels than both the negative control and $\alpha$-tocopherol positive control, and the sample containing the 0.05% methanol extracts had the lowest oxidation. According to the Rancimat method, the methanol extract(320 min) and ethanol extract(316 min) demonstrated longer induction periods as, compared to the control(253 min), $\alpha$-tocopherol(255 min), and BHT(309 min) samples. For the Maejakgwa, acid values increased over the storage period, however, the samples made with STP had lower values than the control group. Peroxide values increased rapidly for 30 days and then decreased. The TBA values of the Maejakgwa samples made with 3% and 9% STP were lower than those of the 15% STP sample and the control. In conclusion, the oxidative stability of soybean oil containing solvent extracts of STP and Maejakgwa made with STP were increased.

Preparation of MWCNTs/Poly(methyl methacrylate) Composite Particles via the Emulsion Polymerization of Methyl Methacrylate Using MWCNTs Modified by Silanization Reaction and Their Morphological Characteristics (실란화 반응으로 표면 개질된 다중벽 탄소나노튜브(MWCNTs)와 Methyl Methacrylate의 유화중합을 통한 MWCNTs/Poly(methyl methacrylate) 복합 입자 제조 및 그 형태학적 특성)

  • In this study, multi-walled carbon nanotubes (MWCNTs) were oxidized with a mixture of nitric acid and sulfuric acid. After oxidation, oxidized MWCNTs were treated with thionyl chloride ($SOCl_2$) and 1,4-butanediol (BD) in sequence at room temperature to introduce hydroxyl groups on the surface of MWCNTs. The prepared MWCNT-OH was silanized with 3-methacryloxypropyltrimethoxylsilane (MPTMS) to make MWCNT-MPTMS. The MWCNT-MPTMS was used as fillers in emulsion polymerization to make MWCNT-MPTMS/PMMA composite particles with 3 kinds of emulsifiers, hexadecyltrimethylammoniumbromide (CTAB) as a cationic, sodium dodecylbenzene sulfonate (SDBS) as an anionic and polyethylene glycol tert-octylphenyl ether (Triton X-114) as a nonionic emulsifier. Morphologies of composite emulsions were confirmed by a particle size analyzer (PSA) and a scanning electron microscope (SEM). Morphologies of emulsion polymerized MWCNT-MPTMS/PMMA with SDBS showed more uniform particle size distribution compared to those of other two emulsifiers used emulsions. MWCNT-MPTMS/PMMA showed $3.4^{\circ}C$ higher $T_g$ compared to pristine MWCNT/PMMA due to covalent bond formation at interface of MWCNT-MPTMS and PMMA.

Effect of DA-6034, a New Flavonoid Derivative, on TNBS-Induced Colitis in the Rats (새로운 플라보노이드 유도체인 DA-6034의 TNBS 유발성 염증성대장염 모델에서의 치료효과)

  • The efficacy of DA-6034, a new flavonoid derivative, was investigated in comparison with sulfasalazine in a trinitrobenzene sulfonic acid (TNBS)-induced rat colitis. Under light anaesthesia with ether, rats were subjected to intracolonic administration of 30mg TNBS in 50% ethanol (0.5ml) and were then sacrificed at 7 or 21 days after colitis induction. The TNBS control group (the saline treated colitic rat) exhibited ulceration and inflammation of the distal colon with formation of granuloma and pathologic connections. Moreover, an increase in colonic myeloperoxidase (MPO) activity (investigated as an index of leukocyte adhesion and accumulation) and an elevated colonic leukotriene $B_4$ ($LTB_4$) level were observed. The colitic rats received DA6034 (0.3-30mg/kg) or sulfasalazine (50-100mg/kg), prednisolone (0.3-3mg/kg) after the induction of colitis until they were sacrificed. Oral treatment with DA-6034 resulted in significant reductions of macroscopic colonic damage, colonic inflammation. DA6034 had a more potent effect than sulfasalazine and prednisolone on macroscopic colonic damage, while it has similar effect with prednisolone on the reduction of colonic $LTB_4$ synthesis and MPO activity. This study show, therefore, that DA-6034 is effective m attenuating the colonic lesion in an TNBS-induced colitis model. Furthermore, the results suggest that the effect of DA-6034 is partially related to its action on $LTB_4$ synthesis and MPO inhibition.
