Evaluation of Cell Cytotoxicity on the extractives of Magnoliaceae (목련과 수목의 추출성분에 대한 세포독성평가)

  • The 90% methanol extracts of eight magnoliaceae plants were collected and tested the cytotoxicity against SK-OV-3 and SiHa cells. Also six pure compounds such as magnonol, honokiol, dihydroxybiphenyl ether, linodenine, anonaine, asimilobine which were previously isolated from Magnolia obovata Thunb. were evaluated the cytotoxicities and their mechanism study using the Lactate dehydrogenase assay(LDH) and FACScan analysis system. Of the tested six compounds, magnonol, honokiol, dihydroxybiphenyl ether showed high cytotoxicities against human cancer cell lines, SK-OV-3 and SiHa cells. In addition, one of the plausible mechanisms of their antitumor activities suggested that they could induce the early stage of apoptosis. For the quantitative analysis, the methanol extractives were fractionated with chloroform, ethylacetate, $H_2O$ and then the ethylacetate fraction was chromatographed on silica gel using n-Hexane ; Acetone(4:1, v/v) as eluent. This fraction was subjected for the quantitative analysis in the HPLC system. The result suggested that the methanol extractives of Magnolia obovata Thunb. contained with magnonol, honokiol, dihydroxybiphenyl ether, 0.9%, 0.3% and 0.24%, respectively.

Antimicrobial Activity and Antioxidative Activity in the Extractives of Quercus dentata Thunberg (떡갈나무 추출물의 항균활성 및 항산화활성)

  • Antimicrobial activity and antioxidative activity of the organosoluble extractives from the leaves, bark and xylem of Quercus dentata were investigated. Antimicrobial activity was tested by paper disk method and bioautography methods, The most pronounced antimicrobial activities of leaves parts were ethyl acetate solubles fraction against the Klebsiella pneumoniae by the paper disk method. The strongest activities of bark parts were ethyl acetate solubles fraction against Bacillus subtilis, Klebsiella pneumoniae. Ethanol extractives from xylem parts showed high activities against Bacillus subtilis, Staphylococcus aureus, Klebsiella pneumoniae and Vibrio parahaemolyticus. These antimicrobial activities of ethanol extractives from Quercus dentata were in order to xylem > bark > leaves. The strong inhibition zones of the ethanol extractives and its fractions of xylem showed Rf values in 0.41~0.63. In leaves extractives, the petroleum ether solubles showed lower antioxidative activity and ethyl acetate insolubles showed higher antioxidative activity of 70% compared with the $EC_{50}$ values of the control. Antioxidative activity of bark and xylem extractives showed higher approximately 2 times than the control except the petroleum ether solubles.

Extractives from the Needles of Torreya (벼자나무 잎의 추출성분)

  • The needles of torreya(Torreya nucifera S. et Z) were collected, extracted with acetone-$H_2O$(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. The ethylacetate soluble mixture was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Most of the needle extractives were flavan and its methyl ether compounds such as (+)-catechin, (-)-epicatechin, (+)-gallocatechin, (-)-epigallocatechin, 3'-O-methyl-(+)-catechin and 3'-O-methyl-(-)-epicatechin including protocatechuic acid, one of benzoic acids. $^1H$- and $^{13}C$-NMR, NOE and CI-MS analyses were performed to elucidate the structures of the isolated compounds.

  • As a way to revive the traditional technology of Korean golden varnish(Hwangchill), the exudates of D. morbifera, this study was carried out at first to investigate the anatomical characteristics and the chemical composition of D. morbifera stems and their exudates. Trees of more than 20 years old were harvested at Wando and Jejudo islands in the southwestern part of Korean peninsula during different physiological seasons of winter, spring, and summer, The results obtained are as follows: 1. In the anatomical aspect of wood, Hwangchil woods is ring-porous wood, has alternate inter-vessel pittings and horizontal intercellular canal in xylem. 2. In the chemical aspect of wood and bark, the general compositions appeared hot to differ from those of other hardwoods, and ash and alcohol-benzene extractives showed little increase with the increase of atmospheric temperature of harvesting season. 3. In the solvent-sequential extraction of bark, wood and exudate, the exudates was extracted up to 80% by ether, but the bark and wood contained a very small amount of ether extractives, about 2% in the bark and 0.5% in the xylem.

  • In order to elucidate chemical structure of the wood extractives, softwood (Pinus koraiensis Sieb. et Zucc.) meal was extracted with 95% ethanol at room temperature for 72 hours. The extract was fractionated with organic solvents such as n-hexane, ether, ethylacetate, and acetone. From the n-hexane and ether soluble fraction of the wood extractives, four flavonoid compounds were isolated and identified as 5-hydroxy-7-methoxyflavone (I), 5-methoxy-7-hydroxyflavone (II), 5-hydroxy-7-methoxyflavanone (III), and 5-methoxy-7-hydroxyflavanone (IV) by UV, IR, $^1$H-NMR spectroscopy and MS spectrometry.

  • In order to reconstruct the traditional technology of Korean golden varnish(Hwangchil) made from the exudates of D. morbifera, this study in the second step was carried out to analyze the chemical composition and oil properties of the exudates for coatings and bark extractives made from the trees of D. morbifera more than 20 years old grown in Wando and Jejudo islands in the southwestern part of Korean peninsula according to the physiological seasons. The results obtained are as follows: 1. The exudates appeared to be a phytoalexin because it exudated after several days in tapping only in summer season. 2. Both the exudates and bark-extracts with acetone contained 50~60% unsaponifiables and 30~50% saponifiables 3. A major component of essential oil in bark-extractives was ${\beta}$-cubebene and reached to 60~80%, while that of exudates included only 34% ${\beta}$-cubebene and some other compounds such as ${\beta}$-cadinene of 12%, aromadendrene of 9%, ${\beta}$-selinene of 9%, CAS030021-74-0 of 10%, etc. in GC-MS spectrometer. 4. A mjor component of fatty oil fraction in bark-extractives was linoleic acid and reached to more than 60%, while the exudates had mostly other components such as terpenes and phenolics instead of fatty acids accordint to GC-MS spectrometer. 5. Iodine value of samples after oil refining had 214mg/g in the exudates and more than than 150mg/g in bark-extractives, so the latter belonged to a drying oil.. 6. Therefore, the exudates from D. morbifera for traditional Korean golden varnish seems to have beeen used to a good varnish because it has some specific compounds different from its bark-extractives or general varnishes.

  • 2kg of pollen extracted with EtOH(98%), concentrated, and fractionated with a series of hexane, CH$_2$Cl$_2$ EtOAc and $H_2O$ on a separately funnel. Each fraction was freeze dried to give dark-brown powder and EtOAc soluble portion of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents. Spectrometric analysis such as NMR and FAB-MS including TLC were performed to characterize the structures of isolated compounds. Pollen contained a small amount of flavonol derivatives such as quercetin-3-O-$\beta$-D-glucopyranoside and kaempferol-3-O-$\beta$-D-rutinoside in addition to a small amount of flavanonol compound such as aromadendrin-5-methyl ether and acid compound such as.

  • The chemical compositions of the root bark of Ulmus species and mucilage were investigated. The methanol extractives were fractionated with n-hexane, ethyl ether, ethyl acetate and water and antibacterial activities of each fraction were investigated. Although differences of chemical compositions of the Ulmus species were not noticeable, nitrogen and lignin contents of xylem were higher than those of phloem. However, ash and extractive contents showed adverse results. The mucilage of root bark was slowly gushing out by water and similar to pectin substance with high acidic sugar content. In fractionation experiment, the yield of methanol extraction was about 7% and the portion of water-soluble materials in methanol extractives was 78%. These water- soluble materials had the most high antibacterial activity on Staphylococcus aureus and Salmonella typhimurium.

  • Xylem, bark and flower buds of Magnolia kobus DC. var. borealis Sarg. were extracted with ethanol. And then the ethanol extracts were fractionated with diethyl ether. The lignans structure of the diethyl ether fractions were determined by spectroscopic methods. The results are summarized as follows : 1. Three compounds, aschantinn(X-I), yangambin(X-II), (-)syringaresinol(X-III), tetrahydrofurofuranoid lignans were isolated from the xylem. 2. Five compounds were isolated from the bark. These are all tetrahydrofurofuranoid lignans, aschantin(B-I), yangambin(B-II), medioresinol(B-III), (+)syringaresinol(B-IV), and tetrahydrofuranoid, kobusinol B(B-V). 3. Three tetrahydrofurofuranoid lignans, kobusin(F-I), aschantin(F-II), yangambin(F-III) were isolated from the flower buds. 4. Isolated lignans from the each tissue were two types of tetrahydrofurofuran type and tetrahydrofuran type. 5. The tetrahydrofurofuranoid lignans were five compounds such as kobusin, aschantin, yangambin, medioresinol, and syringaresinol, while the tetrahydrofuranoid lignans was kobusinol B. 6. Most of these lignans were substituted with the methylenedioxy or methoxyl groups.

  • The methanol extractives from root bark of Ulmus davidiana var japonica nakai were fractionated with n-hexane, ethyl ether, ethyl acetate and waster, the water soluble fraction was also fractionated with silicagel column chromatograhy. The chemical structure of purifided compounds were identified with UV, IR, 1H-NMR and 13C-NMR spectra and the antibacterial activities also were investigated. Two different antibacterial compounds (compound A and B) were fractionated with silicagel chromatography and TLC. Compounds B was identified as a catechin rahmnoside. The both of compounds had antibacterial activity on Staphylococcus aureus and Salmonella typhimurium.
