  • For the purpose of evaluating protective components against alcohol-induced toxicity, the active components enhancing alcohol metabolism was pursued from water soluble fraction by ethanol precipitation and DEAE-cellulose chromatographic technique. As a result, various high molecular weight fractions from Aloe vera and Aloe arborescens, on a single oral administration in rats were found to cause a significant decrease in the blood ethanol concentration as well as enhancement of liver cytosolic ADH and ALDH activities and among which, a strong acidic high molecular weight fraction was demonstrated to exhibit the most potent enhancing activity on ethanol metabolism.

Antioxidant and Suppressive Effects of Ethanolic Extract Fractions from Safflower (Carthamus tinctorius L.) Flower on the Biosynthesis of Inflammatory Mediators from LPS-stimulated RAW 264.7 Cells

  • The aim of this study was to elucidate the anti-inflammatory activity of safflower (Carthamus tinctorius L.) ethanolic extract fractions (CFEFs). Butanol fraction had the strongest antioxidant activity, and all CFEFs, except for chloroform fraction, partly inhibited lipopolysaccharide (LPS)-induced nitrite production in RAW 264.7 cells. In the cell-free system, hexane and butanol fractions chemically quenched nitric oxide (NO). In addition, the iNOS mRNA transcription was suppressed by ethanol extract and hexane fraction in LPS-stimulated RAW 264.7 cells. Taken together, the inhibitory effect of CFEFs on NO production from LPS-stimulated RAW 264.7 cells, might be due to both the chemical NO quenching activity and the suppression of iNOS mRNA transcription partially. The synthesis of prostaglandin $E_2$ ($PGE_2$) was potently inhibited by ethanol extract to below basal label, and the transcription of cyclooxygenase-2 (COX-2), an enzyme involving in $PGE_2$ synthesis, was partially suppressed by ethanol extract and hexane fraction. Based on these results, CFEFs may be useful as an alternative medicine for the relief and retardation of immunological inflammatory responses through the reduction of inflammatory mediators, including NO and $PGE_2$ production.

들기름에 대한 칡추출물 분획의 항산화 효과 (Antioxidant Effect of Ether and Ethylacetate Fractions of Pueraria thunbergiana Extract on Perilla Oil)

  • The objective of this study was to determine antioxidant effect of ether and ethylacetate fractions of 70% ethanol extract of some food (acid treated or not) on perilla oil. Each fraction of food extract was added to perilla oil and stored for 0,3,6.9,11 days at 60$^{\circ}C$. Then, the peroxide value (POV) of perilla oil samples were analyzed. Perilla oil contained ${\gamma}$-tocopherol 0.6800 $\mu\textrm{g}$/mg, ${\alpha}$-tocopherol 0.3189 $\mu\textrm{g}$/mg. $\delta$-tocopherol 0.0463 $\mu\textrm{g}$/mg, but it was easily oxidized due to high linolenic acid content. To increase yield of ether and ethylacetate fractions from each food extract, the 70% ethanol extract was treated with 0.2% H$_2$SO$_4$ and fractionized by ether and ethylacetate. Among ether and ethylacetate fractions of 70% ethanol extracts of some food, the yield of ethylacetate fraction of acid treated Pueraria thunbergiana extract was 5 times more than that of ethylacetate fraction untreated with acid. Perilla oil which added 100 ppm ethylacetate fraction of acid treated Pueraria thunbergianan extract showed low POV (44.8 meq/kg) compared to POV (80.0 meq/kg) of control.

해삼의 생리활성물질 추출 용매분획에 따른 연구 (A Study on the Solvent Fraction for Extraction of Bioactive Substances in Sea Cucumber)

  • 본 연구에서는 해삼속의 폴리페놀과 플라보노이드의 생리활성물질을 추출하기 위한 용매분획의 수율을 확인하고자 연구를 시도하였다. 이미 보고된 사례에서 50%에탄올 추출 용매분획은 해삼 항산화 물질의 높은 수율 결과로 확인되었다. 해삼의 항산화물질 추출량을 결정짓는 것은 추출에 적용된 추출 용매분획의 결과로 확인되었다. 또한 '추출용매에 따라 해삼생리활성 항산화물질 추출함량이 크게 차이가 있다.'는 선행연구를 통해 용매분획추출에 관한 필요성의 결과를 얻었다. 50%에탄올 추출 용매분획의 해삼추출물에 포함된 페놀물질의 높은 항산화성분 추출 결과가 증명되었다. 추출용매분획 연구사례에서 수율이 가장 저조한 아세트산에틸 용매분획은 다른 용매분획에 비해 높은 페놀함량을 수확하여 항산화효과가 확인되었다. 이에, 추출 용매분획물의 적용에 따른 수율변화를 통해 항산화추출물의 높은 수율에 미치는 영향을 확인 하였다. 따라서 본 연구를 통해 50%에탄올 용매의 최적화된 해삼생리활성물질 추출 용매분획으로 검증되었다.

  • Rate Constants for the solvolysis of p-methylbenzyl chloride in various ethanol-water mixtures were studied at 30 and $40^{\circ}C$ under various pressures up to 1600 bar. The rates of reaction were increased with increasing temperature and pressure, and decreased with increasing solvent composition of ethanol mole fraction. From the rate constants, the values of the activation parameters (${\Delta}V^{\neq},{\Delta}{\beta}^{\neq},{\Delta}H^{\neq}\;and\;{\Delta}S^{\neq}$) were evaluated. The values of ${\Delta}V^{\neq}\;and\;{\Delta}{\beta}^{\neq}$ exhibit the extremum behavior at about 0.30 mole fraction of ethanol. This behavior is discussed in terms of solvent structure variation (electrostriction). From the relation between the reaction rate and the dielectric constant of solvent or the number of water molecule participated in the transition state, it could be postulated that the reaction proceeds through $S_N1$ mechanism.

우리밀 Ethanol-Acetic Acid 추출물에 함유된 대식세포의 식작용 활성 증강 물질의 분리.정제 (Purification of Macrophage Phagocytic Activity-Enhancing Component from Ethanol-Acetic Acid Extract of Korean Wheat)

  • 본 연구에서는 우리밀로부터 대식세포 식작용 활성화시키는 물질을 탐색하는 과정에서 고분밀의 EA 추출물이 가장 높은 식작용 활성 증강효과를 보임을 알았고, EA 추출물을 정성한 후 순수 분리 정제하여 대식세포 활성 증강효과를 나타내는 단일 물질을 찾고자 하였다. TLC 방법을 이용하여 ASW와 고분밀의 EA 추출물을 정성한 결과 약 5개 정도의 band를 확인한 후, column chromatography를 이용하여 EA 추출물을 분획하였다. 1차 column chromatography만으로는 single fractionation이 어려워 분획물을 fraction A, B, C, D로 분류하여 식작용 활성 증강효과를 측정하였다. 대분류된 각 fraction의 식작용 활성 증강효과를 측정한 결과 고분밀의 fraction b가 대부분의 활성을 가진 분획으로 사료되었다. 2차 column chromatography를 통해 fraction b만응 분리 하였으며, 구조결정 실험과 동물실험을 추가로 시행하여 fraction b의 효과를 입증할 것이다.

Effects of Chenopodium album Linne on Gastritis and Gastric Cancer Cell Growth

  • In our previous study, we investigated Chenopodium album Linne (CAL) ethanol extract and its fractions on anti-gastritic actions using the HCl/ethanol and indomethacin induced gastric lesion model and Helicobacter pylori (H. pylori). Based on the results, butanol fraction was most effective among fractions obtained from CAL. This study aims to elucidate the mechanisms of butanol fraction, and betaine as a constituent of the butanol fraction, on gastritis and anti-gastric cancer cell growth. First, we examined antioxidant properties using hydrogen peroxide and superoxide radical, and we found that butanol fraction and betaine may be good antioxidants. Second, cytotoxicity was assessed by measuring cell viability and 4,6-diamidino-2-phenylinodole dihydrochloride (DAPI) staining of human gastric cancer cells (AGS cells). We also examined the relationship between the cytotoxicity and intracellular $Ca^{2+}$ signaling mechanism. The butanol fraction demonstrated cell viability 71.49% at the concentration of 100 ${\mu}g/ml$ and increased intracellular $Ca^{2+}$ concentration in a dose dependent manner. Finally, we observed the mucus content as a defensive factor and gastric secretion as an aggressive factor, and found that the mucus content noticeably increased when treated with butanol fraction and betaine and gastric secretion decreased when treated with betaine in vivo study. From these results, we suggest that CAL butanol fraction and betaine may have protective effects on gastritis.

우엉 뿌리의 항혈전 및 항산화 활성 (Anti-thrombosis and anti-oxidative activity of the root of Arctium lappa L.)

  • 식용 및 약용으로 이용되고 있는 우엉 지하부의 혈전 관련 활성과 항산화 활성을 평가하고자, 우엉의 ethanol 추출물 및 이의 순차적 유기용매 분획물을 조제하여 혈액응고 저해 활성, 혈소판 응집저해 활성, 인간 적혈구 용혈활성 및 in-vitro 항산화 활성을 평가하였다. 먼저 우엉 추출 수율은 10.94%로 다른 식용 및 약용식물보다 높았으며, ethanol 추출물의 경우 total polyphenol 함량은 5.01 mg/g으로 낮았으나, 694.53 mg/g의 높은 총당 함량을 나타내었다. 순차적 유기용매 분획의 경우, n-hexane, ethylacetate(EA), n-butanol 분획 수율은 각각 1.62, 0.42 및 5.98%로 나타났으며, 물 잔류물은 85.38%였다. 우엉 시료들의 혈액응고 저해활성을 TT, PT, aPTT를 측정한 결과, ethanol 추출물에서는 유의적인 활성이 나타나지 않았으나, EA 분획에서 강력한 TT, PT, aPTT 연장효과를 확인하였으며, 농도 의존적 혈액응고 저해활성을 확인하였다. 한편 물 잔류물에서는 내인성 혈액응고인자 활성화에 의한 aPTT 감소효과를 확인하여 혈전생성 촉진효과가 있음을 확인하였다. 혈소판 응집저해 활성평가의 경우, 우엉 ethanol 추출물 및 분획물들은 임상에서 사용하는 항혈소판제인 아스피린보다 강력한 저해효과를 나타내었으며, 특히 EA 분획은 정제되지 않은 상태에서도 아스피린의 2배 이상의 강력한 혈소판 응집저해능을 나타내었다. 또한 항산화 활성 평가 결과, EA 분획물은 DPPH 음이온, ABTS 양이온, nitrite에 대해 우수한 소거능 및 환원력을 나타내었다. 또한 우엉 추출물과 분획물들은 0.5 mg/mL 농도까지 인간 적혈구에 대한 특이한 용혈활성을 나타내지 않았다. 상기 결과들은, 우엉 EA 분획물이 천연물 유래의 안전한 항혈전제로 개발 가능함을 제시하고 있다.

복분자 에탄올 추출물 및 분획물의 항산화 효과 (Antioxidant Capacity of Ethanol Extracts and Fractions from Rubus coreanus Miq.)

  • 본 연구는 복분자 에탄올 추출물과 용매 분획물의 총 폴리페놀 함량 및 총 플라보노이드 함량과 라디칼 소거능과 환원력을 이용한 항산화 활성 측정하였다. 총 폴리페놀 함량은 70% 에탄올 추출물 238.46 mg GAE/g 으로 나타났으며, ethyl acetate 분획물 413.59 mg GAE/g 으로 다른 분획물보다 높은 함량을 나타내었다(p<0.05). 또한 총 플라보노이드 함량도 ethyl acetate 분획물에서 52.83 mg QE/g으로 높은 함량을 나타내었다(p<0.05). DPPH 라디칼 소거능과 ABTS 라디칼 소거능의 경우 ethyl acetate 분획물의 RC50값이 각각 3.95와 6.25 ㎍/mL 으로 대조군인 ascorbic acid와 유사한 소거능을 나타내었다(p<0.05). FRAP에 의한 환원력 측정 결과에서도 ethyl acetate 분획물에서 높은 활성을 나타내었다(p<0.05). 이상의 결과 복분자 70% 에탄올 추출물과 용매 분획물 중 ethyl acetate 분획물이 다량의 폴리페놀과 플라보노이드를 함유하고 있으며, 항산화 효과가 높게 나타났다. 따라서 본 연구의 결과는 복분자를 기능성 신소재 개발 원료로 활용하는데 기초 자료가 될 것으로 기대된다.

원료백미의 분쇄정도가 무증자 당화 탁주의 발효에 미치는 영향 (Effect of Ground Rice Particle Size on the Brewing of Uncooked Rice Tsckju)

  • Uncooked rice Tackju was prepared from the groud rice with different particle size such as 12, 20, 35 and 50 mesh, and its characteristics were analysed during the brewing. Reducing sugar and ethanol contents as well as yeast cell counts were the highest in main mash and Tackju prepared from 20 mesh fraction. Especially, reducing sugar content was about. 1.5-2.5 times as much as that obtained from 35 or 50 mesh fraction. The final concentration of ethanol in main mash prepared from 20 mesh fraction was 14.8%(v/v). It was about 1.5%(v/v) higher than the lowest one [13.35%(v/v)] which was prepared from 12 mesh fraction. No significant difference In total acidity was observed. Sensory evaluation revealed 20 or 35 mesh Tackju to be the best in overall preference, especially in sweet and refreshing tastes.
