소어분(小魚粉)의 주정추출성분(酒精抽出成分)이 백서(白鼠)의 핵산대사(核酸代謝)에 미치는 영향 (The Effect of Ethanol Extracts from Fish Flour on the Nucleic Acid Metabolism in Rats)

  • This study was designed to observe the effect of ethanol extracts from fish flour on the nucleic acid metabolism in rats. Young rats, weighing 75-85g were used as the experimental animals and diet used were 8 kinds; diet supplemented with 10% fish flour, diets which were supplemented with the extracts and or remainders of fish flour after extracting by either 76% or 96% ethanol to the rice diet, respectively, and diet supplemented with 6% casein. After feeding corresponding diet for 40 days, RNA and DNA contents, and DNase activities in the liver, kidney and braid were determined. The results obtaioed from this study are summarized as follows: 1. The RNA contents of the ethanol-treatment groups are, in the liver and kidney, similar to, and in the brain, generally higher than, that of the control group. 2. The DNA contents of each organ show no difference between ethanol-treatment groups and control group, but in the liver, of ethanol extrat groups are lower than casein group. 3. the DNase activity of each organ in the ethanol-treatmeut groups, is generally lower than the control group. The above results indicate that ethanol extracts from fish flour have influence on the nucleic acid metabolism.

$\beta$-Carotene 첨가식이가 알코올성지방간 쥐의 지질과산화물 수준과 항산화효소 활성에 미치는 영향 (Effects of $\beta$-Carotene Supplementation on Lipid Peroxide Levels and Antioxidative Enzyme Activities in Alcoholic Fatty Liver Rats)

  • This study was performed to investigate the effect of dietary $\beta$-carotene supplementation on lipid peroxide levels and antioxidant enzyme activities in alcoholic fatty liver rats. Forty five Sprague-Dawley male rats aging 8 weeks were used as experimental animals, which were divided into the control diet (CD) and the ethanol diet (ED) and the ethanol + $0.02\%$ $\beta$-carotene diet (EPD) groups and fed the experimental diet respectively for 5 weeks. After the feeding, rats were sacrificed to get blood and liver to analyze lipid and lipid peroxide levels and antioxidant enzyme activities. The mean body weight and food intake of the ethanol diet group was significantly lower than that of the control diet. The liver index (LI) of the ethanol diet group was significantly higher than those of the control diet and the $\beta$-carotene supplementation group. Serum levels of total lipid, triglyceride of the ethanol diet group were significantly higher than those of the control diet and the $\beta$-carotene supplementation group. Total cholesterol levels were not significantly different among all groups. HDL-cholesterol of the ethanol diet group was significantly lower than those of the control diet and the $\beta$-carotene supplementation group. Liver TBARS of the ethanol diet group was significantly higher than those of the control diet and the $\beta$-carotene supplementation group. Liver lipofuscin and conjugated diene levels were not significantly different among all groups. The superoxide dismutase activity of the ethanol diet group was significantly lower than those of the control diet and the $\beta$-carotene supplementation group. Catalase and glutathione peroxidase activities were not significantly different among all groups. Because v-carotene supplementation significantly decrease the serum total lipid, triglyceride, liver TBARS revels and increase the superoxide dismutase activity in alcoholic ratty liver rats, $\beta$-carotene supplementation seems to give beneficial effect for the alcoholics.

에탄을 공급이 흰쥐 조직중의 Glutathione 및 지질산화 수준에 미치는 영향 (Effects of Ethanol Administration on Glutathione and Lipid Peroxide Levels in Rat Liver and Cerebellum)

  • 에탄올의 급성적 및 만성적 투여가 흰쥐의 간 및 소뇌 중의 glutathione(GSH) 양상과 지질과산화물 수준에 미치는 영향을 알아 본 결과는 다음과 같다. 간조직에서는, 만성적 에탄을 투여 (6~9g/kg, per day, 10% 음료수로서, 4주간)에 의해 총 GSH 농도가 14. 5% 저하되었고, 산화형 GSH(GSSG)는 변화가 없었으며, 따라서 GSSG/총 GSH 비율은 증가하였다. 그러나 지질과산화 수준은 변함이 없었다. 급성적 에탄올 투여에 의해 간 조직 중 지질과산화 수준이 상승되고 총 GSH 농도가 감소함은 이미 보고되고 있다. 본 실험에서는 이와 관련시켜 급성적 에탄을 투여 (50mmole/kg, i.p.)후 post-hepatic 혈장 중의 총 GSH 수준을 측정한 결과 현저히 상승하였다. 이러한 GSH의 간에서 혈액으로의 유출은, GSH의 항산화적 소모 이외에도, 급성적 그리고 아마도 만성적인 에탄을 투여에 의한 간 조직 중의 총 GSH의 감소의 한 가능한 원인으로서 간접적으로나마 추정될 수 있겠다. 소뇌에서 는 급성적 에탄을 투여는 지질과산화 수준을 증가시켰으나 GSH 양상은 변화시키지 않았으면, 만성적인 경우엔 모두 변동시키지 못하였다.

뇌 조직에서 알코올 투여에 대한 녹차 건분의 항산화 효과 (Antioxidative Effects of Green Tea Powder Diet Against Ethanol-Induced Oxidative Damage in Rat Brain Regions)

  • The present study investigated the protective effects of green tea against acute ethanol-induced lipid peroxidation and the change of antioxidative enzyme activities in various regions of rat brain : cortex, cerebellum, striatum and hippocampus. The following parameters were examined : malondialdehyde(MDA) levels and activities of superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px). Male Sprague-Dawley rats were given the experimental containing 1% green tea powder or control diet for 4 weeks, and at the end of feeding diet group received acute ethanol(5g/kg body weight) or equicaloric sucrose solution intragastrically. Green tea powder significantly decreased MDA levels in the striatum compared to control-non alcohol treated group to 1% green tea-non alcohol treated group without altering the antioxidative enzyme activities. Green tea resulted in a significant increase in GSH-Px activities in the hippocampus compared to either control-non alcohol treated group(0.043units/mg protein) or 1% green tea-non alcohol treated group(0.071units/mg protein). In conclusion, these results suggest that moderate consumption of green tea leaves can exert protective effects against ethanol-induced oxidative stress in brain regions, by reducing MDA concentrations in the striatum and enhancing GSH-Px activities in the hippocampus. How to Treat Peripheral Arteriovenous Malformations

  • Arteriovenous malformations (AVMs) are direct communications between primitive reticular networks of dysplastic vessels that have failed to mature into capillary vessels. Based on angiographic findings, peripheral AVMs can be classified into six types: type I, type IIa, type IIb, type IIc, type IIIa, and type IIIb. Treatment strategies vary with the types. Type I is treated by embolizing the fistula between the artery and the vein with coils. Type II (IIa, IIb, and IIc) AVM is treated as follows: first, reduce the blood flow velocity in the venous segment of the AVM with coils; second, perform ethanol embolotherapy of the residual shunts. Type IIIa is treated by transarterial catheterization of the feeding arteries and injection of diluted ethanol. Type IIIb is treated by transarterial or direct puncture approaches. A high concentration of ethanol is injected through the transarterial catheter or direct puncture needle. When the fistula is large, coil insertion is required to reduce the amount of ethanol. Type I and type II AVMs showed the best clinical results; type IIIb showed a satisfactory response rate. However, type IIIa showed the poorest response rate, either alone or in combination with other types. Clinical success can be achieved by using different treatment strategies for different angiographic AVM types.

고정화 Pichia stipitis 를 이용한 글루코오스/자일로오스 혼합당으로부터 에탄올 생산 (Ethanol Production with Glucose/Xylose Mixture by Immobilized Pichia stipitis)

  • 리르노셀룰로오스로부터 생산된 글루코오스와 자일로 오스의 혼합당을 동시에 발효하여 에탄올 생산을 증가시키며, 또한 에탄올 생산에서의 세포고정화의 영향과 ICR (immobilized cell reactor)을 이용한 혼합당에서의 에탄올 연속생산을 수행하였다. 고정화 P. stipitis를 이용한 플라스크에서 에탄올을 생산에 대한 혼합당과 질소원의 영향으로부터 5% 혼합당 (글루코오스/자일로오스 = 3:1)과 1% 질소원이 최적으로 타나났으며, 이때 생산된 에탄올 농도는 약 19-20 g/L이었다. 고정화된 P. stipitis을 이용하여 반복적 유가식배양 (repeated fed-batch)으로 에탄올을 생산하였을 때는 모든 당 농도에서 글루코오스는 빠르게 소비되었지만, 혼합당의 농도가 높아질수록 자일로오스의 소비속도는 점차적으로 감소하였다. 즉 혼합당 농도가 증가하면서 더불어 당 소비속도는 감소하였다. 또한 ICR에서 1% 혼합당을 연속적으로 공급하면서 에탄올을 안정적으로 생산하여, 에탄올 농도는 5.6 g/L이었고 에탄올 생산 속도는 0.13 g/$L{\cdot}h$이었다.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. 흰쥐의 에탄올성 간장해에 미치는 식이 단백질과 섬유소의 영향 (Effect of Dietary Protein and Fiber on Ethanol-induced Hepatotoxicity in Rats)

  • 체내에서 영양소의 흡수에 영향을 미치는 에탄올과 섬유소 및 단백질의 급여수준에 따른 이 들 상호작용을 구명하기 위해 에탄올을 투여한 횐쥐에게 단백질 급여 수준을 7%와 20%로 달리 하고 섬유소를 5%와 10% 첨가한 실험식이를 5주간 급여하므로써 이들 영양소가 에탄을 대사효소와 항산화 물질에 미치는 영향을 관찰하였다. Aldehyde dehydrogenase(ADH)와 microsomal ethanol oxidizing system(MEOS) 활성은 에탄을 투여시 대조군에 비해 유의적으로 증가하였으며, 특히 MEOS 활성은 단백질 정상 급여와 정상섬유소 급여시 그 증가가 현저하였다. 에탄을 투여로 감소된 aldehyde de-hydrogenase(AIDH) 활성은 정상섬유소군의 경우 단백질 적정 급여시 유의적인 감소를 나타내었다. Cytoch-rome P-450(P-450) 활성은 에탄을 투여로 증가되었으며 에탄을 투여군에서 단백질 정상급여와 과량의 섬유소 급여로 유의하게 감소되었다. Xanthine oxidase (XO) 활성은 에탄을 투여로 증가하는 경향이었으며, 저 단백군에 비하여 단백질 적정 급여시 유의적인 증가를 나타내었다. 또한 섬유소 급여수준에 따른 XO 활성은 에탄올 투여군 중 단백질 정상군에서만 유의적인 차이가 관찰되었다. 에탄을 투여는 간조직 중의 글루타티온 함량을 유의적으로 감소시켰으며, 단백질 결핍에 따른 영향이 현저하게 나타났다. 간조직 중의 지질과 산화 함량은 단백질 결핍시 에탄올 투여로 증가하였다. 이상의 결과에서 에탄올을 해독키 위하여 간의 ADH와 MEOS 활성이 증가되었으며 이로 인해 생성된 아세트알데히드는 AIDH 보다 XO를 통해 해독된 것으로 나타났다. 또한 에탄올 대사효소 활성에 요구되는 단백질 공급과 10% 즉, 과량의 섬유소 급여 보다 5% 섬유소 급여시 에탄올성 간손상을 경감시킬 수 있을 것으로 사료된다.

인삼사포닌이 동물생체의 주정대사에 미치는 영향 (Effect of Ginseng Saponin on Alcohol Metabolism in the Animal Body)

  • Unlike carbohydrats and fats, alcohol is essentially foreign to the body and it is known that the body get rid of it by oxidizing alcohol maily in the liver. Acetaldehyde is produced during ethanol metabolism and is known to be oxidized mainly by aldehyde dehydrogenase (ALDH). ALDH activity was found mainly in the mitochondrial fraction but a significant ALDH activity was also present in microsomal and cytosol fraction. Wistar rats (150~200 g, male) were given freely with 12% ethanol (Control) and/or 12% ethanol containing 0.1% ginseng saponins (Test) instead of water for 6 days and the liver was analyzed. ALDH activities of both control and test group were lower than that of normal group but test AkDH was less inhibited than control. ADH activies of both control and test were slightly higher than that of normal group but our previous data showed that it became gradually steady after prolonged ethanol feeding. MEOS activities of both control and test group were much higher than that of normal group. MEOS enzymes are inducible but the activity of test group was greatly higher than that of control. Ethanol containing [1-i4C] ethanol (5 $\mu$Ci) was injected to the above three groups and 30 min later, the distribution of radioactivity of hepatic lipids was investigated. Radioactivities of hepatic lipids of both control and test group were higher than that of normal group, however, that of test group was much lower than that of control. Analysis of individual lipids showed that phospholipid biosynthesis was significantly impaired and fatty acid and triglycerides biosynthesis were greatly stimulated. However, it was realized that the saponin prevented phospholipid biosynthesis depression and the increase of triglyceride biosynthesis considerably. It seemed that the saponin might stimulate ADH, ALDH and MEOS and the acetaldehyde formed would be removed faster. The excess hydrogen can be shunt more quickly into lipid biosynthesis. Electron microscopic observation showed that the hepatic cell of control group was si gnificantly damaged. Mitochondria were swollen and rough endoplasmic reticulum were dilated, however, hepatocytes of test group were not damaged.

Chronic saponin treatment attenuates damage to the pancreas in chronic alcohol-treated diabetic rats

  • Background: Chronic heavy alcohol consumption may raise the risk of developing type 2 diabetes mellitus. Saponins inhibit apoptosis of pancreatic islet cells and reduce lipid parameters. The present study was designed to investigate the effect of saponin on chronic ethanol-treated diabetic rats. Methods: Long-Evans Tokushima Fatty (LETO) and Otsuka Long-Evans Tokushima Fatty (OLETF) rats were pair-fed a Lieber-DeCarli diet with and without 5% ethanol for 12 wks. Two weeks after starting the pair-feeding with the Lieber-DeCarli diet, intraperitoneal injection of saponin was performed for 10 wks. To perform the experiments, rats were divided as follows: LETO-Control (LC), LETO-Ethanol (LE), LETO-Ethanol-Saponin (LES), OLETF-Control (OC), OLETF-Ethanol (OE), and OLETF-Ethanol-Saponin (OES). Results: The weights of epididymal and mesenteric fat tissue in LES and OES rats were the lightest from among the LETO and OLETF groups, respectively. The secretion of alanine aminotransferase and cholesterol in OES rats decreased significantly compared to their secretion in OC and OE rats, respectively. The islets of the pancreas in LE and OE rats showed clean, unclear, and smaller morphology compared to those of LC, LES, OC, and OES rats. In addition, the expression of insulin in the islets of the pancreas in LC, LES, OC, and OES rats was higher than in LE and OE rats. Conclusion: Saponin may not only be helpful in alleviating the rapid progress of diabetes due to chronic alcohol consumption in diabetic patients, but may also show potential as an antidiabetic drug candidate for diabetic patients who chronically consume alcohol.
