Antioxidant and Antimicrobial Activity of Rosa multiflora Thunberg Fruits Extracts

  • In this study, we selected some material to have potential bioactivity from natural plants, confirmed as basic data for industrializing and tried to develope the food materials using them. DPPH, ABTS, antioxidant protection factor, TBARs and antimicrobial activity of extracts from Rosa multiflora Thunberg fruits were determined. The total phenolics extracted from Rosa multiflora were 12.08, 11.82, 11.1 and 12.6 mg/g when using water, 70% ethanol, 70% methanol and 70% acetone as the solvent, respectively. The optimum conditions for extracting the phenolic compounds were 70% ethanol over for 12 hrs(11.82 mg/g). The electron donating ability and inhibition rate on ABTS of the 70% ethanol extracts were 97% and 92.2%, respectively while the antioxidant protection factor(PF) of the water extracts and 70% ethanol extracts were 1.79 and 1.34 PF, respectively. The TBAR (thiobarbituric acid reactive substance) value were $1.3{\mu}M$ for the control and $0.15{\mu}M$ for the 70% ethanol extracts. The inhibitory activity against ${\alpha}$-amylase was 26% for the 70% ethanol extracts. The 70% ethanol extracts from Rosa multiflora Thunberg fruits exhibited antimicrobial activity against H. pylori, S. epidermidis, S. aureusand and E. coli with clear zone diameters of 14, 25, 14 and 13 mm, respectively when using $200{\mu}g/mL$ of the phenolic compounds. An HPLC analysis identified 6 major phenolic metabolites in the Rosa multiflora Thunberg fruits extracts: rosmarinic acid, caffeic acid, chlorogenic acid, courmaric acid, protocatechuic acid and quercetin. In particular, the content of rosmarinic acid was the highest in the 70% ethanol extracts. Therefore these results indicate that 70% ethanol extracts from Rosa multiflora Thunberg fruits can be useful as a natural antioxidant and in functional foods.

소어분(小魚粉)의 주정추출성분(酒精抽出成分)이 백서(白鼠)의 핵산대사(核酸代謝)에 미치는 영향 (The Effect of Ethanol Extracts from Fish Flour on the Nucleic Acid Metabolism in Rats)

  • This study was designed to observe the effect of ethanol extracts from fish flour on the nucleic acid metabolism in rats. Young rats, weighing 75-85g were used as the experimental animals and diet used were 8 kinds; diet supplemented with 10% fish flour, diets which were supplemented with the extracts and or remainders of fish flour after extracting by either 76% or 96% ethanol to the rice diet, respectively, and diet supplemented with 6% casein. After feeding corresponding diet for 40 days, RNA and DNA contents, and DNase activities in the liver, kidney and braid were determined. The results obtaioed from this study are summarized as follows: 1. The RNA contents of the ethanol-treatment groups are, in the liver and kidney, similar to, and in the brain, generally higher than, that of the control group. 2. The DNA contents of each organ show no difference between ethanol-treatment groups and control group, but in the liver, of ethanol extrat groups are lower than casein group. 3. the DNase activity of each organ in the ethanol-treatmeut groups, is generally lower than the control group. The above results indicate that ethanol extracts from fish flour have influence on the nucleic acid metabolism.

  • 추출조건에 따른 지골피의 몇가지 물리적 성질 및 화학성분조성을 조사하였다. 1. 고형분과 총당함량은 물추출이 에탄올 추출물보다 높았으나 반면에 탄닌산함량은 에탄올 추출물에서 높았다. 2. 지골피 물추출물이 탁도와 갈색도가 에탄올추출물보다 높았으며 색도 중 적도와 황색도는 유사하였다. 3. 용매별 분획추출결과 수율은 물 > n-부탄올 > 에틸에텔 > 초산에 틸 분획순으로 증가하였다.

가압형 마이크로웨이브 추출장치를 이용한 가용성 인삼성분의 추출조건 최적화 (Optimization of Extraction Conditions for Soluble Ginseng Components Using Microwave Extraction System under Pressure)

  • Microwave extraction system equipped with closed vessels was applied to confirm its capability of extracting ginseng solubles including saponin, which was monitored to optimize extraction con ditions by response surface methodology. Total yield increased with the decrease in particles size and more extraction steps. Soluble components were completely extracted by operating the system within 6 min per one step, which should be repeated by 3 or 4 times. Optimized conditions for maxi mum extraction of response variables(total yield, crude saponin) were 40~50% of ethanol concentration, about 140oC of extraction temperature, and within 6 min of the extraction time. The extraction effi ciency of total yield was dependent on the decrease of ethanol concentration, while crude saponin content was favored on the higher degree of ethanol concentration. According to the temperature guideline, below 90oC of extraction temperature, the optimal ranges of extraction conditions were predicted as 30~50% of ethanol concentration and 2.5~6.5 min of extraction time. Estimated values of total yield and crude saponin were in good agreement with experimental values.

  • 홍미삼을 추출하여 홍삼엑기스를 제조할 때 추출용매인 에탄올의 농도와 추출온도, 그리고 시간이 홍삼엑기스의 물리적 성질에 미치는 영향을 비교조사 하였다. 홍삼엑기스의 고유점도(固有粘度)는 에탄올의 농도가 증가함에 따라 현저히 감소되었고 추출온도의 상승과 추출시간외 경과에 따라 다소 증가되는 경향이었으나 그 폭은 크지 않았다. 한편 측정온도의 상승은 고유점도를 크게 감소시키나 에탄올 농도가 50% 이상되면서 그 감소폭은 현저히 줄어들었다. 홍삼엑기스의 탁도와 pH는 에탄올의 농도 증가와 추출시간의 결과에 따라 낮아졌고 온도 상승에서는 큰 차이가 없었다.

음양곽의 항산화활성 (Antioxidant Activity of the Aerial Part of Epimedium koreanum NAKAI)

  • 본 연구의 목적은 음양곽의 기능성 연구의 일환으로 폐놀 성 화합물의 추출방법과 DPPHdp 의한 수소공여능, linoleic acid의 산화방지활성 및 LDL에 의한 항산화활성 등을 중심으로 연구한 결과이다. 음양곽에 60% ethanol를 첨가하여 추출회 수별 폐놀성 화합물을 조사한 결과 3회까지 추출했을 때 약 96%가 추출되었기 때문에 추출 횟수는 3회가 적당한 것으로 판단되었다. 60% ethanol 추출물의 수율은 3회까지 추출했을 때 약 22%이었다. 추출 용매에 따른 폐놀성 화합물의 추출 효율은 물 추출구를 100%로 했 을 때, 60% ethanol 추출구는 140%, 그리고 60% ethanoldyddor으로 추출한 뒤 농축하여 물 로 부피를 재조정한 구는 133%로 조사되었다. 항산화활성 조사에서 DPPHdp 의한 수소\ulcorner 여활성은 반응초기에 높게 나타났으며 반응시간이 경과함에 따라 완만하게 증가하였다. Linoliec acid 산화에 대한 산화방지 효과는 700 ppm 농도에서 약95% 산화저해율을 나타났 으며, LDL에 대한 산화방지 효과는 500 ppm 농도에서 산화저해율이 약 70%로 나타났다.

  • $\beta$-cyclodextrin adsorption and saponification methods were applied to isolate and purify cholesterol from the by-product of the low-cholesterol egg yolk product. They by-product was prepared from processing low-cholesterol egg yolk followed by extracting with chloroform to remove $\beta$-cyclodextrin and concentrated to 3,069 mg% cholesterol. When $\beta$-cyclodextrin method between two purification methods was applied, 50% ethanol as a solvent showed higher cholesterol concentration of 5.82% rather than the other solvents. Repeated purification of 3 times could not improve the cholesterol concentration significantly(p<0.05). In case of purification using saponification method, hexane as a solvent for extraction of unsaponificated materials was more efficient to increase cholesterol concentration than chloroform and ether. 60 times(v/w) saponification solution (95% ethanol:33% KOH = 94:6) of sample weight was most effective to increase the cholesterol concentration of 35.7%. Repeated purification process by saponification method could increase cholesterol concentration to 95.7% by 4 times repetition.

  • 쑥으로부터 Clostridium perfringens의 생육 저해 물질의 추출 조건을 조사하였다. 쑥의 유용성분을 최대한 추출하기 위하여 C. perfringens에 대한 생육 억제 정도를 기준으로 추출용매 농도, 추출온도 및 추출시간을 조사한 결과 100% ethanol, $40^{\circ}C$, 4시간 추출한 것이 C. perfringens의 생육 저해에 효과가 가장 좋았다.

고추 Oleoresin의 추출에 관한 연구 (Extraction of Oleoresin from Korean Red Pepper)

  • This study was conducted to investigate the optimal condition of the extraction of red pepper oleoresin. The yield of oleoresin form red pepper was 24~28% when it was extracted by ethanol and methanol. and 8~10% by acetone, ethylene dichloride and n-hexane. But more than 60% of portion in alcohol extracts were moisture and sugars. Capsaicin and pigment were extracted more than 70% of those in red pepper when it was extracted by ethylene dichloride Those useful components were extracted most efficiently by mixed solvents of ethanol and ethylene dichloride at the ratio of 50:150. other extracting conditions were investigated in this studies, such as optimal extraction time was more than 3 hours, temperature was 55$^{\circ}C$ and particle size was finer than 250 mesh

  • 추출조건(抽出條件)에 의(依)한 알로에 베라의 barbaloin 함량(含量)을 측정(測定) 비교(比較)하였다. 용매별(溶媒別) barbaloin 함량(含量)은 메탄올>에탄올>물의 순으로 증가(增加)하였다. 정치(定置) 추출(抽出)은 메탄올과 에탄올 추출의 경우 $60^{\circ}C$에서 4시간까지 가장 많은 함량(含量)이 추출(抽出)되었고, 그 이상은 감소(減少)하였다. 물추출의 경우 추출시간(抽出時間)과 온도(溫度)에 따라서 barbaloin 함량(含量)이 각각 다르게 나타났다. 물추출의 경우 온도(溫度)와 추출시간(抽出時間)이 증가(增加)할수록 추출량(抽出量)이 감소(減少)하는 것은 일반적으로 유기용매(有機溶媒)에서는 화합물(化合物)이 안정(安定)하지만, 물에서는 가수분해(加水分解) 등으로 분해(分解)된 것으로 판단(判斷)된다. 초음파(超音波) 추출(抽出)의 경우 메탄올에서 $60^{\circ}C$/40분이 가장 함량(含量)이 많았다. 진탕 추출(抽出)시 추출함량(抽出含量)은 물추출은 $40^{\circ}C$/8시간, 메탄올은 $60^{\circ}C$/4시간, 에탄올은 $60^{\circ}C$/2시간이 최대(最大) 함량(含量)을 나타냈고, 그 이상은 약간씩 감소(減少)하였으며, 특히 물추출은 $20^{\circ}C$$40^{\circ}C$에 비하여 $60^{\circ}C$에서 급격(急激)히 감소(減少)하였다. 환류추출(還流抽出)은 온도증가(溫度增加)에 따라 성분(成分) 함량(含量)도 증가(增加)하였다. 제반(諸般) 추출조건(抽出條件)을 비교한 결과(結果) 동시다수(同時多數)의 시료(試料)를 간편(簡便)하게 분석(分析)하기 위하여 메탄올 $60^{\circ}C$/40분간 초음파(超音波) 추출(抽出)하는 것이 양호(良好)하였고, 혼합용매(混合溶媒)의 경우 메탄올 : 에탄올(1 : 1) 혼합용액(混合溶液)을 사용하는 것이 능률적(能率的)이었다.
