식중독세균에 대한 솔잎 Ethanol 추출물의 항균작용 (Antibacterial Activity of Ethanol Extract of Pine Needle against Pathogenic Bacteria)

  • 5종류의 식중독세균 (Aeromonas hydrophila, Escherichia coli O157:H7, Listeria monocytogenes, Staphylococcus aureus 196E, Salmonella typhimurium)에 대하여 솔잎 ethanol 추출물의 항균작용을 조사하였다. 솔잎 ehanol 추출물을 0-2% (w/v) 함유한 tryptic soy broth(TSB)에 각 식중독세균을 약 $10^4$-$10^{6}$ CFU/ml 정도 되게 접종하여 35$^{\circ}C$에서 48시간 배양하면서 증식억제 정도를 생균수 변화로서 비교하였다. Gram 양성균인 L. monocytogenes, S. aureus와 Gram 음성균인 A. hydrophila는 솔잎 ehanol 추출물에서는 0.5% 이상의 농도에서 증식이 억제되어 솔잎 ethanol 추출물의 항균활성이 우수하였다. 그러나 Gram 음성균인 E. coli O157:H7과 S. typhimurium은 2%의 솔잎 ethanol 추출물에서 약간 증식이 억제되었으나 대체로 두 균주 모두 솔잎 ethanol 추출물에 대하여 강한 내성을 나타내었다. 솔잎 ethanol 추출물에 의한 증식억제 효과는 S. aureus > A. hydrophila> L. monocytogenes > E. coli O157:H7의 순으로 나타났으며 S. typhimurium에서 가장 강한 내성을 나타내었다.

솔잎 추출물의 항균성 검색

  • To develope natural food preservatives of pine needle (Pinus densiflora Seib et Zucc.) extract, pine needle sap, ethanol and ether extracts were prepared for investigation of antimicrobial activities against food-related bacteria and yeasts. All extracts exhibited growth inhibiting activities for most of microorganisms tested. However, in general, growth inhibiting activities were higher in ethanol extract than in sap or ether extract. Minimum inhibitory concentrations (MIC) of ethanol extract for Lactobacillus casei, Pseudomonas aeruginosa or Escherchia coli were as low as 0.1 mg/ml, whereas MIC of sap or ether extract for most bacteria and yeasts were 0.25-0.8mg/ml, indicating that the ethanol extract showed the antimicrobial activity by 2.5 $\sim$8 times higher than the sap and ether extract. The antimicrobial activity of the ethanol extract was reduced by heating or alkali treatment. Moreover, growth of Pseudomonas aeruginosa was completely inhibited within 24 hours by the addition of at least 50ppm of ethanol extract. These findings suggest that pine needle, specially the ethanol extrat may play a role for natural food preservatives.

에탄올 및 증류수 추출에 의한 솔잎 염색 견직물의 색채 이미지 및 선호도 (Color Image and Preference of the Silk Fabrics Dyed of Extract from Pine Needle by Ethanol and Distilled Water)

  • The purpose of this study was to evaluate the color image of the natural-dyed silk fabrics. The dye was extracted from pine needle by boiling pine needle with ethanol at $78^{\circ}C$ for 3hours and distilled water at $100^{\circ}C$ for 2hours. The 100% silk fabric was dyed of extract in pH 5 at $90-100^{\circ}C$ for 1 hr. As mordants used were compounds of Al, Sn, Fe, and Cr, color image of pine-needle dyed silk fabrics was classified into 5 factors (pure, gentle, sophisticate, comfortable, pastorale) and the factor pure is most important one of those. Most cheerful image in pure factor was from the fabrics dyed with ethanol extract and then, none and Cr mordanting. Dignified image was from the fabrics dyed with ethanol extract and then, Cu or Fe mordanting. In production, products dyed with ethanol extracts was preferred to those dyed with distilled water extracts. Color image and preference of the silk fabrics dyed with pine needles extracted was affected by extraction solvents and mordants.

감잎, 녹차, 솔잎의 건분 및 에탄올추출물이 흰쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Dried Leaf Powders and Ethanol Extracts of Persimmon, Green Tea and Pine Needle on Lipid Metabolism and Antioxidative Capacity in Rats)

  • The effects of dried leaf powders and ethanol extract of persimmon, green tea and pine needle on lipid metabolism, lipid peroxidation and antioxidative activity were investigated in rats. Forty-nine male Spargue-Dawley rats weighing 107.8$\pm$1.8g were blocked into seven groups according to body weight. Rats were raised for four weeks with diets containing either 5%(w/w) dried leaf powders of one of three different Korea traditional teas, persimmon(Diospyros kaki Thunb), green tea(Camellia Sinensis O.Ktzc)or pine needle(pinus Koreansis Sieb. Et Zucc), or ethanol from equal amounts of each dried tea powder. Food intake, weight gain, food efficiency ratio, and weights of liver, kidney and epididymal fat were significantly higher in the green-tea-powder group, and significantly lower in the pine-needle-powder and pine-needle-extract groups. Persimmon-leaf powder was found to decrease plasma total lipid, triglyceride and cholesterol concentration by increasing fecal total lipid triglyceride and cholesterol excretions. Liver cholesterol concentration was significantly lower in the green-tea and pine-needle-extract groups. Red-blood-cell superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) activities were significantly increased in rats fed green-tea extract. Liver SOD activity was increased in rats fed pine-needle powder or extract, and liver GSH-px activity was increased in rats fed greentea powder. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentration were both decreased in rats fed dried leaf posers or extracts of persimmon or green tea. It is believed that high vitamin E levels in persimmon leaf, and high flavonoid, beta-carotene and vitamin C levels in green tea effectively inhibited lipid peroxidation. In conclusion, persimmon and green tea leaves were effective in lowering lipid levels and inhibiting lipid peroxidation in animal tissue, while pine needles were effective and lowering body weight gain. From these results, persimmon and green tea leaves can be recommended in the treatment and prevention of chronic discorders such as cardiovascular disease, cancer and aging. As ethanol extracts from these teas were also effecitive in lowering tissue lipid levels and inhibiting lipid peroxidation, we recommend the use of discarded tea grounds for this.

소나무 부위별 추출물의 항산화 활성 (Antioxidant Activities of Extracts from Different Parts of the Pine Tree)

  • This study was conducted to investigate the antioxidant activities of extracts from various parts of the pine tree, which is known as a good source of functional food material. While ethanol extraction yields of pine bud and cone were higher than water extraction yields of pine bud and cone, water extraction yield of pine needle was higher than ethanol extraction yield of the pine needle. The content of polyphenols in the pine cone ethanol extract was 5 times higher than that in the pine bud and needle. Further, the content of flavonoids in the pine cone ethanol extract was 8 times higher than that in the pine bud and needle. DPPH radical scavenging effect of the pine cone ethanol extract was 3~5 times higher that of the pine bud and needle extract. Regardless of the extraction solvents, trolox equivalent antioxidant capacity (TEAC) and ferric reducing antioxidant power (FRAP) of the pine cone were stronger than those of the other parts of the pine tree. Taken together, it can be expected that the pine cone can be practically used as an antioxidant substance in food and beauty industries.

쑥과 솔잎의 항산화작용 및 아질산염 소거작용 (Antioxidative and Nitrite Scavenging Activities of Mugwort and Pine Needle Extracts)

  • 쑥과 솔잎을 각각 물과 70% 에탄올로서 추출하여 전자공여능과 아질산염 소거능을 조사하였다. 쑥 추출물의 전자공여능은 300-1,000ppm 농도에서 물추출물은 50-57%였으며, 에탄올추출물은 51-64%로서 에탄을 추출물이 우수하였다. 솔잎 추출물의 전자공여능은 물추출물은 100-500ppm에서 52-60%, 1,000ppm에서는 70%의 전자공여능을 나타내었으며 에탄을 추출물은 100-500ppm에서 68-71%, 1,000ppm에서는 약 77%로서 물추출물보다 높은 활성을 나타내었다. 아질산염 소거능은 pH 1.2, 3.0, 4.2, 6.0에서 측정한 결과, pH 1.2에서 가장 높았으며 pH 증가에 따라 감소하였다. 쑥추출물 1,000ppm의 아질산염 소거능은 pH 1.2에서 물추출물은 37%, 에탄올추출물은 27%였다. 솔잎 추출물 1,000ppm의 경우에는 pH 1.2에서 물추출물은 65%, 에탄올추출물은53%의 아질산염 소거능을 나타내었다. 추출물의 전자공여능과 아질산염소거능은 솔잎추출물이 쑥추출물에 비하여 우수하였다. 쑥과 솔잎 추출물에서 전자공여능은 에탄을 추출물이 물추출물보다 우수하였으나 아질산염소거능은 물추출물이 에탄을 추출물보다 우수하였다.

동충하초, 쑥 및 솔잎 추출물의 항균작용 (Antibacterial Activities of Cordyceps spp., Mugwort and Pine Needle Extracts)

  • 동충하초(눈꽃 동충하초, 번데기 동충하초), 쑥 및 솔잎 추출물의 항균작용을 조사하기 위하여 동충하초의 자실체와 균사체, 쑥 및 솔잎을 물과 70% 에탄올로 추출하였다. 항균실험에는 3종류의 그람양성균(Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus)과 3종류의 그람음성(Escherichia coli, Shigella sonnei, Salmonella typhimurium) 식중독 및 병원성 세균을 사용하였다. 동충하초의 추출수율은 물과 에탄올 모두 자실체의 수율(39∼58%)이 균사체(9∼24%)에 비하여 2.4∼4.4배 높았으며, 쑥과 솔잎 추출물의 수율은 9%미만이었다. 눈꽃 동충하초의 에탄올추출물(JME)은 1%농도에서 L. monowtogenes에 대하여 항균활성을 나타내었으며 번데기 동충하초의 에탄올추출물(MFE)은 3%농도에서 S. aureus에 대하여 항균활성을 나타내었다. 쑥의 에탄올추출물(MGE)은 1% 농도에서 L. monocygenes와 S. aureus에 대하여 항균활성을 나타내었으나 눈꽃 동충하초, 번데기 동충하초 및 쑥의 물추출물은 시험균주에 대하여 항균활성을 나타내지 않았다. 솔잎 물추출물은 E. coli를 제외한 모든 균주에 대하여, 솔잎 에탄올추출물은 실험에 사용한 모든 균주에 대하여 항균활성을 나타내어 솔잎추출물들은 가장 폭넓은 항균스펙트럼을 나타내었다. 액체배지내에서 솔잎추출물의 세균증식 억제작용은 에탄올추출물이 물추출물에 비하여 큰 편이었다.

솔잎 추출물과 고추냉이의 Vibrio에 대한 항균활성 (Antimicrobial Activity of Pine Needle Extract and Horseradish on the Growth of Vibrio)

  • 시판 꽃게와 가자미에서 Vibrio를 분리하여 Api 20E kit로 동정한 결과 V. parahaemolyticus HY I과 V. vulnificus FST I으로 동정되었다. 분리 Vibrio와 V.parahaemolyticus ATCC 17802에 대한 솔잎 추출물의 항균활성 검색을 실시한 결과 솔잎 추출물 1%를 첨가하였을 경우 뚜렷한 성장억제현상을 나타내었다. 시판 고추냉이와 솔잎 ethanol 추출물을 혼합하여 각 Vibrio에 대한 성장억제효과를 검토한 결과 l%고추냉이 단독 첨가구보다 1% 솔잎 추출물과 혼합치리구가 뚜렷한 성장억제효과를 나타내었다. 시판 고추냉이를 간장에 첨가하여 고추냉이 간장을 제조한 다음 Vibrio균의 생존을 실험한 결과 솔잎 추출물 1%를 첨가한 고추냉이 간장에서 Vibrio 생균수의 감소는 뚜렷하였다 고추냉이 간장을 제조 한 다음 관능검사를 실 시 한 결과 5% 수준에서 대조구와 솔잎 추출물 첨가구 간의 유의적인 차이는 나타나지 않았다.

Antioxidant activity and analysis of proantbocyanidins from pine (Pinus densiflora)needles

  • In this study, we evaluated the antioxidant activity of pine needle extracts prepared with hot water, ethanol, hexane, hot water-hexane (HWH), and hot water-ethanol (HWE), using the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical method. The hot water extract possessed superior antioxidant activity than the other extracts. We also compared the antioxidant activity of pine needle extracts through ROS inhibition activity in a cellular system using MC3T3 E-1 cells. The hot water extract exhibited the lowest ROS production. The pattern of HPLC analysis of each extract indicated that the hot water extract contained the highest proanthocyanidin level. The pine needle hot-water extract was then isolated and fractionated with Sephadex LH-20 column chromatography to determine the major contributor to its antioxidant activity. The No.7 and 12 fractions had high antioxidant activities, that is, the highest contents of proanthocyanidins and catechins, respectively. These results indicate that the antioxidant activity of procyanidins from the hot water extract of pine needles is positively related to not only polymeric proanthocyanidins but also to monomeric catechins. Moreover, the antioxidant activity of the pine needle hot water extract was similar to well-known antioxidants, such as vitamin C. This suggests that pine needle proanthocyanidins and catechins might be of interest for use as alternative antioxidants.

Growth Inhibition on the Strain Isolated from Spoiled Red Bean Paste

  • Growth inhibitory effects of ethanol extracts of green tea and pine needles on Bacillus stearothermophilus isolated from spoiled red bean paste were detected at concentrations higher than 750 ppm, and antimicrobial activity of pine needle extract was slightly higher than that of green tea exract. Growth inhibitory effect of pine needle extract in nutrient broth adjusted to pH 6.0, water-activity 0.92, and $45\;^{\circ}$Brix was observed at 500 ppm. These results indicated growth of B. stearothermophilus could be inhibited by adding pine needle and green tea extracts.
