고본(藁本)의 추출용매에 따른 항염, 항산화 및 항균 효과에 대한 비교 연구 (The Comparative Study on Anti-inflammatory Antioxidant and Antibacterial Effects with Regard to the Extraction Solvents of Ligustici Rhizoma)

  • Objective : This study was performed to compare anti-inflammation, anti-oxidation and anti-bacterial effects of Ligustici Rhizoma (LR) extracted with two kinds of solvents, ethanol and distilled water. Methods : It is prepared two kinds of LR extracts 20, 50, 100 ${\mu}l/mg$ by first. MTT assay way to measure cytotoxicity is formed in Raw 264.7 cell. The anti-inflammation effect is measured by ability to inhibit production of NO in Raw 264.7 cell. The anti-oxidation effect was measured by DPPH Radical scavenging ability in HaCaT cell. The anti-bacterial effect was measured by inhibition zone diameter on Propionibacterium acnes. Results : 1. LR (20 ${\mu}l/mg$) extracted with ethanol was showed 80% cytotoxicity, LR (50 ${\mu}l/mg$) extracted with ethanol and LR (20, 50 ${\mu}l/mg$) extracted with water were showed 70% cytotoxicity, LR (100 ${\mu}l/mg$) extracted with ethanol and LR (100 ${\mu}l/mg$) extracted with water were showed 60% cytotoxicity in Raw 264.7 cell. 2. LR (100 ${\mu}l/mg$) extracted with ethanol was showed more significantly inhibitory effect on NO production than the water extraction. 3. Two kinds of LR extraction groups did not show significantly scavenging effect of DPPH radicals. 4. Two kinds of LR extractions did not have a inhibitory effect on Propionibactrium acnes. Conclusion : Two kinds of LR extracts have not cytotoxicity, statistically significant ability to scavenge DPPH radicals and effect to inhibit Propionibactrium acnes. LR extracted with ethanol only have a little effect to inhibit NO production. This study proposes that LR extracted with ethanol is more effective in anti-inflammation.

Effects of Polygala tenuipolia on Expression of Fos-protein and Ethanol Amnesia in Rat

  • Effect of single administration of Polygala tenuipolia was examined on short-term memory in step through test and the intensity of the immunoreactive c-Fos protein induced by oral administration of ethanol. The acquisition of memory was significantly reduced by ethanol, and ethanol amnesia was remarkably reversed following oral administration of Polygala tenuifolia. c-Fos protein in normal rat brain was highly expressed in order of thalamus, pariental cortex, hippocampus, hypothalamus, amygdaloid and cingulate cortex. The expression of Fos protein was remarkably suppressed by single administration of ethanol. The inhibitory effect of ethanol on expression of Fos protein was reversed by single administration of Polygara tenuipolia, especially tissues of limbic areas such as amygdala, parietal cortex and CA3 of hippocampus. These results suggested that the amelioration process of Polygala tenuipolia on ethanol amnesia seems to be involve the expression of c-Fos protein in partly.

정상 성인에 있어서 미배아 발효액 (Glumate)이 에탄올의 혈중농도에 미치는 영향 (Effect of Glumate on the Blood Concentrations of Ethanol in Healthy Adults)

  • The extracts fermented of rice germ are called GMT (Glumate) which is a main ingredient of Condition$^{\circledR}$. It has been reported that GMT reduced the blood concentrations of alcohol in heathy adults. The purpose of the study is to examine the effect of GMT on blood concentrations of ethanol and on heart rate increase induced by ethanol. GMT was administered orally to 15 healthy adults before (10 min) and after (1 h) ethanol ingestion. The oral administration of GMT before (10 min) and after (1 h) ethanol ingestion reduced the area under the blood concentration of ethanol-time curve (p<0.12) and tended to inhibit the ethanol-induced heart rate increase.

Rat에 있어서 ethanol투여가 체조직의 무기질 함량에 미치는 영향 (The effect of ethanol on zinc, calcium and copper concentrations of tissues in rats)

  • This study was designed in order to clarify the effect of ethanol drinking on the mineral contents on liver, kidney, muscle and hair. Forty-five rats were divided into 2 groups and a control group. The control group received tap water and the other 2 groups were given 8% and 40% ethanol as drinking source. Liver, kindney, muscle and hair samples were taken and analyzed for zinc, calcium and copper contents by atomic absorption spectrophotometric methods. The results obtained are summrized as follows; 1. The zinc content of muscle showed significant (p<0.01) decrease in both groups. 2. The calcium content of hair showed significant (p<0.1) increase in 8% ethanol group. 3. The copper contents of kidney and muscle in 8% ethanol group and liver in 40% ethanol group showed significant (p<0.1) decrease.

흰쥐에게 Ethanol과 Xylene 병행 투여시 간손상에 미치는 영향 (Effect of Liver Damage by Combined Administration of Ethanol and Xylene in Rats)

  • 흰쥐에게 ethanol과 xylene을 투여시 간손상 정도를 알아보기 위해 정상적인 S-D계 수컷 흰쥐를 4군으로 나누어 실험을 수행하였다. 생화학적 검사 결과에서 혈청 AST, ALT, LDH활성은 대조군보다 ethanol 투여군, xylene투여군, ethanol+xylene투여군으로 갈수록 활성이 유의하게 증가되었으며, 그 증가율은 ethanol+xylene투여군에서 가장 높게 나타났다. 산화적 손상정도를 알아보기 위한 혈청 MDA함량은 대조군보다 ethanol투여군과 xylene투여군 그리고 ethanol+xylene투여군으로 갈수록 활성이 증가되었으며, 그 증가율은 ethanol+xylene투여군에서 가장 높게 나타났다. 그러나 혈청 ALP는 대조군에 비해 모두 감소하였다. Xylene의 투여시 최종대사산물인 요 중 m-methylhippuric acid는 대조군에 비해 xylene투여군과 ethanol+xylene투여군에서 증가되었으며, 그 증가율은 xylene투여군이 ethanol+xylene투여군보다 더 높게 나타났다. 간 조직검사에서는 xylene투여군은 지방변화를, ethanol+xylene 투여군은 경도~중등도의 괴사와 염증을 보였다. 이상의 혈청학적 검사와 조직학적 검사결과 xylene 단독 투여에 비하여 ethanol과 xylene을 병행 투여했을 때 간손상이 더 크게 초래된 것으로 나타났다.

갈화(Puerariae flos)추출물이 Rat 혈중 Ethanol 농도에 미치는 영향 (Effect of Arrowroot Flower (Puerariae flos) Extract on Lowering of Ethanol Concentration in Rat Blood)

  • 갈화(Puerariae flos)추출물이 rat의 혈액, 뇌 및 간의 ethanol농도를 유의차있게 감소시켰다. 혈중 ethanol의 감소 효과는 ethanol을 투여한지 1시간 경과한 때에 대조군과 가장 많은 차이를 보였으며 열수 추출물이 80% ethanol 추출물보다 혈중 ethanol 농도를 더 크게 감소시킴이 관찰되었다. Ethanol 투여 10분 전에 갈화추출물 투여가 1시간 전 혹은 10분 후에 투여한 군에 비해서 ethanol 농도 저하에 더 효과적이었으며, 간과 뇌 조직에서도 ethanol의 농도가 저하되는 현상이 관찰되었다. 그러나 이 때 acetaldehyde는 검출되지 않았다. 혈중 ethanol 농도를 가장 효율적으로 감소시키는 갈화 추출물의 적정량은 55.7 mg/kg body weight이었다. 이번 실험을 위해 새로이 시도된 dichloromethane에 의한 시료 추출과 GC를 이용한 분석법은 간단, 신속 하면서도 좋은 분석결과를 보였다.

와송(Orostachys japonicus) 추출물의 항산화 및 생리 활성 (Antioxidative and biological activity of extracts from Orostachys japonicus)

  • 와송에 함유된 phenol성 물질은 50% ethanol을 용매로 하여 추출하였을 때 $10.56{\pm}0.32mg/g$의 함량을 나타내었다. 와송 추출물의 항산화력을 측정한 결과 전자공여능은 $200{\mu}g/mL$ phenolic 농도에서 열수 추출물과 50% ethanol 추출물 모두 80.0% 이상으로 높은 효과가 확인되었다. ABTS radical cation decolorization을 측정한 결과에서 와송 열수와 50% ethanol 추출물 모두 95.0% 이상의 높은 항산화 활성이 측정되었다. Antioxidant protection factor (PF)는 열수 추출물이 50% ethanol 추출물에 비해 높은 PF값을 나타내었으며, thiobarbituric acid reactive substance (TBARs) 저해효과는 50% ethanol 추출물이 열수 추출물 보다 높은 항산화력을 나타내었다. 와송 추출물의 항고혈압 효과를 살펴보기 위하여 angiotensin converting enzyme 저해활성을 측정한 결과 $200{\mu}g/mL$ phenolic의 처리농도에서 열수와 50% ethanol 추출물이 각각 6.67%과 7.98%로 낮게 나타났다. 와송 추출물의 XOase 저해활성은 50% ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 60.85%의 저해율을 나타내었으며, tyrosinase 억제활성은 50% ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 64.59%로 나타나 대조구인 kojic acid 보다 미백 효과가 더 우수함을 입증하였다. 주름 생성과 관련된 collagenase 억제효과는 50% ethanol 추출물의 $50200{\mu}g/mL$ phenolic 농도에서 75.9585.02%로 매우 우수하였으며, 대조구인 epigallocatechin-gallate의 68.9176.64% 보다 다소 높았다. Elastase 저해활성도 50% ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 최대 63.65%의 저해효과를 나타내어 와송 50% ethanol 추출물의 주름개선 효과가 매우 우수함을 확인하였다. 이상의 결과를 통해 와송 추출물의 항산화 활성, 통풍 억제효능, 미백 및 주름개선 등의 기능성 검증을 위한 기초 연구 자료로 활용이 가능할 것으로 예상되며, 향후 추가적인 연구를 통해 와송 추출물을 기능성 식품 및 화장품, 의약품 소재로 적용이 가능할 것으로 판단되었다.

원기생맥산(元氣生脈散)의 추출물별 항산화작용과 쥐의 운동피로 회복효과 (Effect of Wongisaengmaek-san Extract on Antioxidative and Anti-Fatigue Activity)

  • Objectives : This study aimed to evaluate the effect of Wongisaengmaek-san (extracted with water or 50% ethanol) on antioxidative activity and recovery of fatigue induced by weight-loaded forced swimming exercise. Methods : Antioxidant activity of Wongisaengmaek-san was evaluated in terms of total amount of polyphenol, 1.1-diphenyl-2-picrylhvdroxyl (DPPH) radical scavenging activity and xanthine oxidase inhibition. The anti-fatigue effect of Wongisaengmaek-san was investigated using an acute weight-loaded forced swimming test by monitoring swimming test times and blood biochemical parameters creatinine. BUN, lactic acid dehydrogenase (LDH), free fatty acid (FFA) and lactic acid (LA). Results : 1. 50% EE (ethanol extract) had 2.3 times higher amount of total polyphenol compared to water extract. 2. It was identified that 50% ethanol extract showed enhancement of xanthine oxidase inhibition and DPPH scavenging effect in vitro. 3. According to anti-fatigue effect in the scale of 5% weight loaded mouse swimming test, both water and 50% ethanol extract showed significant improvement of swimming time elongation respectively, and 50% ethanol extract induced a more positive result. 4. After swimming for 1% weight-loaded mouse. water extract and 50% ethanol showed significant anti-fatigue effect with manifestation of blood sampling among both of the intervention: 50% ethanol extract showed a greater result. Conclusions : 50% ethanol extract of Wongisaengmaek-san has more effective antioxidant activity and anti-fatigue than water extract.

MODELLING STUDY OF THE EFFECT OF CHEMICAL ADDITIVES ON SOOT PRECURSORS REDUCTION

  • The effect of chemical additives, such as dimethyl ether(DME), ethanol, carbon disulfide on the soot formation were examined numerically. ill this study, the Frenklach soot mechanism was used as a base mechanism to predict the soot formation in the ethane flame. The combination of Westbrook's DME mechanism, Marinov's ethanol mechanism, and chemical kinetic mechanism for hydrogen sulfide and carbon disulfide flames was made with the base mechanism because the DME, ethanol, $CS_2$ additives are added into the ethane fuel. CHEMKIN code was used as a numerical analysis software to simulate the effect of chemical additives on reduction of the polycyclic aromatic hydrocarbons(PAH's) which are soot precursors. From the numerical results it is observed that addition of DME, ethanol and $CS_2$ into ethane fuel can reduce PAH species significantly. That means theses additives can reduce soot formation significantly. Results also strongly suggest suppression of soot formation by these additives to be mainly a chemical effect. Hand OH radicals may be the key species to the reduction of PAH species for additives.

에탄올-유발 위점막손상에 대한 애엽추출물 (DA-9601)의 방어효과 및 기전에 관한 연구 (Studies on Protective Effect of DA-9601\ulcorner an Artimisiae Herba Extract, against Ethanol-induced Gastric Mucosal Damage and its Mechanism)

  • Protective effect of DA-9601, an extract of Artemisia Herb, against ethanol-induced gastric mucosal injury was evaluated in rats. In the prophylactic study, DA-9601 exhibited total protection (99.4%) against absolute ethanol-induced gastropathy, And the protective effect of DA-9601 lasted up to 2 hours, which was longer than those of other contemporary mucoprotectants. In the treatment study, DA-9601 significantly facilitated the healing of 70% ethanol-induced mucosal damage, which was superior to cetraxate, a commonly used anti-ulcer drug. The mechanisms of mucoprotection of DA-9601 were also assessed. DA-9601 increased the release of prostaglandin E$_2$ from murine neutrophils in a dose-dependent manner in vitro. The cytoprotective effect of DA-9601 against ethanol-induced mucosal damage was significantly diminished by the concommitant injection of N$\omega$-nitro-L-arginine methyl ester (L-NAME, 5 mg/kg, i.v.), a non-specific nitric oxide (NO) synthase inhibitor, while it was not affected by preinjection of indomethacin (5 mg/kg, s.c.), a prostaglandins-depletor. And it was found that DA-9601 significantly enhanced adaptive cytoprotective action of 10% ethanol against absolute ethanol (56.9$\pm$6.5 vs 23.0$\pm$3.3 mm$^2$, p<0.05, mean$\pm$SEM), though its exact underlying mechanism remains to be clarified. The present fin[lings demonstrate that DA-9601 exerts gastroprotecticv actions for the stomach against ethanol through several different underlying mechanisms, in which prostanglandins and NO are involved. In conclusion, the results obtained suggest that DA-9601 can be useful both in prevention and treatment of ethanol-induced gastric damage.
