Comparison of Antioxidant and Physiological Activities of Various Solvent Extracts from Hizikia fusiformis (추출용매에 따른 톳(Hizikia fusiformis) 추출물의 항산화 및 생리활성 비교)

  • The seaweed Hizikia fusiformis is rich in protein, carbohydrates, vitamins, and minerals. This study investigated the antioxidant and physiological activities of H. fusiformis extracts prepared with 70% ethanol, 80% methanol, or distilled water. The extraction yields of these various solvent extracts were as follows: ethanol extract, 15.26%; methanol extract, 17.95%; and water extract, 45.62%. The methanol extract showed the highest total polyphenol content (24.06 mg GAE/g), but total flavonoid content was similar in all extracts. ABTS (2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) radical scavenging activity was highest in the ethanol extract (IC50: 0.90 mg/mL), while the methanol extract exhibited the strongest DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity (IC50: 8.09 mg/mL), reducing power (EC50: 0.40 mg/mL), and ferric reducing antioxidant power (0.28 mM). By contrast, tyrosinase and α-glucosidase inhibitory activities were higher in the ethanol extract than in the other extracts. The high BACE1 (β-secretase) inhibitory activity was observed in the ethanol extract (IC50: 1.03 mg/mL). These results indicate that H. fusiformis ethanol extracts may be useful for their antioxidant and functional properties in food and pharmaceutical materials.

Functional Biological Activity of Hot Water and Ethanol Extracts from Taraxaci Herba (포공영의 열수 및 에탄올 추출물의 기능적 생리활성)

  • This study was conducted to investigate the functional biological effects of hot water and ethanol extracts from Taraxacum mongolicum (TM). Then, the hot water and ethanol extracts of TM were measured for total flavonoids content, total phenolics content, electron donating ability, nitrite-scavenging ability, SOD-like activity, tyrosinase inhibitory effect, and elastase inhibitory effect. Total flavonoids contents of hot water and ethanol extracts from TM were 7.80$\pm$0.97 mg/g and 9.12$\pm$0.51 mg/g, respectively, and total phenolics contents were estimated as 54.20$\pm$1.95 mg/g for water extract and 79.43$\pm$4.44 mg/g for ethanol extract. The $RC_{50}$ values for electron donating ability of hot water and ethanol extracts were 943.98 $\mu$g/mL and 309.41 $\mu$g/mL. SOD-like activity and nitrite-scavenging ability were dependent on concentration of hot water and ethanol extracts, and the activity of ethanol extract was higher than that of hot water extract. However, hot water and ethanol extracts from TM showed no inhibitory activities on the elastase and tyrosinase inhibitory activities. Based on the above results, the ethanol extract of TM seems to be the most pertinent for use as functional food and cosmetic.

Analysis of nutritional compositions and biological activity of Agrocybe aegerita (버들송이(Agrocybe aegerita) 영양성분 및 생리활성 분석)

  • The mushroom Agrocybe aegerita was analyzed to evaluate the nutritional value of this potential food and to enhance the cultivation and consumption. Among the nutritional compositions of Agrocybe aegerita, contents of crude proteins, crude fats and ashes were 38.3%, 3.0% and 8.2% respectively. The contents of potassium and phosphorus were higher than that of other minerals. Total phenolics contents were $65.2{\mu}g/m{\ell}$ and $46.0{\mu}g/m{\ell}$ in water extract and ethanol extract, respectively. Total flavonoids were estimated as $12.5{\mu}g/m{\ell}$ contents of water extract and ethanol extract were $7.1{\mu}g/m{\ell}$. Free radical scavenging activity against DPPH of ethanol extract, 79.2% was higher than that of water extract(58.2%). The contents of total phenolics, flavonoids and antioxidant activity by the DPPH scavenging activity of Agrocybe aegerita were higher than those of Pleurotus eryngii and Pleurotus ostreatus. The tyrosinase inhibitory activity showed 63.2% and 65.0% in water extract and ethanol extract, respectively. Angiotensin converting enzyme inhibitory activity of water extract was 78.7% and 75.0% in ethanol extract. In this study, Agrocybe aegerita has abundant essential nutrients and thus is good source of functional health food.

Comparison and Analysis of Several Main Components between Hot Water Extract and Alcohol Extract of Gamichungsangbohatang (가미청상보하탕(加味淸上補下湯) 열수추출물(熱水抽出物)과 알코올추출물(抽出物)의 지표물질(指標物質) 및 패턴의 비교분석(比較分析))

  • Objectives : To find out more pharmacologically efficient way of extraction of herbal dicoction, Gamichungsangbohatang (GMCSBHT). Methods : Several main components of GMCSBHT was compared and analysed between hot water extract of GMCSBHT and Alcohol(70% ethanol) extract of GMCSBHT via HPLC method. Results : Hot water extract of GMCSBHT showed relatively more component content than ethanol extract of GMCSBHT. Also weighted mean of main components of hot water extraction of GMCSBHT was higher than that of alcohol extract of GMCSBHT. But from chromatographic pattern analysis of matching ratio and similarity ratio showed that these two forms of extracts might have different chemical composition, and 3D PDA plot of alcohol extract of GMCSBHT showed high peaks near UV $190{\sim}220nm$ which was invisible in hot water extract of GMCSBHT. Conclusion : Alcohol extract of GMCSBHT may have some special components which do not exist in hot water extract of GMCSBHT.

Comparison of Immuno-stimulatory Activities by Purification Process of Schizandra chinensis Baillon Fruits

  • Two compounds from Gomisin N and Gomisin A were isolated from the fruits of Schizandra chinensis Baillon. The highest extraction yield as 21.36% was observed in the ethanol extract, compared to the yield obtained form the water extract. The extraction yields of the single compounds were measured to be 0.13 and 0.014 Gomisin A and Gomisin N, respectively. Approximately, 90% of the growth of human stomach adenocarcinoma cancer cells was inhibited after adding 1.0 g/l of the ethanol extract. The growth of the human normal lung cell was limited to 24% after adding the ethanol extract. The water extract lowered the specific secretion of $TNF-\acute{a}$ and IL-6 from T cells, $10.3{\times}10^{-4}\;pg/cell\;and\;12.1{\times}10^{-4}\;pg/cell$, respectively, compared to the ethanol extracts. On the other hand, a treatment with the ethanol extract increased the specific secretion of $TNF-\acute{a}$ and IL-6 from human T cells, to $11{\times}10^{-4}\;pg/cell\;and\;14.3{\times}10^{-4}\;pg/cell$, respectively. The crude ethanol extract had the highest effect on the differentiation of human promyelocytic leukemia cells compared to the other extracts and Gomisin A and N. In general, the biological activities of the extracts gradually decreased as the purification process proceeded, which suggests that higher immunostimulatory activities can be maintained by adding the crude extracts of the fruits rather than by adding a single compound.

  • 'Sochungryoung-Tang' has been known to be used in acute edema, bronchitis, pneumonia, nephrosis, and arthritis etc. in traditional chinese medicine. In this study, it was examined on the antimicrobial and anti-inflammatory activities. The following results were obtained; 1) In the antibacterial activities, the ethanol extract of 'Sochungryoung-Tang' showed to be more active than water extract aginst ten bacterials (Table I). Especially, antibacterial activities against Staphylococcus sp. showed the highest among the strains tested. But in the fungi, ethanol extract and water extract showed slightly significant effects (Table II). 2) The water extract showed significant anti-inflammatory effects against the rat hind paw edema induced by 1% carrageenin in the scab and carbuncle in the mice in fected by Staphylococcus aureus CUCC 74371.

Antioxidant Activities of Hot Water and Ethanol Extracts from Portulacae Herba (마치현 열수 및 에탄올 추출물의 항산화 활성)

  • Objectives : This study aims to provide basic data about Portulacae Herba (PH) extracts as natural antioxidants by considering diverse antioxidant activities of PH depending on solvents. Methods : The samples of PH were pulverized, and A hot water and a 70% EtOH were stir-extracted for two hours three times repeatedly in a water bath with a temperature of 95 degrees and at room temperature respectively to measure 7 kinds of antioxidant activities. Results : There were significant differences in total phenol content, because the total phenol content of the 70% EtOH extract was higher than the hot water extract's, and the total flavonoid content of the 70% EtOH extract($4.40{\mu}g/mg$) was nearly 3.8 times higher than the hot water extract's($1.16{\mu}g/mg$). DPPH and ABTS radical scavenging activities were 70% EtOH extract showed a little higher activity than the hot water extract, and at a concentration of $500{\mu}g/mg$, the highest scavenging activity was found in the 70% EtOH extract, not in the control group. Hydroxyl radical and $Fe^{2+}$ chelatingactivities were slightly higher in hot water extract than in 70% EtOH extract, and increased in a dose-dependent manner. Nitrite scavenging activities increased dose-dependently in the hot water and the 70% EtOH extract, regardless of the pH level, and scavenging activity of the 70% EtOH extract was higher at pH 1.2 than at pH 3.0. Conclusions : In conclusion, it is thought that PH hot water and 70% EtOH extract have antioxidant activities, and can be used as natural antioxidants in future.

Biological Activities in roots of Glycyrrhiza uralensis Fisch (감초 세근의 생리활성 탐색)

  • The biological activities of ethanol, ethanol: water(1 : 1v/v) and water extracts from Glycyrrhiza uralensis Fisch, glycyrrhizin and enzymatically hydrolyzed glycyrrhizin were compared. About 50% of the growth of MCF7, A549, Hep3B and AGS cells were inhibited in adding 1.0 g/L of the crude extracts, glycyrrhizin and enzymatically hydrolyzed glycyrrhizin. For example, the ethanol extract inhibited 76%, 66% in MCF7 and Hep3B cells by adding 1.0 g/L. For cytotoxicity on human normal liver cell(WRL-68), the crude extracts were scored as above 26%. For the result of antimutagenecity using CHO V79 cell, the crude extracts proved more effective than other samples. The growth of human immune B and T cells were enhanced up to $1.2{\sim}1.3$ times by adding the crude extracts. In inhibitory effect of ${\alpha}-glucosidase$ activity was showed that the ethanol extract, water extract and ethanol: water (1 : 1v/v) extract were appeared 65%, 68%, 62% in adding 1.0 g/L. The higher enhancement of glutathione -S-transferase activity was observed in the ethanol extract as 257% compared to the control in adding 1.0 g/L. From the results, the biological activities of the crude extracts were equivalent or higher than glycyrrhizin and enzymatically hydrolyzed glycyrrhizin.

  • Physiological effects of curcumin, major yellow-colored pigment in tumeric (Curcuma longa L.), extracted by traditional extracting methods, ethanol and hot-water extractions, and supercritical fluid extraction (SFE) using supercritical carbon dioxide as new extracting method. Antioxidative activity of ethanol extract was higher than those of SFE and hot-water extracts. Results of Ames mutagenicity test on SFE, ethanol, and hot-water extracts revealed no mutagen in the extracts. Antimutagenicity rates of SFE, ethanol, and hot-water extracts against direct mutagen, 2-nitrofluorene (2-NF), were 20.1, 9.3, and 15.2%, respectively. Antimutagenicity rate of SFE extract against TA98 derived from indirect mutagen, 2-acetamidofluorene (2-AF), was 12.2%, whereas none was observed in ethanol and hot-water extracts. Nitrite-scavenging ability of SFE extract was higher than those of ethanol and not-water extracts.

Antioxidant, Antidiabetic, and Anti-Inflammatory Effects of Extracts and Fractions from Parthenocissus tricuspidata Stems (담쟁이덩굴 추출물과 분획물의 항산화, 항당뇨 및 항염증 효과)

  • This study was carried out to analyze the effects on antioxidative, antidiabetic, and anti-inflammatory activities of Parthenocissus tricuspidata (PT) stem extracts. The total phenolic contents of hot water and ethanol extracts from PT stems were 61.5 mg TAE/g and 122.1 mg TAE/g, respectively. The antioxidative activities of hot water and ethanol extracts from PT stem were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD) assay. The DPPH radical scavenging activities of ethanol extract and butanol fraction were approximately 95% and 92% at 1 mg/ml, respectively, and the SOD activities of ethanol extract and butanol fraction were about 91% and 97% at 1 mg/ml, respectively. The DPPH radical scavenging and SOD activities of ethanol extract and butanol fraction from PT stem increased remarkably increased in a dose-dependent manner and were higher than in the hot water extracts. Compared to the acarbose, a known anti-diabetic drug, which was used as a positive control, the ${\alpha}$-glucosidase inhibitory capacity of PT stem showed a strong inhibitory rate in ethanol extract and in butanol and hexane fractions. We investigated the effect of hot water extract from PT stem on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells. Hot water extract from PT stem inhibited LPS-induced NO production up to 40% at a treatment of 1 mg/ml. These results suggest that PT stem extracts have an effect on antioxidative, antidiabetic, and anti-inflammatory activities and thus have great potential as antidiabetic materials and a source for natural health products.
