Change in the Expression of Occludin, a Gene for Blood-Brain Barrier by Phytoestrogens in Hippocampus of Rat Model for Menopause (폐경기모델 백서 해마에서 식물성 에스트로젠에 의한 뇌-혈액장벽 유전자 occludin 발현의 변화)

  • To elucidate the effect of phytoestrogens on the prevention of neurodegenerative disease in postmenopausal women, the expression of occludin which build up the blood-brain barrier was examined in hippocampus following oral administration of estrogen (E2), genistein, diadzein or combination of genistein and diadzein in ovariectomized (OVX) female rats. E2 significantly increased occludin mRNA level in OVX rat hippocampus, suggesting that estrogen is a physiological regulator for structural integrity of the blood-brain barrier in hippocampus. Following isoflavone diet for 4 weeks, there was significant increase in occludin mRNA level in hippocampus, suggesting that isoflavone diet may be effective for protection of structural integrity of blood-brain barrier in hippocampus from degenerative changes in estrogen deficiency.

Effect of Platycodon Grandiflorum A. Extract in Bone Metabolism in Ovariectomized Rats (난소 절제한 흰쥐에서 도라지 추출물이 골 대사에 미치는 영향)

  • Osteoporosis is one of the major health problem affecting postmenopausal women. Estrogen deficiency results in an increase in bone turnover, lead to bone resorption and an increase risk of fracture. The aim of this study was to evaluate the effects of Platycodon grandiflorum A. extract (PG) in bone metabolism in ovariectomized estrogen-deficient rats. Two groups were surgically ovariectomized (OVX). The third group was sham operated. Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with PG at 50mg/kg body wt (OVX-PG50) The Platycodon grandiflorum extracts were orally administrated at 1mL per day. The ovariectomy caused a decreasing in the levels of collagen content in bone, cartilage and skin tissues. However PG group, supplementation with Platycodon grandiflorum extract, were increased the level of collagen content in bone, cartilage and skin tissues than OVX-control group. PG group had a higher content of pyridinoline in collagen than OVX-control group. Alkaline phosphatase activity on serum were decreased after supplemented with the PG extract. These results might be expected that Platycodon grandiflorum is believed to be possible protective effects in postmenopausal bone loss.

Effect of Capsosiphon fulvecense Extract on Collagen Content of Connective Tissues in Ovariectomized Rats (매생이 추출물이 난소를 절제한 흰쥐 결합조직의 collagen 함량에 미치는 영향)

The aim of this study was to evaluate the effects of Capsosiphon fulvecense extract (SCF) on the collagen content of the connective tissues and alkaline phosphatase activity of serum in ovariectomized estrogen-deficient rats. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with CsF at 50mg/kg body wt (OVX-CSF50) and ovariectomized rats supplemented with CsF at 200mg/kg body wt (OVX-CSF200). The Capsosiphon fulvecense extracts were orally administrated at 1mL per day. The ovariectomy caused a decreasing in the levels of collagen content in bone, cartilage, skin and lung tissues. However CSF groups, supplementation with Capsosiphon fulvecense extract, were increased the level of collagen content in bone, cartilage, skin and lung tissues than OVX-control group. Alkaline phosphatase activity also were increased and calcium levels were decreased than OVX-control on serum. These results suggest that Capsosiphon fulvecense supplementation prevents postmenopausal bone loss, thus it may be used possibly to improve the quality of life in menopausal women.

Effects of Phytoestrogen on Cell Growth and Insulin-like Growth Factor-I (IGF-I) Production in MC3T3-El Cells (식물성 에스트로겐이 MC3T3-El 골아세포의 성장과 Insulin-like Growth Factor-1(IGF-1)생성에 미치는 영향)

  • Estrogen is known to play an important role in maintaining bone mass, since the concentration of serum estrogen decrease after menopause and the estrogen deficiency results in bone loss. Phytoestrogens are plant compounds with estrogen-like biological activity, In this study, to investigate the bioactivities of phytoestrogen, which act on bone metabolism, we examined the effect of selected food-borne phytoestrogens (genistein, daidzein and resveratrol) on osteoblast proliferation and IGF-I production using MC3T3-El cells, a mouse calvaria osteoblast-like cell line. Cells were cultured in a serum free medium for 48 hr in the presence of genistein $(10^{-5}\;M)$, daidzein $(10^{-5}\;M)$ and resveratrol $(10^{-5}\;M)$. The effects of genistein, daidzein and resveratrol on the cell proliferation and growth were evaluated by total cell numbers, MTS assay and cell migration assay. Their effect was compared with the $17\beta-estradiol$. Genistein, daidzein and resveratrol exhibited stimulatory effects on the growth of MC3T3-El cells, and the most pronounced effect was shown with daidzein. In addition, these phytoestrogen increased alkaline phosphatase activity of MC3T3-El cells. These effects were similar to that of $17\beta-estradiol$ effects. Moreover, treatment with genistein, daidzein and resveratrol increased production of insulin like growth factor-I (IGF-I) in conditioned media, indicating that the growth promoting effects of these phytoestrogen were related to the changes in production of IGF-I by MC3T3-El cells. These results show that genistein, daidzein and resveratrol have a stimulatory effect on osteoblast function, and that these findings in a cell model may prove relevant to protecting against the loss of bone mass and the development of osteoporosis in human subjects.

  • The use of biochemical markers of bone turnover may be particular interest in the investigation of bone disorders with osteoporosis. Serum osteocalcin(OC), total alkaline phosphatase and procollagen C, reflecting bone formation, and urinary pyridinium cross-links excretion, reflecting bone reabsorption have been measured in hyperthyroidism, postmenopause women, after testosterone supplementation, androgen, testosterone and estrogen deficiency, bone mineral density degree, age duration. Bone marks which is reflect to metabolic bone disorders are biochemical indices method to measure enzyme activity about bone formation, bone absorption and bone components in blood or urine. Bone metabolism biochemical marks are correlated with osteophorotic agents and also represent significantly different between bone mineral density and bone biochemical marks. Therefore if we develope and use bone metabolism marks which have higher sensitivity and specificity in bone formation and bone absorption, I think that these bone biochemical marks can have utility in the clinical application to predict osteoporosis risk group, bone loss, bone fracture and response degree to treatment of osteoporosis risk groups.

  • In order to investigate the effect of the Aqua-acupuncture of the Pakukchun treated on Shinsu(BL23) loci on the estrogen-deficiency osteoporosis induced by ovariectomy in rats, serum osteocalcin estradiol progesterone follicular stimulating hormone(FSH) calcium(Ca) and phosphorous(P) levels were monitored. The Aqua-acupuncture of Pakukchun treated on Shinsu loci have favorable effect on treatment and prevention of osteoporosis induced by ovariectomy in rats.

Effects of a Pueraria lobata-root based combination supplement containing Rehmannia glutinosa and aerobic exercise on improvement of metabolic dysfunctions in ovariectomized rats (갱년기 모델 유도 흰 쥐에서 갈근과 지황 복합물 및 유산소 운동이 대사이상 개선에 미치는 영향)

  • Purpose: There is a fair amount of evidence indicating that increased risk of obesity and insulin resistance is associated with postmenopausal state, but can be modulated by diet and exercise. In this study, we explored whether a Pueraria lobata root-based supplement containing Rehmannia glutinosa (PR) and/or aerobic treadmill exercise can modify the metabolic changes associated with estrogen deficiency. Methods: Seventy rats were randomly assigned to the following groups for 8 weeks (n=10 per group): SHAM, sham-operated; PR0, ovariectomized (OVX) control; PR200, OVX with PR200 mg/kg B.W; PR400, OVX with PR400 mg/kg B.W; EPR0, OVX with exercise; EPR200, OVX with exercise and PR200 mg/kg B.W; EPR400, OVX with exercise and PR400 mg/kg B.W. Results: OVX induced significant increases in body weight, food intake, fat mass, LDL-cholesterol, and fasting blood glucose, confirming induction of menopausal symptoms. PR supplementation or exercise significantly suppressed the above mentioned changes through different regulatory elements in adipose tissue: PR supplement upregulated adiponectin gene expression and aerobic exercise upregulated adiponectin and insulin receptor gene expression and a combination of PR supplement and aerobic exercise showed an additive effect on adiponectin gene expression. Conclusion: Taken together, the results of this study suggest that PR supplement has a potential to provide health benefits in OVX rats through leptin and adiponectin secretion. In addition, the data suggest that combination of exercise and PR would have additive effects on metabolic dysfunction associated with estrogen deficiency.

Effects of Silkworm Pupae on Bone Mineral Density in Ovariectomized Rat Model of Osteoporesis

  • Osteoporosis is characterized by the reduced density of mineralized bone matrix. The loss of bone has been attributed to an imbalance between bone formation and bone resorption. The silkmoth is one of the famous traditional tonic medicines. Silkworm pupa was evaluated for its antiosteoporotic activity in an ovariectomized rat model of osteoporosis. The rats were ovariectomized at 6weeks of age and divided into 7 groups. All the groups were fed without calcium diet. The OVX rats were treated with water and silkworm powder for 6 weeks. In OVX rats, the body weight, feeding and water intake quantity did not show a significant change, but the silkworm pupa powder immediately before the eclosion of Yeonrokjam was significantly increased the bone mineral density in the femoral bone. The silkworm pupa powder increased the bone with increasing calcium in serum. These results also showed its protective action through promotion of bone formation. The silkworm pupa powder has a definite antiosteoporotic effect; similar to estrogen, it is especially effective for the prevention of bone fracture induced by estrogen deficiency. The silkworm pupa powder is a promising anti-osteoporotic agent that can prevent bone fractures in osteoporotic patients. The silkworm pupa powder does not show serious side effects because natural herbal medicine origin.

A Case of Recovery of Ovarian Function in Woman Who was diagnosed as Premature Ovarian Failure (조기난소기능부전 환자 치험 1례)

  • Purpose : Premature ovarian failure, characterized by amenorrhea or oligomenorrhea, estrogen defieciency and elevated gonadotropins in woman less than 40 years of age, affects approximately 1% of woman and is associated with autoimmune phenomena or genetic abnormalities, but mostly idiopathic. This disease is loss of ovarian function seems not to be always permenent. Method : We investigated herb medication was effective in patient of premature ovarian failure. Results: Signs and symtoms of estrogen deficiency were improved and mensturation occurred monthly and the elevated during treatment of herb medication. As a result of our case recovering of ovarian function, we hope more investigtions about treating premature ovarian failure by oriental medicine will be gained.

The effects of sesame oil and different doses of estradiol on testicular structure, sperm parameters, and chromatin integrity in old mice

  • Objective: Studies of the effects of estrogens on the male reproductive system have emphasized the role of these hormones in male fertility. Sesame oil has many phytoestrogenic compounds and may improve male fertility. This study investigated the effects of sesame oil and different concentrations of estrogen on sperm parameters and DNA integrity in male mice. Methods: Twenty old NMRI (The Naval Medical Research Institute) male mice (40 weeks; weight, 30-35 g) were treated with sesame oil or different concentrations of estrogen (estradiol, 1 and 10 μL/kg/day) or received no treatment (controls). After 35 days, sperm parameters and DNA integrity were assessed and analyzed. Results: Sperm count, progressive motility, and morphology were decreased in the group that received 10 μL/kg of estradiol. A remarkably lower percentage of DNA fragmentation and protamine deficiency were detected in the group that received 1 μL/kg of estradiol. In the groups that received sesame oil and 1 μL/kg of estradiol, the numbers of spermatogonia and Leydig cells were higher than in controls. The combination of sesame oil and 1 μL/kg of estradiol led to improved sperm parameters and chromatin and testicular structure. Conclusion: Based on this study, consumption of sesame oil and a low concentration of estradiol may improve testicular function in older mice.
