Influence of Storage Temperature on Levels of Bioactive Compounds in Shiitake Mushrooms (Lentinula edodes)

  • Shiitake mushroom (Lentinula edodes) hold high nutritional and medicinal value as they contain an abundance of health-promoting compounds. However, the effect of long-term postharvest storage on the variation in the levels of health-promoting compounds has not been extensively studied. In this study, we investigated the changes in the levels of phenolic compounds, antioxidants, eritadenine, and ergothioneine in shiitake mushrooms stored at three different temperatures (1, 3, and 5 ℃) for 4 weeks. Compared to mushrooms stored at lower temperatures, those stored at 5 ℃ exhibited a higher level of total phenolics in their pileus after 2 weeks of storage; however, storage at 5 ℃ also increased the deterioration of the fruiting body of these mushrooms. In mushrooms stored at all temperatures, the eritadenine content in the pilei tended to increase up to 2 weeks of storage. In contrast, the ergothioneine content in the pileus decreased during storage, with a significantly lower level detected in mushrooms stored at 5 ℃ for 4 weeks. Together, these results suggest that the mechanisms underlying the accumulation of phenolics and eritadenine may be related to mushroom deterioration during storage. Our findings indicate that the levels of health-promoting compounds in shiitake mushrooms are influenced by storage temperature, suggesting the potential to control adjustments of specific bioactive compounds by regulating storage conditions.

Blood Pressure Reduction of SHR Treated with EA-contaning Lentinus edodes Mycelial Solid Culture Extract (Eritadenine 함유 표고버섯균사체 고체배양 효소추출물(EA-LEMSCEE)의 spontaneous hypertension rat 혈압감소)

  • Eritadenine (EA), derived from Lentinus edodes (LE), reduced low-density lipoprotein (LDL), triglyceride (TG), and phospholipids in bloods, and fatty acid depositions in animals and humans. Previously, we reported that EA inhibited angiotensin-converting enzyme (ACE) activity in vitro. Now, we report that EA reduced blood pressures in spontaneous hypertension rats (SHR). EA-containing LE mycelial culture enzyme extract (EA-LEMSCEE) was prepared from LE mycelial solid cultures and the hot-water extract of LE fruit bodies. Both EA and EA-LEMSCEE inhibited ACE activity in immortalized human umbilical endothelial cells (EA.hy926). EA-LEMSCEE treatments (7.5 mg/kg, 22.5 mg/kg) significantly reduced systolic and diastolic blood pressure in SHR. At five weeks of treatment, EA-LEMSCEE treatment significantly reduced systolic and diastolic blood pressure, similar to the positive control (captopril, CP; 4 mg/kg) treatment. In addition, the LEMSCEE without EA decreased systolic and diastolic blood pressures compared to the control, but not significant. EA-LEMSCEE decreased renin and ACE activities, and angiotensin II (Ang II) contents in SHR compared to the control. After five weeks of treatment, the effect of EA-LEMCEE was similar to that of CP. These results indicate that EA and EA-LEMSCEE reduce blood pressure by inhibiting the renin and ACE activity of SHR. Furthermore, these results imply that EA or EA-LEMSCEE could be used as an antihypertension agent in humans.

Eritadenin Contents Analysis in Various Strains of Lentinula edodes using LC-MS/MS (LC-MS/MS를 이용한 표고 균주별 에리타데닌 함량 분석)

  • Eritadenine, a potent compound of hypocholesterolemic activity, was investigated in relation to its content in domestic cultivars and wild types of shiitake (Lentinula edodes). Eighteens strains of shiitake were tested for the quantification of eritadenine by LC-MS/MS analysis. Among the strains, wild type-40 was highest as the content was 3.912 mg/g. Also, Soohyangko was 3.352mg/g, Sanlim No. 9 3.008mg/g, Chunbaegko 2.832 mg/g, Gaeulhyang and KFRI 675 both 2.792 mg/g as high-content strains. Soohyangko and Chunbaegko are applied strains for registration in 2010. Soohyangko is high-temperature type with concentrated fruiting, and 90% of production occurs in the first year, thus, recovery of cost is very fast. Chunbaegko is mid-temperature type with concentrated flushing, and produces "hwago", the best quality, in spring. Wild type-40 is excellent in productivity and is prepared for registration. Wild type-40 could be used as parent strain to make new strain with high eritadenine content.

Agaricus blazei Mycelial Liquid Culture Extract Containing Eritadenine Improves Benign Prostatic Hyperplasia-related Biochemical Markers in RWPE-1 Cells through Anti-inflammatory and Anti-oxidative Actions (RWPE-1 전립선세포에서 eritadenine을 함유한 신령버섯균사체 액체배양물의 항염증효과 및 항산화효과에 의한 전립선비대증 관련 biochemical marker 개선 효과)

  • Agaricus blazei mycelial liquid culture extract (ABMLCE) promoted the production of testosterone (TS) in TM-3 mouse Leydig testis cells. Now, we report that ABMLCE containing eritadenine (EA) as a minor constituent (15.3 mg/100 g) reduced $5{\alpha}-reductase$ 2 ($5{\alpha}-R2$) enzyme activity and dihydrotestosterone (DHT) content which are key constituents for the benign prostatic hyperplasia (BPH) inductions. RWPE-1 prostate cells were grown in a Keratinocyte serum-free medium (K-SFM) containing ABMLCE (0~50 ppm), EA (0~10 ppm,), and finasteride (FS $10{\mu}M$: a positive control) in a 24-well plate for 24 hr. Supernatants collected from cell-cultured media were used for the assay of $5{\alpha}-R2$, superoxide dismutase (SOD), catalase (CAT) and cyclooxygenase-2 (COX-2) enzyme activities, and for TS, DHT, tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and $interleukin-1{\beta}$ ($IL-1{\beta}$) contents by their assay kits. The $5{\alpha}-R2$ activity and DHT content were proportionally reduced (p<0.05) to concentrations of ABMLCE. The SOD and CAT enzyme activities were significantly (p<0.05) elevated concomitant with ABMLCE concentrations, while COX-2, $TNF-{\alpha}$ and $IL-1{\beta}$ showed reverse results (p<0.05). Similarly, the effects of EA were similar to those of ABMLCE. Efficacies of ABMLCE 50 ppm and EA 10 ppm in $5{\alpha}-R2$ and DHT reduction were similar to those of $10{\mu}M$ FS. These results suggest that ABMLCE and EA reduced $5{\alpha}-R2$ and DHT through their anti-inflammatory and anti-oxidative actions. This implies that ABMLCE containing EA could be a beneficial material in the cure of BPH in humans.

Antioxidative and nutritional characteristics of Shiitake mushrooms when cooked using different methods (경도 기반 조리 조건을 달리한 표고버섯의 산화방지 및 영양특성 연구)

  • The textural and antioxidant properties, as well as the changes in functional constituents of shiitake mushrooms cooked by three different methods (boiling, steaming, and frying) were analyzed. The hardness of the mushrooms did not change significantly at the end time of cooking for each method. The antioxidant properties determined by measuring the free-radical scavenging activity and total phenolic content were the highest in the control (uncooked shiitake), and these properties were adversely affected after cooking, especially by boiling. Both the riboflavin and eritadenine contents were the highest in the control, but decreased upon cooking, in the following order: frying > steaming > boiling. The ergocalciferol content after frying was less than that in other samples. Although uncooked shiitake mushroom showed a higher concentration of functional constituents, it needs to be cooked adequately owing to its tough texture. If the hardness of the mushrooms cooked by different methods is the same, frying is considered the most acceptable method.

Enhancement of Testosterone in TM3 Leydig Cells by an Eritadenine-containing Agaricus blazei Mycelial Liquid Culture Extract (TM-3 cell에서 eritadenine 함유 신령버섯균사체 액체배양물의 testosterone 생성 촉진효과)

  • Enhancement mechanistic actions of testosterone (TS) productions in mouse Leydig TM3 cells by the eritadenine (EA) and/or the Agaricus blazei mycelial liquid culture extract (ABMLCE). Productions of TS in TM3 cells were investigated in normal and oxidative-stressed culture conditions. In the normal culture condition, TM3 cells grown in a Dulbecco's Modified Eagle's Medium (DMEM) were treated with EA (0~100 ppm) and ABMLCE (10 ppm) + EA (0~50 ppm) for 24 hr, and in the oxidative-stressed culture condition, the cells grown in DMEM containing $50{\mu}M$ $H_2O_2$ to induce oxidative stress for 4 h were treated with the same as those in the normal culture condition. TS content, $3{\beta}$-hydroxysteroid dehydrogenase 2 (HSD3B2) enzyme activity, $5{\alpha}$-reductase 2 ($5{\alpha}-R2$) enzyme activity, and free-radical nitric oxide (NO) content in the culture media were measured using their corresponding assay kits. EA, ABMLCE, and ABMLCE + EA significantly, p<0.05, enhanced TS productions in both cultural conditions, relative to control treatment. The activity of the HSD3B2 enzyme, which is involved in the production of precursors for TS production, was elevated by EA, ABMLCE, and ABMLCE + EA treatments in both culture conditions. The activity of the $5{\alpha}-R2$ enzyme, which converts TS to dihydroxytestosterone (DHT), was not significantly affected in either culture condition by EA, ABMLCE, or ABMLCE + EA treatments. The treatments included reduced NO content. These results indicate that EA, ABMLCE, and EA + ABMLCE treatments elevated TS in TM3 cells via the enhancements of HSD3B2 activity and the reduction of NO production, and also imply that EA and ABMLCE or EA + ABMLCE could be useful materials for the production of TS in humans.
