In order to evlauate feasibility of the gene tagging by the maize transposable element Ac in heterologous plant systems, we have investigated physical distances and directions of transposition of the element in Arabidopsis thaliana and tobacco cultured cell line BY-2. We prepared a T-DNA construct that carried a non-autonomous derivative of Ac with a site for cleavage by endonuclease I-Scel (designated dAc-I-RS element). Another cleavage site was also introduced into the T-DNA region outside dAc-I-RS. A number of transgenic Arabidopsis plants were generated, each of which had a single copy of the T-DNA at a different chromosomal location. To examine the pattern of transposition, three out of these transgenic plants were crossed with the Arabidopsis plant that carried the gene for Ac transposase and progeny in which dAc-I-RS had been transposed were isolated. After digestion of the genomic DNA of these progeny with I-SceI, sizes of segment of DNA were determined byd pulse-field gel electrophoresis. We also performed linkage analysis for the transposed elements and sites of mutations near the elements. Our results with three transgenic lines showed that 50% of all transposition events had occurred within 1, 700 kilo-base pairs (kb) on the same chromosome, with 35% within 200 kb, and that the elements transposed in both directions on the chromosome with roughly equal probability. The data thus indicate that the Ac-Ds system is most useful for tagging of genes that are present within 200 kb of the chromosomal site of Ac in Arabidopsis. In addition, determination of the precise localization of the transposed dAc-I-RS element should definitely assist in map-based cloning of genes around insertion sites. In the present paper, we report typical examples of such gene isolation studies.
Majority of transmission line system failures at many locations worldwide have been caused by severe localized wind events in the form of tornadoes and downbursts. This study evaluates the structural response of two different transmission line systems under equivalent F2 tornadoes obtained from real incidents. Two multi-span self-supported transmission line systems are considered in the study. Nonlinear three-dimensional finite element models are developed for both systems. The finite element models simulate six spans and five towers. Computational Fluid Dynamics (CFD) simulations are used to develop the tornado wind fields. Using a proper scaling method for geometry and velocity, full-scale tornado flow fields for the Stockton, KS, 2005 and Goshen County WY, 2009 are developed and considered together with a previously developed tornado wind field. The tornado wind profiles are obtained in terms of tangential, radial, and axial velocities. The simulated tornadoes are then normalized to the maximum velocity value for F2 tornadoes in order to compare the effect of different tornadoes having an equal magnitude. The tornado wind fields are incorporated into a three-dimensional finite element model. By varying the location of the tornado relative to the transmission line systems, base shears of the tower of interest and peak internal forces in the tower members are evaluated. Sensitivity analysis is conducted to assess the variation of the structural behaviour of the studied transmission lines associated with the location of the tornado relative to the tower of interest. The tornado-induced forces in both lines due to the three different normalized tornadoes are compared with corresponding values evaluated using the simplified load case method recently incorporated in the ASCE-74 (2020) guidelines, which was previously developed based on the research conducted at Western University.

An accurate and efficient numerical method for two-dimensional nonlinear radiation problem has been developed. The wave motion due to a moving body is described by the assumption of ideal fluid flow, and the governing Laplace equation can be effectively solved by the higher-order boundary element method with the help of the GMRES (Generalized Minimal RESidual) algorithm. The intersection or corner problem is resolved by utilizing the so-called discontinuous elements. The implicit trapezoidal rule is used in updating solutions at new time steps by considering stability and accuracy. Traveling waves caused by the oscillating body are absorbed downstream by the damping zone technique. It is demonstrated that the present method for time marching and radiation condition works efficiently for nonlinear radiation problem. To avoid the numerical instability enhanced by the local gathering of grid points, the regriding technique is employed so that all the grids on the free surface may be distributed with an equal distance. This makes it possible to reduce time interval and improve numerical stability. Special attention is paid to the local flow around the body during time integration. The nonlinear radiation force is calculated by the "acceleration potential technique". Present results show good agreement with other numerical computations and experiments.
The main goal of this study is to prepare a program for analyzing High Strength Steel Fibrous Reinforced Concrete (HSSFRC) slabs and predict the response and strength of the slab instead of preparing a prototype and testing it in the laboratory. For this purpose, new equations are proposed to represent the material properties of High Strength Steel Fibrous Reinforced Concrete. The proposed equations obtained from performing regression analysis on many experimental results using statistical programs. The finite element method is adopted for non-linear analysis of the slabs. The eight-node "Serendipity element" (3 DoF) is chosen to represent the concrete. The layered approach is adopted for concrete elements and the steel reinforcement is represented by a smeared layer. The compression properties of the concrete are modeled by a work hardening plasticity approach and the yield condition is determined depending on the first two stress invariants. A tensile strength criterion is adopted in order to estimate the cracks propagation. many experimental results for testing slabs are compared with the numerical results of the present study and a good agreement is achieved regarding load-deflection curves and crack pattern. The response of the load deflection curve is slightly stiff at the beginning because the creep effect is not considered in this study and for assuming perfect bond between the steel reinforcement and the concrete, however, a great agreement is achieved between the ultimate load from the present study and experimental results. For the models of the tension stiffening and cracked shear modulus, the value of Bg and Bt (Where Bg and Bt are the curvature factor for the cracked shear modulus and tension stiffening models respectively) equal to 0.005 give good results compared with experimental result.
A phase-change material is a substance with a high heat of fusion which, melting and freezing at a certain temperature, is capable of storing and releasing large amounts of energy. Heat is absorbed or released when the material changes from solid to liquid. Therefore, PCMs are classified as latent heat storage (LHS) units. The purpose of this study is to analyze PCM wallboard design parameters using dynamic energy simulation. Among the factors of PCM, melting temperature, latent heat, phase change range, thermal conductivity are very important element to maximize thermal energy storage. In order to analyze these factors, EnergyPlus which is building energy simulation provided by department of energy from the U.S is used. heat balance algorithm of energy simulation is conduction finite difference and enthalpy-temperature function is used for analyzing latent heat of PCM. The results show that in the case of melting temperature, the thermal energy storage could be improved when the melting temperature is equal to indoor surface temperature. It seems that when the phase change range is wide, PCM can store heat at a wide temperature, but the performance of heat storage is languished.

In this paper, the governing differential equations for the free vibration of uniform parabolic arches are derived on the basis of equilibrium equations of a small element of arch rib and the D'Alembert principle. A trial eigen value method is used for determining the natural frequencies and mode shapes. And the Runge-Kutta fourth order integration technique is also used in this method to perform the integration of the differential equations. A detailed study is made of the first mode for the symmetrical and anti-symmetrical vibrations of hinged arches with the Span length equal to 10 m. The effects of the rise of arch, the radius of gyration and the rotary inertia on free vibrations are presented in detail in curves and table.
The current study was designed to scientifically evaluate the atmospheric particulate pollution in residences relative to their proximity to a Korean major iron/metal industrial complex (IMIC). This objective was achieved by measuring the concentrations and elemental composition of particulate matter with aerodynamic diameters equal to or less than 10 ${\mu}m$(PM10) in industrial ambient air from IMIC and residential ambient air with relative proximities to IMIC. The trace metals were analyzed using an inductively coupled plasma-atomic emission spectroscopy (ICP-AES). The industrial mean values exceeded the Korean year/70-${\mu}g/m^3$ standard for PM10, whereas the residential mean values did not. However, the maximum residential values did exceed or were close to the Korean PM10 year standard. For individual elements, the ambient concentrations ranged widely from values in the order of a few $ng/m^3$ to thousands of $ng/m^3$. The residential mean mass concentrations in the PM10 measured in the present study were higher than or similar to those reported in earlier studies. This study suggests that residents in neighborhoods near the IMIC are exposedto elevated particulate levels compared to residents living further away from such a source.

This paper presents a design process of light-weighted fuel cell vehicle (FCV) frame to meet design target of natural frequency in early design stage. At first, using validated FE model for the current design, thickness optimization was carried out. Next. optimization process, comprised of beam model size optimization, shell model design and shell model thickness optimization, was investigated for two frame types. In addition, in order to ensure hydrogen tanks safety against rear impact load, structural collapse characteristics was estimated for the rear frame model finally produced from the previous optimization process and, with the target of equal collapse characteristics to the current design model, structural modification with small weight increase was studied through static structural collapse analyses. The same attempt was applied to the front side frame. The results explain that the proposed process enables to design light-weighted frames with high structural performance in early stage.

High-rise apartments of shear wall system are governed by flexural behavior like a cantilever beam. Installation of the damper-brace system in a structure governed by flexural behavior is not suitable. Because of relatively high lateral stiffness of the shear wall, a load is not concentrate on the brace and the brace cannot perform a role as a damping device. In this paper, a friction damper applying flexibility of shear wall is proposed in order to reduce the deformation of a structure. To evaluate performance of the proposed friction damper, nonlinear time history analysis is executed by SeismoStruct analysis program and MVLEM(multi vertical linear element model) be used for simulating flexural behavior of the shear wall. It is found that control performance of the proposed friction damper is superior to one of a coupled wall with rigid beam. In conclusion, this study verified that the optimal control performance of the proposed friction damper is equal to 45 % of the maximum shear force inducing in middle-floor beam with rigid beam.

Hydro dynamically developing and simultaneously (hydro dynamically and thermally) developing laminar flows of a Bingham plastic in a circular pipe have been investigated numerically. Solutions have been obtained by using a four-step fractional method combined with an equal order bilinear finite element method. For the hydro dynamically developing flow, shorter entrance length is required to reach fully developed velocity field for larger yield stress and non-monotonic pressure drop along the pipe centerline is observed when the yield stress exceeds a certain critical value. For the simultaneously developing flow, the heat transfer characteristics show the same trends as those predicted for the thermally developing flow (Graetz problem).
