Skin biopsy: an emerging method for small nerve fiber evaluation

  • Skin biopsy and staining the specimens with immuno-reactive markers has been proven to be a useful method to demonstrate the pathologic status of small nerve fibers. Quantification of intraepidermal nerve fiber density using anti-protein gene product 9.5 antibody is a standard method to diagnose small fiber neuropathy. Skin biopsy also makes it possible to differentiate the nerve fibers according to their function by using different markers. Quantification of dermal structures with different types of nerve fibers could reveal the pathophysiologic mechanism of the disease state.

  • Author has studied and reported on taxonomy of Korean sedges, using gross morphology, anatomy and epidermal patterns of the leaf blades(1969, 1971, 1973, 1974). This paper is the 6th report of epidermal patterns of leaf blade on sedges and includes 5 genera, Eriophorum, Fuirena, Kobresia, Rhynchospora and Scirpus. The author proposed to find epidermal patterns of leaf blades as an important taxonomic characteristic of sedges classification. The result of this study, the elements of leaf epidermis, subsidal cells, silica body, cell wall of long cell, prickles, and arrangement of the elements are considered to be significant characteristics for the identification and classification of sedge.

  • Preventive effect of red ginseng saponin on experimentally-induced hyperkeratosis was investigated by ultrastructural observation, skin weight and epidermal lipid analysis. Hexadecane increased skin weight per unit area and epidermal lipids, free fatty acids, cholesterol and triglyceride in guinea pig skin. Topical application of ginseng saponin reduced these hyperkeratotic responses regradless of the concentration and the purity of ginseng saponin. Ultrastructurally, lipids and empty space-containing multiple horny cells were piled and nuclear remnants, desmosome, desmosomal bodies, tight junction were shown in the stratum corneum of hexadecane-treated skin.

  • The cutaneous network of unmyelinated nerve fibers is extremely dense, and closely interacts with the many cell types present in dermis and epidermis, including keratinocytes, fibroblasts, Langerhans cells, and melanocytes. Cell communication involves various neuroendocrine factors, with cell differentiating and proliferative activities, or inflammatory properties. Thus, nervous cells in the skin not only create a sensory system connected to the central nervous system, but also mediate many of the biological activities of the skin.(omitted)

Establishment of an Efficient Agrobacterium Transformation System for Eggplant and Study of a Potential Biotechnologically Useful Promoter

  • An efficient and reliable Agrobacterium transformation procedure based on TDZ (thidiazuron)-induced organogenesis was established and applied to six Brazilian eggp1ant varieties. Optimum transgenic plants recovery was achieved upon the study of the following parameters affecting transformation efficiency, using F-100 variety as a model: i) explant source; ii) pre-culture period; iii) physical state of the pre-culture medium and iv) coculture conditions. The highest frequency of kanamycin-resistant calli derived from leaf explants (5%) was obtained without a pre-culture period and co-cultivation for 24 h in liquid medium followed by five days on solid RM (regeneration medium). For cotyledon explants, best results were achieved upon a pre-culture of 24 h in liquid RM and a co-cultivation period of 24 h in liquid RM followed by three days in solid RM, resulting in a transformation Sequency of 22.7%. Kanamycin-resistant organogenic calli were also obtained from cultivars Emb, Preta Comprida, Round nose Shaded, Campineira and Florida Market. The expression pattern of an epidermis-specific promoter was studied using transformants expressing a chimaeric construct comprised by the promoter Atgrp-5 transcriptionally fused to the coding region of the gus gene. The expression pattern was similar to that previously observed in tobacco and Arabidopsis thaliana, with preferential expression at the epidermis and the stem phloem. These results support the idea that the Atgrp-5 promoter can be used to drive defense genes in these tissues, which are sites of pathogen interaction and spread, in programs for the genetic improvement of eggplant.

  • To explore the possibility of using pyroligneous acid for environmentally friendly ginseng farming, this study observed samples of ginseng whose shoots were treated with pyroligneous acid sprays beginning in mid June, which is after foliation stage. The spongy tissue structure got thickened from triple layers to quadruple layers with the pyroligneous acid regardless of the concentration. The upper and lower epidermis cell of the leaves as well as the leaf mesopyll cells also became thicker. Compared to the no-treatment group, the overall growth and development of ginseng roots treated with pyroligneous acid were excellent. Accordingly, it is believed that pyroligneous acid can be an environmentally friendly alternative to conventional agro-chemicals applied to ginseng that can be used to facilitate the growth and development of ginseng.

Ecotypic Variation of Pulsatilla koreana Nakai Distributed in Korea (韓國에 分布하는 할미꽃 (Pulsatilla Koreana Nakai)의 生態型的 變異)

  • In order to investigate the ecotypic variations of pulsatilla koreana nakai distributed in korea, 8 local populations of the plant were selected in accordance with latitude and compared on the differences of leaf shape, elongstion, stomatal distribution and dhlorophyll content. The incised angle of the plant is 38。32' in cheju, showing a great difference as compared with 23。77'-30。 32' of the other sites. as to the incised rate, mt. togyu and cheju representsed relatively high values of 49.04% and 48.21% the parted leaved individuals were only observed along the warm current coastal area including cheju, sokcho and sunchon and were 54(62.8%), 2(2.5%) and 3(3.3%) in the sites, respectively. Especially, the individuals consisting of only parted learves were 5(5.8%) in cheju alone and none of the other sites showed such individuals at all. So p. cernua distributed in cheju and p. koreana in inland area can be considered to beecotypic variations of the same species from the upper investigations. According to the elongations of leaf and rachis, the local populations could be classified into 3 types, such as warm current coastal area type(cheju,sunchon and sokcho),southern inland area type(andong, chungju and mt. togyu)and central inland ara type(mt. komdan and hongchon). The dentate numbera of 39.80-43.73 in the warm current area were also different from those of 87% in mt.togyu and 15% in hongchon, while opposite leaflets account for 100% in the other sites. The content of chlorophyll ranged from 0.609 mg/g. d.w.(cheju)to 0.924 mg/g.d.w.(hongchon), showing an increasing trends as the latitudes grow higher. The number of stomata on the low epidermis is much larger than that on the upper epidermis in the whole area. But both aspects showed an increasing tendency, too as the latitude becomes higher.

A Dermal Turnover Flap for Treating the Accessory Tragus (부이주에서 진피전환피판술을 활용한 새로운 치료법)

  • Purpose: Accessory tragus is a fairly common congenital malformation and usually located at pretragal area. Surgical removal is a common treatment of accessory tragus irrespective of location and morphology. Most accessory tragi do not have depression site around them, but some do. So in those cases, simple surgical excision was not enough to promote the aesthetic facial appearance. For depression site remodeling, the excess amount of skin and cartilage need to be remained partially instead of total excision. This method can achieve the symmetric contour of pretragal area. The authors excised the epidermis and cartilaginous tissue totally and remained the dermis for reconstruction of the depression site around accessory tragus. The depression site is filled with dermal turnover flap. The purpose of this report is to present new idea to promote cosmetic result in treatment of accessory tragus containing the depression site. Methods: Two patients had a pair of accessory tragi at pretragal area. One was a common featured accessory tragus, but the other was different. Depression site was found around accessory tragus. After epidermis and cartilaginous tissue were removed from it, dermis component was used as turnover flap for reconstruction of depression site. Results: After accessory tragus was removed and depression site was reconstructed, facial contour and cosmetic result was achieved. Complication such as flap necrosis and wound dehiscence was not observed. Conclusion: The accessory tragus has variant morphology and degree of invasive depth. And some has a depression site around them. In those cases, simple surgical removal results in morphological distorsion and do not promote facial symmetry. The authors suggest dermal turnover flap as reconstruction method of the depression site. This method improves both surgical outcome and cosmetic result.

Difference in Percent Penetration, Hyphal Growth and Lesion Formation by Pre­ and Postdisposing Temperatures (기온변동이 벼 도열병균의 엽신에의 침입과 발병에 미치는 영향 II. 접종전 및 접종시의 온도처리에 의한 침입$\cdot$균사신전$\cdot$병반형성 차이)

  • Three rice cultivars, Aichi-asahi, Toyotama and Yamabiko, possessing a resistance gene $Pi-\alpha$ were evaluated for penetration, hyphal growth in the leaf epidermis and lesion formation using 6 isolates of Pyricularia oryzae by treating pre- and post disposing temperatures of $23/15^{\circ}C\;and\;29/21^{\circ}C$ (day/night) regimes, respectively. Percent penetration of the fungus was higher on the seedlings disposed at $29/21^{\circ}C$ regime and more lesions were formed at 7 days after inoculation than at $23/15^{\circ}C$ regime. Degree of hyphal growth and number of host cells with hyphal growth were remarkably increased from 72 to 96 hr after inoculation at $29/21^{\circ}C$ regime. However, lesion formation on the seedlings disposed at $23/15^{\circ}C$ regime was delayed, possibly as a result of the suppressed hyphal growth until 96 hr after inoculation.

Development of a Chronic Skin Wound Defect: A Swine Model (돼지를 이용한 만성피부결손상처의 모델)

  • Purpose: Animal models of a chronic wound are yet to be fully developed, and animal studies on this subject has yet to take place. The purpose of this study is to create the foundation for research on chronic wound healing based on a swine model, the most similar to that of a human. Methods: Three female 2-3 month old 'yolkshires' were used. Total of eight full thickness skin defects, $6{\times}3cm$ sized, were created on the back of each pigs. Three groups were created for comparison; Group I (n=4) was left as they were after full skin thickness excision, while the excised tissues of Group II (n=3) were turned inside out and sutured so that the epidermis would come in contact with the fascia. Group III (n=3) were excised full skin thickness in depth and silicone blocks were implanted in them. Dressing was not practised so that the wounds would be vulnerable to infection. Results: In Group III, the skin contraction rate was the least among the three groups for each three weeks of observation respectively. Also during the three weeks, bacteral colonization was at the highest among the comparison. On the third week, inflammatory cells were still active, but the generations of epidermis and collagen synthesis were detected minimally. Conclusion: The Group III was relatively the most similar model of chronic wounds. and modification of the silicone blocks, could provide us with a very effective chronic skin wound model similar to human.
