Epidermal growth factor(EGF), a potential healing agent for wounds and burns, has been formulated to topical gels with the hydrophilic polymers, Carbopol 934P. Physicochemical c haracteristics of the aqueous gels were evaluated by rheological properties and pH changes on storage. The gels were relatively stable at $4^{\circ}C$ and room temperature showing no changes in pH for two weeks, and revealed the rheogram of shear thinning plastic flow with the yield values in the range of 40 to 70dyne/$cm^2$. In vivo healing effects of different gel formulations have been compared with water-soluble and oleaginous ointments in full-thickness wound mouse model. The gel systems resulted in better wound healing effects than the other ointments. Furthermore, liposomal Carbopol gel has been developed by the addition of EGF-containing liposomal suspension into the Carbopol gel. The enhanced wound healing effects have been observed in the liposomal gel system, compared to the other gels and conventional ointments.

BACKGROUND/OBJECTIVES: Ultraviolet radiation (UV) is a major cause of skin photoaging. Previous studies reported that ethanol extract (PET) of Prunus persica (L.) Batsch flowers (PPF, peach flowers) and its subfractions, particularly the ethylacetate (PEA) and n-butanol extracts (PBT), have potent antioxidant activity and attenuate the UV-induced matrix metalloproteinase (MMP) expression in human skin cells. In this study, we investigated the protective activity of PPF extract against UV-induced photoaging in a mouse model. MATERIALS/METHODS: Hairless mice were treated with PET or a mixture of PEA and PBT either topically or orally along with UV irradiation. Histological changes and biochemical alterations of mouse skin were examined. Major phenolic compounds in PPF extract were analyzed using an ACQUITY UPLC system. RESULTS: The overall effects of topical and oral treatments with PPF extract on the UV-induced skin responses exhibited similar patterns. In both experiments, the mixture of PEA and PBT significantly inhibited the UV-induced skin and epidermal thickening, while PET inhibited only the UV-induced epidermal thickening. Treatment of PET or the mixture of PEA and PBT significantly inhibited the UV-induced MMP-13 expression, but not type I collagen expression. Topical treatment of the mixture of PEA and PBT with UV irradiation significantly elevated catalase, superoxide dismutase (SOD) and glutathione-peroxidase (GPx) activities in the skin compared to those in the UV irradiated control group, while oral treatment of the mixture of PEA and PBT or PET elevated only catalase and SOD activities, but not GPx. Thirteen phytochemical compounds including 4-O-caffeoylquinic acid, cimicifugic acid E and B, quercetin-3-O-rhamnoside and kaempferol glycoside derivatives were identified in the PPF extract. CONCLUSIONS: These results demonstrate that treatment with PET or the mixture of PEA and PBT, both topically or orally, attenuates UV-induced photoaging via the cooperative interactions of phenolic components having anti-oxidative and collagen-protective activities.

Background: Kaempferol (3,4',5,7-tetrahydroxyflavone) is a flavonoid known to have a wide range of pharmacological activities. The 3-OH group in flavonoids has been reported to determine antioxidant activities. Objective: We tested whether kaempferol can affect the expression of integrins and the stem cell fate of interfollicular epidermal stem cells. Methods: Skin equivalent (SE) models were constructed, and the expression levels of stem cell markers and basement membrane-related antigens were tested. The immunohistochemical staining patterns of integrins, p63, and proliferating cell nuclear antigen (PCNA) were compared between kaempferol- and apigenin-treated SE models. Reverse transcription-polymerase chain reaction (RT-PCR) was used to evaluate the mRNA expression of integrins. Results: Kaempferol increased the thickness of the epidermis when added to prepare SEs. In addition, the basal cells of kaempferol-treated SEs appeared more columnar. In the immunohistological study, the expression of integrins ${\alpha}6$ and ${\beta}1$ and the numbers of p63- and PCNA-positive cells were markedly higher in the kaempferol-treated model. However, apigenin showed no effects on the formation of three-dimensional skin models. RT-PCR analysis also confirmed that kaempferol increased the expression of integrin ${\alpha}6$ and integrin ${\beta}1$. Conclusion: Our findings indicated that kaempferol can increase the proliferative potential of basal epidermal cells by modulating the basement membrane. In other words, kaempferol can affect the fate of interfollicular epidermal stem cells by increasing the expression of both integrins ${\alpha}6$ and ${\beta}1$. These effects, in particular, might be ascribed to the 3-OH group of kaempferol.
'Merlot' 포도 품종의 수확전 ethyl oleate 처리는 외표피와 아표피의 두께를 감소시키며 착색을 현저히 향상시킨다. 무처리구의 과피 두께는 $126-189{\mu}m$인데 반하여 처리 과실의 과피두께는 $90-107{\mu}m$로 조사되었으며, 이러한 외표피와 아표피 층의 두께 감소는 처리 후 외표피와 아표피를 구성하는 세포의 빠른 노화에 의하여 세포가 죽거나 탈수가 나타나기 때문인 것으로 조사되었다. 처리 후에 빠르게 과피 표면의 왁스층이 녹은 듯한 형태를 보이며 이는 착색을 증진시키는 것으로 관찰되었으며, 전체적인 안토시아닌 함량 역시 증가하는 것으로 조사되었다. 반면에 각각의 안토시아닌 함량에 있어서는 메톡시기와 결합하거나 유기산과 결합한 안토시아닌이 증가한 반면 수산기와 결합한 안토시아닌은 감소하는 경향이었다.
본 실험은 황금열수추출물이 알레르기성 접촉 피부염 유발 시 항산화활성, serum Ig E농도와 표피회복능에 미치는 영향을 조사하기 위해 행해진 것으로 1-chloro-2,4-dinitrochlorobenzene(DNCB: 2.5% DNCB 1 mL로 감작, 4일 경과 후 1% DNCB 1 mL로 유발) 처리로 알레르기성 접촉 피부염이 유발된 Sprague Dawley rat 수컷 흰쥐에 황금열수추출물(1 mL/kg/day)을 경구투여한 다음 혈장과 간의 항산화활성, 혈중 Ig E 농도, 표피두께변화를 관찰하였다. DNCB 처리에 의해 지질과산화물 함량과 Ig E 농도, 표피두께가 증가하였으며 황금열수추출물의 경구투여에 의해 간의 지질과산화물 함량 감소, 혈중 Ig E 농도 감소, 표피두께가 저하되는 결과를 보였다. 이상의 결과로 미루어 황금열수추출물이 알레르기성 접촉피부염으로 유발된 rat의 체내 항산화활성과 Ig E 농도개선, 표피두께 개선에 효과가 있는 것으로 생각된다.
본 실험은 오가피열수추출물이 알레르기성 접촉 피부염을 유발한 흰쥐의 체내 지질 조성, 항산화활성, serum Ig E 수준과 표피회복능에 미치는 영향을 조사하기 위해 행해진 것으로 1-chloro-2,4-dinitrochlorobenzene(DNCB: 2.5% DNCB 1 mL로 감작, 4일 경과 후 1% ONCB 1 mL로 유발) 처리로 알레르기성 접촉 피부염이 유발된 Sprague-Dawley 수컷 흰쥐에 오가피열수추출물 (100 mg/체중 kg/day)을 경구투여 한 다음 혈장과 간의 지질조성과 항산화활성, 혈중 Ig E 농도, 표피두께변화를 관찰하여 다음과 같은 결과를 얻었다. 혈장의 total cholesterol과 LDL-cholesterol은 정상군에 비해 ACFG군에서 유의적으로 감소하여(p<0.05) 오가피 열수추출물이 지질개선에 효과적인 것으로 나타났다. 혈장과 간의 지질과산화물 함량은 정상군에 비해 CG군과 ACFG군에서 지질과산화물 함량이 유의하게 높았으며(p<0.05), ACFG군이 CG군에 비해 과산화물 함량이 감소하는 경향을 보였다. 간의 SOD효소활성은 CG군에서 정상군에 비해 유의하게 증가하였으며(p<0.05), ACFG군에서 정상군의 수준으로 유의하게 감소하였다(p<0.05). Serum 중 Ig E 수준은 정상군에 비해 CG군에서 유의하게 증가하였으며(p<0.05), ACFG군에서 CG군에 비해 정상군의 수준으로 유의하게 감소하였다(p<0.05). 표피두께는 정상군에 비해 CG군에서 표피두께가 유의적으로 증가하였으며(p< 0.05), ACFG 군에서 CG군에 비해 유의하게 감소하였다(p<0.05). 혈장과 간의 지질과산화물 함량과 표피두께의 상관계수가 각각 r=0.644(p<0.01), r=0.516(p<0.05) 로 비교적 높은 정의 상관관계를 보였다. 이상의 결과로 미루어 볼 때 정상 흰쥐에 비하여 DNCB에 의해 유도된 접촉성 피부염을 가진 흰쥐의 혈장과 간의 지질과산화물 생성이 증가되며 이로 인해 표피의 두께가 증가하고, SOD 활성이 증가하는 것으로 나타났다. 오가피열수추출물의 섭취로 알레르기성 접촉피부염으로 유발된 흰쥐의 체내 지질과산화물 생성 억제, Ig E 농도감소, 표피두께 감소되는 효과가 나타났다.
Ursolic acid (UA) and Oleanolic acid (ONA), known as urson, micromerol and malol, are pentacyclic triterpenoid compounds which naturally occur in a large number of vegetarian foods, medicinal herbs, and plants. They may occur in their free acid form or as aglycones for triterpenoid saponins, which are comprised of a triterpenoid aglycone, linked to one or more sugar moieties. Therefore UA and ONA are similar in pharmacological activity. Lately scientific research, which led to the identification of UA and ONA, revealed that several pharmacological effects, such as antitumor, hepato-protective, anti-inflammatory, anticarcinogenic, antimicrobial, and anti-hyperlipidemic could be attributed to UA and ONA. Here, we introduced the effect of UA and ONA on acutely barrier disrupted and normal hairless mouse skin. To evaluate the effects of UA and ONA on epidermal permeability barrier recovery, both flanks of 8-12 week-old hairless mice were topically treated with either 0.01-0.1 mg/ml UA or 0.1-1 mg/ml ONA after tape stripping, and TEWL (Transepidermal water loss) was measured . The recovery rate increased in those UA or ONA treated groups (0.1 mg/ml UA and 0.5 mg/ml ONA) at 6 h more than $20\%$ compared to vehicle treated group (p<0.05). Here, we introduced the effects of UA and ONA on acute barrier disruption and normal epidermal permeability barrier function. For verifying the effects of UA and ONA on normal epidermal barrier, hydration and TEWL were measured for 1 and 3 weeks after UA and ONA applications (2mg/ml per day). We also investigated the features of epidermis and dermis using electron microscopy (EM) and light microscopy (LM). Both samples increased hydration compared to vehicle group from f week without TEWL alteration (p<0.005). EM examination using RuO4 and OsO4 fixation revealed that secretion and numbers of lamellar bodies and complete formation of lipid bilayers were most prominent $(ONA{\geq}UA>Vehicle)$. LM finding showed that thickness of stratum corneum (SC) was slightly increased and especially epidermal thickening and flattening was observed (UA>ONA>Veh). We also observed that UA and ONA stimulate epidermal keratinocyte differentiation via $PPAR\;\alpha$. Protein expression of involucrin, loricrin, and filaggrin increased at least 2 and 3 fold in HaCaT cells treated with either $ONA\;(10{\mu}M)$ or UA $(10{\mu}M)$ for 24h respectively. This result suggested that the UA and ONA can improve epidermal permeability barrier function and induce the epidermal keratinocyte differentiation via $PPAR\;{\alpha}$. Using Masson-trichrome and elastic fiber staining, we observed collagen thickening and elastic fiber elongation by UA and ONA treatments. In vitro results of collagen and elastin synthesis and elastase inhibitory activity measurements were also confirmed in vivo findings. These data suggested that the effects of UA and ONA related to not only epidermal permeability barrier functions but also dermal collagen and elastic fiber synthesis. Taken together, UA and ONA can be relevant candidates to improve epidermal and dermal functions and pertinent agents for cosmeseutical applications.
소라 (Batillus cornutus) 외투막의 조직화학적 특성 및 미세구조를 광학현미경과 투과전자현미경을 이용하여 기재하였다. 소라의 외투막은 결합조직층을 단층상 피층이 위아래로 둘러싸고 있는 구조였으며, 상피층은 내장 기관계 쪽을 향하고 있는 내부상피층과 패각 쪽의 외부상피층으로 구분된다. 상피층에 의해 둘러싸인 결합조직층은 주로 교원섬유와 근섬유다발로 구성되며, 혈림프동이 발달하고 있었다. 외투막의 각정부 상피층에 분포하는 점액세포들은 산성 및 중성 점액다당류를 함유하고 있었으며, 중간부와 가장자리의 상피층에 분포하는 점액세포들은 비황화 강산성의 당단백질 점액성분을 함유하는 것으로 나타났다. 외투막의 두께, 상피층의 두께, 혈림프동의 면적은 각정부에서 가장자리로 갈수록 낮아지는 경향을 보였다. 투과전자현미경으로 관찰하였을 때 외투막의 상피층은 단층으로 원주형 상피세포, 섬모세포, 흡수세포, 분비세포로 구성되어 있었다. 외투막 상피세포들은 원주형으로 장방형의 핵을 가지며, 자유면에는 미세융모들이 발달되어 있었다. 상피세포 사이에는 폐쇄띠, 폐쇄대와 수지상막구조로 연결되어 있었다. 섬모세포는 자유면에 섬모와 미세융모를 가지며, 세포질의 상부에는 다수의 미토콘드리아를 가지고 있었다. 상피층에서는 흡수기능을 가지는 두 종류의 세포가 관찰되었다. 이들은 원주형으로 미세융모, 음소포, 미토콘드리아 그리고 전자밀도가 다양한 용해소체들을 함유하고 있었다. 소라 외투막에서는 네 종류 (A, B, C, D)의 분비세포를 구분할 수 있었는데, 이들은 모두 단세포선으로 확인되어 대부분의 복족류와 이매패류의 외투막에서 보고된 선세포들과 유사한 특징을 보였다.
TBTCl에 36주 동안 노출된 대복(Gomphina veneriformis)의 성장과 외투막의 조직학적 변화를 관찰하였다. 대복의 성장은 모든 실험구의 개체들이 12주 이후부터 대조구에 비해 유의적인 차이를 보이며 감소하였다(p < 0.05). 대복의 외투막은 가장자리에 4 개의 주름 (내부 안쪽, 외부 안쪽, 가운데, 바깥쪽)을 가지며, 단층의 원주형 상피층이 결합조직층을 둘러싸고 있는 구조였다. 대복의 외투막은 TBTCl 노출 12주 이후부터 혈림프동이 확장과 근섬유 다발의 분절이 관찰되었다. 노출 20주에는 내부상피층에 존재하는 점액세포들이 대조구보다 유의적인 차이를 보이며 감소하였지만(p < 0.05), 외투막 가장자리에 존재하는 주름들에서는 노출 12주까지 감소하였던 점액세포가 증가하면서 AB-PAS (pH 2.5) 반응시 보라색 (2587c)으로 관찰되는 점액물질이 관찰되었다. 이후 노출 28주에는 가운데 주름 부위의 안쪽 분지에서 상피세포의 변형이 관찰되었고, 외부 상피층에서는 상피세포의 부분적인 파괴가 관찰되었다. 실험 종료시기인 36주에는 바깥쪽 주름 부위에 호산성 과립들이 관찰되었으며, 가운데 주름부위의 바깥 분지에서 상피세포의 변형과 파괴로 인한 상피층의 붕괴가 관찰되었다. 또한 외부 상피층의 상피세포들은 입방형 또는 편평형으로의 변화와 함께 부분적인 상피층의 붕괴가 나타났고, 그로 인하여 상피층의 두께가 감소하였다. 따라서 섬모의 탈락과 점액세포의 감소 그리고 상피층의 파괴는 외투강 정화 기능의 장애로 인한 먹이 섭식의 장애를 유발할 것으로 생각되며, 특히 외투막 가장자리에 존재하는 주름들의 구조적인 변화는 각피층 형성을 억제하여 패각 성장 장애의 직접적인 원인으로 생각되어진다.
Purpose: The purpose of this study was to evaluate the effects of essential oil on oxidative stress, immunity, and skin condition in atopic dermatitis (AD) induced mice. Methods: This study was a $3{\times}3$ factorial design. Factors were oil type (Lavender, Thyme, and 2:1 mixture of lavender and thyme oil [blending oil]) and treatment period (0 day, 7 days, and 21 days). The samples were 45 mice with AD and randomly assigned to nine groups of five mice per group. The dependent variables such as superoxide radical, IgE, degranulated mast cells, and epidermal thickness were measured. Data were collected from February to April in 2014. Descriptive statistics, One-way ANOVA, Two-way ANOVA, and Tukey's HSD test were performed using the SPSS WIN 20.0 program. Results: Dependent variables were not statistically significantly different by the three oil types (p >.05). Essential oils such as lavender, thyme, and blending oil were all effective in reducing AD symptoms and especially 2:1 blending oil were most effective. There were statistically significant differences by the three treatment periods in all dependent variables (p <.001). There were statistically significant interactions between oil types and treatment periods in all dependent variables (p <.01). For decreasing superoxide radical, degranulated mast cells, and epidermal thickness, 2:1 mixed oil should be applied for at least 21 days. Otherwise to reduce IgE, 2:1 mixed oil should be used for at least 7 days. Conclusion: These findings provide bases for developing effective interventions for AD patients to manage their AD symptoms.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
