Effect of Dietary Minerals and Ca-Regulating Hormones on Bone Enzyme Alkaline Phosphatase Activity

  • Parathyroid hormone(PTH) is known to stimulate bone resorption and to inhibit bone collagen synthesis. In contrast, as the evidence of stimulation of bone formation by PTH has recently been observed, the study on the role of PTH involved in osteoporosis draws remarkable attention. This study has dealt with the role of alkaline phoshatase(AP), a marker enzyme for bone formation and osteoblast action, Animals(BALS/cmice) were divided into three dietary groups(high and medium Ca and Ca-free) and hormones including PTH, calcitonin(CT), cholecalciferol(citamin D) were i.p. injected. AP in the serum and liver was measured using Sigma 221 alkaline buffer solutions containing 9mM of p-nitrophenyl phoshate. Enzume was reacted at 37$^{\circ}C$ for 10 minutes and the reaction was stopped by 1.8ml of 0.1N NaOH and measured at 410nm. We found that serum and liver AP activity was increased by low dietary Ac. Compared to the control, and serum Ap activity was enhanced by PTH and vitamin D regardless of the dietary Ca. On the other hand, liver AP activity was inhibited by OTH and vitamin D at all levels of dietary Ca. CT inhibited the action of PTH and vitamin D in the serum. But, the inhibition of PTH and vitamin D action by CT was not observed in the liver, unlike in the case of serum.

Pre-harvest ethylene control affects vase life of cut rose 'Carola' by regulating energy metabolism and antioxidant enzyme activity

  • We studied the role of ethylene control in regulating energy metabolism, antioxidant enzyme activity, and vase life of cut rose Rosa hybrida 'Carola'. Rose flowers at stage II were sprayed with one of the following solutions: water (control), $10{\mu}L\;L^{-1}$ 1-methylcyclopropene (1-MCP), or $0.5g\;L^{-1}$ 2-chloroethanephosphonic acid (ethephon). After harvest, ethylene production rate, respiration intensity, energy charge (EC), activities of energy metabolism-related and antioxidant enzymes, and malondialdehyde (MDA) content were measured. Results showed that 1-MCP enhanced the activities of superoxide dismutase, $H^+$-adenosine triphosphatase, $Ca^{2+}$-adenosine triphosphatase, succinic dehydrogenase, and cytochrome c oxidase, increased adenosine triphosphate (ATP) content, maintained high EC levels, inhibited respiration intensity, reduced peroxidase (POD) and polyphenol oxidase (PPO) activity and MDA accumulation, and prolonged vase life. Ethephon promoted ethylene production and respiration intensity, increased POD and PPO activity, reduced ATP content and EC levels, and accelerated senescence. Our results support a novel role for ethylene control in regulating senescence of 'Carola'.

Properties of Porphyran and Hemicellulose extracted with Different extract Solutions and Enzymatic Pretreatments from Porphyra (추출 용매 및 효소 전처리 방법에 따른 포피란과 헤미셀루로오즈의 특성)

  • Laver, Porphyra, is distinctive for its high content of proteins and polysaccharides such as porphyran and hemicellulose. The chemical properties of the polysaccharides extracted with different extraction methods such as hot water, dilute acid(pH 4.0) or alkali solution(2N NaOH) were examined to investigate the suitable extraction conditions for porphyran and hemicellulose from laver. For porphyran extraction, dilute acid solution was more preferable to hot water and alkali solution because of its higher 3,6-anhydrogalactose content and lower protein content. However, alkali solution was more suitable to extract the hemicellulose because of higher mannose content indicating the extraction of mannan. To decrease contamination of the polysaccharides with protein, the dried lavers were pretreated with enzymes (Protamex, Flavourzyme, Alcalase, Viscozyme) before hot water extraction. All enzyme pretreatments increased the yield of polysaccharides by compared with control (enzyme unpretreated) and Flavourzyme pretreatment was most effective to decrease protein contamination in the polysaccharide. All viscosities of porphyran solutions pretreated by enzymes were lower compared to the control porphyran solution and showed pseudoplastic behavior with yield stress. In case of alkali extraction of residues obtained after enzyme hydrolysis and hot water extraction, protease pretreatment increased the mannose contents in the polysaccharide while the xylose content was increased by Viscozyme pretreatment.

DNA Computing Adopting DNA Coding Method to solve Maximal Clique Problem (Maximal Clique Problem을 해결하기 위한 DNA 코딩 방법을 적용한 DNA 컴퓨팅)

  • DNA computing has used to solve MCP (Maximal Clique Problem). However, when current DNA computing is applied to MCP. it can't efficiently express vertices and edges and it has a problem that can't look for solutions, by misusing wrong restriction enzyme. In this paper we proposed ACO (Algorithm for Code Optimization) that applies DNA coding method to DNA computing to solve MCP's problem. We applied ACO to MCP and as a result ACO could express DNA codes of variable lengths and generate codes without unnecessary vertices than Adleman's DNA computing algorithm could. In addition, compared to Adleman's DNA computing algorithm, ACO could get about four times as many as Adleman's final solutions by reducing search time and biological error rate by 15%.

Screening, Characterization, and Cloning of a Solvent-Tolerant Protease from Serratia marcescens MH6

  • solvent-tolerant bacterium strain, MH6, was isolated by hydrophilic organic solvent DMSO enrichment in the medium and identified as Serratia marcescens. The extracellular protease with novel organic-solvent-stable properties from strain MH6 was purified and characterized. The molecular mass of the purified protease was estimated to be 52 kDa on SDS-PAGE. The open reading frame (ORF) of the MH6 protease encoded 504 amino acids with 471 amino acid residues in the mature protease. Based on the inhibitory effects of EDTA and 1,10-phenathroline, the MH6 protease was characterized as a metalloproteinase. The enzyme activity was increased in the presence of $Ni^{2+}$, $Mg^{2+}$, and $Ca^{2+}$. The protease could also be activated by the nonionic surfactants Tween 80 (1.0%) and Triton X-100 (1.0%). The protease showed remarkable solvent stability in the presence of 50% (v/v) solutions of long-chain alkanes and long-chain alcohols. It was also fairly stable in the presence of 25% solutions of hydrophilic organic solvents. Owing to its high stability in solvents and surfactants, the MH6 protease is an ideal candidate for applications in organic catalysis and other related fields.

Characteristics on the Permeation of Protein Through Membrane of Ultrafiltration Reactor (한외여과막 반응기에서 단백질의 막 투과특성)

  • Effects of operating time, pH, temperature, concentration and addition of proteolytic enzyme on permeate flux for the ultrafiltration of gelatin and bovine serum albumin(BSA) solutions were studied. The results showed that permeate flux of gelatin solution was maintained almost constant during the operating time, and that of BSA solution was decreased to 66% of the initial value after 40 min operation. The permeate flux of gelatin solution was increased by increasing temperature. The permeate flux of BSA solution was constant in the temperature range of 30~$50{\circ}C$, but increased at $60{\circ}C.$. The permeate fluxes of gelatin and BSA solution showed minimum values near the isoelectric point of pH 5.0. The permeate fluxes of 1%(w/v) and 6% gelatin solution were $43.0l/m^2\cdot hr$ and $13.5l/m^2\cdot hr$, respectively. Those of 1% and 4% BSA solution were $33.0l/m^2\cdot hr$ and $14.0l/m^2\cdot hr$, respectively. The permeate fluxes of gelatin and BSA solutions were decreased to 68.6% and 57.6% of their initial values by increasing their concentration, respectively. The permeate fluxes of gelatin and BSA solutions were enhanced by 30% with the addition of proteolytic enzyme.

  • 1) Respiratory metabolism patterns and its enzyme systems in the gill tissue of the fresh water mussels, Cristaria plicata were investigated through the examination on the effects of respiratory enzyme inhibitors, (KCN, NAF) and succinoxidase assay, while studying the effects of neutral salts (NaCL, KCL, CaCl2) and pH on oxygen consumption of the gill tissue. 2) In the limited concentration of KCL (0.3mM) and NaCl (0.4mM) solutions, oxygen consumption of the intact gill tissue was accelerated, but in CaCl2(0.5mM) solution, it showed no significant effect. The oxygen consumption was gradually decreased at the above concentrations of these limitations. The optimum pH for the respiration of the gill was 7.3. 3)Cyanide in 10-8M solution inhibited 88.8% of the respiration of the intact gill tissue. Methylene blue accelerated the respiration of the noral gill tissue, and slightly but significantly reversed the cyaniide poisoned respiration. 4)Oxygen consumption of the gill homogenate was apparently increased by the mixed addition of succinate, cytochrome c and activators (AlCl3 and CaCl2). This results suggested that succinoxidase system acts on the respiratory pattern of the gil tissue. 5) It was able to recognize that the enolase, which acts on the anaerobic glycolytic system, participated in the tissue respiration of the gill for NaF in 5$\times$10-2 M solution inhibited 55.5% of the respiration of the same intact tissue.

The Changes in Firmness, Ca Content and Polygalacturonase and Pectinesterase Activities during Oyijangachi Preparation (오이장아찌 제조중 경도, 칼슘 함량, Polygalacturonase 및 Pectinesterase 활성 변화에 관한 연구)

  • Oyijangachi, a traditional Korean brinded cucumber, was prepared by brinning the cucumbers in five different solutions for 48 hrs and then, was dipped into dipping sources(Kochujang, Doenjang and Ganjang) for 30 days of aging. Firmness, calcium content and enzyme activities(pectinesterase and polygalacturonase) changes were measured among the cucumbers which were treated by five different solutions during aging. The firmness of Kochujang Oyijangachi were the lowest after 10 days of aging for all from the five brining solutions because of "hollow phenomena" of cucumbers. Calcium contents of cucumbers after dipping into the five solutiosn increased as calcium content of the solutions increased and also increased when the cucumbers dipped into the dipping bases(Kochujang, Doenjang and Ganjang) because of calcium migration from the dipping sources into the cucumbers during aging. The calcium contents of the three dipping bases were ranged from 70mg% to 120mg% of Ca. The activity of polygalacturonase in the Oyijangachi decreased generally during aging and decreased rapidly during initial 5 days of aging. The activity of pectinesterase of cucumbers treated with 12% salts solutions(treatment 3, 4 and 5) were higher than those of cucumbers treated with 6% salts solutions(treatment 1 and 2).

  • The widely used polyethylene glycol (PEG)-based solutions have been proven effective for bowel preparation when 4 L of the solution is administered before colonoscopy. However, large volumes of the solutions are generally poorly tolerated. A new PEG-based solution consisting of 2 L of PEG and a high dose of ascorbic acid has recently become available. Electrolyte abnormalities caused by PEG-based solutions have rarely been reported. We report on a case of acute severe hyponatremia with associated generalized tonic-clonic seizures after bowel preparation with a low-volume PEG plus ascorbic acid solution in a 74-year-old woman with no history of seizures. She took a beta blocker, an angiotensin-converting enzyme inhibitor, and glimepiride for hypertension and diabetes mellitus. She showed general weakness, nausea, agitation, muscle cramping, and seizures after ingestion of the PEG plus ascorbic acid solution. Her serum sodium level was 112 mEq/L. Her symptoms improved after intravenous administration of hypertonic saline. Physicians should pay attention to screening for electrolytes and development of neurological symptoms during bowel preparation.

Enzymatic Bleaching of Kraft-pulp with Horseradish Peroxidase and Radical Mediator (Horseradish Peroxidase와 라디칼 전달체를 이용한 Kraft 펄프의 표백)

  • The use of 2,2-azinobis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS) as a radical mediator enhanced the bleaching efficiency of kraft pulp by horseradish peroxidase(HRP) and $H_2O_2$. High concentrations of up to 20 mM $H_2O_2$. were used. The bleaching of the kraft pulp increased as the amount of HRP and ABTS concentration were inceased up to 0.3 mg/90 mL and 2 mM, respectively. The bleaching of the kraft pulp was closely related with the HRPs activity and its adsorption onto the pulp. The activity of HRP and bleaching of kraft pulp were maximum at pH 7 and were reduced either in a acidic or alkaline solutions. The adsorption of HRP onto pulp was low in solutions of pH 6-8 and high in an acidic(pH5) and an alkaline solutions(pH 9). The adsorption of the enzyme was greater for alkali-lignin than for crystalline cellulose, the two major components of pulp.
