Performance Properties of Multi-Functional Finishes on the Enzyme-Pretreated Wool/Cotton Blend Fabrics

  • Research information related to application of enzyme as pretreatment and subsequent functional finishing on wool blended textiles for imparting multi-functional properties is still scanty. Yarn-blended wool/cotton fabric was pretreated with both a cellulase (Bactosol-CA) or a protease (Savinase-16.0LEx) in individual, subsequently finished with Synthappret-BAP and ${\beta}$-cyclodextrin based combination to impart anti-shrink, anti-microbial, softening and anti-crease properties. The performance of the finished fabrics depended on type of finishing combinations applied rather than enzyme pretreatment. Savinase pretreatment followed by Synthappret+Ceraperm-MW combination finishing impart both anti-shrink property as well as softening, while Bactosol pretreatment followed by ${\beta}$-cyclodextrin and sanitize combination finishing impart antimicrobial activity as well as anti-shrink finish to the wool/cotton blend fabric.

Effects of Treatments with Two Lipolytic Enzymes on Cotton/Polyester Blend Fabrics

  • This study examined the use of cutinase and lipase to process cotton/polyester blend fabric. Optimum treatment conditions for cutinase and lipase were investigated for cotton/polyester blend fabric. The properties of enzyme-treated fabrics were evaluated and compared in optimal treatment conditions. In addition, the possibility to provide an enzymatic finishing on blend fabrics using mixed enzymes in a two-step process were studied. The weight loss of cotton/polyester blend fabrics with Triton X-100 was 0.8% and the dyeing property of blend fabrics with calcium chloride increased by a factor of 1.2. The use of two enzymes in combination with cutinase and lipase in the presence of auxiliaries resulted in a cotton/polyester blend fabric weight loss of 0.8%. In addition, the dyeing properties of cotton/polyester blend fabrics improved by a factor of 1.5 and the moisture regain of cotton/polyester blend fabrics improved by a factor of 1.16. However, no marked loss was observed in tensile strength. The surface morphology of cotton/polyester blend fabrics is modified through a two-enzyme treatment. The treatment of cotton/polyester blend fabrics with cutinase and lipase maintains cotton strength and improves the moisture regain of polyester fabrics.

사람 난표액의 Caseinolytic Enzyme (A Caseinolytic Enzyme in Human Follicular Fluid)

  • 포유동물의 성숙한 난포의 난포액 속에는 여러 종류의 단백질 분해효소가 있으며 이들은 난포의 형성과 퇴화 및 난자의 성숙과 배란 등의 다양한 변화에 중요한 역할을 하는 것으로 여겨진다. 난포액 속의 단백질 가수분해효소 중에는 serine proteinase가 비교적 잘 알려져 있으나 다른 효소 특히, caseinolytic enzyme에 대해서는 거의 알려져 있지 않다. 본 연구에서는 사람의 난포액을 재료로 하여 caseinolytic enzyme의 존재 여부 및 동 효소의 특성을 알아보고자 하였다. 사람의 난포액과 혈청 그리고 사람의 제대혈을 $\alpha$-casein을 기질로 하는 zymography 방법으로 분석 한 결과 분자량 80 kDa의 매우 강한 caseinolytic activity를 지니는 효소 단백질과 분자량 78 kDa의 비교적 약한 caseinolytic activity를 갖는 단백질 등 두 개의 caseinolytic enzyme이 관찰되었다. 이 caseinase들의 특성을 알아보기 위해 phenylmethylsulfonyl fluoride(PMSF), soybean trypsin inhibitor(SBf), 1,10-phenanthroline, E-64 그리고 ethylenediamine tetraacetic acid(EDTA)를 zymouaphy의 substrate buffer에 처리한 결과 EDTA와 SBTI에 의해서 80 kDa와 78 kDa caseinase의 활성이 억제되었다 이로 미루어 80 kDa와 78 kDa caseinase는 trypsin-like enzyme인 것으로 추측된다. 한편 사람 난포액의 zymouaphy 수행시에 5 mM의 EDTA가 첨가된 substrate buffer에 CaC $l_2$, MgC $l_2$, MnC $l_2$ ZnC $l_2$를 각각 0에서 10 mM의 농도별로 처리한 결과 모두 5 mM 농도에서 가장 높은 caseinase 활성을 보였다. 금속 이온의 첨가없이 EDTA만 처리한 대조군의 경우 caseinase의 활성은 나타나지 않았다. 이로 미루어 80 kDa 및 78 kDa caseinase는 효소 활성을 위해 이가 금속 양이온을 필요로 하는 것으로 여겨진다.

복합효소를 이용한 고추 추출액의 이화학적 및 관능적 특성 (Physicochemical and Sensory Properties of Red Pepper Extract treated with Enzyme Complex)

  • 고추 추출액의 수율 및 생리적 기능성을 향상시키기 위하여 고추에 cellulase(C 처리군), pectinase(P 처리군), amylase(A 처리군)를 각각 또는 이들 효소들을 혼합(CP, CA, PA 및 CPA 처리군) 첨가하여 2~8시간동안 가수분해시킨 후 가열처리하고, 여과시킨 추출액에 대하여 이화학적 및 관능적 평가를 측정하였다. 효소 처리 고추 추출액의 수율은 효소 처리를 하지 않았을 때 38.84% 정도로 매우 낮게 나타났지만, 효소처리군이 높은 수율을 보였고, 효소 단독처리군보다는 병용처리 시 수율이 증가하였으며, 효소 처리 시간이 길어질수록 유의적으로 수율이 증가하는 것으로 나타났다. 특히 cellulase + pectinase + amylase(CPA) 복합처리군은 추출수율이 74.37%까지 증가하였다. 가용성 고형분의 함량변화는 대조군의 경우에는 8.51% 를 나타내었으나, 효소처리군은 대조군보다 높은 함량을 나타내었으며, CA 혼합처리군과 CPA 혼합처리군이 가장 높은 가용성 고형분 함량을 보였다. 환원당의 함량 또한 효소 처리에 의하여 증가하는 것으로 나타났다. 효소 처리에 의한 색도의 변화에서는 대조군과 효소처리군 간에 색도의 변화를 보이지는 않았다. 효소 처리한 고추 추출액에 대한 관능검사 결과에서는 전반적으로 효소 처리를 하지 않은 대조군에 비하여 효소 처리 시 기호도 면에서 우수한 것으로 나타나, 고추 추출물 제조를 위한 효소 처리는 수율 및 기호도 증진을 위한 좋은 방법으로 사료된다.

N-Acetylglycine Side Chain is Critical for the Antimicrobial Activity of Xanthostatin

  • This study was carried out to elucidate the mode of bacteriostatic property of xanthostatin (XS), a novel depsipeptide antibiotic with an N-acetylglycine side chain and selective antimicrobial activity against Xanthomonas spp. Two biotransformed XSs were isolated by the treatment of XS with the cell lysate of Xanthomonas campestris pv. citri, a solvent partition, preparative TLC, and HPLC. Structure determination of those two biotransformed XSs demonstrated deletion of the N-acetylglycine side chain. Noteworthily, they showed no antimicrobial activity against Xanthomonas spp. This result suggests that the N-acetylglycine side chain plays a critical role in the antimicrobial activity of XS, and that the bacteriostatic property of XS is due to susceptibility of the ester bond between the hexadepsipeptide nucleus and the N-acetylglycine side chain to hydrolytic enzyme(s) produced by Xanthomonas spp.

Aspergillus nidulans FGSC 159의 carboxymethylcellulases의 분리 순화 및 그 성질에 관한 연구 (Purification and Properties of Carboxymethylcellulases from Aspergillus nidulans FGSC 159)

  • Washed mycelia of Aspergillus nidulans FGSC159 were incubated in CMC minimal liquid medium and the culture filtrate which contained induced extracellular cellulase was fractionated by a three-step procedure including chromatography on Bio-Gel P-150, chromatography on DEAE-Sephadex A-50 and chromatography on Sephadex G-100. Three CMCase components ; F-I-Ia, F-I-Ib and F-II-Ia were prepared. No enzyme activity toward avicel could be detected in these components. Similarly, there was no ${\beta}-glucosidase$ activity. pH-optima of the three components were all 5.0 in acetate buffer. Temperature-optima for the activities of F-I-Ia, F-Ib and F-II-Ia were $45^{\circ}C,\;40^{\circ}C\;and\;50^{\circ}C$, respectively. F-II-Ia was shown to be more thermostable than the other two components. F-II-Ia was proved to have quite a different substrate specificity and action property and action property from those of F-I-Ia and F-I-Ib by product analysis on liquid chromatography.

Probiotic Property and Anti-Obesity Effect of Lactiplantibacillus plantarum KC3

  • Lactic acid bacteria are representative probiotics that have beneficial effects on humans. Nineteen strains among the 167 single strains from kimchi was selected and their physiological features were investigated. The selection of a strain was based on strong enzyme (lipase, α-amylase, and α-glucosidase) inhibitory activities and anti-obesity effects in the adipocytes. For the final selection, the strain Lactiplantibacillus plantarum KC3 was tested for its potential as a starter. To assess its functionality, a freeze-dried culture of L. plantarum KC3 was administered to a diet-induced obese mouse model receiving a high-fat diet. The animal group administered with L. plantarum KC3 showed significant body weight loss during the 12-week feeding period compared to the high-fat control group. This study investigated the physiological characteristics of selected strain and evaluated its potential as an anti-obesity probiotic in mice.

  • 토양으로부터 분리한 호알카리성 세균 Bacillus sp. No. 1911 파쇄하여 유안투석, DEAE-cellulose column chromatography, Sephadex G-150 gel filtration으로 포도당 이성화효소를 정제하여 효소적 특성을 조사하였다. 이 효소는 Sephadex G-200 gel filtration에서 분자량은 170,000으로 측정되었고 SDS-polyacrylamide 전기영동에서 43,000의 단일 band를 얻어, 이 효소는 4개의 동일한 subunit로 구성되었음을 나타내었다. 이 효소는 pH7.5 및 $65^{\circ}C$에서 최적활성을 나타내었으며 pH7.5에서 7$0^{\circ}C$ 까지 안정하였고 Co$^{++}$ 존재하에 6$0^{\circ}C$에서 30분간 가열하였을 때 pH 6-9까지는 비교적 안정되었다.

  • 한우(韓牛)의 round muscke에 papain을 농도별(濃度別)로 증가(增加)시켜서 longitudinal section과 cross section하여 결체조직(結締組織)의 collagenous fibre와 elastic fibre를 각각(各各) 염색(染色)하여 현미경으로 관찰한 결과(結果)는 다음과 같다. 1. 효소(酵素) 농도(濃度)의 증가(增加)에 따라 비정형 상태로 뭉쳐져 있는 collagenous fibre는 점점 풀어지는 상태(狀態)를 지나 후에는 붕괴되며 또한 염색성(染色性)이 현저하게 약해졌다. 2. elastic fibre의 방향(方向)은 muscle fibre에 평행(平行)하게 존재(存在)하며 효소(酵素) 농도(濃度)의 증가(增加)에 따라 점차 탄성(彈性)을 잃고 loose헤지며 직선적(直線的)으로 보이고 fragment되는 것도 현저하다. 3. collagenous fibre와 elastic fibre 모두 효소(酵素) 처리구(處理區)가 대조구(對照區)에 비하여 조직변화(組織變化)가 현저하다.

효소처리 초당옥수수 식혜의 이화학적 특성 (Physicochemical Characteristics of Enzyme-treated Super Sweet Corn Sikhye)

  • To produce super sweet corn sikhye, substituted for sweetener, the ratio of rice and super sweet corn was adjusted and processed with complex enzymes during saccharification, and their physicochemical and sensory properties were analyzed. The soluble solid content of the control and Corn-5 showed significantly high content at 13.50 °Brix, and the reducing sugar content of Corn-5 showed the highest content at 9.45%. The control showed the lowest free sugar content among all the experimental groups, excluding maltose content. In the enzyme-treated corn sikhye group, as the amount of super sweet corn increased, the content of sucrose decreased and the contents of glucose and fructose increased. The content of ascorbic acid and polyphenol compounds increased as the amount of super sweet corn increased. DPPH and ABTS radical scavenging abilities increased with increasing ratio of super sweet corn and enzyme treatment compared to the control. In the case of sensory evaluation, Corn-3, which substituted 30% of super sweet corn for rice and treated with enzymes, showed higher evaluations in taste, sweetness, and overall preference than those of the control.
