  • Crude enzymes of Trichoderma viride and Phanerochaete chrysosporium were evaluated for their effect to brightness and physical properties of recycled pulp from newspaper and corrugated container board. The brightness of recycled pulp from newspaper increased with crude enzymes from Trichoderma viride and Phanerochaete chrysosporium. The brightness of recycled pulp of corrugated container board increased with crude enzyme from Phanerochaete chrysosporium, but decreased with crude enzyme from Trichoderma viride. The Δbrightness with chemical bleaching of crude enzyme treated pulp was lower than that of heat killed enzyme treated pulp, but the final brightness of pulp was increased. Modification of recycled paper with crude enzymes can result in a substantial increase in physical properties with little no loss in pulp freeness.

  • The properties of purified intracellular adenosine deaminase (adenosine aminohydrolase, EC 3.5.4.4) of Nocardioides sp. J-326TK isolated from soil have been studied. The enzyme deaminated adenosine and 2`-deoxyadenosine and the respective {TEX}$K_{M}${/TEX} values were 4.0×{TEX}$10^{-4}${/TEX} M and 5.0× {TEX}$10^{-4}${/TEX} M, but the enzyme was not active on 8-bromoadenosine, 6-methylaminopurine riboside, ATP, ADP, 2`-AMP, 3`-AMP, 5`-AMP, dAMP, cAMP, NAD, FAD, NADP and adenine. The enzyme activity was strongly inhibited by the addition of {TEX}$Hg^{2+}${/TEX} and {TEX}$Ag^{+}${/TEX}, {TEX}$Cu^{2+}${/TEX}, {TEX}$Co^{2+}${/TEX} and {TEX}$Mn^{2+}${/TEX} also inhibited the activity but much less extent. The effect of alkyl reagents, metal chelating reagents and certain other compounds on the enzyme activity were also examined. No reagent activated the enzyme. On the contrary, the enzyme reaction was slightly inhibited by o-phenanthroline and 6-benzyladenosine.

Combined Effects of Pressure Cooking and Enzyme Treatment to Enhance the Digestibility and Physicochemical Properties of Spreadable Liver Sausage

  • This study aimed to determine the effect of enzyme, guar gum, and pressure processing on the digestibility and physicochemical properties of age-friendly liver sausages. Liver sausages were manufactured by adding proteolytic enzyme (Bromelain) and guar gum, and pressure-cooking (0.06 MPa), with the following treatments: control, without proteolytic enzyme; T1, proteolytic enzyme; T2, proteolytic enzyme and guar gum; T3, pressure-cooking; T4, proteolytic enzyme and pressure-cooking; T5, proteolytic enzyme, guar gum, and pressure-cooking. The pH was high in the enzyme- and pressure-processed groups. The pressure-processed groups had lower apparent viscosity than other cooking groups, and it decreased during enzyme treatment. Hardness was lower in the enzyme- and pressure-processed groups than in the control, and the T4 was the lowest. Digestibility was the highest in T4 at 82.58%, and there was no significant difference with that in T5. The general cooking group with enzyme and guar gum also showed higher digestibility than the control (77.50%). As a result of the sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the enzyme- and pressure-treated groups (T4, T5) were degraded more into low-molecular-weight peptides (≤37 kDa) than the control and other treatments. Viscoelasticity showed similar trends for viscous and elastic moduli. Similarly, combined pressure processing and enzymatic treatment decreased viscoelasticity, while guar gum increased elasticity but decreased viscosity. Therefore, the tenderized physical properties and improved digestibility by enzyme and pressurization treatment could be used to produce age-friendly spreadable liver sausages.

The Study of the Properties Coated Paper by the Enzyme Treatment (효소처리한 도공지의 물성 관한 연구)

  • We studied on the effects of the enzyme treatment of pulp fiber on physical properties of coated paper were elucidated using four types of enzymes. The results of this study showed that the blank in which enzyme was not treated showed the highest $St{\ddot{o}}ckigt$ and Hurcules sizing degrees. And when the enzyme treated base paper was used for coating, the coated paper has average 5.7 % higher gloss and whiteness than when untreated base paper was used. Brightness showed 0.12 % higher results with the enzyme treated base papers than the blank while it is considered to show similar trends. Whiteness of coated paper was not influenced by the enzyme treatments but by the properties of coating color. Coated paper whose base paper was enzyme treated showed improved roughness while smoothness of those samples except the ${\beta}$-Glucosidase treated one was relatively low due to the high fines content. Also the coated paper whose base paper was not enzyme treated showed high porosity.

  • Streptomyces sp. YB-88-20 was Isolated from soil and the properties of chitobiase were investigated. The optimal reaction condition for the enzyme was pH 5.5 and 4$0^{\circ}C$ , and was stable in the range of pH 4. 0 to 5.5 and temperature at 4$0^{\circ}C$, and 40 min, respectively The enzyme was inactivated by heating at 45$^{\circ}C$ for 1 hr. The enzyme was slightly activated by Mna+. Mg2+, but inhibited by Fea+. Km and activation energy was 1.5072 M and 8.314 kcal/mol.

Quality Properties and Carotenoid Pigments of Yellow Sweet Potato Puree (황색고구마 퓨레의 품질과 Carotenoid색소)

  • Two kinds of sweet potato puree were prepared with Benihayato cultivar of yellow sweet potato with or without $\alpha$-amylase enzyme treatment. Chemical and rheological properties of enzyme-treated puree were different from those of control puree. Reducing sugar content and iodine value increased by $\alpha$-amylase enzyme treatment, while alcohol insoluble solids and viscosity decreased by enzyme treatment. However, the changes of carotenoid content were not significantly different. Hunter-b-values(yellowness) were 27.19 and 23.54 for no enzyme-treated puree(NTP) and enzyme-treated puree(ETP), respectively. Hunter-a values(redness) were 2.24 and 6.05 for NTP abd ETP, respectively. Content of total carotenoid of canned puree heated at 13$0^{\circ}C$ for 30 min decreased by 59 percents.

  • The mechanical properties of deinked mixed office waste paper in relation to sorts and dosage of enzymes were investigated for successful enzymatic deinking of mixed office waste paper. The increasing of $\Delta$. freeness was most predominant in pulp of Denimax treated pulp. The tensile and burst properties are decreased with enzyme dosage while $\Delta$. freeness were increased. The fine content in disintegrated pulp was decreased with enzyme dosage of $0.4\%$ on the dried weight of paper. The water absorption of handsheets of enzyme deinked pulp was like as that of market roll tissue papers and the mechanical properties of handsheets of enzyme deinked pulp were improved with enzyme treatment comparing of market roll tissue papers.

  • An enzyme, named GTP cyclohydorlase, that catalizes the hydrolytic removal of carbon No.S of GTP has been partially purified from extracts of Pseudomonas putida (IAM 1506). The enzyme exists in two molecuar weight forms : a high molecular weight form (150,000) and a low molecular weight from (40,000). The high molecular weight form has been purified 25-fold. Some of the properties of the enzyme are as follows : It functions optimally at pH8.0, and at $52^{\circ}C$. The Km value for GTP is $20{\mu}M$. Divalent cations $(Cd^{2+}\;and\;Hg^{2+})$ 2+/) at a concentration of 5mM inhibit completely the enzyme activity. No metal ion including $Mg^{2+}$ is needed for the catalysis. The enzyme is heat labile ; its half at $57^{\circ}C$ is 1.5 min. Of a number of nucleotides tested, only GDP was used to any extent as substrbte in place of GTP. One of the products of the enzyme is determined to be a dihydro-neopterin compound.

  • It is very difficult to compare directly the research results of enzymatic process in pulp and paper industry because commercial enzymes have diversity in its property. The chemical and biological properties of commercial enzymes were Investigated to help comparison of various commercial enzymes each other. In most case, the solid content of liquid enzymes was about 20%. The higher protein content in enzyme product does not mean the higher enzyme activity. Enzymes for paper process should selected by basis of enzyme activity, not by price of enzyme products. The chemical composition of fiber was not so much change with enzyme treatment. The enzymatic hydrolysis of fiber might negligible in paper process.

  • Glucose oxidase was immobilized in polypyrrole by electrosynthesis. The enzyme had an influence on the redox properties of a complex enzyme electrode. In the cyclic voltammograms of the enazyme electrode new peaks were appeared at the potential around 0.7V vs. Ag/AgCl in additional to the typical peaks for polypyrrole. The more immobilized the stronger the peaks became. During the cycling the pH of electrolyte solution was decreased to about 4.4 The reason for that is to be the proton released from the carboxyl in the glucose oxidase in order to keep on a charge neutrality of the oxidized enzyme. This fact suggests that the new peaks in the voltammograms are caused by the redox of glucose oxidase. In the AC impedance spectrum analysis of the electrode the diffusion of electrolyte anion was limited because of chained structure of the enzyme. The faradic impedance was large since the glucose oxidase is an insulator. Therefore when glucose oxidase is entrapped the enzyme should be limited in amount. Because the growth of the polypyrrole is accompanied both charge transfer and mass transport. For the traditional electrosynthesis that means amount of enzyme present in the electrode is limited to as much as film growable.
