Effects of grape pomace on the antioxidant defense system in diet-induced hypercholesterolemic rabbits

  • The effects of grape seeds extract and grape peels extract prepared from grape pomace on the activity of antioxidant enzymes, degree of lipid peroxidation in serum and liver tissue were investigated in rabbits fed on high cholesterol diet. New Zealand white rabbits were divided as follows ; 1) NOR (normal group); 2) CHOL (cholesterol group); 3) GSH (cholesterol + grape seed extract group); 4) GPE (cholesterol + grape peel extract); 5) GSP (cholesterol + grape seed powder); 6) GPP (cholesterol + grape peel powder); 7) GE (cholesterol + grape seed and peel extract); 8) GP (cholesterol + grape seed and peel powder). Eight groups of rabbits were studied for 8 weeks. At the end of the experimental period, rabbits were sacrificed and the liver tissue were removed. Then, GSH, GPx, GST, CAT and MDA in the liver were measured. In liver tissues, total glutathione contents (GSH), glutathione peroxidase (GPx) and catalase (CAT) activity, which was significantly higher by grape seed extract supplementation. The level of malondialdehyde (MDA) was lower in the serum of rabbits fed grape seed extract or grape peel powder plus cholesterol than in the serum of rabbits fed cholesterol alone. It is therefore likely that grape seed extract prepared from grape pomace functioned as antioxidants in vivo, negating the effects of the oxidative stress induced by 1% cholesterol diet. The grape seed extract was found effective in converting the oxidized glutathione into reduced glutathione, and in removing $H_2O_2$ that is created by oxidative stress. The grape peel powder was found to have small influence on reduced glutathione content, CAT and GPX activity, but it increased GST activity in liver tissues, resulting in promoting the combination of lipid peroxide and glutathione (GSH), and further, lowering the formation of lipid peroxide in the serum. Therefore, grape pomace (grape seed extract and grape peel powder) supplementation is considered to activate the antioxidant enzyme system and prevent damage with hypercholesterolemia.

Anticancer effect of joboksansam, Korean wild ginseng germinated from bird feces

  • Background: Joboksansam, Korean bird wild ginseng, is an artificially cultivated wild ginseng germinated from bird feces. Although numerous pharmacologic activities of wild ginsengs have been reported, the beneficial effect of joboksansam in cancer has not been elucidated. In this study, we investigated the in vivo and in vitro anticancer activities of joboksansam powder. Methods: To evaluate the in vivo anticancer activity of joboksansam, we established a xenograft mouse model bearing RMA cell-derived cancer. Direct cytotoxicity induced by joboksansam powder was also investigated in vitro using (3-4-5-dimethylthiazol-2-yl)-2-5-diphenyltetrazolium bromide (MTT) assay. The inhibitory activity of this powder on the activation of cell survival signaling involving Akt and Src was examined with immunoblot analysis. Results: Joboksansam powder displayed strong inhibitory activity against the increased tumor size, increased weight of total body and cancer tissues, and mortality of tumor-bearing mice. Joboksansam powder also suppressed the activation of survival regulatory enzymes Akt and Src, as assessed by phosphorylation levels in the immunoblot analysis of tumor tissues. Interestingly, the viability of RMA cells in vitro was directly decreased by joboksansam treatment. Conclusion: Overall, our results strongly suggest that joboksansam powder has the potential to protect against cancer generation by direct cytotoxic effects on cancer cells resulting from suppression of cell survival signaling.

시멘트 분말 노출에 따른 참갈겨니의 스트레스 저항성 (Stress Resistance of Zacco koreanus due to Exposure of Cement Powder)

  • 본 연구는 시멘트 노출에 따른 참갈겨니 조직의 형태 생리적 변화를 분석하여 용해된 시멘트 분말이 어류에 미치는 영향을 조사하고자 하였다. 용해된 시멘트 분말에 노출된 아가미는 노출기간이 길어질수록 이차새변의 간격이 불규칙해지고, 점액세포의 활성도 증가하고 있으며, 곤봉화, 부종, 상피세포의 박리가 이차새변에서 관찰되었다. 신장 조직에서는 보우만주머니 공간이 넓게 관찰되었고, 표피 조직은 표피층의 두께가 감소하고 진피층의 배열이 불규칙해지는 것으로 관찰되었다. 항산화효소와 LDH의 활성은 조직 및 노출기간에 따라 활성에 차이가 있는 것으로 나타났다. 표피조직에서 용해된 시멘트 분말에 의해 발현이 증가되는 단백질은 근단백질 생성과 관련된 스트레스 단백질로 확인되었고 발현이 저하된 단백질들은 해당과정과 에너지 대사에 관여하는 단백질로 조사되었다. 이러한 결과로 보아 시멘트 노출에 따른 스트레스는 참갈겨니 조직의 형태적 변형과 생리적 기능의 약화를 초래하여 어류의 생존에 커다란 위협이 될 요인으로 작용할 수 있을 것으로 사료된다.

농축 및 건조방법에 따른 생강 추출액 분말의 품질변화 (Quality of Ginger Powder as Affected by Concentration and Dehydration Methods of Ginger Extracts)

  • 생강 추출액의 저장안정성을 제공하기 위하여 적정농축 및 건조방법을 선정코자 감압증류 및 RO(역삼투압)에 의한 농축과 냉동 및 분무건조방법별로 분말화한 다음 관능특성을 비롯한 이화학적인 품질특성을 비교하여 보았다. 농축 및 건조방법별 분말제품을 제조한 결과 갈변도는 감압증류법이 RO보다. 분무건조가 냉동건조보다 갈변이 덜 진행된 상태인 반면에 당 함량이나 유리아미노산은 농축방법의 영향을 인식할 수 없었으나 건조방법에 있어서는 냉동건조시 대체로 이들 성분의 손실율을 적게할 수 있었다. 효소적 액화추출액 분말은 생강 추출액 분말보다 단백질과 조회분함량이 낮은 반면 조지방 함량은 약간 높게 나타났으며, 농축 및 건조 중 갈변도와 유리 아미노산의 변화율을 생강 추출액 분말보다 억제하면서 용해도를 증가시킬 수 있는 효과가 있었다. 또한 생강분말에 대한 기호도 검사결과, 농축방법에서는 생강 추출액 및 효소적 액화추출액에서 RO농축한 제품이 우수한 결과를 보였으며 건조방법에서는 냉동건조가 우수한 결과를 나타내었다. 특히 효소적 액화 추출액의 경우에는 RO농축한 후 냉동건조하면 종합 기호도에 있어서 생강추출액 분말에 대한 기호도 특성과 거의 차이가 없는 것으로 나타났다.

뽕잎과 누에가루 혼합환의 Streptozotocin유발 당뇨쥐에서의 혈당강하 효과 (Hypoglycemic Effects of Pills Made of Mulberry Leaves and Silkworm Powder in Streptozotocin-Induced Diabetic Rats)

  • 본 연구에서 는 뽕잎과 누에가루의 함량 비를 달리 하여 제조한 환의 혈당강하 효과를 in vitro및 동물실험을 통해 검정 하고자 하였다. 실험 동물은 체중 100 g내외 의 Sprague-Dawley종 수컷 흰쥐를 이용하였으며, 제조환은 식이 내에 0.4%(4 ghg)씩 공급하였다. 실험군은 제조환의 종류에 따라 제조환을 공급하지 않은 당뇨 대조군(DM group), 뽕잎 100%로 제조한 군(M group), 뽕잎에 누에가루를 각각 25%(25SM group), 50%(50SM group), 75%(75SM group) 및 100%(100S group)씩 혼합하여 제조한 군으로 나누었다. 식이와 식수는 자유섭식시켰으며,3주 간 사육한 후 STZ로 당뇨를 유발시켰으며, 9일 만에 희생시켰다 제조된 혼합 환의 DNJ의 함량은 누에가루의 혼합 비율이 높을수록 증가되었다. GABA및 rutin의 함량은 뽕잎의 혼합 비율이 증가함에 따라 함유량이 증가되었다. in uitro에서 소장 점막의 $\alpha$-glucosidase의 활성 저해 효과도 뽕잎 에 누에가루를 50% 이상 혼합시켰을 때 혼합하지 않은 환과 2SSM 환보다 유의적으로 증가되었다. 혈당강하 효과는 DM군과 M군에 비해 누에가루를 혼합한 군에서 더 높았으며 특히 50%, 75% 및 100% 혼합한 군들에서는 유의적 인 혈당강하 효과를 나타내었다. 소장 점막 maltase 활성은 proximal 부분에서 DM군에 비하여 제조환 공급군 모두가 유의적으로 감소되었으며, 제조환 공급군 간에는 유의적인 차이는 없었다. Middle부분에서는 실험군 간에 유의적인 차이가 없었다. Distal 부분에서는 DM군에 비하여 제조환 공급군에서 감소되었으며, 특히 50SM군,75SM군 및 100S군에서 현저하게 감소하였다. Sucrase와 lactase의 경우에는 prorimal부분에서 DM군에 비하여 제조환 공급군에서 유의적으로 감소되었으며, middle 및 distal부분에서는 실험군 간에 유의적인 차이가 없었다. 결론적으로 뽕잎과 누에가루를 혼합하여 제조한 환은 in vitro실험 에서 $\alpha$-glucosidase 활성 저해효과를 증가시켰으며, STZ 유발 당뇨 쥐의 소장 proximal 부분의 이 당류 분해 효소 활성을 억제함으로써 급속한 혈당상승을 억제하는 효과가 있었다. 이러한 효과는 뽕잎과 누에가루를 50 : 50의 동량으로 혼합시켰을 때 가장 좋은 것으로 관찰되었다.

각종 미강 추출물들의 항고혈압성 엔지오텐신 전환효소 저해 활성 (Antihypertensive Angiotensin I-Converting Enzyme Inhibitiory Activity of Various Extracts from Some Rice Brans)

  • 각종 미강 추출물들의 항고혈압성 엔지오텐신 전환효소 저해 활성을 측정하고 이들의 추출 최적조건을 검토하였다. 각종 미강 추출물들중에서 일품벼 미강의 물 추출물이 77%의 가장 높은 안지오텐신 전환효소 저해활성 보였다. 일품벼 미강중의 안지오텐신 전환효소 저해물질은 물을 1:10(w/v)으로 미강 분말에 첨가한 후 $30^{\circ}C$에서 12시간 추출하였을 때 가장 많이 용출되었다.

Studies on the Ligninolytic Enzyme Activities During Biological Bleaching of Kraft Pulp with Newly Isolated Lignin-Degrading Fungi

  • A screening has been performed to find hyper-ligninolytic fungi, which degtrade beech and pine lignin extensively in order to broaden the understanding of the ligninolytic enzymes elaborated by various white-rot fungi. One hundred and twenty two ligninolytic strains were selected from decayed woods with a selective medium for screening ligninolytic wood-rotting fungi. Two of them, Phanerochaete sordida YK-624 and YK-472, showed much higher ligninolytic activity and selectivity in beech-wood degradation than typical lignin-degrading fungi, phanerochaete chrysosporium and Coriolus versicolor. They also degraded birch dioxane lignin and residual lignin in unbleached kraft pulp(UKP) much more extensively than P. chrysosporium and C. versicolor. During fungal treatment of beech wood-powder, the fungus strain P. sordida YK-624 showed higher activity of extracellular manganese peroxidase (MnP) in the medium than P. chrysosporium. It also showed MnP activity, which would not be lignin peroxidast during treatment of oxygen-bleached kraft pulp(OKP) and under enzyme-inducing conditin.

Electrochemical Investigation of Animal Tissue Embedded Biosensor Bound with Ethylene-Propylene Rubber

  • When rubber dissolved in toluene was used as a binding material of graphite powder, the mechanical robustness of the carbon paste was guaranteed by the fast volatility of the solvent immediately after electrode construction. This characteristic of the rubber solution met qualifications for practical use of carbon paste electrodes and enabled the design of a new enzyme electrode bound with EPDM. In order to confirm whether the electrode shows quantitative electrochemical behaviors or not, its kinetic parameters, e. g. the symmetry factor (0.2), the exchange current density ($3.66\;{\mu}A/cm^2$), the capacity of the double layer ($2.0{\times}10^{-5}\;F$), the Michaelis constant ($4.39{\times}10^{-3}\;M$), the diffusion coefficient of substrate ($2.58{\times}10^{-12}\;cm^2/sec$), the time constant (0.018 sec) and other factors were investigated.

Enzyme-linked Immunosorbent Assay for the Detection of Hen's Egg Proteins in Processed Foods

  • The Hen's egg is widely used in many processed foods as an ingredient and is one of the most prevalent food allergens in children. To detect egg proteins in processed foods, we developed a competitive indirect enzyme-linked immunosorbent assay (ciELISA) using an anti-ovomucoid (OM) antibody, which was produced by immunization of rabbits with OM, the most heat-stable component of the egg proteins. The detection limit of this quantitative assay system was 30 ng/mL. Cross-reactivity of the anti-OM antibody toward OM, ovalbumin, skim milk, casein, whey protein isolate, and isolated soy protein was 100, 0.4, 0.2, 0.04, 0, and 0%, respectively. In the spike test of egg white powder in milk replacer, commercial sausage, and in-house sausage, the assay recoveries ($mean{\pm}SD$) were $129{\pm}13.7%$, $73.9{\pm}12.5%$, and $65.5{\pm}13.6%$, respectively. When egg white in a commercial crab meat analog and sausage was determined by ciELISA, the assay recovery was found to be 108% and 127%, respectively. The combined results of this study indicate that this novel ciELISA for OM detection could be applied for the quantification of hen's egg proteins in processed foods.

Effect of Diets Supplemented with Pharbitis Seed Powder on Serum and Hepatic Lipid Levels, and Enzyme Activities of Rats Administered with Ethanol Chronically

  • The levels of $\gamma$-aminobutyric acid (GAGA) have been analyzed from pharbitis seeds by an AccQ-Tag amino acid analysis procedure. The GABA level of the pharbitis seeds was 125 nmole per gram fresh weight. To investigate the effects of pharbitis seed diets on serum and hepatic lipid levels, as well as enzyme activities of rats administered with ethanol chronically, Sprague-Dawley male rats were fed with either a AIN-76 diet (control), a control diet plus ethanol, a control plus pharbitis seed diet, or a control plus pharbitis seed diet plus ethanol for 30 days. Pharbitis seed diets decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and $\gamma$-GTP levels that were increased by the chronic ethanol administration. In addition, pharbitis seed diets decreased the liver triglyceride and total lipid levels that were increased by the ethanol administration. However, ethanol metabolism was not retarded by the pharbitis seed supplemented diets. The present Endings, plus previous data showing the differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the enzyme activities of rats (Cha and Oh [2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505), raise the possibility that GABA in plants could have a nutraceutical role in the recovery of chronic alcohol-related diseases.
