인삼 Saponin 분획의 고혈당강하작용에 관한 연구(II) (Study on the Hypoglycemic Action of Ginseng Saponin on Streptozotocin Induced Diabetic Rats (II))

  • The decreased activities of liver enzymes relating to carbohydrate metabolism such as glucose- 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of streptozotocin injected rats were significantly modified by the intraperitoneal injection of ginseng saponin mixture and/or purified ginsenosides. However, several enzymes such as pyruvate kinase, malic enzyme and glycogen phosphorylase were not modified appreciably by the saponin administration, suggesting that the effect of ginseng saponin might be depend upon individual enzymes. Examination of liver enzymes by liver professing technique using perfusion buffer containing saponin (10-3%) showed that the ginseng saponin might stimulate insulin biosynthesis as well as the related enzyme activities.

우라늄 투여후 간조직에서의 효소활성도의 변화 (Enzyme activity changes by intraperitoneal injection of uranium in the carp liver)

  • 우라늄 피폭후 효소활성도가 내부피폭의 biomarker로서의 가능성을 연구하기 위하여 수서생물인 잉어(Caprinus carpio)의 복강내 우라늄 투여시 간조직에서 일어나는 여러가지 효소계의 변화를 조사하였다. 이러한 효소활성도의 변화는 우라늄 내부피폭의 biomarker로서 가능성을 가지고 있으며 연구결과는 다음과 같다. 1) 6일째까지 우라늄을 주사하여 적출한 간에서의 전체단백질량은 계속 감소하고 있다. 2) 세포내의 lysosome내에 함유하고 있는 acid pretense와 ${\beta}-glucuronidase$의 활성도는 6일째 투여후까지 활성도가 감소하고 있다. 3) Alkaline phosphatase의 경우 6일째 우라늄 투석후까지 증가하고 있으며 반대로 acid phosphatase의 경우 6일째 우라늄 투여후까지 활성도가 급격히 증가하였고 glutamate oxaloacetate transaminase의 활성도는 완만하게 증가하고 있다. 4) Creatine kinase의 활성도는 완만한 감소를 보이고 있으며 malate dehydrogenase는 첫번 우라늄 투여후에 활성도가 급격히 감소하였고 3일째 우라늄 투여후에는 활성도가 거의 나타나지 않았다.

Fatty acid compositions, free radical scavenging activities, and antioxidative enzyme activities of high-preference and low-preference beef cuts of Hanwoo (Bos taurus coreanae) cows

  • Objective: This study compared fatty acid compositions and antioxidant activities of high-preference cuts (loin, tenderloin, and rib) and low-preference cuts (brisket, topside, and shank) of Hanwoo (Bos taurus coreanae) cows to obtain potentially useful information for promoting the consumption of various low-preference cuts. Methods: Individual 500 g samples of fresh beef were collected from each of the six cuts from 10 Hanwoo cows (quality grade 1) and immediately freeze-dried. The dried samples were evaluated for fatty acid composition, free radical scavenging activities (hydroxyl, alkyl, and 2, 2'-diphenyl-1-picrylhydrazyl [DPPH] radical), and antioxidative enzyme activities (glutathione peroxidase [GPx], glutathione-S-transferase [GST], and superoxide dismutase [SOD]). Results: The percentages of total polyunsaturated fatty acids were significantly higher in low-preference cuts than in high-preference cuts (p<0.05). Hydroxyl, alkyl, and DPPH radical scavenging activities were significantly higher in low-preference cuts than in high-preference cuts (p<0.05). In addition, the activities of antioxidant enzymes, such as GPx, GST, and SOD, were significantly higher in low-preference cuts compared with high-preference cuts (p<0.05). Conclusion: These results may influence consumers to include more low-preference cuts in their selections based on the nutritional facts, which could help to balance the beef market in South Korea.

  • Effect of furnish on enzyme activity was investigated by using the three components (cellulose, enzyme, and cationic polyelectrolyte) model papermaking system. Avicel was used as a cellulose model compound to observe the effect of adsorption and desorption of enzyme with other component and the resultant change of particle size. As an experimental result, the enzyme loses considerably its apparent activity due to the adsorption onto cellulose and cationic polyelectrolyte. Activities of enzyme applied to the actual papermaking stocks having controlled fiber length showed different behavior in terms of pulp species UKP and KOCC stocks. That is, the enzyme activity in UKP was increased as fines content increased, however, vice versa in KOCC stock . This result can be considered to be the existence of various contaminants included in the fines of KOCC . The effect of possible contaminants such as inorganic materials, calcium ion, surfactant, and conductivity on enzyme activity were also investigated.

  • As a part of pharmacological studies of saikosaponins, which were reported to exhibit diverse biological activities especially concerning with liver function, effects of saikosaponin on metabolizing enzymes and lipid peroxide contents in liver were examined. As the result, UDP-glucose dehydrogenase activity and lipid peroxidation which were due to acetaminophen were inhibited by saikosaponin treatment. But other metabolizing enzyme activities were not modified.

  • An extremely cadmium tolerant yeast, Hansenula anomala B-7 used to determine the modification of the intracellular enzyme activities by cadmium ion. The activities of alcohol dehydrogenase, phosphofructokinase, and cytidine deaminase were decreased up to 90%, 40%, and 86% compa- red with the control by 1 mM cadmium nitrate respectively, but the activities of malate dehydrogenase, 6- phosphogluconate dehydrogenase, cytochrome c oxidase, and alkaline phosphatase were increased up to 440%, 136%, 260% and 155% compared with the control by 1 mM cadmium nitrate respectively. These results show that the activities of the enzymes participating in Embden-Mayerhof pathway (e.g. anaerobic metabolism) were reduced by cadmium, but those involved in hexose monophosphate pathway and tricarboxylic acid cycle (e.g. aerobic metabolism) were stimulated in contrast. It has been suggested that the diminished activity of cytidine deaminase in pyrimidine nucleotide dissimilation occured due to the inhibited nucleotide dissimilation by cadmium ion; the enhanced activity of cytochrome c oxidase was specifically required in order to oxidize a raised amount of NADH and NADPH due to the increased aerobic metabolism.

Neurotoxicant (fenitrothion) alters superoxide dismutase, catalase, and peroxidase activities in Chironomus riparius Mg. (Diptera, Chironomidae) larvae

  • Effects of exposure to a neurotoxicant, fenitrothion on antioxidant enzyme activities in Chironomus riparius Mg. (Diptera, Chironomidae) larvae were evaluated under laboratory conditions. Exposure to this chemical led to an increase of cupper, zinc type superoxide dismutase and manganese type superoxide dismutase activities and to a decrease of glutathion peroxidase activity. An activation of catalase was observed in the larvae exposed to high fenitrothion concentration. The response of superoxide dismutase was rapid and sensitive to low chemical concentrations, but changes in catalase, total peroxidase and glutathion peroxidase were less sensitive. In this study, antioxidant enzyme activities in Chironomus riparius larvae were identified as pertinent biomarkers for environmental monitoring.

동백 발효 추출물 단기 투여의 활성에 대한 연구 (Biological Activities of Extracts of Fermented Camellia japonica Leaf and Flower)

  • Objectives : This study was carried out to investigate the biological activities of methanol extracts of fermented Camellia japonica leaf and flower. Methods : Methanol extracts of fermented Camellia japonica leaf and flower were prepared and a dose of 100 and 400mg/kg/day was administered orally into mice. And after appropriate weeks, changes of serum enzyme activities were investigated to confirm its effects on serum glucose, cholesterol and short term administration safety. Results : Fermented flower extract showed significant decrease of serum level of cholesterol. And showed no toxicity on kidney and liver within the dose of 400mg/kg/day. Conclusion : Thus above result showed no toxicity on kidney and liver in male and female mice.

Phenanthrene-induced changes in blood organic components, transaminase and ALP activities of Paralichthys olivaceus

  • Phenanthrene-induced changes in blood organic components. transaminase and ALP activities of Paralichthys oIivaceus were investigaled. The aim of this study was to obtain a holistic view of the toxic responses, and compensations of fish exposed to waterborne phenanthrene. Fish were exposed to 0, 0.5, 1, 2$\mu$M of phenanthrene for 4 weeks. The most noticeable changes were concentration-dependent increase in levels of blood GOT and GPT activities. At concentrations 1.0 and 2.0 $\mu$M of phenanthrene GOT, GPT and ALP enzyme activities in plasma were increased. Plasma albumin concentration was stable in phenanthrene-exposed fish, however, plasma total protein level was reduced significantly at the 4-week sampling points (2.0$\mu$M). Plasma glucose levels were found to be increased significantly over the cootrol throughout duration of the experiment with phenanthrene. Overall the results suggest that phenanthrene has the potential to alter some physiological functions in olive flounder.

Isolation and Characterization of Soil Streptomyces Involved in 2,4-Dichlorophenol Oxidation

  • Over 50 morphologically distinctive soil Streptomyces were isolated from various Jocations in the Yongin area in Korea and visually screened for dye-decoloring activities on an agar plate. Two Streptomyces species (AD001 and ND002) showed strong dye-decoloring activities on the plate containing congo-red and new-fuchin dyes, respectively. Also, the liquid culture supernatants of these species showed 2,4-dicholophenol (DCP) oxidation activities only in the presence of hydrogen peroxide, a characteristic of Actinomycetes lignin-peroxidase (ALiP)-P3 isoform found in dye-degrading S. viridosporus T7A and S. badius 252. Based on their dye-decoloring capabilities and the 2,4-DCP oxidation kinetic data, it is suggested that these Streptomyces secrete not-yet-characterized extracelluar enzyme(s), whose activities are very similar to the ALiP-P3 enzyme.
