Objectives: Bisphenol A, or BPA, is a chemical component in polycarbonate plastic with which many people come into contact every day. A great deal of controversy has arisen over its safety since this material, which is known to disrupt the human endocrine system and cause neurological difficulties and cancer, is commonplace in beverage containers, food can liners, and receipt paper rolls. In this study, we determined the levels of exposure to BPA of workers in the service industry depending on the number of receipts contacted. Methods: The participants were 16 male and 18 female workers employed in the service industry. Using a questionnaire, we investigated general and job characteristics. Urine samples were collected and analyzed by the LC-MS/MS technique after enzymatic hydrolysis and solid phase extraction (SPE). Results: The geometric mean (GM) concentration of urinary BPA from all subjects was 1.02 ng/ml. Workers were exposed significantly to more BPA according to the number of receipts they contacted, their work experience, and working hours per day. The BPA concentration of those who touched more than 100 receipts per day was 3.09 ng/ml, while that of the other participants was 0.61 ng/ml. It was shown that wearing gloves can protect from BPA exposure. Conclusion: We determined the urinary BPA concentrations of workers in service industry and found that the contact with receipts could increase the BPA exposure of service workers.
The misuse of anabolic androgenic steroids is of particular concern in sports and society. Thus, it is of great importance to discriminate endogenous steroids such as testosterone or testosterone prohormones from their chemically identical synthetic copies. In this study, gas chromatography-combustion/isotope ratio mass spectrometric (GC-C/IRMS) method has been developed and validated for discriminating the origin of anabolic androgenic steroids. The method involves the solid-phase extraction, enzymatic hydrolysis with ${\beta}$-glucuronidase, HPLC-fractionation for the cleanup and analysis by GC-C/IRMS. The difference(${\Delta}^{13}C$) of urinary ${\delta}^{13}C$ values between synthetic analogues and endogenous reference compounds (ERC) by GC-C/IRMS was used to elucidate the origin of steroids, and intra- and inter-day precision, specificity and isotope fractionation were evaluated. The present GC-C/IRMS method combined with HPLC cleanup was accurate and reproducible enough to be successfully applied to the test of urine sample from suspected anabolic steroid abusers.
알긴산 제거시 전반적으로 고형분 및 단백질 수율이 감소하였고 UF처리가 $CaCl_2$ 첨가시 보다 더 뚜렸한 감소를 보였으며 점도는 알긴산 제거전 $84{\sim}94\;cps$이었던 것이 10cps로 감소하였고 탁도 또한 현저히 감소하였다. 아미노태 질소의 회수량은 $CaCl_2$ 첨가보다 UF처리한 것이 적었고 특히 NaCl을 첨가시 가장 적게 나타났으며 mannitol은 추출방법에 따른 변화가 없었다. 유기산은 oxalic acid의 함량이 높았으며 효소처리 후 NaCl 1.5%를 첨가하여 추출한 시료가 전반적으로 함량이 높게 나타났으며 핵산 조성은 분말과 2시간 열수추출의 경우 4가지 핵산이 비교적 균등한 분포를 보였으나 알긴산 제거시 일부 핵산이 함께 제거되어 핵산조성비율에 영향을 주었다. 특히 $CaCl_2$에 의한 침전제거는 IMP가 전부 그리고 UMP가 많이 손실되었으며 UF처리가 $CaCl_2$ 방법보다 핵산손실이 훨씬 적었다. 추출액의 아미노산 조성은 전체함량 중 aspartic acid와 glutamic acid가 80% 이상을 차지하고 있고 그 밖의 아미노산 함량은 대단히 적게 나타났으며 tryptophan, proline등 일부 아미노산은 검출되지 않았다.
In this study, we investigated the bioactive substances of the Alcalase hydrolysate obtained from fishery processing by-products in Jeju by measuring bioactivities including radical scavenging acitivty, cytoprotective activity against 2,2-azobis-(2-amidino-propane) dihydrochloride (AAPH), and ACE inhibitory activity. This study is important because of utilization of unused fishery processing by-products in Jeju. The Alcalase hydrolysate was prepared through the hot water extraction and enzymatic hydrolysis, and then further separation of the Alcalase hydrolysate was performed by ultrafiltration using 10 kDa molecular weight cut-off membrane. The Alcalase hydrolysate showed the relatively higher DPPH and peroxyl radical scavenging activity ($IC_{50}$ value; 1.30 mg/ml and 0.888 mg/ml, respectively). Also, the Alcalase hydrolysate showed the ACE inhibitory activity with 1.87 mg/ml of $IC_{50}$ value. These biological activities are increased over 1.2 or 2.5 times through the ultrafiltration of the Alcalase hydrolysate. Therefore, the Alcalase hydrolysate obtained from fishery processing by-products in Jeju and the different molecular weight fractions should be given consideration for food and cosmetics ingredient. Furthermore, this research on the utility of fishery processing by-products might be a useful tool into the industry.
Red cell glycolytic intermediates, metabolites and metabolic ratios were studied. Glycolytic intermediates were measured in neutralized perchloric acid extracts of red cell suspensions after 3 hr incubation at $37^{\circ}C$ in the presence and absence of saponin. Adenosine triphosphate(ATP), adenosine diphosphate(ADP), pyruvate and lactate were measured by enzymatic procedures involving stoichiometric oxidation or reduction of a pyridine nucleotide. Glucose was determined using glucose oxidase after zinc hydroxide extraction. The redox state was calculated from the lactate dehydrogenase equilibrium. Adenosine triphosphatase activity(ATPase) was measured by determining the amount of phosphate released from ATP by washed erythrocyte membranes(ghost) during 20 min. incubation. Both total hydrolysis and the amount of hydrolysis that occured in the presence of ouabain were measured. The second measurement yields Mg-ATPase and represents nonspecific ATPase activity of the membranes. The difference between total and Mg-ATPase activity can be attributed to Na-K-ATPase. For the measurement of sodium fluxes, human erythrocytes were preincubated in $^{22}Na$ for 3 hr at $37^{\circ}C$, washed and suspended in a tracer-free medium. The amount of $^{22}Na$ transported out of cells at any time was determined by analysis of supernatant samples taken at various time after addition of the labeled cells to isotope-free medium. The cells and medium were separated and the radioactivity appearing in the medium was measured. From the total radioactivity in the suspension and the radioactivity appearing in the medium at known time, the rate constant for sodium release was computed. The results are summarized as follows: 1) ATP and ATP/ADP were found to increase at every concentration of saponin tested whereas ADP declined at every cone. of saponin. The increase in pyruvate and lactate were observed at every cone, of saponin and thus $NAD^+/NADH$ computed from pyruvate/lactate also increased. Glucose utilization was stimulated by saponin. 2) $Na^+-K^+-ATPase$ activities showed a biphasic response to saponin, first increasing in lower concentration and then decreasing in higher concentration of saponin. 3) The efflux of sodium was significantly increased by saponin in the range of 5 to 10 mg%. The stimulatory effect of saponin on the rate constants for active(ouabain-sensitive) sodium efflux was inhibited by addition of ouabain.
본 연구는 소, 돼지, 닭의 부산물인 신장, 간, 기관으로부터 황산콘드로이틴을 추출하고 추출수율을 극대화할 수 있는 추출조건을 탐색하고자 실시하였다. 추출방법은 일반추출방법, 열수추출방법 및 효소가수분해 추출방법을 이용하였으며 각 추출방법에 따른 수율 및 추출물내의 황산 콘드로이틴의 양을 측정하였다. 추출방법간의 비교에서 일반추출법(대조구) 보다 열수추출과 효소가수분해 추출의 수율이 더 높게 나타났으며 유의적인 차이를 보였다(P<0.05). 효소가수분해 추출에서 식육부산물 중 가장 높은 값을 나타낸 것은 닭 기관, 소 기관, 소 간 순이었다(P<0.05). 모든 추출방법 간에 있어서 소기관의 황산콘드로이틴 함량이 가장 높았으며 효소가수분해 추출방법에 의한 소기관의 황산콘드로이틴 함량이(21.19±1.82) 가장 높은 수치를 나타내었다(P<0.05). 추출물의 수율에 영향을 미치는 에탄올에 의한 추출물 분별침전에서는 에탄올 농도가 증가할수록 황산콘드로이틴 수율이 증가하였다. 결론적으로 식육부산물로부터 황산콘드로이틴을 추출하는데 있어 효소가수분해방법이 가장 좋은 결과를 나타내었다.
본 연구에서는 국내산 상황버섯의 효소 가수분해 전처리를 통한 ${\beta}-glucan$의 최적 추출조건을 확립하고 그에 따른 활성을 알아보고자 추출 조건에 따른 생이화학적활성을 측정하였다. 효소가수분해 조건을 최적화하기 위해 실시한 반응표면분석법의 결과 0.66%(v/v)의 viscozyme 농도에서 6.08시간 반응하는 것이 최적이라 예측되었으며($R^2=0.9245$), 이에 따라 최적 추출 조건에서 추출한 시료의 ${\beta}-glucan$ 함량은 1.9594 g/100 g으로 측정되었다. 추출 수율(0.76-16.40%)은 EBE가 NEBE에 비해 약 3배 높았다. ${\beta}-glucan$ 순도(11.15-59.05%)로 가장 높았으며, ${\beta}-glucan$ 함량 또한 0.26-3.38 g/100 g으로 EB (3.38 g/100 g)가 가장 높았다. 총당 함량(0.61-1.17 mg/mL)은 NEB, EB가 NEBE, EBE보다 높았으며, EB가 가장 높았다. 구성당 분석 결과, 모든 추출물에서 glucose의 함량이 가장 높았으며, 대조구와 효소 전처리구 모두 정제하면서 그 비율이 증가하였다. 단백질 함량(0.44-11.73 mg/mL)은 NEBE, EBE가 NEB, EB보다 높았으며, EBE가 가장 높았다. FT-IR 분석 결과 $890cm^{-1}$ 부근에서 peak가 확인되었기에 ${\beta}-glycosidic$ linkage를 가지고 있는 것으로 판단하였다. MTT assay를 통해 B6F10과 SK-MEL-5 세포 독성을 측정한 결과 B6F10의 경우 대조구의 세포 생존율을 100%로 하였을 때 세포 생존율이 80% 이상으로 나타나 세포독성을 보이지 않았으나, SK-MEL-5에서는 EBE를 $100{\mu}g/mL$의 농도로 처리하였을 때 세포 생존율이 75%로 나타나 약간의 세포독성을 보였다. Wound healing assay를 통해 암세포 증식 억제활성 측정 결과, 정제한 NEB, EB가 NEBE, EBE보다 활성이 높았으며, 특히 12시간일 때 EB $30{\mu}g/mL$를 처리한 경우 B6F10과 SK-MEL-5 모두에서 가장 높은 활성을 나타내었다.
가공 및 이용율이 낮은 패류를 이용한 정미성분의 효과적인 추출방법과 분말 조미료의 제조조건을 검토한 결과를 요약하면 다음과 같다. 패류에 함유된 정미성분의 추출은 열수추출보다 단백질 분해효소를 이용한 가수분해 처리가 효과적이있으며, 분말제품의 수율을 높이기 위해서는 원료육을 완전히 액화시키는 것이 효과적이었다. 분말조미료는 굴, 키조개 및 새조개 육중량에 $50\%$의 물과 Alcalase를 각각 $4\%$ 첨가하여 $60^{\circ}C$에서 3시간동안 가수분해시키고 여과한 후 다시 막을 통과시켜 분자량 500 dalton 범위로 분획한 액을 진공동결건조하여 제품으로 하였다. 이들 제품의 수분함량이 각각 $3.5\%,\;3.8\%$ 및 $3.7\%$일 때 총질소량과 아미노질소량은 각각 $69.4\%,\;78.8\%$ 및 $74.2\%$와 $45.5\%,\;48.9\%$ 및 $45.4\%$이었다. 그리고 용해도는 각각 $78.4\%,\;76.3\%$ 및 $76.9\%$이었고 건조제품의 수율은 각각 $11.7\%,\;8.2\%$ 및 $9.8\%$로 나타났다.
동물용의약품은 2007년부터 급격한 잔류허용기준 신설에 따라 많은 수의 분석법도 함께 신설하였으며, 국제식품규격위원회(CODEX), EU 등에서 동물용의약품에 대한 기준이 국제적으로 엄격해지고 있어, 낮은 농도의 정량한계 및 재현성이 높은 분석법이 요구되어지고 있다. 하지만 국내 식품공전에서의 클렌부테롤 및 락토파민 분석법은 각각 개별 분석법으로 나뉘어져 있고, 시간적 및 경제적으로 손실이 있을 뿐 아니라 추출 효율 및 재현성이 낮아 분석에 어려움이 있다. 따라서 본 연구는 물리화학적 특성이 유사한 ${\beta}$-agonist계 동물용의약품인 클렌부테롤 및 락토파민의 기존 개별 분석법을 동시 분석법으로 개선하고 검사 효율성을 증대시키고자 하였다. 분석에 사용된 검체는 소와 돼지의 근육을 이용하였다. 검체에 내부표준물질인 클렌부테롤-$d_9$과 락토파민-$d_3$을 각각 첨가하고 ${\beta}$-글루쿠로니다제/아릴설파타제 효소를 사용하여 가수분해한 후 에틸아세테이트로 추출하였다, 추출액을 농축한 후 헥산과 메탄올을 포화시킨 용매를 적용하여 지방 제거과정을 거친 뒤 MIP 카트리지로 정제한 후 액체크로마토그래피-질량분석기(LC-MS/MS)에 주입하였다. 기기분석은 ESI(Electro-Spray Ionization) 및 positive MRM(Multiple Reaction Monitoring) 모드로 하였고, 검증은 CODEX 가이드라인 규정에 따라 실시하였다. 그 결과, 클렌부테롤과 락토파민의 LOQ는 각각 0.2 및 0.5 ${\mu}g/kg$ 수준이었고, 평균회수율은 각각 104.2-113.5% 및 107.6-118.1%로 나타났다. 또한, 분석오차는 각각 2.8-10.5% 및 1.6-5.2%로 CODEX 가이드라인 규정에 만족하는 수준이었다. 따라서 개선된 동시 분석법은 잔류동물용의약품의 분석에 있어 보다 신속하고 경제적인 분석 및 모니터링에 적용 가능할 것으로 기대된다.
