Plants are frequently exposed to numerous environmental stresses such as dehydration and high salinity, and have developed elaborate mechanisms to counteract the deleterious effects of stress. The phytohormone abscisic acid (ABA) plays a critical role as an integrator of plant responses to water-limited condition to activate ABA signal transduction pathway. Although perception of ABA has been suggested to be important, the function of each ABA receptor remains elusive in dehydration condition. Here, we show that ABA receptor, pyrabactin resistance-like protein 8 (PYL8), functions in dehydration conditions. Transgenic plants overexpressing PYL8 exhibited hypersensitive phenotype to ABA in seed germination, seedling growth and establishment. We found that hypersensitivity to ABA of transgenic plants results in high degrees of stomatal closure in response to ABA leading to low transpiration rates and ultimately more vulnerable to drought than the wild-type plants. In addition, high expression of ABA maker genes also contributes to altered drought tolerance phenotype. Overall, this work emphasizes the importance of ABA signaling by ABA receptor in stomata during defense response to drought stress.
The recombinant bioluminescent Escherichia coli strains, DPD2511 and TV 1061 containing the katG and grpE promoters, respectively, from Vibrio fischeri fused to luxCDABE, were used to detect the adaptive and repair responses to oxidative damage caused by hydrogen peroxide $(H_2O_2)$, and protein damage due to phenol. The response ratio, represented as the bioluminescence induced in subsequent inductions of DPD2511 and TV1061 with the mother cells previously induced by each chemical, i.e., $H_2O_2$ and phenol during the previous induction stage, decreased suddenly compared with the ratio of the control culture of each strain, meaning there is a possible adaptive response to stress caused by chemicals. Protein damage due to phenol was completely repaired by the second culturing after the initial induction, as was oxidative damage caused by $H_2O_2$ which was also rapidly repaired, as detected by the recovery of bioluminescence level. This result suggests that E. coli promptly adapt and repair oxidative and protein damage by $H_2O_2$ and phenol completely.
The purpose of this study was to investigate changes in the external thermal environment, following the application of evaporative cooling systems in buildings, in response to climate change. In order to verify changes in the external thermal environment, a T-test was performed on the microclimate, Thermal Comfort Index (TCI), and building surface temperature. Differences in microclimate, following the application of the evaporative cooling system in the building, were significant in terms of temperature and relative humidity. In particular, temperature decreased by more than 7% when the evaporative cooling system was applied. According to the results of the Thermal Comfort Index analysis, the Wet-Bulb Globe Temperature (WBGT) was below the limit of outdoor activities, indicating that outdoor activities were possible. The Universal Thermal Climate Index (UTCI) values were within the very strong heat stress range when the evaporative cooling system was not applied, When the system was applied, the UTCI values were within the strong heat stress range, indicating that they were lowered by one level. The building surface temperature decreased by ~10% or more when the evaporative cooling system was applied, compared to when it was not applied. Finally, the outside surface temperature of the building decreased by ~12% or more when the system was applied, compared to when it was not applied. We conclude that the energy saving effect of the building was significant.

The present study was undertaken in investigate the response to salinity and effect of plant growth regulators and proline under salinity stress on the germination and seedling growth of Vigna angularis. The protective effect of external Ca2+ on root elongation under saline conditions was also investigated. The seed germination of Vigna angularis decreased with an increase in salinity. The growth regulators GA3 was more effective than kinetin. At a higher salinity, low concentrations of kinetin and high concentrations of GA3 were more effective. The external application of proline and betaine improved germination under saline conditions. At a low salinity proline and betaine alleviated the salinity-induced inhibition of germination, yet at higher NaCl concentrations, proline and betaine were both ineffective. Exposure to salinity during germination was accompanied by an increase in the proline content, thereby suggesting that one compatible solute in the germinating seed would seem to be proline. The inhibition of germination by high NaCl concentrations was relatively more severe in scarified seeds than in intact seeds, indicating that the seed coat acts as a partial barrier to an Na2+ ameliorated the adverse effect of salinity stress.
A bacterial isolate (strain JS-2) characterized as Bacillus sp. was challenged with high concentrations of toxic selenite ions. The microbe was found to transform the toxic, soluble, colorless selenite (${SeO_3}^{2-}$) oxyions to nontoxic, insoluble, red elemental selenium ($Se^0$). This process of biotransformation was accompanied by cytoplasmic and surface accumulation of electron dense selenium ($Se^0$) granules, as revealed in electron micrographs. The cells grown in the presence of selenite oxyions secreted large quantities of extracellular polymeric substances (EPS). There were quantitative and qualitative differences in the cell wall fatty acids of the culture grown in the presence of selenite ions. The relative percentage of total saturated fatty acid and cyclic fatty acid increased significantly, whereas the amount of total unsaturated fatty acids decreased when the cells were exposed to selenite stress. All these physiological adaptive responses evidently indicate a potentially important role of cell wall fatty acids and extracellular polymeric substances in determining bacterial adaptation towards selenite-induced toxicity, which thereby explains the remarkable competitiveness and ability of this microbe to survive the environmental stress.

In this study, in order to examine whether salt and heat shock stress would alter or not contraction and relaxation of isolated rat aorta. Under anesthesia with sodium pentobarbital(50 mg Kg$^{-1}$ i.p.), male Sprague Dawley rats weighing 300-330 g were subjected to 0, heat shock combined salt stress, where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsps) by means of western blotting. The combination group of heat and 50 mM NaCl group increased vascular contractility, and the heat and 150 mM NaCl group decreased vascular contractility for 5 hours, and then recovered for 8 hours compared to that of control. Expressin of Hsp 70 of vascular muscle of rat aorta more increased by combination of heat and NaCl treatment than those of single treatment of heat or NaCl treatment, and vascular Hsp 70 showed a little decrease at 8 hours compared at 5 hours. These result indicate that mixed environmental stress either increased or decreased in vascular contractility by combination of heat and NaCl concentration.

Transcription factor Nrf2 is essential for the antioxidant responsive element (ARE)-mediated induction of phase II detoxifying and oxidative stress enzyme genes. Detailed analysis of differential Nrf2 activity displayed in transfected cell lines ultimately led to the identification of a new protein, which we named Keap1, that suppresses Nrf2 transcriptional activity by specific binding to its evolutionarily conserved amino-terminal regulatory domain. The closest homolog of Keap1 is a Drosophila actin-binding protein called Kelch, implying that Keap1 might be a Nrf2 cytoplasmic effector. We then showed that electrophilic agents antagonize Keap1 inhibition of Nrf2 activity in vivo, allowing Nrf2 to traverse from the cytoplasm to the nucleus and potentiate the ARE response. We postulate that Keap1 and Nrf2 constitute a crucial cellular sensor for oxidative stress, and together mediate a key step in the signaling pathway that leads to transcriptional activation by this novel Nrf2 nuclear shuttling mechanism. The activation of Nrf2 leads in turn to the induction of phase II enzyme and antioxidative stress genes in response to electrophiles and reactive oxygen species.
To examine whether salt stress would alter or not contractility of isolated rat aorta, under anesthesia with sodium pentobarbital(50 mg kg-1 i.p.), male Sprague Dawley rats(300-330 g) were subjected to 0, 50, and 150 mM of sodium chloride at 37$^{\circ}C$ for 60 min. where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsp) by Western blotting. One, three and eight hours later, we measured vascular contractility of isolated rat aorta treated with KCI, phenylephrine from organ bath study. The dose-vascular responses of potassium chloride and phenylephrine showed a little augmentation by NaCl concentration in the strips exposed to NaCl for 8 hours. And the response of relaxation induced by nitroprusside and acetylcholine was not influenced by NaCl stress in isolated aorta ring for 8 hours, respectively. Expression pattern of Hsp 70 of vascular muscle in isolated rat aorta showed a little increase in 150 mM NaCl group at 8 hours after NaCl treatment but not at 3 hours, and Hsp 60 expression of rat aorta was markedly increased in 50 mM NaCl group at 8 hours after NaCl treatment. Taken together, NaCl induced dose-and time dependent accumulation of the Hsp but not affected contraction of rat aorta. These data suggest that short term high salt stress was not sufficient to induce hypertension of rat aorta.

Cell response to genotoxic agents is complex and involves the participation of different classes of genes including cell cycle control, DNA repair and apoptosis. In this report, we presented a approach to characterize the cellular functions associated with the altered transcript profiles of MCF-7 exposed to low-dose in vitro gamma-irradiation. We used the method of human 2.4 k cDNA microarrays containing apoptosis, cell cycle, chromatin, repair, stress and chromosome genes to analyze the differential gene expression characterization that were displayed by radiation-exposed cell, human breast carcinoma MCF-7 cell line, such as 4 Gy 4 hr, 8 Gy 4 hr, and 8 Gy 12 hr. Among these genes, 66 were up-regulated and 49 were down-regulated. Specific genes were concomitantly induced in the results. Cyclin dependent kinase 4 (Cdk4) is induced for starting the cell cycle. This regulation is required for a DNA damageinduced G1 arrest. In addition to, an apoptotic pathways gene Bcl-w was concomitantly induced. Mismatch repair protein homologue-l (hMLH1), a necessary component of DNA mismatch protein repair (MMR), in G2-M cell cycle checkpoint arrest. The present study provides new information on the molecular mechanism underlying the cell response to genotoxic stress, with relevance to basic and clinical research.
