Structural Characterization of the Genome of BERV γ4 the Most Abundant Endogenous Retrovirus Family in Cattle

  • The genome of replication-competent BERV ${\gamma}4$ provirus, which is the most abundant ERV family in the bovine genome, was characterized in detail. The BERV ${\gamma}4$ genome showed that BERV ${\gamma}4$ harbors 8576 nucleotides and has the typical 5'-long terminal repeat (LTR)-gag-pro-pol-env-LTR-3' retroviral organization with a long leader region positioned before the gag open reading frame. Multiple sequences analysis showed that the nucleotide difference between 5' and 3' LTRs was 4.2% (mean value 0.042) in average, suggesting that the provirus formed at most 13.3 million years ago. Gag separated by a stop codon from pro-pol in the same reading frame, while env resides in another reading frame lacking of a functional surface domain. According to the current bovine genome sequence assembly, the full-length BERV ${\gamma}4$ provirus sequences were only found in the chromosomes 1, 2, 6, 10, 15, 23, 26, 28, X, and unassigned, although the partial sequences almost evenly distributed in the entire bovine genome. This is the first detailed study describing the genome structure of BERV ${\gamma}4$, the most abundant ERV family present in bovine genome. Combined with our recent reports on characterization of ERVs in bovine, this study will contribute to illuminate ERVs in the cattle of which no information was previously available.

Taxonomic Characteristics of Peyssonnelia capensis Montagne(Peyssonneliaceae, Rhodophyta) from Jeju Island in Korea (제주도산 홍조 아프리카바다표고(Peyssonnelia capensis Montagne)의 분류학적 특성)

  • The characteristics of Peyssonnelia capensis Montagne (Peyssonneliaceae, Rhodophyta) from Jeju Island were investigated in order to clarify the species entity and re-evaluate taxonomic position. The morphological variations of the Peyssonnelia capensis from Jeju Island were divided into two types, the thin thallus type and the thick thallus type. The thin thallus type is characterized by thin, soft, orbicular, slightly lobed thallus with entire to undulate margin, and compact rhizoids. The thick thallus type has thick and stiff thallus with sinuate margin and scattered rhizoids, and has more branches than the thin thallus type. The basal layer and the epithallium of thin thallus type are similar to those of thick thallus types in their structure. However, the hypothallial cells of the thick thallus type are longer than those of the thin thallus type. The erect filament of thick thallus type has more cells than those of the thin thallus type. Tetrasporangial nemathecia are distinctly protruded. Mature tetrasporangia of this species are divided cruciately into four spores as those of other Peyssonneliaceae. In addition to their morphological and anatomical distinctness between two types, the nucleotide sequence analyses of SSU and ITS2 region make no differences among populations from Jeju and other localities in Korea. Therefore these two morphological variation may not base on genetic variation.

A Study on Phase Error of Orthogonal MC DS-CDMA Using Hybrid SC/MRC-2/4 (하이브리드 SC/MRC-2/4기법을 적용한 직교 MC DS-CDMA 시스템의 위상 에러에 관한 연구)

  • In this paper, the Hybrid SC/MRC-2/4 method in which bit synchronization and phase synchronization were not required was applied to the orthogonal MC DS-CDMA system in which each normalized subcarrier interval and processing gain had the same value, respectively, and the direct sequence spread code of each subcarrier was orthogonal. In the broadband wireless system in which multi-carrier transmission was used, a Doppler frequency shift occurred, which was caused by the difference between the highest subcarrier frequency md the lowest subcarrier frequency. In order to complement phase error caused by the shift, the orthogonal MC DS-CDMA system was analyzed so that the receiving signal could be perfectly synchronized by adjusting the PLL gain suitable for the entire system. As a result of simulations, as the PLL gain was increased, the change in the intervals was close to the case of perfect synchronization however, it became less when the PLL gain reached more than a certain value. Therefore, by selecting a proper PLL gain suitable for the system the orthogonal MC DS-CDMA can be designed in which the Hybrid SC/MRC method is applied.

A Numerical Study on the Simulation of Power-pack Start-up of a Staged Combustion Cycle Engine (다단연소 사이클 엔진의 파워팩 시동 모사를 위한 해석적 연구)

  • In this study, the start-up characteristics of a staged combustion engine were analyzed numerically based on relational equation modeling of the entire engine components. The start-up characteristics were extensively analyzed considering the transient period of the total engine system from the start-up sequence till the steady-state of the engine. The performance characteristics of the engine components such as RPM of engine power-pack, chamber pressure and O/F ratio of pre-burner, and mass flow of propellants in the start-up period were investigated. Furthermore, the calculated engine data were compared satisfactorily with the experimental data. Through the comparison of data, successful validation of present engine start-up analysis has been obtained.

Effects of new construction technology on performance of ultralong steel sheet pile cofferdams under tidal action

  • Cofferdams made of teel sheet piles are commonly utilized as support structures for excavation of sea-crossing bridge foundations. As cofferdams are often subject to tide variation, it is imperative to consider potential effects of tide on stability and serviceability of sheet piles, particularly, ultralong steel sheet piles (USSPs). In this study, a real USSP cofferdam constructed using new construction technology in Nanxi River was reported. The design of key parts of USSP cofferdam in the presence of tidal action was first introduced followed by the description of entire construction technology and associated monitoring results. Subsequently, a three-dimensional finite-element model corresponding to all construction steps was established to back-analyze measured deflection of USSPs. Finally, a series of parametric studies was carried out to investigate effects of tide level, soil parameters, support stiffness and construction sequence on lateral deflection of USSPs. Monitoring results indicate that the maximum deflection during construction occurred near the riverbed. In addition, measured stress of USSPs showed that stability of USSP cofferdam strengthened as construction stages proceeded. Moreover, the numerical back-analysis demonstrated that the USSP cofferdam fulfilled the safety requirements for construction under tidal action. The maximum deflection of USSPs subject to high tide was only 13.57 mm at a depth of -4 m. Sensitivity analyses results showed that the design of USSP cofferdam system must be further improved for construction in cohesionless soils. Furthermore, the 5th strut level before concreting played an indispensable role in controlling lateral deflection of USSPs. It was also observed that pumping out water before concreting base slab could greatly simplify and benefit construction program. On the other hand, the simplification in construction procedures could induce seepage inside the cofferdam, which additionally increased the deflection of USSPs by 10 mm on average.

Molecular Authentication and Phylogenetic Analysis of Plant Species for Breeae and Cirsii Herba based on DNA barcodes (DNA 바코드 분석을 통한 소계(小薊) 및 대계(大薊) 기원식물 감별과 종간 유연관계 분석)

  • Objectives : The origin of Breeae Herba (So-gye) and Cirsii Herba (Dae-gye) is differently prescribed in Korean and Chinese modern pharmacopoeia. Since the similar morphological characteristics and chaotic plant names, moreover, the aerial part of Carduus crispus have been used as the Cirsii Herba. To develop a reliable method for correct identification of these herbal medicines and to evaluate the genetic relationship of these closely related plant species, we analyzed sequences of DNA barcode regions. Methods : Thirty-one samples of 6 medicinal plants (B. segeta, B. setosa, C. japonicum var. maackii, C. setidens, C. chanroenicum, and C. crispus) were collected from different habitate and nucleotide sequences of DNA barcode regions (rDNA-ITS, matK, and rbcL) were analyzed after amplification using appropriate primers reported in previous studies. The nucleotides of species-specific authentic marker and phylogenetic relations were estimated based on the entire sequences of DNA barcodes by the analysis of ClastalW and UPGMA, respectively. Results : In comparative analysis of DNA barcode sequences, we obtained specific nucleotides to discriminate the medicinal plant of Breeae/Cirsii Herba in species level and evaluated the phylogenetic relationship of these species. Futhermore, we identified distinct marker nucleotides enough to authenticate respective species. These sequence differences at corresponding positions were avaliable genetic markers to determine the botanical origins of Breeae Herbal as well as Cirsii Herba. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by providing of definitive information that can identify each plant species and distinguish from unauthentic adulterants and substitutes.

Development of Basic Practice Cases for Recurrent Neural Networks (순환신경망 기초 실습 사례 개발)

  • In this paper, as a liberal arts course for non-major students, a case study of recurrent neural network SW practice, which is essential for designing a basic recurrent neural network subject curriculum, was developed. The developed SW practice case focused on understanding the operation principle of the recurrent neural network, and used a spreadsheet to check the entire visualized operation process. The developed recurrent neural network practice case consisted of creating supervised text completion training data, implementing the input layer, hidden layer, state layer (context node), and output layer in sequence, and testing the performance of the recurrent neural network on text data. The recurrent neural network practice case developed in this paper automatically completes words with various numbers of characters. Using the proposed recurrent neural network practice case, it is possible to create an artificial intelligence SW practice case that automatically completes by expanding the maximum number of characters constituting Korean or English words in various ways. Therefore, it can be said that the utilization of this case of basic practice of recurrent neural network is high.

  • Industrial facilities that use modern IT technologies require the ensured reliability and security of information in automated enterprise management. Concurrently, so as to ensure a high quality of communication, it is necessary to expand the bandwidth of communication channels, which are limited by the physical parameters of the radio frequency spectrum. In order to overcome this contradiction, we propose the application of technology fundamental to ultra-wideband signals, in which the ratio between the bandwidth and its central part is greater than "one". For this reason, the information signal is emitted without a carrier frequency - simultaneously within the entire frequency band - provided that the signal level is lower than the noise level. For the transmission of information content, the method of positional-time coding is used, in which each information bit is encoded by hundreds of ultrashort pulses that arrive within a certain sequence. Mathematical models of signals and values observed in wireless communication systems with autocorrelation reception of modulated ultra-wideband signals are furthermore recommended. These assist in identifying features of the dependence of the error probability on the normalized signal-to-noise ratio and the signal base. Comparative analysis has shown that the best noise immunity of the systems considered in this paper is the communication system, which uses the time separation of the reference and information signals. During the first half of the bit interval, the switch closes the output of the transmitter directly to the generator of the ultra-wideband signal - forming a reference signal. In the middle of the bit interval, the switch alternates the output to one of two possible positions depending on the encoding signal - "zero" or "one", forming the information part of the ultra-wideband signal. It should also be noted that systems with autocorrelation reception and separate transmission of reference and information signals, provide a high level of structural signal secrecy. Furthermore, they provide the reliable transmission of digital information, especially in interference conditions.

Characterization of Salmonella species from poultry slaughterhouses in South Korea: carry-over transmission of Salmonella Thompson ST292 in slaughtering process

  • Importance: Salmonella outbreaks linked to poultry meat have been reported continuously worldwide. Therefore, Salmonella contamination of poultry meats in slaughterhouses is one of the critical control points for reducing disease outbreaks in humans. Objective: This study examined the carry-over contamination of Salmonella species through the entire slaughtering process in South Korea. Methods: From 2018 to 2019, 1,097 samples were collected from the nine slaughterhouses distributed nationwide. One hundred and seventeen isolates of Salmonella species were identified using the invA gene-specific polymerase chain reaction, as described previously. The serotype, phylogeny, and antimicrobial resistance of isolates were examined. Results: Among the 117 isolates, 93 were serotyped into Salmonella Mbandaka (n = 36 isolates, 30.8%), Salmonella Thompson (n = 33, 28.2%), and Salmonella Infantis (n = 24, 20.5%). Interestingly, allelic profiling showed that all S. Mbandaka isolates belonged to the lineage of the sequence type (ST) 413, whereas all S. Thompson isolates were ST292. Moreover, almost all S. Thompson isolates (97.0%, 32/33 isolates) belonging to ST292 were multidrug-resistant and possessed the major virulence genes whose products are required for full virulence. Both serotypes were distributed widely throughout the slaughtering process. Pulsed-field gel electrophoretic analysis demonstrated that seven S. Infantis showed 100% identities in their phylogenetic relatedness, indicating that they were sequentially transmitted along the slaughtering processes. Conclusions and Relevance: This study provides more evidence of the carry-over transmission of Salmonella species during the slaughtering processes. ST292 S. Thompson is a potential pathogenic clone of Salmonella species possibly associated with foodborne outbreaks in South Korea.

Development and growth of the human fetal sacroiliac joint revisited: a comparison with the temporomandibular joint

  • The human fetal sacroiliac joint (SIJ) is characterized by unequal development of the paired bones and delayed cavitation. Thus, during the long in utero period, the bony ilium becomes adjacent to the cartilaginous sacrum. This morphology may be analogous to that of the temporomandibular joint (TMJ). We examined horizontal histological sections of 24 fetuses at 10-30 weeks and compared the timing and sequences of joint cartilage development, cavitation, and ossification of the ilium. We also examined histological sections of the TMJ and humeroradial joint, because these also contain a disk or disk-like structure. In the ilium, endochondral ossification started in the anterior side of the SIJ, extended posteriorly and reached the joint at 12 weeks GA, and then extended over the joint at 15 weeks GA. Likewise, the joint cartilage appeared at the anterior end of the future SIJ at 12 weeks GA, and extended along the bony ilium posteriorly to cover the entire SIJ at 26 weeks GA. The cavitation started at 15 weeks GA. Therefore, joint cartilage development seemed to follow the ossification of the ilium by extending along the SIJ, and cavitation then occurred. This sequence "ossification, followed by joint cartilage formation, and then cavitation" did not occur in the TMJ or humeroradial joint. The TMJ had a periosteum-like membrane that covered the joint surface, but the humeroradial joint did not. After muscle contraction starts, it is likely that the mechanical stress from the bony ilium induces development of joint cartilage.
