Ku complex suppresses recombination in the absence of MRX activity during budding yeast meiosis

  • During meiosis, programmed double-strand breaks (DSBs) are repaired via recombination pathways that are required for faithful chromosomal segregation and genetic diversity. In meiotic progression, the non-homologous end joining (NHEJ) pathway is suppressed and instead meiotic recombination initiated by nucleolytic resection of DSB ends is the major pathway employed. This requires diverse recombinase proteins and regulatory factors involved in the formation of crossovers (COs) and non-crossovers (NCOs). In mitosis, spontaneous DSBs occurring at the G1 phase are predominantly repaired via NHEJ, mediating the joining of DNA ends. The Ku complex binds to these DSB ends, inhibiting additional DSB resection and mediating end joining with Dnl4, Lif1, and Nej1, which join the Ku complex and DSB ends. Here, we report the role of the Ku complex in DSB repair using a physical analysis of recombination in Saccharomyces cerevisiae during meiosis. We found that the Ku complex is not essential for meiotic progression, DSB formation, joint molecule formation, or CO/NCO formation during normal meiosis. Surprisingly, in the absence of the Ku complex and functional Mre11-Rad50-Xrs2 (MRX) complex, a large portion of meiotic DSBs was repaired via the recombination pathway to form COs and NCOs. Our data suggested that Ku complex prevents meiotic recombination in the elimination of MRX activity.

Design of Six-Component F/T Sensor with Flexible Fixed Ends (유연한 고정단을 가진 6축 F/T 센서의 설계)

  • This paper describes the design process of a six-component force/torque (F/T) sensor. The new six-component F/T sensor having leaf spring ends has been developed using a cross beam structure as the basic sensing element. Fundamental strain analysis of both ends fixed beam having a leaf spring structure is performed by finite element analysis. In order to obtain similar output sensing strains from the six component loads and minimize coupling strains, the optimal location of strain gages is determined and the strain gages are connected so that the bridge circuits with four strain gages would be balanced. Using leaf spring ends instead of rigid fixed ends, remarkable increment in output sensing strain can be achieved for two component forces. Several modifications in design result in a similar sensing strain of approximately $400\;{\mu}m/m$ for the six-component forces and moments, and a reduced coupling strain of $0\;{\mu}m/m$ between the forces and moments.

  • In this study, it induced to a conclusion below by experiment consideration to regarding an effective supercooling ends method of the flow cooling water in a tube of continuous ice making method and the static cooling water in a tube of continuous ice making method which used magneticfluid in a dynamic type ice storage system. Continuous ice making in a tube of the flow cooling water was shortened about 12 minutes until supercooling ends that case which gave vertical eccentricity rotation magnetic field 120rpm than did not provide magnetic field by experimental result that was tested to supercooling ends effect from shape control of magneticfluid. Continuous ice making method in a tube of the static cooling water compared with and reviewed the case that was not provided with the magnetic field and exposed cooling surface instantaneously by magnetic field. It confirmed that supercooling degree $\Delta$ $T_{c}$, $\Delta$ $T_{s}$, and $\Delta$ $T_{w}$ became lower because of heat transfering increasing by the occurrence of natural convection between after cooling starting progress time 1∼3 minutes if it did not give a magnetic field, and peformed the supercooling ends when natural convection occurred confirmed that refrigerating capacity was better. That relation $\Delta$ $T_{c}$, and $t_{e}$/($\Delta$ $T_{c}$-$\Delta$ $T_{s}$) after convection occurred, was not depended on $T_{b}$ and initial temperature if the depth of water and thickness of magneticfluid were regular and it was possible to verify conjecture of tp from $\Delta$ $T_{s}$ and $\Delta$ $T_{c}$.lar and it was possible to verify conjecture of tp from $\Delta$ $T_{s}$ and $\Delta$ $T_{c}$.c}$.>.

  • To develop the detailing methods at the ends of flexural shear walls. which are satisfying the ductility requirement corresponding to R = 3.5 and capable of improving the consturctibility, tests of ten isolated cantilever shear walls were carried out under the load condition comprising the cyclic lateral and constant vertical loads. major test parameters include the way of detailing and arrangement of transverse reinforcement in the boundary elements at the ends of walls, and placement of vertical reinforcement. From tests, comparable ductile behavior in test specimens incorporating the channel type of open hoops, compared with those incorporating the regular type of closed hoops, was observed.

  • The purpose of this study is to optimize Nd:YAG laser welding parameters to seaz a Rf source into a Ti micro capsule. Ti tube ends can be sealed as some length of ぉbe end is melted and coalesced. The exact control of the melted length is the most important to get sound sealing. The Nozzle type, tube rotating speed, tilt angle, focal position, pumping voltage, pulse frequency and pulse width were selected as the Nd:YAG laser welding Parameters. These Parameters were optimized by the Taguchi experimental method using 115 orthogonal array. Appearance and cross section of the seated tube ends were examined by SEM.

  • It is expected, in the near future, that the use of distributed generation systems should be increased considerably. In this case, distance relay algorithm should be developed in the manner that it can reduce the error due to the in-feed effect. This paper presents a method to consider the fault current rushed from the remote end of a line. In the steady-state, the relays in both ends exchange the voltages of upper nodes. If a fault is perceived, the relay calculates fault location taking the fault current from the remote end by using voltage data of the remote ends obtained just before the fault. Even though this method has inevitable error, it can show more precise fault allocation. The suggested method is applied to a typical transmission system with two power sources in both ends to verify its effectiveness.

Numerical Analysis of Tapered Circular Arch with Fixed Ends (양단고정 변단면 도호아-치의 수치해석에 관한 연구)

  • The governing differential equations for the tapered circular arch with fixed ends have been derived, and a numerical procedure for the solution of these equations have been developed. The governing differential equations were solved numerically by an initial value integration procedure and Shooting Methods for boundary value problems. The Rungekutta fourth order integration technique was used. The methods was programmed for a Cyber 73-18 computer System, and all esults were obtained on this computer. A detailed study has been made for a fixed arch with an angle of opening equal to 0.7 radian, and the results are presented in detail in tables and curves. It is hoped that the results presented herein is applied to the deformations of gives point from the tri-axial direction of tapered circular arch with fixed ends, bending moment, and torsional moment, and that at the same time results to be used for archwise structures in steel structure.

Thermal post-buckling analysis of a laminated composite beam

  • The purpose of this study is to investigate thermal post-buckling analysis of a laminated composite beam subjected under uniform temperature rising with temperature dependent physical properties. The beam is pinned at both ends and immovable ends. Under temperature rising, thermal buckling and post-buckling phenomena occurs with immovable ends of the beam. In the nonlinear kinematic model of the post-buckling problem, total Lagrangian approach is used in conjunction with the Timoshenko beam theory. Also, material properties of the laminated composite beam are temperature dependent: that is the coefficients of the governing equations are not constant. In the solution of the nonlinear problem, incremental displacement-based finite element method is used with Newton-Raphson iteration method. The effects of the fibber orientation angles, the stacking sequence of laminates and temperature rising on the post-buckling deflections, configurations and critical buckling temperatures of the composite laminated beam are illustrated and discussed in the numerical results. Also, the differences between temperature dependent and independent physical properties are investigated for post-buckling responses of laminated composite beams.

  • A 6-year-old male mongrel dog with a 7-month history of ulnar-radial nonunion fracture was treated with implantation of recombinant human bone morphogenetic protein-2 (rhBMP-2). The dog had received surgical correction three times prior to the admission but radiography of the affected limb revealed a typical figure of nonunion fracture. Glossly, the fractured ends were sclerotic and the area between the ends was filled with fibrous tissue. After debridement the shaft was fixed by an 10-hole plate. rhBMP-2 at a total dose of 256 micrograms was implanted with a synthetic carrier into the 10-mm defect formed by the debridement. Callus formation responding to rhBMP-2 was radiographically observed at 4 weeks after implantation and the defect bridged both fracture ends by 8 weeks after implantation. The plate was removed at 12 months after implantation. Any complications were not observed for 5 months after removal of the plate.

  • This study concerned with the steel beam of bonding method and bonded steel beams by this method and both ends of different height, steel beams and steel beams in the center makes the junction. Both ends and the central part of steel beams connecting the lower flange by additional combining steel plates to convey stress, the stress to focus on the beam connections are passed to both ends of steel beams, and strength of beam connections is improved and steel structural beams is proposed to minimize the loss by Incision. If you use the developed method, the construction period is shortened, and reducing the amount of material can decrease the cost and reduction in floor height can be maximized business feasibility.
