  • Endometriosis is classically defined as the growth of endometrial glands and stroma at extra-uterine sites. Although it is a common gynecological problem accompanied by chronic pelvic pain, infertility, and adhesion formation, the etiology of this disease is unknown. Endometriosis pathogenesis may involve endocrine and immune dysregulation since uterine endometrial growth is regulated by sex hormone in concert with bioactive mediators produced by uterine immune and endocrine cells.   • Endocrine disruptors have been implicated in carcinogenesis in animal studies, but carcinogenetic effects on human remain controversial. In order to examine the genotoxicity of two common endocrine disruptors, Bisphenol A and Diethylstilbestrol, cytogenetic endpoints including chromosome aberration (CA), sister chromatid exchange (SCE), micronuclei (MN) analyses and DNA damage by single cell gel electrophoresis (SCGE) were assessed. The effects of Bisphenol A and Diethylstilbestrol on the frequencies of CA and MN were increased in a dose-dependent manner and that of Bispheol A was more significant by Kendall'$\tau$test. Bisphenol A and Diethylstilbestrol also increased the frequency of SCE. Bisphenol A and Diethylstilbestrol induced DNA damage in a dose-dependent manner and the DNA damage induced by Diethylstilbestrol in human blood lymphocytes was more significant.

  • Toxicology is under challenge from several new trends in science and technology, namely computer, the Internet, genome projects, genomic technologies, and combinatorial chemistry. These new trends will drastically change research style of toxicology. In addition to conventional uni cellular tests and animal tests using rodents, computer simulation, DNA chips (microarrays), in vivo tests using simple model organisms such as nematodesor flies become important routine screening tests. How to arrange these tests in tiers will become a new problem. Endocrine disruptors hypothesis is a good example for this kind of futuristic approach. Computer, particularly the Internet, is also enabling toxicologists and regulatory experts to collaborate more closely. The IPCS (International Program for Chemical Safety) which is ajoint project of WHO, ILO and UNEP, is a well-known international collaborative research for chemical risk assessments. The GINC project of IPCS is an effort to utilize the Internet for such collaborations. Some efforts were also made to establish regional collaboration network in East Asia under this project.

Endocrine Disruptors in Developing Embryo on Daphnia magna

  • In crustaceans, as in other arthropods, the molt cycle and the physiological process of growth are controlled by molting hormones (MH) which are steroid hormones, the ecdysteroids. Ecdysteroids are major arthropod hormones which control both development (embryonic and larval molts, metamorphosis) and reproduction. The purpose of the present study was to evaluate both fenarimol and methoprene for embryotoxicity to daphnids. The embryotoxicity associated with each compound was assessed to discern whether the embryotoxicity of methoprene might be due to ecdysone agonist and the ecdysone antagonistic effects of fenarimol on Daphnia embryo. Exposure of daphnids for three weeks to 50 M methoprene resulted in a significantly high incidence of offspring that exhibited general toxicity. This exposure concentration had significant effects on the overall number of embryo death. However, exposure to 3 or 1 $\mu$M fenarimol were no significant effects on the embryo toxicity. The incidence of both of these toxicity increased with methoprene exposure. This observation suggest that methoprene showed embryonic general toxicity during embryo development, while, only fenarimol showed weak general toxicity with early stages of embryonic development.

Vitellogenin Production of Marine Fishes Exposed to E2 (17$\beta$-estradiol 처리에 따른 해산 어류의 vitellogenin 생성)

  • A common used endpoint in bioassays testing the estrogenicity of chemicals including endocrine disruptors is the induction of egg yolk Precursor vitellogenin in male fish. Two marine fishes (Sebastes schlegeli and Paralichthys olivaceus) were exposed to the 17$\beta$-estradiol(E2) to determine the vitellogenin production. Vitellogenin was measured in fish blood using SDS-PAGE and Densimetry. Results showed that exposure to E2 caused vitellogenin in male fish. Especially, vitellogenin levels in young fish were about 4 times higher than in adult fish, which means young fish are more sensitive to E2 exposure. And plasma vitellogenin in fish increased related to E2 concentration and exposure duration.

  • The contents of heavy metals in boiled Sibjeonaebotang with those in its herbal ingredients are compared. The herbal medicines of Sibjeonaebotang were bought at 10 defferent markets. The contents of 14 heavy metals (Ag, As, Ba, Cd, Cr Cu, Fe, Hg, Mn, Ni, Pb, Sb, Sn, Zn) were analysed using ICP It was found out that 5 heavy metals (Pb, As, Ba, Fe, Sb) were detected in only one boiled Sibjeonaebotang But they were detectable in all Sibjeonaebotang and its herbal before boiling. For example the content of Hg in one pack of Sibjeonaebotang before boiling was 0.064$\pm$0.016 me and Hg in boiled one was 0.002$\pm$0.005mg. These results suggest that boiled Sibjeonaebotang which we take is less harmful than herbal medicine itself by heavy metals.

  • Bisphenol A(BPA) is a monomer widely used in the manufacturing polycarbonate plastic or epoxy resin and Mancozeb (MCZ), a polymeric complex of zinc and manganese salts of ethylene bisthiocarbamate, is widely used in agriculture as fungicide, insecticide and herbcide. These chemicals have been recently known as endocrine disruptors. To investigate the effects of BPA and MCZ on LPS-induced cytokine production (TNF-${\alpha}$) in vivo, female ICR mice were administered to various concentration of these materials (BPA; 100, 500, 1000 mg/kg/day, and MCZ; 250, 500, 1000, 1500 mg/kg/day) for 30 days and serum cytokine levels were measured at 1h post LPS injection (day 32) in BPA- or MCZ-administered mice.
