정상임부(正常姙婦)의 혈청단백분획량(血淸蛋白分劃量)의 변동(變動)에 관(關)한 연구(硏究) (Electrophoretic Study of Serum Protein Changes in Pregnancy)

  • Changes in the concentration of total protein, albumin, ${\alpha}_1-,{\alpha}_2-,{\beta}-,$ and ${\gamma}-globulin$ in Serum from 138 healthy, normal pregnant woman were studied by the method of cellulose-acetate electrophoresis. 1. The concentration of total serum protein decreased gradually during the first 7 month, and showed a tendency to increase thereafter. 2. The concentration of serum albumin showed a steady continuous fall untill term. 3. During pregnancy,${\alpha}_1$ and ${\beta}-globulin$ value rose, ${\gamma}-globulin$ value fell and ${\alpha}_2-globulin$ value showed no significant change.

Evaluation of Pyunkang-tang® Administration on Hematological, Biochemical, and Protein and Lipid Profiles in Dogs

  • $Pyunkang-tang^{(R)}$ is a kind of herbal medicine and has been used for the treatment of atopy and allergic disease in humans over forty years. The purpose of this study was to evaluate the hematological, biochemical, and protein and lipid electrophoresis profiles after the oral administration of $Pyunkang-tang^{(R)}$ in healthy dogs. Fifteen clinically healthy beagle dogs were selected and orally administered either 33 ml of $Pyunkang-tang^{(R)}$ (group I, n = 5), 16.5 ml of $Pyunkang-tang^{(R)}$ (group II, n = 5), or 33 ml of distilled water (group III, n = 5) 3 times a day for 4 weeks. The results of the hematological, serum biochemical, and urine analysis did not differ significantly among the 3 groups. The oral administration of $Pyunkang-tang^{(R)}$ for 4 weeks was associated with significant changes in the serum globulin levels and an elevation in the high-density lipoprotein (HDL) concentrations in groups I and II (p < 0.05), compared to group III. The ${\alpha}1-globulin$ and ${\gamma}-globulin$ levels were significantly increased in group I, and the ${\alpha}1-globulin$ and ${\beta}-globulin$ levels were significantly increased in group II. The ${\alpha}2-globulin$ levels were significantly decreased in both groups. During the short-term evaluation of $Pyunkang-tang^{(R)}$ administration, we did not detect any specific adverse effects in the dogs. However, further evaluation of the safety and efficacy of $Pyunkang-tang^{(R)}$ for the treatment of atopic disease in dogs is needed.

콩 종실 단백질의 유전변이 (Genetic variation of 7S and 11S globulins in soybean seed)

  • 콩 저장 단백질의 대부분은 globulin이며, 이중 7S와 11S가 70% 이상을 차지한다. 따라서 콩 단백질의 조성개량을 위해서는 11S/7S비율 조정이 우선되는데, 본 연구에서는 전기영동(SDS-PAGE)법을 사용하여 콩 단백질 7S와 11S를 분리 확인하고, 이들 분획 단백질의 유전변이를 분석하였다. 국내 3개지역에서 재배된 콩 장려품종 6계통들의 평균 7S 함량은 38.9% 이었고 11S는 61.2%의 함량을 나타내었다. 분산분석 결과 품종간에 는 유의성이 있었지만 지역간에는 변이가 없었으며, 품종 x 지역의 상호작용은 고도의 유의성을 나타내었다. 유전력은 7S분획중의 $\beta$함량이 72.7%로 높게 나타났다. 공분산을 이용한 상관계수 추정에서는 유전상관이 표현형 상관 보다 다소 높게 나타났다. 따라서 7S와 11S의 분획간 함량을 조정함으로써 콩 단백질의 조성을 개량할 수 있으리라 판단된다.

유청 및 대두 단백질의 상호작용 (Interaction between Whey and Soybean Proteins)

  • 유청 및 대두 단백질의 상호작용을 규명하기 위하여 두 단백질용액 및 1 : 1 혼합용액에 대한 겔투과크로마토그래피와 전기영동에 의하여 열처리 중 조성단백질의 변화 및 상호작용에 관여한 조성단백질을 조사하였다. 크로마토그래피 결과 $80^{\circ}C$ 이상에서 열처리할 경우, 대두단백질 및 혼합물에서 저분자량의 단백질과 고분자량의 응집물이 증가한 것으로 나타났는데, 이는 열처리에 의하여 대두의 11S globulin이 subunit로 해리되고, 이것이 thiol기, disulfide bond 등을 함유한 유청단백질과 가용성 응질물을 형성하기 때문으로 생각되었다. 전기영동에 의하여, 가열시 유청조성단백질 중의 ${\beta}-Lactoglobulin$, ${\alpha}-Lactalbumin$ 및 proteose-peptone 3이 대두단백질과 상호작용을 일으키는 것으로 나타났다. 그리고 용액의 염환경에 따라 Bovine Serum Albumin, Immunoglobulin G(H) 및 Lactoferrin도 상호작용을 일으킬 수 있으며, 대두조성단백질 중의 11S globulin의 basic subunit와 acidic subunit, 7S globulin의 ${\alpha}'$ subunit가 유청단백질과 상호작용을 일으킬 수 있음을 추측할 수 있었다.

양미리를 급여한 사육 황새에서 ${\beta}$-글로불린 분획의 증가 (Increased ${\beta}$-globulin Levels in Captive Oriental White Storks Fed a Diet of Sand Eels)

  • 이 연구는 사육황새에 급여하는 먹이(양미리와 전갱이)의 차이에 따른 혈청 단백질 분획의 변화를 평가하기 위한 것이다. 사육황새를 먹이에 따라 두 그룹(그룹 1과 2)으로 나누어 연구를 진행하였다. 그룹 1에는 22마리, 그룹 2에는 29마리의 황새가 포함되었고, 각각 양미리와 전갱이를 급여하였다. 두 그룹에서 전혈구검사, 혈청화학검사, 단백질분획검사(${\alpha}$-, ${\beta}$- 및 ${\gamma}$-글로불린) 및 지단백질 분획검사(HDL 및 LDL)를 진행하고 결과를 비교하였다. 검사 결과 그룹 1에서 ${\alpha}$-글로불린의 감소와 ${\beta}$-글로불린의 증가가 유의적으로 관찰되었다. 그룹 1에서 LDL의 농도는 그룹 2에 비해 유의적으로 증가하였다. 결과적으로 양미리를 급여한 사육황새에서 ${\beta}$-글로불린의 증가가 LDL의 증가에 의한 것임을 확인하였다.

여지전기영동법(濾紙電氣泳動法)에 의(依)한 한우(韓牛) 급(及) 돈(豚)의 정상혈청단백질분획(正常血淸蛋白質分劃)에 대(對)한 연구(硏究) (PAPER ELECTROPHORETIC SEPARATION OF SERUM PROTEINS IN CATTLE AND SWINE)

  • The ratios of cattle and swine serum proteins taken from the slaughter house were studied by Paper Electrophoresis. 1. Of 79 cattle and 53 swine, 49 cattle and 32 swine were observed in this studying as normal animals, the rest which was over 60% of albumin, globulin values and 1/2 of A/G (albumin/globulin) ratio was observed separately as abnormalities, because physiological examination was not made before slaughter. The ratios of the normal serum proteins were A (albumin) 58.8, ${\alpha}$(alpha-globulin) 13.7, ${\beta}$(beta-globulin) 11.9, ${\gamma}$(gamma-globulin) 28.6, G(total globulin) 49.2, A/G 1.03 in cattle and A 48.4, ${\alpha}$ 18.0, ${\beta}$ 13.6, ${\gamma}$ 20.0, G 51,6, A/G 0.93 in swine, the result including abnormalities showed A 45.5, ${\alpha}$ 14.8, ${\beta}$ 12.5, ${\gamma}$ 26.7, G 54.5, A/G 0.83 in cattle and A 44.5, ${\alpha}$ 19.8, ${\beta}$ 13.7, ${\gamma}$ 21.8, G 55.3, A/G 0.80 in Swine. 2. The A/G ratio of cattle and swine were 1.03 and 0.93 respectively, the A/G ratio of Korean cattle and swine are higher than the ration reported of others. Although A/G ratio of swine was below 1.00, and its value showed slightly higher than the others. The A/G ratio in this result including the abnormalities was relatively low but this ratio was higher than that values obtained by other reporters. 3. Twenty nine percent of cattles and 34 per cent of swines in this study, fluctuation of A/G ratio was great. The values of ${\alpha}$ and ${\gamma}$ globulins thought to be influenced by the amount of total globulin except ${\beta}$-globulin in swine. To obtain more occurate results, more sample size is required, in other hand some animals that is in subclinical condition might influence the values of this study. 4. The ratios of each fraction mobility which were regarded albumin as 100 were A 100, ${\alpha}$ 73, ${\beta}$ 47, ${\gamma}$ 30 in Cattle and A 100, ${\alpha}$ 71, ${\beta}$ 46, ${\gamma}$ 30 in Swine.

원반전기 영동법을 이용한 전립선액의 단백분리에 관한 연구 (Analysis of the Proteins in Human Prostatic Juice by Disc Electrophoresis)

  • Despite a considerable amount of investigation there continues to be disagreement concerning the proteins present in human seminal plasma. Recently their identification has assumed a greater importance following evidence that infertility in men and women may have an immunological cause (Katsh, 1959; Quinlivan, 1969). Seminal plasma is composed of fluids secreted by the prostate, seminal vesicles, ampullae, ducti deferentes, bulbourethral (Cowper's) glands, urethral(Littre's) glands and the epididymes. Prostatic juice, one of the major components of seminal plasma, has an important role in secretion of acid phosphatase and prostaglandin. A few studies have been reported of human prostatic juice, since, in human subjects, there were some problems in studying prostatic juice due to quite small amount of secretion and possibility of contamination with fluids from the seminal vesicles and ejaculatory ducts. The purpose of the present study was to determine the basic components of proteins in human prostatic juice. Prostatic juice was obtained from normal healthy man of $20{\sim}30\;year-old$ by massage of the prostate, and protein components were separated by means of disc electrophoresis. The results are summarized as follows; 1) Total numbers of protein fractions of normal human serum and prostatic juice are $14{\sim}18$ bands and $9{\sim}12$ bands, respectively. Prostatic juice produces two deeply staining bands which appear similar to those formed by $beta-_1$ globulin and albumin. 2) $Alpha-_1$ globulin area in the fractions of prostatic juice shows 4 bands and one more band is found than that of serum. On the other hand, the fractions of immunoglobulin and $alpha-_2$ globulin areas are eight in serum and it has three bands more than that of prostatic juice. 3) $Alpha-_1$ globulin area in the prostatic juice is more deeply stained than that of serum. In contrast with $alpha-_1$ globulin area, immunoglobulin and $alpha-_2$ globulin areas in the prostatic juice show weaker staining than serum.

Protein Electrophoresis Fraction in Serum of the Rural Elderly Patient

  • We analyzed total protein (TP), albumin (AL), aspartic aminotransferase (AST), alanine aminotransferase (ALT), urea nitrogen (UN), creatinine (CRE) and serum protein electrophoretic fractions in sera of rural elderly patients to evaluate the health status in rural elderly patients. We observed that the frequencies of patients showed lower level of total protein, albumin, and both total protein and albumin than them of reference range were 20.3%, 22.8% and 19.0%, respectively. The rates of patients showed higher level of AST, ALT, both of AST and ALT, UN, creatinine and both of UN and creatinine than them of reference range were 33.8%, 40.0%, 30.0%, 17.7%, 15.2% and 7.9%, respectively. Comparison of protein fractions of each patient to reference range showed that 77.2% of patients showed normal in all of patterns. Few of patients showed abnormal pattern in albumin (13.6%), ${\alpha}1-globulin (0.0%),\;{\alpha}_2-g10bulin(1.1%),\;{\beta}-globulin(1.9%)\;and\;{\gamma}-globulin(6.2%)$. These data suggest that many of rural elderly patients may suffer from heart, liver and kidney diseases. The serum protein fractions are not typical criterion to evaluate the disease, but production of proteins in rural elderly patients may be affected by liver disease and kidney disease.

쌀단백질의 분획 및 전기영동 (Fractionation and Electrophoretic Patterns of Rice Proteins)

  • 네가지 쌀단백군의 양적 관계는 주로 추출조건에 의하여 좌우되며 albumin 및 globulin의 추출양은 교반방법보다 추출온도에 의하여, glutelin은 사용용매의 종류와 pH에 의하여 추출양이 크게 좌우되었다. 적합한 쌀단백질의 추출로는 albumin 및 globulin은 0.5M-NaCl용액으로 $0{\sim}5^{\circ}C$에서, prolamin은 70%(v/v) ethanol용액으로 상온에서, glutelin은 0.1N acetic acid보다는 0.5% SDS-borate buffer (pH 8.3) 과 0.6% ${\beta}-mercaptoethanol$을 포함한 동일용액을 사용하는 것이 효과적이었다. 상기방법으로 추출한 각 단백군 구성성분의 분자량 분포는 albumin은 29,000, $18,000{\sim}15,000$ 및 12,000의 세가지이며 globulin은 27,000, 21,000 17,000 및 12,000의 4가지 이외에 고분자량의 단백질($10^5$이상)로 되어 있고 prolamin은 16,000, 0.5% SDS-borate buffer에 가용성인 glutelin은 35,000, 21,000, 17,000 및 15,000의 4가지와 이외에 globulin의 경우와 같이 고분자 단백질로 구성되어 있었다. Prolamin을 제외한 세 단백군은 각각 disulfide bond로 연결된 subunit를 가지고 있는 구성성분을 포함하고 있음을 알 수 있었다. SDS-glutelin은 Sephadex G-150 column chromatography에 의하여 세가지 fraction으로 분리할 수 있었다. Albumin과 globulin은 starch gel 전기영동상 산성 (pH 3.1)에서는 총 13개 및 21개의 band를 각각 나타내었고 염기성 (pH 8.95)에서는 총 4개 및 6개의 band를 확인할 수 있었다. 쌀품종들은 albumin과 globulin의 starch gel전기영동 pattern에 의하여 구별할 수 있었다. 산성조건에서는 특수 band의 존재 또는 band의 조합양상에 의하여 각 품종의 구별이 가능하였고 염기성조건에서는 각 품종별 구별은 할 수 없었으나 Japonica종을 Indica 또는 Japonica와 Indica교배종과 분리 식별할 수 있었다.
