  • We have launched an investigation for Embryonic Development, Larvae and Juvenile Morphology, of Buenos aires tetra in order to build basic data of Characidae and fish seeding production. We brought 50 couples of Characidae from Bizidduck aquarium in Yeosu-si, Jeollanamdo, from Korea on March of 2015. We put them in the tetragonal glass aquarium ($50{\times}50{\times}30cm$). Breeding water temperature was $27.5{\sim}28.5^{\circ}C$ (mean $28.0{\pm}0.05^{\circ}C$) and being maintained. The shape of fertilized egg was round shape, and it was adhesive demersal egg. The egg size was 0.63~0.91 mm (mean $0.74{\pm}0.07mm$, n=20). After getting fertilized egg, the developmental stage was gastrula stage, and embryo covered almost two-thirds of Yolk. Incubation was happened after 16 hours 13 minutes from gastrula stage, and the tail of juvenile came out first with tearing egg capsule. Immediately after the incubation, prelarvae had 3.78~3.88 mm length (mean $3.84{\pm}0.04mm$, n=5), and it had no mouth and anus yet. 34 days after hatching from the incubation, juvenile had 8.63~13.1 mm (mean $10.9{\pm}1.66mm$), and it had similar silver-colored body shape with its mother.

  • The egg development, early life history and spawning site characters of Korean endemic fish, Coreoleuciscus splendidus (Gobioninae), were investigated at the part of Jo-jong stream in Korea from May 2020. The fertilized eggs were 2.05~2.23 mm (mean, 2.13 mm) in diameter and had no oil globules. The embryo began to hatching about 98 hrs after fertilization under water temperature of 20±1℃. The newly-hatched larvae were 5.03~5.68 mm (mean, 5.31 mm) in total length (TL), and their mouth and anus were not opened. 4 days after hatching, Several rod-like cupulae were observed on the head and lateral side of the body at 6.95~7.89 mm (mean, 7.51 mm). 7 days after hatching, the post-larva stage were 8.39~9.29 mm (mean, 8.78 mm) in total length, and their york were completely absorbed. The cupulae were completely distinguished. They entered the juvenile stage when all fin-rays were formed at 29 days after hatching, and their TL were 14.16~17.04 mm (mean, 14.99 mm). Squamation was initiated on the caudal body at approximately 18.21~23.74 mm (mean, 30.28 mm), 38 days after hatching, and completed at 26.82~33.33 mm (mean, 19.42 mm), 73 days after hatching, the external characteristics from of juveniles were same to adults. The spawning site was characterized by bottom structure of pebble (64~16 mm) and gravel (16~2 mm), environmental conditions of the spawning sites were 6~18 cm in water depth, 0.43~0.73 m/sec in bottom water velocity. In spawning sites, an egg mass or separated eggs were located a small gap of under the pebble and gravel.

  • The egg development and early life history of the Korean endangered fish Koreocobitis naktongensis (Cobitidae) were investigated. Eggs from the females were obtained by injecting 10 IU/g of human chorionic gonadotropin and inseminated with sperm by the dry method in the laboratory. The fertilized eggs were 1.0~1.2 mm in diameter and had no oil globules. The embryo began to hatch about 38 hrs after fertilization under water temperature of $20{\pm}1^{\circ}C$. The newly-hatched larvae were 2.6~2.8 mm in total length, and their mouth and anus were not yet opened. Seven days after hatching, the postlarvae were 4.5~5.2 mm in total length, and their yolk sacs were completely absorbed. They entered the juvenile stage when all fin-rays were formed at 50 days after hatching, and their total lengths were 21~27 mm.

  • The egg development and early life history of Acheilognathus macropterus from Japan which is an exotic bitterling from China were observed under the controlled water temperature, $20.0{\pm}1.0^{\circ}C$. Fertilized eggs are opaque yellow in color and long elliptic globe shaped measuring $2.78{\pm}0.12mm$ in length and $1.44{\pm}0.04mm$ in breadth. The number of egg averaged 151 per an oviposition. The eggs of this species began to hatch about seventy eight hours after insemination and the mean of total length of larvae were 3.8 mm. S form moving of larvae were observed from three days after hatching. The larvae reached at the heterotrophic stage about twenty-five days after hatching. Morphological character and analysis of cytochrome DNA of this species from Japan were relatively similar to Korean but spawned egg shape was different remarkably. Taxonomical research is necessary in the future.

Influence of Food and Density on the Growth and Survival of Sea Cucumber, Stichopus japonicus (해삼, Stichopus japonicus 유생의 성장과 생존에 미치는 먹이 및 수용밀도의 영향)

  • In order to obtain the basic biological data for artificial seedling production of sea cucumber, Stichopus japonicus, the influence of water temperature and air dry for spawning induction, egg development and larvae rearing was investigated during the period from April, 1995 through September, 1995. Spawning induction rate by the water temperature was 6.0~17.5% and air dry was responsed 1.4~4.0%. Number of eggs spawned of Stichopus japonicus were $50~500\times10^4$ individuals, the fertilization and hatching rate were ranged 84.0~96.0%, 71.4~84.6% respectively. The fertilized egg of Stichopus japonicus appeared mean diameter of $154{\mu}m$. At a constant water temperature of $23^{circ)C$, it become 4 tell stage from fertilization after 2 hours 10 minutes, hatching larvae after 14 hours half, auricularia larvae after 3 days, doliolaria larvare after 11 days and pentactula larvae after 15 days ready for settlement. The suitable food in the larvae reared for 17 days after fertilization were shown the best growth and survival in the larvae food of Chaetoceros calcitrans. Optimum density for larvae rearing were maintained of the larval density lower than 2 individuals/ml.

Egg Development and Morphology of Larva and Juvenile of the Chub Mackerel, Scomber japonicus (고등어(Scomber japonicus)의 난발생 및 자치어 형태발달)

  • In order to monitor the developmental features of embryos, larvae, and juveniles of chub mackerel, Scomber japonicus, the fertilized eggs were obtained by artificial insemination. The fertilized eggs were spherical in shape and floated separately. Their membrane and yolk having 1 oil globule were transparent. The transparent eggs, having one oil drop in the yolk, were $0.94{\sim}1.02\;mm$ (mean, $0.95{\pm}0.03\;mm$) in diameter. The fertilized eggs started hatching at 51 hrs after fertilization in $20{\pm}0.5^{\circ}C$ water. The total length of the hatched larvae was $2.52{\sim}3.0\;mm$ (mean, $2.75{\pm}0.04\;mm$). At hatching, the larva, with the mouth and anus not opened yet, had yolk sack, $28{\sim}31$ myomeres and eyes with melanophore. Yolk completely 2 days after hatching and the total length of post-larvae $3.12{\sim}3.63\;mm$ (mean, $3.39{\pm}0.05\;mm$). At the 18 days after hatching, the tip of tail became curved at the end and the stems for pectoral, dorsal, and caudal fins appeared. Juveniles, having all firm rays, was $44.12{\sim}58.72\;mm$ (mean $55.95{\pm}6.74\;mm$) in TL 25 days after hatching.

Development of the Eggs, Larvae and Juveniles by Artificially-Matured Pacific Mackerel, Scomber japonicus in the Korean Waters (성숙 유도한 한국산 고등어 (Scomber japonicus)의 난발생과 자치어의 형태 발달)

  • Development of egg, larvae and juveniles for the Pacific mackerel, Scomber japonicus are described following natural fertilization in the indoor tank of $25^{\circ}C$ water temperature. Following a routine hormone treatment technique for the brood stock, male and female mackerels were artificially matured by intramuscular injections of LHRHa at a dosage of $400{\mu}g/kg$ body weight (BW)+Domperidone at a dosage of $4{\mu}g/kg$ body weight (BW) to induce maturation in a separate aquarium and induced natural spawning. Fertilized eggs were ca. 1.0 mm in diameter; spherical in shape with a single oil globule; pelagic and non-adhesive. Hatching occurs 41 hours after fertilization at $23-24^{\circ}C$. The newly hatched larvae was 3.03 mm in average total length (ATL), the mouth and anus were not open, oil globule located in posterior end of yolk sac, and preanal length was 42.8% of TL. The larvae measuring 2.89 mm ATL, almost absorbed yolk sac and oil globule material in 2 days after hatching, in which the mouth and anus were open. Melanophores, branch or star in shape were observed on the top of head, peritoneal region and along the ventral contour. In 13 days after hatching, the larvae was 6.88 mm ATL, its posterior end of notochord began to flex upward, finfold of caudal fin appeared, jaw teeth were already formed. In 19 days after hatching, the larvae was 7.71 mm ATL completed only caudal fin rays (9+8), and preanal length was 49.4% of TL. In 37 days after hatching, the larvae was 27.4 mm ATL already completed all the fins, and preanal length was 59.9% of TL.

Environmentally-friendly Control Methods and Forecasting the Hatching Time Lycorma delicatula (Hemiptera: Fulgoridae) in Jeonnam Province (전남지역에서 꽃매미 부화시기 예측과 친환경 방제방법)

  • This study was conducted to predict the hatching time of eggs of Lycorma delicatula, to select an effective environmentally-friendly agriculture material (EFAM) and to evaluate the attraction effect of brown sticky traps for controling of Lycorma delicatula nymph and adults. Eggs hatched 55.9, 26.8, 21.6 days after incubation at 15, 20, $25^{\circ}C$ with 14L:10D condition and the hatching rates of egg were 61.9, 57.8, 30.4%, respectively. At high temperature conditions, egg development periods were shorter and the hatching rate was lower. The relationship between temperature and developmental rate was expressed by the linear equation Y=0.0028X-0.0228, $R^2$=0.9561. The low temperature threshold of eggs was $8.14^{\circ}C$ and the thermal constant required to reach larva was 355.4 DD. According to this relationship, the mean estimated hatching date was $22^{nd}$ May. The effective EFAM was natural plant extract, sophora extract, derris extract to nymph and natural plant extract, pyrethrum extract, sophora extract to adult. Among three colors of sticky trap : brown, blue and yellow, the brown sticky trap was the most attractive to nymphs and adults of L. delicatula over a 2 weeks trial period. It suggested that the brown sticky trap could be a very useful and environment-friendly control method for nymphs and adults of L. delicatula.

Morphological of Development Eggs, Larvae and Juveniles Gymnogobius urotaenia in Hwangbocheon, Korea (황보천에 서식하는 꾹저구 Gymnogobius urotaenia의 난발생 및 자치어 형태발달)

  • This study compared the results of observing the early life history of Gymnogobius urotaenia, which lives in Hwangbocheon Stream, an inflow stream on the east coast, with the differences between fish of the same Gobiidae fishes. In May 2022 and 2023, fertilized eggs and brood stork scattered under rocks were captured twice in Hwangbocheon Stream. The spawning amount was 827~1,540 and the orchid was a elliptical in shape with a size of 3.21×1.07 mm. The stage of ovulation observed in the laboratory was 16 cells, and hatching began after 193 hours. The breeding water temperature range was 18.8~19.3℃. Newly after hatching larvae, the yolk sac with a total length of 3.84~4.33 (average 4.10±0.17, n=30) mm, and the anus was not open. 6 days after hatching of incubation, the total length was 5.32~6.11 (average 5.67±0.25, n=30) mm, absorbing all egg yolk and transitioning to the preflexion larvae, ingesting food, and developing a keynote on the tail fin. 15 days after hatching, the end of the urostyle end was completely bent at 45° with a total length of 7.33~8.52 (average 7.81±0.46, n=30) mm and transitioned to postflexion larvae, and melanophore developed throughout the body. 38 days after hatching, the total length is 22.1~26.1 (23.8±1.36, n=30) mm and the number of fins (6 first dorsal fins, 11 second dorsal fins, and 11 anal fins, 12 ventral fins) is all the number of fin base became integer and transferred to the juvenile. As a result of the study, it was possible to distinguish the melanophore of postflexion larvae of G. urotaenia from other postflexion larvae Gobiidae fish in that they were distributed throughout the body of half the body and tail.

Gene expression changes in silkworm embryogenesis for prediction of hatching time

  • The silkworm's dormancy and embryonic development are accomplished through the interaction of various genes. Analysis of the expression of several interacting genes can predict the embryonic stage of silkworms. In this study, we analyzed the changes in the expression level of genes at each stage during the embryonic development of dormant silkworm eggs and selected genes that can predict the hatching time. Jam123 and Jam124 silkworms were collected after egg laying, and the silkworm eggs were preserved using a double refrigeration method and expression analysis was performed for 23 genes during embryogenesis. There were 5 genes showing significant changes during embryogenesis: UDP-glucuronosyltransferases (BmUGTs), heat shock protein hsp20.8 (BmHsp20.8), Cytochromes b5-like proteins (BmCytb5), Krüppel homolog 1 (BmKr-h1), and cuticular protein RR-1 motif 41 (BmCpr41). As a result of quantitative comparison of the expression levels of these 5 genes through real-time PCR, the BmUGTs gene showed a difference between Jam123 and Jam124, making it difficult to see it as an indicator for predicting hatching time. However, the BmHsp20.8 gene had a common expression decreased at the imminent hatching stage. In addition, it was confirmed that the expression level of the BmCytb5 gene decreased to the lowest level at the time of imminent hatching, and the expression of the BmKr-h gene was made only at the time of imminent hatching. The expression of the last BmCpr41 gene can be confirmed only at the time of imminent hatching, and it was confirmed that it shows a rapid increase right before hatching. Taken together, these results suggest that expression analysis of BmHsp20.8, BmCytb5, BmKr-h1, and BmCpr41 genes can determine the stage of embryogenesis, predict hatching time, which facilitate better management of silkworm eggs.
