Expression Patterns of the chgH:rfp Transgene in Response to 17α-Ethinylestradiol (EE2) Exposure in Marine Medaka Oryzias dancena

  • The functional utility of a transgenic marine medaka Oryzias dancena strain carrying the red fluorescent protein (RFP) gene driven by an endogenous choriogenin H (chgH) promoter was evaluated for its ability to detect waterborne $17{\alpha}$-ethinylestradiol (EE2), a synthetic estrogen derivative. The chgH:rfp transgenic marine medaka larvae showed an age-dependent tendency in the efficiency of EE2-mediated transgene expression, in which transgenic larvae older than 6 days post-hatching displayed a more effective response in their transgene expression to EE2 than did younger hatchlings. During experimental exposures to high concentrations of EE2 (200 to 1,000 ng/L), the transgenic responses in the hatchlings were broadly dose- and duration-dependent. With exposures using lower doses of EE2 (25, 50 and 100 ng/L), EE2-induced transgenic RFP was also observed in the transgenic larvae, although the lower doses required exposure of longer duration. Under the EE2 exposure and microscope assay conditions used in our study, transgenic marine medaka larvae exhibited a similar degree of EE2-mediated RFP phenotype expression at various salinity levels (0, 15 and 30 ppt).

Efficacy of Soil Amendment with Medicinal Plant Materials for the Control of Root-knot Nematode (Meloidogyne incognita) in Tomato

  • Soil amendments with oriental herbal medicines such as fruit of Anethum graveolens, flower buds of Syzygium aromaticum, rhizome of Cnidium officinale, rhizome of Coptis chinensis, root bark of Paeonia suffructicosa, stem bark of phellodendron amurense, and stem bark of Cinnamomum cassia at the rate of 0.2% (weight by volume of soil) significantly reduced Meloidogyne incognita infection (root gall formation) of tomato seedlings compared with the control. The most effective treatments were root bark of p. suffructicosa and stem bark of C. cassia as they gave minimum numbers of galls on tomato roots (4.7% and 8.9%, respectively, relative to control) as compared to other treatments. Another study with root bark of p. suffructicosa and C. cassia at different application doses also showed consistent results in reducing gall number. The control efficacy decreased as the application doses were lower-ed, indicating their dose-dependent control activities. These treatments significantly enhanced aboveground plant growths (total masses).

  • It is known that ion-assisted reaction method is effective for the surface modification of polymers. The surface treatment of Spectra fibers using the ion-assisted reaction method was investigated in the present study The Spectra fibers were treated by $Ar^{+}$ / ion irradiation under oxygen environment. The treatment was carried out at different $Ar^{+}$ ion doses. The$ Ar^{+ }$ /ion doses used were $6$\times$10^{15}$ , $1$\times$10^{16}$ , $5$\times$10^{16}$ , $1$\times$10^{17}$ / ions/$\m^2$. Optimal $Ar^{+}$ ion dose in the treatment of Spectra fibers was determined by measuring the tensile strength and modulus of Spectra/vinylester composites as a function of ion dose. It was found that the optimal ion dose was $1$\times$10^{16}$ions/$\m^2$. It was also found from the scanning electron microscope examination that the surface-treatment improved adhesion between fibers and vinylester resin.

  • The purpose of this study is to evaluate the feasibility, advantages/disadvantages of patient-controlled sedation (PCS) compared to anesthesiologist-controlled sedation (ACS) during neurolytic pain block and regional anesthesia. Forty patients were divided randomly into two groups of 20 patients each. Group 1(ACS) received 0.01 $mg{\cdot}kg^{-1}$ intravenous midazolam and 0.5 ${\mu}g{\cdot}kg^{-1}$ fentanyl intravenously by anesthesiologist just before, 30, and 60 minutes after the procedure to acheive sedation; Group 2 (PCS) patients self-administered a mixture of midazolam (0.4 mg) and fentanyl ($20{\mu}g$) using a syringe type infusion pump (Terumo, Japan) to acheive sedation. Considering the dermographics of patients, the types and durations of procedure performed, the level of average sedation the comfort level were similar in both groups. But the doses of midazolam and fentanyl administerd in group 2 were smaller than those in group 1 (p<0.01). Patients in PCS group showed their level of sedation more proper than did those in ACS group. However, patients in ACS group rated their level of comfort higher than did those in PCS group. The findings of this study indicate that PCS using a combination of midazolam and fentanyl is a fafe and effective technique. More studies are, however, needed to determinc the best choice of drug(s), doses, lock-out intervals, and possible use of continuous infusion with patient-controlled sedation.

Synthesis of Allylthiopyridazine Derivatives and Inhibition of Aflatoxin ${B_1}-Induced$ Hepatotoxicity in Rats

  • Five kinds of allylthiopyridazine derivatives were synthesized and their chemoprotective activities examined in rats exposed to aflatoxin $B_1$-toxicant. Rats were pretreated with five allylthiopyridazine derivatives at daily oral doses of 50 mg/kg for 10 consecutive days, and during this period with one or three repeated doses of the potent hepatotoxin, aflatoxin $B_1$. The hepatoprotective effects of the allylthiopyridazine derivatives against aflatoxin $B_1$ (1 mg/kg, three times at intervals of 3 days, i.p., or at 3 mg/kg, once at final days, i.p.) administration were showed the significantly normal as compared with control in body and liver weights. Aspartate aminotransferase and alanine aminotransferase levels were markedly elevated after aflatoxin $B_1$ administration, and pretreatment with allylthiopyridazine derivatives, before aflatoxin $B_1$ administration, resulted in decreased levels of these enzymes. In addition, the allylthiopyridazine derivatives, K6 (3-methoxy-), K8 (3-chloro-), K16 (3-ethoxy-) and K17 (3-n-propoxy), induced elevated hepatic GSH levels. Four kinds of allylthiopyridazine derivatives investigated were effective against aflatoxin $B_1$ -induced hepatotoxicity.

  • This study was carried out to establish an effective and practical system for commercialization of embryo production techniques by analyzing several factors influencing in vivo embryo production on condition of donors treated FSH-P and SUPER-OV in Korean native cattle. In vivo embryos were flushed 226 times from 128 donors. The results obtained for the factors influencing in vivo embryo production by conditionof donors treated FSH-P and SUPER-OV were as follows :1. Age and parity of donors did not affect the transferable and freezable embryos among the treatments in FSH-P but the transferable and freezable embryos were decreased after 6 years old and 4th parity in SUPER-OV(P<0.01). 2. The production of embryos on the weight of donors was higher in over 400kg of body weight in FSH-P(P<0.01) and was higher in over 450kg than 400~450kg of body weight in SUPER-OV(P<0.05). For FSH-P embryo production was better responded in 350~450kg of body weight with 30~32mg doses, and showed a better result in over 450kg body weight with 32~34mg doses.(Key Words : in vivo embryo, donors, FSH-P SUPER-OV)

Assessment of Microorganism-derived Adjuvants for Scuticociliate Miamiensis avidus Vaccine (스쿠티카충Miamiensis avidus 주사백신용 미생물유래 면역보조제의 평가)

  • Microorganism-derived compounds, such as peptidoglycan, lipoteichoic acid, and β-glucan were supplemented in the scuticociliate Miamiensis avidus (M. avidus) vaccine to verify the specify component contribution to the adjuvant effect. Vaccine was formulated with the inactivated M. avidus antigen (YS2, 4.44×105 cells/fish) in combination with either peptidoglycan (10 ㎍ and 100 ㎍/fish), lipoteichoic acid (5 ㎍ and 50 ㎍/fish), or β-glucan (10 ㎍ and 100 ㎍/fish). Olive flounder injected with peptidoglycan supplemented vaccine (10 ㎍ and 100 ㎍/fish) exhibited significant protection, and the relative percent survival (RPS) was 55% and 65% at 4 weeks post vaccination (wpv), respectively, at the corresponding doses. The vaccine groups with added lipoteichoic acid (5 ㎍ and 50 ㎍/fish) exhibited RPS of 40% and 5%, respectively. Additionally, the group with added β-glucan (100 ㎍/fish) exhibited RPS of 35%, but no effect was observed in the group with added 10 ㎍/fish β-glucan. At 8 wpv, olive flounder injected with peptidoglycan and lipoteichoic acid supplemented vaccines exhibited protection with RPS range of 11/11% and 5/21%, respectively, at the respective doses. M. avidus vaccine containing 10 ㎍ and 100 ㎍/fish of β-glucan exhibited the RPS of 32% and 37%, respectively. Conclusively, peptidoglycan contributed in high protection of the M. avidus vaccine, and thus, it can be used as an effective adjuvant in the M. avidus vaccine.

Superovulation in Rabbits with a Single Injection of FSH Dissolved in Polyvinylpyrrolidone (토끼에서 FSH제의 Single Injection에 따른 과배란유기 효과)

  • A series of experiments were designed to determine the feasibility of single dose of FSH as an effective superovulation method in rabbits. The superovulation treatments examined comparatively were as follows: 1) a single injection of 100 IU PMSG, 2) Multiple injections of 1 IU Super-OV of 2X daily for 3 days, 3) Multiple injections of 6.6 rng Folltropin-V 2X daily for 3 days, 4) Multiple injections of 13.3 rng Folltropn-V l$\times$ daily for 3 days, 5) Single injection of 40 mg Folltropin-V solved in 10% PVP, and 6) Single injection of 40 mg Folltropin-V dissolved in 25% PVP. The results obtained were as follows: The nurnher of ovulation points in the single injection of 40 mg Folltropin-V dissolved in 25% PVP and multiple injections of 6.6 mg Folltropin-V 2X daily for 3 days was averaged 44.6 and 39.2, respectively, which were significantly (P<0.05) more than in a single dose of PMSG or multiple doses of Super-OV. The number of total ova recovered in the single injection of 40 mg Folltropin-V dissolved in 25% PVP and multiple injections of 6.6 mg Folltropin-V 2X daily for 3 days was averaged 30.4 and 25.9, respectively, which were significantly (P<0.05) more than in the single dose of PMSG or multiple doses of Super-OV. A similar result was obtained for the number of fertilized ova recovered from the above six treatments. From the above results it was concluded that a single injection of 40 mg Folltropin-V dissolved in 25% PVP could be an effective and simple method for superovulation in rabbits.

Comparative Efficacy of Clove oil, MS-222 and 2-Phenoxyethanol as Anesthetics in Juvenile Scomber japonicus (고등어(Scomber japonicus) 치어의 마취제로서 Clove oil, MS-222 및 2-Phenoxyethanol의 평가)

  • The efficiency of clove oil, MS-222, and 2-phenoxyethanol was evaluated as anesthetics in juvenile Scomber japonicus. Stage A5 of anesthesia was assumed to be sufficient for conducting routine aquaculture procedures in less than 3 min, with recovery (stage R5) in less than 5 min. The lowest effective doses of the three anesthetics were 50 mg $L^{-1}$ clove oil (anesthetic time of 71.3 s and recovery time of 167.0 s), 100 mg $L^{-1}$ MS-222 (anesthetic time of 70.7 s and recovery time of 115.7 s), and 400 mg $L^{-1}$ 2-phenoxyethanol (anesthetic time of 86.7 s and recovery time of 95.0 s). Anesthetic times decreased with increasing doses for all three anesthetic agents, and fish anesthetized with clove oil exhibited the longest recovery times. After 30 min, the highest plasma cortisol and lactate levels were detected with the use of clove oil, whereas the lowest values were observed with 2-phenoxyethanol. In addition, high glucose levels were maintained during recovery with clove oil, but the treatments did not significantly differ. The most effective of the three anesthetic agents was 2-phenoxyethanol, although all were considered acceptable for use in cultures of juvenile Scomber japonicus.

Interpretation of Uranium Bioassay Results with the ICRP Respiratory Track and Biokinetic Model (ICRP 호흡기 및 생체역동학적 모델을 이용한 우라늄 생물분석 결과의 해석)

  • This study describes a practical method for interpretation of bioassay results of inhaled uranium to assess the committed effective doses both for chronic and acute intake situations. Organs in the body were represented by a series of mathematical compartments for analysis of the behavior of uranium in the body according to the gastrointestinal track model, respiratory track model and biokinetic model recommended by the ICRP. An analytical solutions of the system of balance equations among the compartments were obtained using the Birchall's algorithm, and the urinary excretion function and the lung retention function of uranium were obtained. An initial or total intakes by intake modes were calculated by applying excretion and retention functions to the urinary uranium concentration and the lung burden measured with a lung counter. The dose coefficients given in ICRP 78 are used to estimate the committed effective doses from the calculated intakes.
