Effect of Ginseng Saponin on the Activity, Phosphorylation, $[^3H]$Ouabain Binding of Purified$Na^+$ $K^+$-ATPase Isolated from the Outer Medulla of Sheep Kidney (인삼 Saponin이 양신장에서 정제한 $Na^+$ $K^+$-ATPase의 활성, 인산화 및 $[^3H]$Ouabain결합에 미치는 영향)

  • The effects of ginseng saponin on the activity, phosphorylation, [$^{3}$H] ouabain binding and light scattering (disruption) of purified $Na^{+}$ ,$K^{+}$ -ATPase isolated from the outer medulla of sheep kidney were compared to those of gypsophila saponin, sodium dodecylsulfate (SDS), and Triton X-100 on the same parameters. $Na^{+}$ , $K^{+}$ -ATPase activity, phosphorylation, and [$^{3}H$] ouabain binding were inhibited by ginseng saponin (triol>total>diol), SDS, or Triton X-100, but increased by gypsophila saponin. Low doses of ginseng saponin (3.mu.g saponin/.mu.g protein) decreased phosphorylation sites and ouabain binding site concentration (Bmax) without any change of turnover number and affinity for ouabain binding which were decreased by high dose of ginseng saponin (over 10.mu.g saponin/.mu.g protein), SDS or Triton X-100. On the other hand, gypsophila saponin increased the affinity without any change of Bmax for ouabain binding. Inhibition of $Na^{+}$ ,$K^{+}$ -ATPase activity by ginseng saponin and SDS or Triton X-100 appeared before and after decrease in light scattering, respectively. These data suggest that ginseng saponins (total, diol, triol saponin) inhibit $Na^{+}$ , $K^{+}$ -ATPase activity by specific direct and general detergent action at low and high concentrations, respectively, and this inhibitory action of ginseng sapornin to $Na^{+}$ , $K^{+}$ -ATPase is not general action of all saponins.

Changes of SDS-PAGE Pattern and Allergenicity of BSA and BGG in Beef Extract Treated with Heat and High Pressure (물리적 처리에 따른 우육추출물중의 BSA와 BGG단백질의 SDS-PAGE패턴 및 항원성의 변화)

  • In our previous report, we indicated that not only BSA but also BGG played an important role in the allergenicity of beef. In this study, the effect of heat or high-pressure treatments to beef extract on the SDS-PAGE patterns was examined. The antigenicity of each treated samples was also investigated by Western blots assay with the sera of BGG-positive beef allergic patients. The BGG band and its antigenicity slightly disappeared but not generally in $100^{\circ}C$ group, indicating $100^{\circ}C$ treatment is not sufficient to totally eliminate the antigenicity of beef allergens. Compared with BGG band, BSA band significantly disappeared in SDS-PAGE with $100^{\circ}C$ treatment, indicating BSA is more heat- sensitive than BGG. When the beef extract was heated at $120^{\circ}C$, not only BSA but also BGG bands was largely disappeared in both SDS-PAGE and Western blots. High pressure (HP) treatment even at 600 MPa did not affect SDS-PAGE and Western blots pattern of BSA. On the contrary, BGG treated with HP showed visible changes in SDS-PAGE. 600 MPa treatment significantly reduced the antigencity. Interestingly, these behaviors of BGG were not found in the same experiments with pure BGG treated with HP. From these results, it was speculated that some kinds of proteolytic enzymes in beef extracts were involved in the BGG molecular degradation by HP treatment. The aging experiments of beef extracts treated with HP supported this hypothesis. Further studies are needed to clarify the function and working mechanism of enzymes associated with BGG degradation in beef extracts by HP treatment.

Effect of Gamma Irradiation on Egg White Protein (卵白단백질에 대한 감마선 조사의 영향)

  • The ovalbumins obtained from hen's and duck's egg white were irradiated with up to 100 kGy at room temperature. The purified proteins were evaluated for their in vitro digestibility by incubating successively with pepsin and pancreatin conjugate. Amino acid compositions and SDS-PAGE pattern in these proteins were also analyzed. The obtained results indicated that gamma irradiation within the tested dose range(up to 100kGy) produced no statistically significant changes in the in vitro digestibility an amino acid compositions. Analysis of gamma-irradiated ovalbumins by SDS-PAGE revealed radiolysis of ovalbumin into proteins or peptides of low molecular weights.

  • Perilla [Perilla frutescens (L.) Britt] seedlings are easy to grow and eaten as the health vegetable sprout. Two day old perilla seedlings since germination were given a mild wounding using cell wall lytic NaOH/SDS solution for infiltration with recombinant Agrobacterium cells treated with phenolic compounds. In the analysis of fluorometric GUS gene expression for the transformed perilla seedlings, GUS enzyme activity was the highest by the combined treatments of 50 mM acetosyringone and 0.5% NaOH solution containing 0.01% SDS implying a synergic effect. This result could be successfully applied for demonstrating hepatitis B virus antigen (HBsAg) protein expression.

  • There are many methods to synthesize polystyrene latex. Emulsion polymerization technique is commonly used commercially, but it requires a new technology to replace a traditional polymerization method because of the disadvantage of chemical initiator for environmental pollution. Since free radicals can be produced by ultrasound energy effect, polystyrene latex was synthesized using ultrasound energy instead of chemical initiator. As the ultrasonic irradiation time was increased, average molecular weight was increased and polydispersity was decreased. The degree of polymerization was increased with the concentration of SDS and maximum degree of polymerization was shown at 2wt.% SDS concentration and the reaction temperature of $40^{\circ}C$. During the course of polymerization, molecular weight was repeatedly fluctuated because of occurrence of depolymerization. Narrow molecular weight distribution polystyrene latex having controlled molecular weight was synthesized by controlling ultrasonic irradiation time and the concentration of SDS.

Effect of Ginseng Saponin on the $Na^{+}$, $K^{+}$-ATPase of Dog Cardiac Sarcolemma

  • The effects of ginseng saponins on the sarcolemmal $Na^{+}$, $K^{+}$-ATPase were compared to gypsophila saponin, sodium dodecylsulfate (SDS), and Triton X-100 to elucidate whether the effects are due to the membrane distruption, using a highly enriched preparation of cardiac sarcolemma prepared from dog ventricular myocardium. About 26% and 29% of vesicles in the preparation, enriched in ouabain-sensitive $Na^{+}$, $K^{+}$-ATP ase, $\beta$-adrenergic and muscarinic receptors are rightside-out and inside-out orientation, respectively. Ginseng saponins (triol>total> diol) inhibited $Na^{+}$, $K^{+}$-ATP ase activity, $Na^{+}$, $K^{+}$-ATPase activity and [$^{3}$H]ouabain binding of sarcolemmal vesicles. However, gypsophila saponin, SDS (0.4$\mu$g/$\mu$g protein) and Triton X-100 (0.6 $\mu$g/$\mu$g protein) caused about 1.35 and 1.40-fold increase in $Na^{+}$, $K^{+}$-ATPase activity and [$^{3}$H] oubain binding, respectively. Especially, the activating effect of gypsophila saponin on membrane Na+, K+ ATPase was detected at gypsophila saponin to sarcolemmal protein ratios as high as 100. Low dose of ginseng saponin (3$\mu$g/$\mu$g protein) decreased the phosphorylation sites and the concentration of ouabain binding sites (Bmax) without affecting the turnover number and affinity for ouabain binding, while gypsophila saponin, SDS(0.4 ug/ug protein), ahd Triton X-100 (0.6$\mu$g/$\mu$g protein) increased the Bmax. The results suggest that ginseng saponins cause a decrease in the number of active sites by interacting directly with $Na^{+}$, $K^{+}$-ATPase before disruption of membrane barriers of sarcolemmal vesicles.

  • In this study, sodium dedocyl sulfate (SDS), which was anionic surfactant, was added for forming micelles to remove ferrous ions that could be contained with a small amount in industrial water. Then aggregates were formed by adsorption or binding of ferrous ions on the surface of micelles, and then rejected by ceramic membranes to remove ferrous ions. Ferrous concentration was fixed at 1mM and SDS was changed as $0{\sim}10mM$ to investigate the effect of the anionic surfactant. As a result, rejection rate of ferrous was the highest to 88.97% at 6mM. And we used ELS (Electrophoretic Light Scattering Spectrometer) to investigate particle size distribution of micellar aggregates depending on SDS concentration. Then distribution of large aggregates was the highest at 6mM. And we investigated effects of $N_2$-back-flushing time (BT) during periodic $N_2$-back-flushing on ceramic membranes. Finally optimal $N_2$-BT for NCMT-723l (pore size $0.10{\mu}m$) membrane was 20 sec.

  • The aquation and base hydrolysis of [Co(en)$_2$NH$_3$Cl]$^{2+}$ were studied by UV spectroscopic method in various SDS aqueous solution. The base hydrolysis of [Co(en)$_2$NH$_3$Cl]$^{2+}$ with the addition of 0, 0.05, 0.1 mol dm$^{-3}$ sodium chloride was studied. For the aquation of the complex, the rate constant in the micellar phase(kH$^M$) was a little larger than that in the aqueous phase(kH$^W$). With the increase of SDS concentration, the second order rate constant(kOH) for the base hydrolysis unchanged below the CMC and sharply decreased down to a limiting value after the CMC was reached. The effect of added NaCl on the rate behavior of the complexes in the micellar solution were investigated by using an ion-exchanged model.

Effects of Polyelectrolytes on the Charge Transfer Complexing between Indole derivatives and Methylviologen: Hydrophobic and Electrostatic Interactions

  • The effect of anionic polyelectrolytes, poly(styrenesulfonate) (PSS) and poly(vinylsulfonate) (PVS), on the charge transfer complexing between indole derivatives and methyl viologen($MV^{++}$) cation was investigated. The results were compared with effect of NaCl and an anionic surfactant, sodium dodecylsulfate (SDS). Both PSS and PVS enhanced the complex formation of neutral species (indole and indole acetate at low pH), zwitter ionic tryptophan, and positively charged tryptamine and tryptophan at low pH with $MV^{++}$. This result was attributed to the contribution of hydrophobic interaction, in addition to electrostatic interaction. The enhancing effect of PSS was much higher than that of PVS reflecting the higher hydrophobicity of PSS. The interaction between indole acetate anion and $MV^{++}$ was greatly reduced by addition of PVS and PSS. The higher charge density of PVS was appeared as greater reducing effect indicating the importance of electrostatic force in this case. In all cases, the effect of polyelectrolytes showed maxima, and further addition of PVS and PSS decreased the effect. This behavior was explained in terms of distribution of indole derivatives and $MV^{++}$ in domain of polyanions. The complex formation constants and molar absorptivities of complexes were determined, and the values were compared with those in water and SDS solutions.

  • This study was carried out to develop a practical enzyme-linked immunosorbent assay(ELISA) for the determination of soy protein in processed meat products as a preliminary study. The titer of antiserum raised in rabbit by injection of SDS-treated whole buffer extract(WBE) from isolates soy protein(ISP) was above 1:10,000 in indirect ELISA. When the SDS concentration was higher than 0.03% the antibody-antigen reaction was inhibited significantly. However, the antibody-antigen reaction inhibition was not observed when the SDS concentration was less than 0.02%. The antibodies used in this experiment also reacted with renatured antigen after removing SDS by dialysis, though not better than with SDS-denatured antigen(immunogen). The calibration curve with $100\;{\mu}g/100\;ml$ of sensitivity was obtained in indirect competitive ELISA.
